Cumulative Exposure and Health Risk Assessment of PFAS in Animal-Derived Foods Using the Relative Potency Factor Approach.

Per- and polyfluoroalkyl substances (PFASs) are persistent and bioaccumulative contaminants frequently detected in animal-derived foods, raising concerns for consumer health. In 2020, the European Food Safety Authority (EFSA) established a group tolerable weekly intake (TWI) of 4.4 ng/kg bw per week for four PFAS (PFOA, PFNA, PFOS, PFHxS) based on immunotoxicity, prompting the European Commission to set maximum levels in food. However, many other PFAS are present in the diet, and their cumulative risk is poorly characterized. This study applied the Relative Potency Factor (RPF) approach, using

Control of Listeria monocytogenes in food industry by a combination treatment of natural aromatic compound with Listeria-specific bacteriophage cocktail.

Most Listeria monocytogenes found in the food industry are listeriosis-causing pathogens and possess the ability to form biofilms on food and food contact materials (FCMs). This study aims to evaluate the efficacy of the combination treatment of natural aromatic compounds (thymol, eugenol, carvacrol, and citral) with a Listeria-specific phage cocktail in mitigating the threat posed by L. monocytogenes in the food industry. In vitro combination treatment of 1 minimal inhibitory concentration (MIC) of natural aromatic compound with phage cocktail at multiplicity of infection (MOI) 100 reduced mo

Exposure to methylmercury and inorganic mercury in the food does not lead to trophic magnification in the sea star Asterias rubens.

Methylmercury accumulated at the top of aquatic food chains constitutes a toxicological risk to humans and other top predators. Biomagnification of methylmercury takes place among vertebrates at the higher trophic levels, but this process is less elucidated in benthic invertebrates at the lower trophic levels. Therefore, we investigated the accumulation from food and elimination of methylmercury and inorganic mercury in the benthic sea star Asterias rubens (L.) - a representative of trophic level ~3 - in laboratory experiments. Sea stars fed over 49 days with contaminated mussels (Mytilus edul

The Role of Stress and Stress Adaptations in Determining the Fate of the Bacterial Pathogen Listeria monocytogenes in the Food Chain.

The foodborne pathogen Listeria monocytogenes is a highly adaptable organism that can persist in a wide range of environmental and food-related niches. The consumption of contaminated ready-to-eat foods can cause infections, termed listeriosis, in vulnerable humans, particularly those with weakened immune systems. Although these infections are comparatively rare they are associated with high mortality rates and therefore this pathogen has a significant impact on food safety. L. monocytogenes can adapt to and survive a wide range of stress conditions including low pH, low water activity, and lo

Scientific Opinion on the risks to animal and public health and the environment related to the presence of nickel in feed.

Following a request from the European Commission, the risks to animal and human health and the environment related to the presence of nickel (Ni) in feed were assessed by the EFSA Panel on Contaminants in the Food Chain (CONTAM Panel). The presence of Ni in feed can arise from both natural and anthropogenic sources. Additionally, certain feed materials contain metallic Ni, since it is used as a catalyst in their production. Based on the differences observed between the Ni exposure levels estimated for different animal species and identified no observed adverse effect levels (NOAELs) and lowest

Real-time PCR using SYBR Green for the detection of Shigella spp. in food and stool samples.

Shigella spp are exquisitely fastidious Gram negative organisms that frequently get missed in the detection by traditional culture methods. For this reason, this work has adapted a classical PCR for detection of Shigella in food and stool specimens to real-time PCR using the SYBR Green format. This method follows a melting curve analysis to be more rapid and provide both qualitative and quantitative data about the targeted pathogen. A total of 117 stool samples with diarrhea and 102 food samples were analyzed in Public Health Regional Laboratory of Nabeul by traditional culture methods and rea

A scoping review of the prevalence of antimicrobial-resistant pathogens and signatures in ready-to-eat street foods in Africa: implications for public health.

BACKGROUND AND OBJECTIVE: Despite its critical role in individual and societal health, food hygiene remains underexplored. Antibiotic-resistant pathogenic bacteria in ready-to-eat (RTE) food threaten public health. This scoping review collected data on the epidemiological prevalence of RTE food-contaminated pathogens resistant to antimicrobial drugs and resistance genes in Africa. METHOD: Using electronic databases, such as PubMed, Scopus, and Web of Science (WoS), handpicked from references, pre-reviewed published articles were retrieved and analyzed according to the PRISMA-ScR guidelines.

Structural basis of a CDR3-embedded binding mechanism in a nanobody for sensitivity enhancement toward tenuazonic acid.

Nanobodies (Nbs) have shown great potential for use in immunoassays targeting small-molecule contaminants in food safety monitoring. However, a limited understanding of their recognition mechanisms has hindered the development of high-performance Nbs and the improvement of assay performance. Herein, a previously developed nanobody (Nb) 3F9 against tenuazonic acid (TeA) was selected as a model to resolve its X-ray crystal structure. Notably, Nb3F9 adopts a typical immunoglobulin fold, with TeA deeply inserted into the complementary-determining region 3 (CDR3) and buried in a binding pocket form

Emerging multimodal lateral flow immunoassays: Rational signal synergy design for enhanced food safety monitoring.

Lateral flow immunoassay (LFIA) has become a core tool for rapid on-site screening of food hazards. However, traditional single-modal LFIA exhibits insufficient sensitivity and is susceptible to matrix interference, posing significant challenges in practical applications. In recent years, the systematic integration of multimodal signals, including colorimetric, catalytic, magnetic, fluorescence, and photothermal, has emerged as a critical strategy to improve detection sensitivity, anti-interference capability, and quantitative accuracy. Notably, the performance of multimodal LFIA depends on th

Salmonellosis Including Enteric Fever.

Salmonella is a gram-negative, motile, nonsporulating, facultative anaerobic bacillus, belongs to the family Enterobacteriaceae. The bacteria were first identified in 1884. It is transmitted through direct contact with an infected person or indirect contact by the consumption of contaminated food and water. More than 2500 serotypes of Salmonella enterica have been identified but less than 100 serotypes are known to cause infections in humans. S. enterica serovar typhi (S. typhi) and S. enterica serovar paratyphi (S. paratyphi A B C) cause enteric fever, whereas nontyphoidal Salmonella serotype

Botulism in Spain: Epidemiology and Outcomes of Antitoxin Treatment, 1997-2019.

BACKGROUND: Botulism is a low incidence but potentially fatal infectious disease caused by neurotoxins produced mainly by Clostridium botulinum. There are different routes of acquisition, food-borne and infant/intestinal being the most frequent presentation, and antitoxin is the treatment of choice in all cases. In Spain, botulism is under surveillance, and case reporting is mandatory. METHODS: This retrospective study attempts to provide a more complete picture of the epidemiology of botulism in Spain from 1997 to 2019 and an assessment of the treatment, including the relationship between a

Estimates of Toxoplasmosis Incidence Based on Healthcare Claims Data, Germany, 2011-2016.

Toxoplasmosis is a zoonotic infection contracted through Toxoplasma gondii-contaminated food, soil, or water. Seroprevalence in Germany is high, but estimates of disease incidence are scarce. We investigated incidences for various toxoplasmosis manifestations using anonymized healthcare claims data from Germany for 2011-2016. Patients with a toxoplasmosis diagnosis during the annual observational period were considered incident. The estimated incidence was adjusted to the general population age/sex distribution. We estimated an annual average of 8,047 toxoplasmosis patients in Germany. The ave

The role of occupational Aspergillus exposure in the development of diseases.

Aspergillus spp. have a high nutritional versatility and good growth on a large variety of construction materials. They also colonize soil or food, but decaying vegetation is their primary ecological niche. Therefore, exposure to fungi may occur at home, during hospitalization, during specific leisure activities, or at the workplace. The development of Aspergillus infections depends on the interplay between host susceptibility and the organism. Environments with high counts of fungal elements (conidia, hyphal fragments and others), high levels of bioarerosols, and elevated concentrations of my

Aluminum Effects in Infants and Children.

Aluminum has no known biological function; however, it is a contaminant present in most foods and medications. Aluminum is excreted by the renal system, and patients with renal diseases should avoid aluminum-containing medications. Studies demonstrating long-term toxicity from the aluminum content in parenteral nutrition components led the US Food and Drug Administration to implement rules for these solutions. Large-volume ingredients were required to reduce the aluminum concentration, and small-volume components were required to be labeled with the aluminum concentration. Despite these rules,

Plant and Microalgae Derived Peptides Are Advantageously Employed as Bioactive Compounds in Cosmetics.

Bioactive peptides (BP) are specific protein fragments that are physiologically important for most living organisms. It is proven that in humans they are involved in a wide range of therapeutic activities as antihypertensive, antioxidant, anti-tumoral, anti-proliferative, hypocholesterolemic, and anti-inflammatory. In plants, BP are involved in the defense response, as well as in the cellular signaling and the development regulation. Most of the peptides used as ingredients in health-promoting foods, dietary supplements, pharmaceutical, and cosmeceutical preparations are obtained by chemical s

1,3-dichloro-2-propanol induced lipid accumulation in HepG2 cells through cAMP/protein kinase A and AMP-activated protein kinase pathways via Gi/o-coupled receptors.

1,3-dichloro-2-propanol (1,3-DCP) is a food born hepatoxic chloropropanol contaminant that has been detected in a wide range of foods. In the present study, we investigated the effects and mechanisms of 1,3-DCP on lipid accumulation in HepG2 cells. The data showed 1,3-DCP significantly increased intracellular content of triglyceride (TG) and total cholesterol (TC) at 0.5-2μg/mL. Further results showed that 1,3-DCP greatly decreased cyclic AMP (cAMP) level. In addition, 1,3-DCP inhibited PKA and AMPK signaling pathway, but had no influence on intracellular calcium and regulated proteins. Moreov

Determination of chlorophenols in red wine using ionic liquid countercurrent chromatography as a new pretreatment method followed by high-performance liquid chromatography.

A countercurrent chromatography method for the enrichment and cleanup of chlorophenols from food samples was successfully established by using an ionic-liquid-modified two-phase solvent system composed of dichloromethane containing 2% 1-butyl-3-methylimidazolium bis(trifluoromethanesulfonyl)imide and water. The column was firstly filled with the organic stationary phase, and then a large volume of sample was pumped into the column after it was equilibrated with pure water at the rotation speed. Finally, the trace amounts of chlorophenols extracted and enriched in the stationary phase were elut

[Listeriosis in pregnancy--case report].

Listeriosis is a rather rare infectious disease but its incidence in pregnancy is over 20 times higher than in the general population. Pregnant women with listeriosis comprise one-third of all listeriosis cases. Listeriosis is a foodborne disease. Sporadic as well as epidemic cases of listeriosis are usually related to contaminated processed food, especially meat dishes served in fast-food restaurants and dairy products. Pregnant women are at an increased risk for listeriosis infection. Unfortunately the symptoms are not specific and the diagnosis presents a considerable challenge. Although th

Rapid detection of Listeria monocytogenes in raw milk and soft cheese by a redox potential measurement based method combined with real-time PCR.

The incidence of outbreaks of foodborne listeriosis has indicated the need for a reliable and rapid detection of the microbe in different foodstuffs. A method combining redox potential measurement and real-time polymerase chain reaction (PCR) was developed to detect Listeria monocytogenes in artificially contaminated raw milk and soft cheese. Food samples of 25 g or 25 ml were homogenised in 225 ml of Listeria Enrichment Broth (LEB) with Oxford supplement, and the redox potential measurement technique was applied. For Listeria species the measuring time was maximum 34 h. The absence of L. mono

Staphylococcus aureus food-poisoning outbreak associated with the consumption of ice-cream.

In April 2013, a food poisoning outbreak caused by staphylococcal enterotoxins (SEs) in ice-cream occurred in Freiburg, Germany, among the 31 participants of a christening party. Of the 13 cases, seven were hospitalized or obtained ambulatory treatment. Different types of ice-cream, which was freshly produced at the hotel where the party took place, were found to contain SE and high amounts of coagulase positive staphylococci. Enterotoxigenic Staphylococcus aureus strains isolated from ice-cream and human cases were of the same spa-type (t127), harboured the sea gene and displayed identical ph
