The Renin-Angiotensin System and Skeletal Muscle.

The renin-angiotensin system (RAS) plays a key role in the control of blood pressure and fluid homeostasis. Emerging evidence also reveals that hyperactivity of the RAS contributes to skeletal muscle wasting. This review discusses the key role that the RAS plays in skeletal muscle wasting due to congestive heart failure, chronic kidney disease, and ventilator-induced diaphragmatic wasting.

Comparative effects of carrageenan on systemic candidiasis and listeriosis in mice.

Carrageenan, a toxic substance for the mononuclear phagocyte system (MPS), increase drastically the susceptibility of mice against Listeria monocytogenes challenge but induces concurrently an increasing resistance against systemic candidiasis and granulocytosis. These results corroborate the minor role played by MPS and suggest that polymorphonuclear cells play a major role in non-specific resistance of mice against systemic candidiasis.

Noncanonical Roles of RAD51.

Homologous recombination (HR), an evolutionary conserved pathway, plays a paramount role(s) in genome plasticity. The pivotal HR step is the strand invasion/exchange of double-stranded DNA by a homologous single-stranded DNA (ssDNA) covered by RAD51. Thus, RAD51 plays a prime role in HR through this canonical catalytic strand invasion/exchange activity. The mutations in many HR genes cause oncogenesis. Surprisingly, despite its central role in HR, the invalidation of RAD51 is not classified as being cancer prone, constituting the "RAD51 paradox". This suggests that RAD51 exercises other noncan

Glial-mediated dysregulation of neurodevelopment in Fragile X Syndrome.

Astrocytes are highly involved in a multitude of developmental processes that are known to be dysregulated in Fragile X Syndrome. Here, we examine these processes individually and review the roles astrocytes play in contributing to the pathology of this syndrome. As a growing area of interest in the field, new and exciting insight is continually emerging. Understanding these glial-mediated roles is imperative for elucidating the underlying molecular mechanisms at play, not only in Fragile X Syndrome, but also other ASD-related disorders. Understanding these roles will be central to the future

The critical roles of histone deacetylase 3 in the pathogenesis of solid organ injury.

Histone deacetylase 3 (HDAC3) plays a crucial role in chromatin remodeling, which, in turn, regulates gene transcription. Hence, HDAC3 has been implicated in various diseases, including ischemic injury, fibrosis, neurodegeneration, infections, and inflammatory conditions. In addition, HDAC3 plays vital roles under physiological conditions by regulating circadian rhythms, metabolism, and development. In this review, we summarize the current knowledge of the physiological functions of HDAC3 and its role in organ injury. We also discuss the therapeutic value of HDAC3 in various diseases.

Role of chemokines in Leishmania infection.

Chemokines are a growing group of chemoattractant cytokines that play important roles in physiological as well as pathological processes. Their roles in various aspects of pathogenesis and inflammation have come to light in the past decade or so. It is becoming increasingly clear that chemokines play a major, perhaps decisive role in Leishmania infections. In this review, we recapitulate important works linking the chemokine system with relation to visceral and cutaneous leishmaniasis over the past decade and attemptto put it all together to propose a single yet unfinished model to account for

Translational approaches to the study of eosinophils in vasculitis.

Eosinophil basic biology, including immunoregulatory functions, plays a complex role in a myriad of disorders where eosinophils play a role. Established in vivo and in vitro models and novel emerging techniques for studying eosinophils are integral to parsing the involvement of eosinophils in the pathogenesis of disorders including vasculitis. Knowledge of translational approaches from eosinophil-associated diseases and their application to eosinophilic vasculitis are highlighted. It is becoming increasingly evident that the roles of eosinophils in disease are complex. The use of targeted biol

Investigating the underlying mechanism of resistance to Ascaris infection.

The generative mechanism(s) of predisposition to Ascaris infection are currently unknown. While many factors play a role in interindividual infection intensity, much focus has been placed on the host's immunological response to infection and the underlying genetics. The present review describes the research conducted that has examined various immunological parameters and genetic factors that may play a role in resistance to ascariasis. We also discuss the contribution that animal models have made to our understanding of resistance to the parasitic roundworm and their role in possible future wo

Progesterone receptors--animal models and cell signalling in breast cancer. Diverse activation pathways for the progesterone receptor: possible implications for breast biology and cancer.

Progesterone and estradiol, and their nuclear receptors, play essential roles in the physiology of the reproductive tract, the mammary gland and the nervous system. Estrogens have traditionally been considered associated with an increased risk of breast cancer. There is, however, compelling evidence that progesterone plays an important role in breast cell proliferation and cancer. Herein, we review the possible role of progestins and the progesterone receptor-associated signaling pathways in the development of breast cancer, as well as the therapeutic possibilities arising from our growing kno

Synovium in haemophilic arthropathy.

Synovium is an essential component of the joint and plays a critical role in maintaining a balance between physiological processes and pathological changes in the joint. Recurrent intra-articular bleeding as occur in haemophilia induce pathological synovial changes in the joint. From a certain point on, synovitis inevitably plays a major role in joint destruction, although in the early phase of haemophilic arthropathy its role may be secondary to cartilage damage as a result of the direct effects of blood on cartilage. The changed haemosiderotic, synovial tissue produces catabolic cytokines an

The role of stearoyl-CoA desaturase in obesity, insulin resistance, and inflammation.

Stearoyl-CoA desaturase 1 (SCD1) is an essential lipogenic enzyme that has been shown to play an intrinsic role in the development of obesity and related conditions, such as insulin resistance. Through the generation of various mouse models of SCD1 deficiency, we have come to understand that SCD1 plays a role, directly or indirectly, in diverse metabolic processes, including lipogenesis, fatty acid oxidation, insulin signaling, thermogenesis, and inflammation. This review will address recent advances in our understanding of this key regulator of cellular metabolic processes, including the role

The role of adipokines in the pathogenesis and course of selected respiratory diseases.

Adipose tissue is also a secretory organ producing active substances called adipokines. Some of them (apelin, resistin, adiponectin, leptin, chemerin, or visfatin) may play a role in the pathogenesis and course of respiratory diseases, e.g. COPD, asthma, pulmonary hypertension, or lung cancer. There are limited and conflicting data on the role of adipokines in asthma. It has been confirmed, however, that visfatin and leptin can be markers of inflammation in COPD. Elevated concentrations of leptin and resistin play a pro-inflammatory role in the development of cancer cachexia. The role of adipo

Role of IL-13 in a model of Strongyloides venezuelensis infection in rats.

IL-13 is a cytokine known to play a role in several pulmonary diseases, including asthma and fibrosis. The role of IL-13 in the context of pulmonary changes induced by helminth infection is unclear. Rats experimentally infected with Strongyloides venezuelensis and treated with anti-IL-13 neutralizing antibody were used to evaluate the role of IL-13 on functional and inflammatory changes of host lungs, and on parasite control. S. venezuelensis-induced airway hyperreactivity was IL-13-independent, but IL-13 played an essential role in driving airway mucus production and eosinophil infiltration.

Amylin decreases food intake in mice.

The isolation of amylin from pancreatic islets has stimulated interest in its potential role in the pathogenesis of type II diabetes mellitus and in its possible physiological roles. Amylin administered intraperitoneally decreased food intake in non-food-deprived and food-deprived diabetic and nondiabetic mice. Amylin also decreased feeding induced by insulin administration without significantly affecting blood glucose levels. Amylin also decreased food intake following intracerebroventricular administration. It is possible that amylin plays a physiological role in appetite regulation and may

Getting the guts to expand stroke treatment: The potential for microbiome targeted therapies.

AIMS: This review focuses on the recent literature regarding the role of the gut-brain axis (GBA) following ischemic stroke. DISCUSSION: Stroke is the 5th leading cause of death and disability in the United States; however, few therapies have been developed to improve prognoses. There is a plethora of evidence suggesting peripheral inflammatory responses play a large role in the pathogenesis of stroke. Additionally, hyperglycemic conditions may play a significant role in worsening stroke outcomes due to microbiome dysbiosis. CONCLUSION: Recent research has illuminated the vital role of the G

Targeting immunometabolism during cardiorenal injury: roles of conventional and alternative macrophage metabolic fuels.

Macrophages play critical roles in mediating and resolving tissue injury as well as tissue remodeling during cardiorenal disease. Altered immunometabolism, particularly macrophage metabolism, is a critical underlying mechanism of immune dysfunction and inflammation, particularly in individuals with underlying metabolic abnormalities. In this review, we discuss the critical roles of macrophages in cardiac and renal injury and disease. We also highlight the roles of macrophage metabolism and discuss metabolic abnormalities, such as obesity and diabetes, which may impair normal macrophage metabol

Toll-like receptor 2 is required for innate, but not acquired, host defense to Borrelia burgdorferi.

Borrelia burgdorferi lipoproteins activate inflammatory cells through Toll-like receptor 2 (TLR2), suggesting that TLR2 could play a pivotal role in the host response to B. burgdorferi. TLR2 does play a critical role in host defense, as infected TLR2(-/-) mice harbored up to 100-fold more spirochetes in tissues than did TLR2(+/+) littermates. Spirochetes persisted at extremely elevated levels in TLR2-deficient mice for at least 8 wk following infection. Infected TLR2(-/-) mice developed normal Borrelia-specific Ab responses, as measured by quantity of Borrelia-specific Ig isotypes, the kinetic

Mitochondrial and peroxisomal Lon proteases are functionally divergent in environmental adaptation and pathobiology of the entomopathogenic fungus Beauveria bassiana.

Lon protease belongs to the AAA‑ATPase family and plays comprehensive physiological roles via maintaining cellular protein homeostasis. This study seeks to explore the functions of Lon protease in biocontrol potential of the filamentous entomopathogenic fungus Beauveria bassiana. B. bassiana contained mitochondrial and peroxisomal Lon proteases (BbLon1 and BbLon2) which were functionally analyzed via gene disruption and complementation. BbLon1 contributed to fungal tolerance to oxidative, thermal, hyperosmotic, and cell wall-perturbing stresses as well as iron deficiency. Both BbLon1 and BbLon
