Early iron deficiency has brain and behavior effects consistent with dopaminergic dysfunction.

To honor the late John Beard's many contributions regarding iron and dopamine biology, this review focuses on recent human studies that test specific hypotheses about effects of early iron deficiency on dopamine system functioning. Short- and long-term alterations associated with iron deficiency in infancy can be related to major dopamine pathways (mesocortical, mesolimbic, nigrostriatal, tuberohypophyseal). Children and young adults who had iron deficiency anemia in infancy show poorer inhibitory control and executive functioning as assessed by neurocognitive tasks where pharmacologic and neu

The logopenic/phonological variant of primary progressive aphasia.

OBJECTIVE: Primary progressive aphasia (PPA) is characterized by isolated decline in language functions. Semantic dementia and progressive nonfluent aphasia are accepted PPA variants. A "logopenic" variant (LPA) has also been proposed, but its cognitive and anatomic profile is less defined. The aim of this study was to establish the cognitive and anatomic features of LPA. METHODS: Six previously unreported LPA cases underwent extensive neuropsychological evaluation and an experimental study of phonological loop functions, including auditory and visual span tasks with digits, letters, and word

The superior temporal asymmetrical pit remains asymmetric in autism spectrum disorder.

UNLABELLED: Neuroimaging studies identify the superior temporal sulcus (STS) as an anatomical and functional target in autism spectrum disorder (ASD). Yet, the STS is a heterogeneous structure containing unique subregions, such as the superior temporal asymmetrical pit (STAP), which is a recently identified human-specific landmark. To date, no study has compared the STAP between ASD individuals and neurotypical controls (NTs) in a large sample. Here, we analyzed the mean intra-hemispheric sulcal depth and inter-hemispheric laterality of the STAP, as well as the anterior and posterior portions

Mixture-of-Experts Electroencephalography-Functional Magnetic Resonance Imaging Fusion for Interpretable Computer-Assisted Brain Disorder Diagnosis.

Electroencephalography (EEG) and functional magnetic resonance imaging (fMRI) provide complementary information about brain function, and have shown significant promise in detecting functional abnormalities in various brain disorders. However, the distinct signal characteristics and disparate representational spaces of EEG and fMRI pose severe challenges to effective multimodal fusion, thereby hindering accurate computer-assisted diagnosis of brain disorders with conventional fixed models. This study presents Brain Mixture-of-Experts (BrainMoE), an adaptive and interpretable EEG-fMRI fusion fr

Feasibility and exploratory outcomes of immersive VR intervention on brain functional networks and clinical symptoms in ADHD children: A pilot study.

BACKGROUND: Current evaluations of immersive virtual reality (VR) interventions for attention-deficit/hyperactivity disorder (ADHD) primarily rely on subjective clinical scales. This pilot study investigated the effects of an immersive VR intervention on brain functional networks in children with ADHD using resting-state functional magnetic resonance imaging (rs-fMRI) degree centrality (DC) and how they correlate with clinical symptoms. METHODS: Fourteen children with attention-deficit/hyperactivity disorder (ADHD group) and 13 age- and sex-matched healthy controls (HC group) were recruited.

Insights Into Subacute Combined Degeneration of the Cord: 'Rabbit Ear Sign' as a Radiodiagnostic Clue in Case of Vitamin B12 Deficiency Associated With Schizophrenia.

BACKGROUND: Subacute combined degeneration manifests as the degeneration of the dorsal and lateral columns of the spinal cord. Typical symptoms include sensory deficits, weakness, ataxia, and disturbances in gait which can progress to spasticity and paraplegia. Prolonged vitamin B12 deficiency can result in irreversible neurological damage and psychiatric manifestations such as depression, irritability, hallucinations, dementia, and delirium in the affected individuals. To date, only a few cases of schizophrenia with vitamin B12 deficiency with SCD have been reported in the literature. OBJECT

Gait and graphology changes after ventriculoperitoneal shunting for idiopathic normal pressure hydrocephalus: an exploratory pilot study.

BACKGROUND: Idiopathic normal pressure hydrocephalus (iNPH) is a potentially reversible neurological syndrome characterized by gait impairment, cognitive decline, and urinary dysfunction. While gait improvement after ventriculoperitoneal shunting (VPS) is well established, postoperative changes in handwriting-related graphomotor function remain poorly investigated. This exploratory pilot study aimed to evaluate whether a multidimensional motor-oriented assessment, including clinical gait scores, instrumental gait analysis, and handwriting-related graphomotor features, may capture postoperative

Left precuneus ALFF is associated with arterial oxygenation and cognitive impairment in patients with COPD: a resting-state fMRI study.

BACKGROUND: Cognitive impairment is a common but underrecognized comorbidity in chronic obstructive pulmonary disease (COPD). Lower arterial oxygenation may be associated with altered spontaneous brain activity, but the regional functional correlates linking oxygenation and cognition remain incompletely understood. This study examined amplitude of low-frequency fluctuation (ALFF) abnormalities in COPD patients with cognitive impairment and explored their associations with arterial oxygenation and cognitive performance. METHODS: This cross-sectional study included 90 participants: 30 healthy c

Immediate-Term Dapoxetine Administration Suppressed Functional Connectivity of Subcallosal Cingulate Cortex in Premature Ejaculation: A Resting-State fMRI Study.

INTRODUCTION: Dapoxetine, a short-acting selective serotonin reuptake inhibitor (SSRI), is commonly prescribed to treat premature ejaculation (PE). Functional connectivity (FC) between dorsolateral prefrontal cortex (DLPFC) and subcallosal cingulate cortex (SCC) has been found to serve as neuroimaging biomarker for predicting SSRI treatment response. Despite the fact that altered FC has been observed in PE patients, the neural mechanisms underlying the rapid-acting effects on brain of immediate-term dapoxetine administration remain unknown. This study aimed to explore the mechanisms underlying

Temporal order of clinical symptoms in Alzheimer's disease.

BACKGROUND: Understanding the temporal order of clinical symptoms is critical for improving disease progression assessment in biomarker-defined Alzheimer's disease (AD). However, the sequential emergence of cognitive, neuropsychiatric, and functional manifestations remains incompletely characterized. The purpose of this study is to characterize the population-level sequence in which Alzheimer's disease symptoms appear, to help understand how the disease is progressing. METHODS: We analyzed 932 participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI), including 546 amyloid-posi

Amyloid-β modulates APOE ε4 effects on cognition but not on targeted structural or functional connectivity measures over 2 years in pre-dementia adults.

BACKGROUND: The apolipoprotein E (APOE) ε4 allele is the strongest genetic risk factor for late-onset Alzheimer's disease (AD), yet emerging evidence suggests its cognitive effects may be age- and pathology-dependent. Whether and how amyloid-β (Aβ) pathology modifies the longitudinal cognitive trajectory associated with APOE ε4 in the pre-dementia stage remains unclear. METHODS: We conducted a 2-year longitudinal study in 70 pre-dementia adults from the Alzheimer's disease Neuroimaging Initiative (ADNI), assessing APOE genotype, Aβ status via positron emission tomography (PET), multi-domain c

Walking as a Window to the Brain: Redefining Gait in Neurology.

Walking is not merely locomotion but a window into the nervous system, integrating cortical, subcortical, cerebellar, spinal, and peripheral networks into a unified motor behavior. Across neurological diseases-including Parkinson's disease, atypical parkinsonism, cerebellar ataxias, stroke, multiple sclerosis, neuropathies, neuromuscular disorders, and functional gait syndromes-gait disturbances are among the most disabling clinical features, contributing to falls, loss of independence, institutionalization, and premature mortality. Traditional bedside observation remains indispensable, but it

Volumetric Changes in Cerebellar Transverse Zones: Age and Sex Effects in Health and Neurological Disorders.

Cerebellar volumetric changes are intricately linked to aging, with distinct patterns across its transverse zones, the functional subdivisions characterized by unique cytoarchitectural and connectivity profiles. Despite research efforts, the cerebellar aging process in health and neurological disorders remains poorly understood. In this study, we investigated the effects of age and sex on total cerebellum, transverse zone, and lobule volumes using MRI data from over 45,000 participants compiled from six neuroimaging datasets. We also propose a framework for estimating cerebellum age as an indi

The Energetic Collapse of the Alzheimer's Brain: Metabolic Inflexibility Across Cells and Networks.

Alzheimer's disease (AD) is more than just amyloid and tau. While often described as a disease of metabolic dysfunction, AD can more accurately be described as a disorder of metabolic inflexibility that leads to bioenergetic failure. In the healthy brain, neurons, glia, and vascular cells dynamically share and switch between different fuel sources (e.g., glucose, lactate, ketones, and fatty acids) to match functional demand. In AD, this adaptability is progressively lost because cellular metabolism is actively reprogrammed to support neuroinflammatory and disease-associated processes at the co

Neuroimaging and plasma evidence of early white matter loss in Parkinson's disease with poor outcomes.

Parkinson's disease is a common and debilitating neurodegenerative disorder, with over half of patients progressing to postural instability, dementia or death within 10 years of diagnosis. However, the onset and rate of progression to poor outcomes is highly variable, underpinned by heterogeneity in underlying pathological processes. Quantitative and sensitive measures predicting poor outcomes will be critical for targeted treatment, but most studies to date have been limited to a single modality or assessed patients with established cognitive impairment. Here, we used multimodal neuroimaging

Lesions to different regions of frontal cortex have dissociable effects on voluntary persistence.

Deciding how long to keep waiting for uncertain future rewards is a complex problem. Previous research has shown that choosing to stop waiting results from an evaluative process that weighs the subjective value of the awaited reward against the opportunity cost of waiting. In functional neuroimaging data, activity in ventromedial prefrontal cortex (vmPFC) tracks the dynamics of this evaluation, while activation in the dorsomedial prefrontal cortex (dmPFC) and anterior insula (AI) ramps up before a decision to quit is made. Here, we provide causal evidence of the necessity of these brain region…
