INTRODUCTION: CD73 is a membrane-bound enzyme crucial in adenosine generation. The adenosinergic pathway plays a critical role in immunosuppression and in anti-tumor effects of immune checkpoint inhibitors (ICI). Here, we interrogated CD73 expression in a richly annotated cohort of human lung adenocarcinoma (LUAD) and its association with clinicopathological, immune, and molecular features to better understand the role of this immune marker in LUAD pathobiology.
MATERIALS AND METHODS: Protein expression of CD73 was evaluated by immunohistochemistry in 106 archived LUADs from patients that und

Background: Stem-cell-based therapies have recently been explored in the field of erectile dysfunction (ED). However, the cellular and molecular phenotype of adipose derived stem cells (ADSCs) stromal vascular fraction (SVF) from ED patients remains largely unknown. Herein we compared the global gene expression profile in the SVF from ED patients and healthy individuals and identified altered signaling pathways between the two groups. Methods: Samples (2-5 g) of abdominal adipose tissue from ED patients (n = 6) and healthy individual controls (n = 3) undergoing elective cosmetic liposuction wer

Chronic liver fibrosis is a progressive and globally prevalent pathological condition that poses a substantial threat to human health, frequently culminating in cirrhosis and hepatocellular carcinoma (HCC). A range of etiological factors, including metabolic disorders such as diabetes and obesity, chronic alcohol consumption, prolonged medication use, and exposure to hepatotoxic agents, contribute to the rising incidence and persistence of this disease. Despite considerable progress in therapeutic development, effective curative interventions remain limited, underscoring the need for novel reg

Disturbance in redox homeostasis always leads to oxidative damages to cellular components, which inhibits cancer cell proliferation and causes tumor regression. Therefore, synergistic effects arising from cellular redox imbalance together with other treatment modalities are worth further investigation. Herein, a metal-organic framework nanosystem (NMOF) based on coordination between Fe (III) and 4,4,4,4-(porphine-5,10,15,20-tetrayl) tetrakis (benzoic acid) (TCPP) was synthesized through a one-pot method. After surface capping of silk fibroin (SF) to form NMOF@SF nanoparticles (NPs), this nanop

BACKGROUND: Imaging studies are an integral and important diagnostic modality to stage, monitor, and follow-up patients with metastatic urogenital cancer. The currently available guidelines on diagnosis and treatment of urogenital cancer do not provide the clinician with evidence-based recommendations for daily routine. It is the aim of the current manuscript to develop scientifically valid recommendations with regard to the most appropriate imaging technique and the most useful time interval in metastatic urogenital cancer patients undergoing systemic therapy.
RESULTS: Therapeutic response o

INTRODUCTION: Traditional drug discovery faces challenges, including high costs, time-intensive processes, and inherent risks. The disease-specific signature-based Connectivity Map (CMap) approach is widely utilized. However, the commonly employed method for constructing disease-specific signatures, known as Differentially Expressed Genes (DEGs), suffers from inconsistencies between dysregulated genes and their prognostic implications in tumor tissue, as well as discrepancies in prognosis genes between tumor and normal tissues. In this study, we present predictive analyses of CMap-based drug r

Clustered regularly interspaced short palindromic repeats (CRISPR)-based genome editing has expanded from experimental biology to early clinical application, raising the possibility of durable therapies for cardiovascular disease. Because many cardiac conditions are monogenic, they provide clear targets for allele-specific correction or modulation. In hypertrophic cardiomyopathy, preclinical research has shown that base editing of pathogenic MYH7 and MYBPC3 mutations can restore sarcomere function; concurrently, RNA-targeting approaches selectively suppress mutant transcripts. Dilated cardiomy

PURPOSE: Sickle Cell Disease (SCD) is a monogenic, autosomal recessive disorder caused by mutations in the β-globin gene, resulting in the production of abnormal hemoglobin S (HbS). When deoxygenated HbS polymerizes, it causes red blood cells to become sickle-shaped, and prone to hemolysis. These abnormal and sickled cells obstruct small blood vessels, leading to vaso-occlusion and tissue ischemia, contributing to multiple complications. SCD impacts nearly 8 million people worldwide, and is estimated to affect 500,000 newborns annually. Historically, potentially curative therapies have been li

Mast cells (MCs) are versatile immune cells that play a key role in neuroimmune interactions due to their strategic localization in barrier tissues and perineuronal sites, including the meninges, choroid plexus, thalamus, hypothalamus, and hippocampus within the central nervous system (CNS), as well as peripheral sites such as the skin, gastrointestinal tract, and dorsal root ganglia. They rapidly release stored and newly synthesized mediators, including histamine, tryptase, tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), substance P, and prostaglandins, which regulate nerve signaling

BACKGROUND: Efferocytosis is a critical physiological process in which phagocytes clear apoptotic cells to maintain tissue homeostasis. However, within the tumour microenvironment (TME), this process is systematically hijacked by tumour cells, transforming it into a key pathological mechanism that drives immunosuppression, tumour progression and therapeutic resistance.
KEY FINDINGS: This review systematically elucidates the central role of metabolic reprogramming in this functional reversal, emphasising that efferocytosis is essentially an immunometabolic intersection process precisely regula

Multiple sclerosis (MS) is an autoimmune condition that affects the central nervous system (CNS) and leads to neuroinflammation. The chronic inflammatory cascades of the immune system damage the functions of the nerves and myelin tissue. The heterogeneity of MS symptoms depends on the anatomical location of the lesion within the CNS. The identification of the gut microbiota as a crucial modulator of neuroinflammation via the microbiota-gut-brain axis has been the subject of new studies. Butyrate, one of the well-known short-chain fatty acids (SCFAs) produced by gut bacteria through the ferment

Activation of CD40, a pivotal component of immune oncology, could facilitate the infiltration of immune cells into the tumor microenvironment. In this investigation, we characterized SHR-2005, a second-generation CD40 agonistic mAb, as a potential candidate for cancer immune therapy. Our results showed that SHR-2005 possessed high affinity in binding to both human and rhesus CD40 and could effectively compete with CD40L for CD40 binding. Notably, SHR-2005 was engineered to exert a strong affinity for human FcγRIIb, consistent with its observed Fc receptor-dependent agonistic activity. Treatmen

Glioblastoma (GBM) is the most common and aggressive adult brain cancer with an average survival rate of around 15 months in patients receiving standard treatment. Oncolytic adenovirus expressing therapeutic transgenes represent a promising alternative treatment for GBM. Of the many human adenoviral serotypes described to date, adenovirus 5 (HAdV-C5) has been the most utilised clinically and experimentally. However, the use of Ad5 as an anti-cancer agent may be hampered by naturally high seroprevalence rates to HAdV-C5 coupled with the infection of healthy cells via native receptors. To explor

Imaging technologies that allow non-radiative visualization and quantification of apoptosis have a great potential for assessing therapy response, early diagnosis, and disease monitoring. Magnetic particle imaging (MPI), the direct imaging of magnetic nanoparticles as positive contrast agent and sole signal source, enables high image contrast (no tissue background signal), potential high sensitivity, and quantifiable signal intensity. These properties confer a great potential for application to tumor apoptosis monitoring. In this study, a simple and robust method was used to conjugate Alexa Fl

BACKGROUND: Mast cells (MCs) are pivotal immune cells. Beyond soluble mediators, mast cell-derived extracellular vesicles (MC-EVs) are crucial for intercellular communication. MC-EV heterogeneity, influenced by cellular activation and microenvironment, drives diverse biological roles. This review synthesizes current MC-EV biology, cellular interactions, and translational potential in immunity, inflammation, and disease.
METHODS: We systematically reviewed literature (2001-2024) from PubMed, Web of Science, and Scopus on MC-EV biogenesis, cargo, and functional roles from in vitro, in vivo, and

Rheumatoid arthritis (RA) exhibits significant sexual dimorphism, with females being two to three times more susceptible than males. CD8+ T cells are key effector immune cells driving synovial inflammation, bone erosion, and autoimmune injury within RA joints; nevertheless, their sex-specific pathogenic characteristics in early-stage RA (ERA) remain poorly defined. This study aimed to identify gender-specific pathogenic factors in RA. Based on a well-characterized patient cohort containing 46 RA individuals, we combined peripheral flow cytometry and synovial transcriptomic profiling to systema

BACKGROUND: The cardiac hormone ANP (atrial natriuretic peptide) maintains intravascular volume homeostasis through coordinated endothelial and renal effects. Since in vitro studies suggested that this circuit involves the inhibition of adrenal aldosterone release, here we investigated whether ANP indeed mediates endocrine heart-to-adrenal communication in vivo.
METHODS: Mouse models with zona glomerulosa (ZG)-restricted deletion knockout of the ANP receptor, GC-A (guanylyl cyclase-A), or the downstream cyclic GMP-stimulated PDE (phosphodiesterase) 2A were established through Cre/LoxP technol

Although patients with advanced ovarian cancer may respond initially to treatment, disease relapse is common, and nearly 50% of patients do not survive beyond five years, indicating an urgent need for improved therapies. To identify new therapeutic targets, we performed single-cell and nuclear RNA-seq data set analyses on 17 human ovarian cancer specimens, revealing the oncostatin M receptor (OSMR) as highly expressed in ovarian cancer cells. Conversely, oncostatin M (OSM), the ligand of OSMR, was highly expressed by tumor-associated macrophages and promoted proliferation and metastasis in can

CD38 serves as a marker for defining subsets of immune cells; however, being an ectoenzyme, it also plays a crucial part in modulating immunometabolic homeostasis, has immense potential to influence the microenvironment of tissues, functions as a prognostic indicator, as well as a therapeutic target. Despite the extensive information on CD38 and the scale of efforts in developing and optimizing therapies targeting various B cell-related diseases, there is still ongoing fundamental research addressing its basic functions on healthy human B cells, which is well needed. Pheochromocytomas and paragangliomas (PPGLs) are rare neuroendocrine tumors derived from chromaffin cells, with 80-85% originating in the adrenal medulla and 15-20% from extra-adrenal chromaffin tissues (paragangliomas). Approximately 30-40% of PPGLs have a hereditary component, making them one of the most genetically predisposed tumor types. Recent advances in genetic research have classified PPGLs into three molecular clusters: pseudohypoxia-related, kinase-signaling, and WNT-signaling pathway variants. Specifically, the detection of SDHB-related tumors indicates an increased risk of metasta
