Pseudomonas aeruginosa is responsible for several infections due to its remarkable antibiotic resistance, facilitated by its ability to form biofilms. In this study, Aloin, an anthraquinone present in the plant Aloe vera, was tested for antibiofilm activity against P. aeruginosa PAO1. Aloin demonstrated efficient biofilm-inhibiting capacity at a sub-MIC dose of 4.8 mM, along with/without GEN at 0.002 µg/µL (sub-MIC), in an additive manner, determined by the FICI test. Furthermore, it reduced biofilm-associated carbohydrates by 66.9%, proteins by 66.22%, and extracellular DNA by 69.57%. In sili

The spread of antibiotic resistance has become a major challenge in global public health, and there is an urgent need to develop novel antibacterial agents with low resistance. Peptide-based antibacterial agents have attracted increasing attention for their membrane-targeting mechanisms, which may reduce the risk of the development of resistance. Herein, a series of quaternary ammonium-modified cyclic dipeptides (named cHHn-m) were designed and synthesized through a quaternization reaction and their antibacterial activity and cytotoxicity were systematically investigated. The optimal cyclic di

The global spread of NDM-1-producing Klebsiella pneumoniae poses a serious therapeutic challenge because these strains are resistant to nearly all β-lactams and many other antibiotics. Glutathione (GSH) is essential for maintaining intracellular redox homeostasis, thereby protecting cells from oxidative damage. However, exogenous GSH may disrupt this homeostasis and increase bacterial vulnerability to oxidative stress, such as that induced by antibiotic exposure. This study investigates whether exogenous GSH exerts antibacterial activity and increases β-lactam susceptibility in NDM-1-producing

BACKGROUND AND OBJECTIVES: Mycobacterium abscessus can cause severe infections in at-risk patients. Treatment efficacy for M. abscessus infections remains low, and better treatment options are needed. Factors hampering antibiotic potency may include the ability of M. abscessus to form biofilms and to endure in nutrient-deprived environments. These factors are underrepresented in current preclinical drug activity assays. Diversifying preclinical models by incorporating characteristics of these harsh environments may be important to better predict drug efficacy in patients. We aimed to develop a

Bedaquiline has transformed the treatment of multidrug-resistant tuberculosis (MDR-TB). However, bedaquiline resistance now threatens these gains. Emerging surveillance data demonstrate both baseline and acquired resistance across high-burden settings, with bedaquiline-resistant disease associated with poor clinical outcomes, high mortality, and growing evidence of transmission. In this Perspective, we examine the converging factors driving this trend, including bedaquiline's unique pharmacokinetic properties and limitations of current drug-susceptibility testing (DST) approaches. We describe

Fungal infection, particularly that caused by Candida albicans, pose emerging challenges in aquaculture, leading to significant economic and health losses in cultured fishes. However, limited research has explored eco-friendly antifungal alternatives for Clarias gariepinus, a species of high commercial value and susceptibility to fungal pathogens. This study aimed to isolate and identify marine-derived Actinobacteria capable of producing potent antifungal compounds effective against C. albicans. An initial screening of 30 Actinobacteria isolates obtained from thirty marine samples collected fr

Antimicrobial resistance (AMR), largely driven by the inappropriate and excessive use of antibiotics, requires rapid and reliable bacterial detection and antibiotic susceptibility testing (AST), as conventional culture-based methods remain time-intensive. Here, we report a real-time, label-free interdigitated wave electrode array (IWEA) that combines dielectrophoretic (DEP) enrichment with impedance spectroscopy for rapid bacterial detection and AST. The proposed IWEA was designed and optimized via COMSOL Multiphysics to enhance DEP-relevant electric-field strength, thereby improving DEP-assis

Basidiomycota is a cosmopolitan fungal phylum recognized for its nutritional properties and as a natural source of bioactive compounds. Within this group, the family Marasmiaceae Roze ex Kühner remains comparatively understudied. In this study, we performed a metabolic characterization and bioactivity screening of four marasmioid fungal strains using untargeted metabolomic and bioinformatic tools to analyze the metabolic profiles of broth extracts from strains Montiel04, FGR1289, FGR1276, and CC45, detecting 296, 447, 167, and 99 spectrometric features (m/z_RT), respectively. <b>Background and Objective:</b> The extensive use of antibiotics has created selective pressure leading to antimicrobial resistance, with biofilm formation serving as a major defense mechanism. Nearly 80% of infections involve biofilm-producing microorganisms that conventional antibiotics fail to eradicate. This study aimed to evaluate the antimicrobial and antibiofilm properties of ethanol extract from songga wood (<i>Strychnos lucida</i>) against <i>Bacillus</i> sp., <i>Salmonella typhi</i> and <i>Candida albicans</i>. <b>Mat

We report two such cases. Case 1: A 30-year-old male presented with right forearm swelling and pain; examination revealed a non-mobile, fluctuant intramuscular swelling. Case 2: A 30-year-old female had a 3-month history of midline chest pain with a mobile, fluctuant swelling over the xiphisternum. In both cases, aspirated pus showed numerous pus cells with few Gram-negative bacilli. Culture revealed non-lactose fermenting, smooth convex colonies identified as Salmonella Typhi by biochemical tests, Vitek 2 (bioMérieux), and serology. The isolates were susceptible to ceftriaxone, ciprofloxacin,

INTRODUCTION: Typhoid fever is a potentially fatal systemic infection caused by Salmonella typhi (S. typhi), a Gram-negative, rod-shaped, facultative anaerobe. This study aimed to characterize the efflux pump genes, molecular mutations, and antibiotic susceptibility patterns in clinical isolates.
METHODS: A total of 2950 blood samples were collected from suspected typhoid patients. Of these, 380 (12.88%) bacterial isolates were recovered, of which 236 (62.10%) were Gram-negative. S. typhi was detected in 95 isolates (25% of all bacterial isolates), corresponding to an overall prevalence of 3.

Mycotoxins are produced by some filamentous fungi that can contaminate food products and are toxic to humans and animals. The aim of this study was to evaluate the efficiency of extracts rich in bioactive compounds from one coffee industrial processing by-product (coffee husk) against two mycotoxigenic fungi and their mycotoxins. In vitro experiments showed that extracts from coffee husk had anti-fungal effect against Fusarium verticillioides and Aspergillus flavus, and the inhibition was directly proportional to the concentration of the extract. In addition, the extract had a significant anti

Historical processes in community assembly, such as species arrival order, influence interactions, causing priority effects. Candida albicans and Pseudomonas aeruginosa often co-occur in biofilm-based infections of the skin, lungs, and medical devices. Their predominantly antagonistic relationship involves complex physical and chemical interactions. However, the presence and implications of priority effects among these microorganisms remain largely unexplored. Here, we investigate the presence and impact of priority effect in dual-species biofilms using clinical isolates. By varying inoculatio

Three new phthalide derivatives, denominated Cajanthalides A, B and C (1-3), were identified from the leaves of Cajanus cajan. Their structures were meticulously elucidated through comprehensive analysis utilizing UV, IR, NMR (1D and 2D), HRESIMS, chiral analysis and DP4+ analysis. Notably, these new phthalide derivatives are present in racemic forms, with compounds 2 and 3 sharing identical planar structures. A plausible biogenetic pathway is proposed to account for the biosynthesis of compounds 1-3. Biological evaluation revealed that compound 2 exhibited moderate antifungal activity against

OBJECTIVES: Enterococcus spp. are typically found in multiple settings and are sometimes responsible for difficult-to-treat infections. In this context, very little is known about Enterococcus devriesei (E. devriesei), a rare species first isolated from a river lamprey. Importantly, no complete genome of E. devriesei currently exists in public repositories.
METHODS: An E. devriesei strain (Ed-CK-24) was isolated from homogenized Zophobas morio (Z. morio) larvae. Initial species identification (ID) was performed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (M
