Heat shock protein 90 inhibitors suppress pyroptosis in THP-1 cells.

Pyroptosis is a recently discovered inflammatory form of programmed cell death which is mostly triggered by infection with intracellular pathogens and critically contributes to inflammation. Mitigating pyroptosis may be a potential therapeutic target in inflammatory diseases. However, small chemicals to reduce pyroptosis is still elusive. In the present study, we screened 155 chemicals from a microbial natural product library and found Geldanamycin, an HSP90 inhibitor, profoundly rescued THP-1 cells from pyroptosis induced by LPS plus Nigericin treatment. Consistently, other HSP90 inhibitors,

Sphingolipids Signaling in Lamellipodia Formation and Enhancement of Endothelial Barrier Function.

Sphingolipids, first described in the brain in 1884, are important structural components of biological membranes of all eukaryotic cells. In recent years, several lines of evidence support the critical role of sphingolipids such as sphingosine, sphingosine-1-phosphate (S1P), and ceramide as anti- or pro-inflammatory bioactive lipid mediators in a variety of human pathologies including pulmonary and vascular disorders. Among the sphingolipids, S1P is a naturally occurring agonist that exhibits potent barrier enhancing property in the endothelium by signaling via G protein-coupled S1P1 receptor.

White Paper: Developing Antimicrobial Drugs for Resistant Pathogens, Narrow-Spectrum Indications, and Unmet Needs.

Despite progress in antimicrobial drug development, a critical need persists for new, feasible pathways to develop antibacterial agents to treat people infected with drug-resistant bacteria. Infections due to resistant gram-negative bacilli continue to cause unacceptable morbidity and mortality rates. Antibacterial agents have been historically studied in noninferiority clinical trials that focus on a single site of infection (eg, complicated urinary tract infections, intra-abdominal infections), yet these designs may not be optimal, and often are not feasible, for study of infections caused b

Functional characterization of TRPM7 in nasopharyngeal carcinoma and its knockdown effects on tumorigenesis.

The aim of this study was to evaluate the association of functional expression of TRPM7 with nasopharyngeal carcinoma (NPC) growth. We examined the correlation of TRPM7 expression with cell growth and proliferation, cell cycle, and apoptosis in vitro in NPC cell lines and NPC tumorigenesis in mice by conducting experiments in mice and by further analyzing the tumor volume and growth. We further explored to see whether there is any positive correlation with the TRPM7 knockdown in NPC cells with their sensitivity to radiation. We found that the functional expression of TRPM7 in nasopharyngeal ca

Reduced antimony accumulation in ARM58-overexpressing Leishmania infantum.

Antimony-based drugs are still the mainstay of chemotherapy against Leishmania infections in many countries where the parasites are endemic. The efficacy of antimonials has been compromised by increasing numbers of resistant infections, the basis of which is not fully understood and likely involves multiple factors. By using a functional cloning strategy, we recently identified a novel antimony resistance marker, ARM58, from the parasite Leishmania braziliensis that protects the parasites against antimony-based antileishmanial compounds. Here we show that the Leishmania infantum homologue also

Caspase-2 is required for dendritic spine and behavioural alterations in J20 APP transgenic mice.

Caspases have critical roles in Alzheimer's disease pathogenesis. Here we show that caspase-2 is required for the cognitive decline seen in human amyloid precursor protein transgenic mice (J20). The age-related changes in behaviour and dendritic spine density observed in these mice are absent when they lack caspase-2, in spite of similar levels of amyloid beta (Aβ) deposition and inflammation. A similar degree of protection is observed in cultured hippocampal neurons lacking caspase-2, which are immune to the synaptotoxic effects of Aβ. Our studies suggest that caspase-2 is a critical mediator

Effect of insulin on the inflammatory and acute phase response after burn injury.

After a severe burn, the liver plays a pivotal role by modulating inflammatory processes, metabolic pathways, immune functions, and the acute phase response. Therefore, liver integrity and function are important for recovery. A thermal injury, however, causes hepatic damage by inducing hepatic edema, fatty infiltration, hepatocyte apoptosis, and metabolic derangements associated with insulin resistance and impaired insulin signaling. In preliminary studies, we found that these pathophysiological processes are related to hepatic inflammation, altered intracellular signaling, and mitochondrial d

Glucose utilization and the PI3-K pathway: mechanisms for cell survival in preimplantation embryos.

The maintenance of optimal glucose utilization during the preimplantation period is critical for embryo survival. A decrease in glucose transport during preimplantation development has been linked to the early steps of programmed cell death in these embryos. Decreased glucose transport is not thought to be simply a consequence of cell death, rather it is thought to be a trigger that can initiate the apoptotic cascade. Extensive apoptosis during the preimplantation period may manifest later in pregnancy as a malformation-- or miscarriage, if cell loss is excessive. Phosphatidylinositol 3-kinase

Development of the mammalian gonad: the fate of the supporting cell lineage.

Sex determination in mammals is mediated via the supporting cell lineage in the fetal gonad. In the very early stages of gonadal development, the fate of the supporting cell population is critically dependent on the expression of the male-determining gene on the Y chromosome. If this gene is absent or fails to be expressed, or is expressed too late or in too small a number of supporting cells, all supporting cells (XX or XY) differentiate as pre-follicle cells and development proceeds along the female pathway. Supporting cells in which the male-determining gene is expressed in a timely manner

From Repeat Expansion to Circuit Disruption: 35 Years of Fragile X Pathophysiology.

Fragile X syndrome (FXS) is caused by CGG repeat expansion in FMR1, leading to gene silencing and loss of fragile X messenger ribonucleoprotein (FMRP). As the most common inherited cause of intellectual disability and a leading monogenic cause of autism spectrum disorder, FXS has provided an unusually tractable framework for linking a single-gene defect to abnormalities in RNA regulation, synaptic development, circuit function, and behavior. In this review, we synthesize current understanding of FMRP function, dysregulated signaling pathways, synaptic and network phenotypes, and emerging contr

HuR modulates disease-relevant alternative splicing of MAPT and other Alzheimer's disease-related genes.

Alternative splicing of MAPT exon 10 generates 4R- and 3R-Tau isoforms, and its dysregulation is associated with tauopathies including Alzheimer's disease (AD), for which effective treatments remain lacking. Here, we identify HuR as a novel repressor of MAPT exon 10 alternative splicing. Mechanistically, the RRM1 domain of HuR is critical for mediating its splicing inhibitory effect. HuR binds to the U-rich region downstream of intron 9 in MAPT together with FUBP1, KHSRP, hnRNPD, hnRNPAB, hnRNPDL, and TIAL1. The expression of HuR shows a negative association with that of FUBP1, KHSRP, hnRNPD,

Synthesis of Bulk Superconducting LiNbO2 Crystals Through CaH2 Reduction.

We have synthesized layered superconducting LiNbO2 crystals through a bulk phase transformation from LiNbO3 single crystals via CaH2 reduction. As the Nb valence is reduced from 5+ to 3+, the material undergoes a structural transformation to the resulting product, LiNbO2, which is accompanied by metallic behavior and a superconducting transition, Tc onset, as high as 14.4 K. Secondary ion mass spectroscopy (SIMS) and x-ray photoelectron spectroscopy (XPS) show that the resulting phase is hole-doped through de-lithiation during the reduction. Meta-analysis of Genes and Pathways that Protect Against Hypoxia.

Oxygen is essential for all terrestrial animals, but there is dramatic variability in how well different animals and even different cell types can adapt to reduced oxygen availability. We used a meta-analysis of the literature, with a focus on mouse studies, to identify pathways that might act to protect animals in low oxygen environments. We identified 108 genes whose mRNA levels change under hypoxia, and 55 genes critical for mounting a response to hypoxia. With this data, we developed a list of conserved genes, and we tested three C.elegans genes previously uncharacterized in hypoxia, mxl-3

Research progress on the mechanisms and intervention strategies of microbiota-gut-brain axis dysfunction under heat stress.

Heat stress (HS), as an important environmental stressor in the context of global warming, profoundly affects body health by disrupting the Microbiota-Gut-Brain Axis (MGBA). This review provides a critical synthesis of the mechanisms underlying MGBA disruption under HS. HS initially induces gut microbiota imbalance, which subsequently disrupts the intestinal mechanical and immune barriers, thereby interfering with the bidirectional gut-brain signaling mediated by pathways such as the vagus nerve. Based on the above mechanisms, this review further explores MGBA-centered therapeutic strategies,

Laser-Driven Structural Transformation from a Bulk Crystal to a Layered Material.

Laser-induced phase transitions offer pathways of phase transitions that are inaccessible by conventional stimuli. In this Letter, we conduct ab initio simulations to numerically demonstrate a novel laser-induced structural transformation: converting a bulk crystal into a layered van der Waals material using intense light pulses. The transition is driven by a nonlinear phononic mechanism, where selectively exciting polar and antipolar phonon modes with polarized terahertz light breaks targeted interlayer bonds while preserving intralayer ones. We identify that strong anisotropy in bond sensiti

PI3K at the crossroads: choosing B-cell activation or tolerance.

Phosphoinositide 3-kinase (PI3K) mediates signaling downstream of many receptors expressed by B cells. Studies in both mice and humans have shown that PI3K plays a critical role in B-cell development, activation, and tolerance. Indeed the key role of PI3K in regulating activation versus tolerance is demonstrated by patients with genetic variants that cause increased PI3K signaling, which results in an immune dysregulatory disorder characterized by a range of manifestations including lymphoproliferation and autoantibody production. Interestingly, PI3K signaling also plays an essential role in m

Bisphenol A and its analogues in thyroid diseases: Evidence from dysfunction, autoimmunity, and cancer.

Bisphenol A (BPA) and its analogues are widely used industrial chemicals in the production of polycarbonate plastics, epoxy resins, and related polymeric materials. Their extensive production and application have led to pervasive environmental contamination, enabling human exposure via ingestion, inhalation, and dermal absorption. As endocrine disruptors, BPA and its analogues (such as bisphenol S and bisphenol F) can influence thyroid homeostasis through interference with hormone synthesis, metabolism, and receptor signaling pathways. Recent studies suggest that exposure to bisphenols may con

MAIT Cells in Tumor Contexts: Functional Diversity and Regulatory Hierarchies.

Mucosal associated constant T (MAIT) cells are a unique subset of innate T lymphocytes that play a critical role in the host's defense against bacterial and viral pathogens. However, its function in the tumor microenvironment (TME) remains controversial. This review systematically explores the dual role of MAIT cells in tumor immunity by integrating existing literature evidence, and proposes a three-level causal hierarchy that determines the functional fate of MAIT cells. In order to solve the prognostic paradox across cancer types, we introduced the barrier and systemic tumor classification c
