Melanoma is one of the cancers of fastest-rising incidence in the world. Inducible nitric oxide synthase (iNOS) is overexpressed in melanoma and other cancers, and previous data suggest that iNOS and nitric oxide (NO) drive survival and proliferation of human melanoma cells. However, specific mechanisms through which this occurs are poorly defined. One candidate is the PI3K-AKT-mTOR pathway, which plays a major role in proliferation, angiogenesis, and metastasis of melanoma and other cancers. We used the chick embryo chorioallantoic membrane (CAM) assay to test the hypothesis that melanoma gro

The canonical (β-catenin dependent) Wnt signaling pathway has emerged as a likely candidate for regulating tooth replacement in continuously renewing dentitions. So far, the involvement of canonical Wnt signaling has been experimentally demonstrated predominantly in amniotes. These studies tend to show stimulation of tooth formation by activation of the Wnt pathway, and inhibition of tooth formation when blocking the pathway. Here, we report a strong and dynamic expression of the soluble Wnt inhibitor dickkopf1 (dkk1) in developing zebrafish (Danio rerio) tooth germs, suggesting an active repr

BACKGROUND AND PURPOSE 3-Hydroxy-octanoate, recently identified as a ligand for, the orphan GPCR, HCA(3), is of particular interest given its ability to treat lipid disorders and atherosclerosis. Here we demonstrate the pathway of HCA(3)-mediated activation of ERK1/2. EXPERIMENTAL APPROACH Using CHO-K1 cells stably expressing HCA(3) receptors and A431 cells, a human epidermoid cell line with high levels of endogenous expression of functional HCA(3) receptors, HCA(3)-mediated activation of ERK1/2 was measured by Western blot. KEY RESULTS HCA(3)-mediated activation of ERK1/2 was rapid, peaking a

In the preceding manuscript (Zhao, H., Xu, X., Diaz, J., and Muallem, S. (1995) J. Biol. Chem. 270, 19599-19605), we described a Kout(+)-dependent H+/HCO3- and Na+ influx pathway in the luminal membrane of salivary duct cells. In the present studies, we further characterized this pathway to show that the Kout(+)-dependent Na+ influx was not mediated by the luminal amiloride-sensitive Na+ channel, the Na+/H+ exchangers, or any electroneutral or conductive Cl(-)-dependent transport pathway. Thus, K+ efflux probably maintained electroneutrality during Na+ influx induced by removal of Kout+. Accor

Endothelial dysfunction is a critical initiating event in atherosclerosis. The regulatory role of the AKT signaling pathway in the transition between injury and repair remains unclear. This study developed a multiscale model to elucidate how AKT signaling modulates endothelial repair under shear stress. The framework integrates hemodynamic stimuli, cellular behaviors, and nonlinear AKT/NO/ROS biochemical kinetics coupled with endothelial phenotypic transitions. Results demonstrate that endothelial dysfunction is driven by a spatial mismatch between NO production and ROS consumption rather than

Hydrophilic perovskite quantum dots (PQDs) have emerged as promising materials for biomedical diagnostics due to their exceptional optical tunability and high photoluminescence quantum yield. However, their instability, toxicity, and degradation in aqueous and biological environments remain critical barriers to practical applications. This review integrates predictive modeling and bioinspired stabilization strategies to provide a comprehensive understanding of the mechanisms governing PQD instability and the design of robust, biocompatible nanostructures. Multiscale computational approaches-in

The mitogen-activated protein kinases (MAPKs) are critical signaling molecules that regulate T cell development, activation, and function. While the MAPK activation is well-characterized, the role of phosphatases as negative regulators remain incompletely understood. This study aimed to comprehensively investigate the role of phosphatases which regulate MAPK pathway activity in human T cells. The dual-specificity phosphatase 2 (DUSP2) was determined as a key negative regulator of ERK1/2, p38, and to a lesser extent of JNK1/2 in human T cells. Loss of DUSP2 is partly compensated by the inducibl

Dialysis membrane-related hemolysis (DMH) is an important factor influencing blood purification efficacy in critically ill patients. However, the pre-hemolysis fatigue process of RBCs and whether RBC fatigue can induce thrombus formation remains largely unexplored. In this study, rhein-modified polysulfone (PSf) hemodialysis membranes (RH membranes) were fabricated, and their impact on RBC fatigue and thrombus formation was investigated. In this study, the RH membranes demonstrated potent physicochemical properties and hemodialysis performance, effectively inhibiting RBC fatigue and RMPs forma

Green electrospinning is an emerging approach to fabricating environmentally responsible and biocompatible nanofibrous materials for biomedical applications. Reducing or eliminating the use of hazardous organic solvents through aqueous, alternative-solvent, and solvent-free processing approaches has the potential to minimise environmental and occupational concerns associated with conventional electrospinning. However, the sustainability of an electrospinning process cannot be determined solely by the polymer's origin or biodegradability; the entire processing pathway, including solvents, addit

The atmospheric degradation of volatile organic compounds (VOCs) begins through oxidation by hydroxyl or nitrate radicals, producing highly reactive dehydrogenated radicals. While the stability-and thus, the reactivity-of these radicals critically depends on their isomeric form, directly monitoring individual isomers under laboratory conditions remains a major analytical challenge. Here, we combine high-level ab initio calculations to unambiguously identify radicals via their structures, energies, and internal dynamics with a very sensitive high-resolution millimeter wave spectroscopy (150-500

Thiamine (Vitamin B1), an essential water-soluble vitamin, is composed of a pyrimidine and a thiazole ring. Owing to its functional roles as a coenzyme and its anti-inflammatory and antioxidant properties, it plays a critical role in disease prevention and therapeutic interventions. Currently, industrial production of thiamine relies primarily on chemical synthesis-a process that generates significant amounts of hazardous waste and byproducts. In contrast, microbial biosynthesis represents a more sustainable and environmentally friendly alternative. This review first outlines thiamine metaboli

Ferroptosis, an iron-dependent form of programmed cell death driven by toxic lipid peroxide accumulation, plays a critical role in various diseases, making its modulation a promising therapeutic strategy. In this study, we identified several L-type calcium channel blockers as novel inhibitors of ferroptosis. We further elucidated that calcium-dependent activation of PKCβ drives ferroptosis by phosphorylating two key enzymes, ACSL4 and ALOX15, at multiple sites. We generated phosphorylation-specific antibodies targeting these sites and confirmed their specificity in the context of ferroptosis.

Cachexia is a complex multiorgan syndrome associated with various chronic diseases, characterized by anorexia and increased tissue wasting in the context of chronic inflammation. A specific form of this syndrome, known as cancer cachexia (CC), occurs alongside different types of tumors. The pathogenesis of CC is multifactorial. Inflammatory mediators and hormones released by both tumor and host cells have a relevant role in driving the peripheral catabolic process through several direct mechanisms. Accumulating evidence indicates that the central nervous system (CNS) plays an integral role in

Replicative senescence is a powerful tumor suppressor pathway that curbs proliferation of human cells when a few critically-short telomeres activate the DNA damage response (DDR). We show that ATM is the sole DDR kinase responsible for the induction and maintenance of replicative senescence and that ATM inhibition can induce normal cell divisions in senescent cells. Compared to non-physiological atmospheric (∼20%) oxygen, primary fibroblast cells grown at physiological (3%) oxygen were more tolerant to critically short telomeres, explaining their extended replicative lifespan. We show that thi

Metabolomics is emerging as a significant approach to reflect the individual's response to pathophysiological conditions. Nuclear magnetic resonance (NMR) spectroscopy has evolved as a tool to identify metabolic dysregulations in critically ill patients afflicted with conditions like acute respiratory distress syndrome (ARDS), severe acute pancreatitis (SAP), acute kidney injury (AKI), and sepsis. The spectral data from the serum sample of the study and control group are recorded using an 800 MHz NMR spectrometer and processed using NMR processing and analysis tools. Furthermore, a rigorous st

Acute respiratory disease syndrome (ARDS) is a common critical disease with high morbidity and mortality rates, yet specific and effective treatments for it are currently lacking. ARDS was especially apparent and rampant during the COVID-19 pandemic. Excess reactive oxygen species (ROS) production and an uncontrolled inflammatory response play a critical role in the disease progression of ARDS. Herein, we developed molybdenum nanodots (MNDs) as a functional nanomaterial with ultrasmall size, good biocompatibility, and excellent ROS scavenging ability for the treatment of acute lung injury (ALI

The malaria parasite Plasmodium falciparum synthesizes significant amounts of phospholipids to meet the demands of replication within red blood cells. De novo phosphatidylcholine (PC) biosynthesis via the Kennedy pathway is essential, requiring choline that is primarily sourced from host serum lysophosphatidylcholine (lysoPC). LysoPC also acts as an environmental sensor to regulate parasite sexual differentiation. Despite these critical roles for host lysoPC, the enzyme(s) involved in its breakdown to free choline for PC synthesis are unknown. Here, we show that a parasite glycerophosphodieste

In this paper, we present a review of critical concepts, and produce recommendations on management issues in cutaneous T-cell lymphomas (CTCLs) of adults. A panel of nine experts was selected for their expertise in research and clinical practice of CTCLs. During an initial meeting, the areas of major concern in the management of CTCLs were selected by generating and rank-ordering clinical key questions using the criterion of clinical relevance, through group discussion. Recommendations were achieved by multiple-step formalized procedures to reach a consensus after a comprehensive analysis of t

Respiratory failure and acute kidney injury (AKI) are associated with high mortality in SARS-CoV-2-associated Coronavirus disease 2019 (COVID-19). These manifestations are linked to a hypercoaguable, pro-inflammatory state with persistent, systemic complement activation. Three critical COVID-19 patients recalcitrant to multiple interventions had skin biopsies documenting deposition of the terminal complement component C5b-9, the lectin complement pathway enzyme MASP2, and C4d in microvascular endothelium. Administration of anti-C5 monoclonal antibody eculizumab led to a marked decline in D-dim
