A digital twin framework for forensic reconstruction of alcohol intake via fast and slow metabolite kinetics.

Accurately determining the timing and/or amount of consumed alcohol is critical in healthcare and forensic contexts, yet self-reported data are often unreliable due to stigma and legal implications. Objective biomarkers are therefore essential. Commonly used markers such as blood- and breath alcohol concentrations (BAC, BrAC) decline rapidly, limiting their utility for reconstructing drinking scenarios. Such reconstructions are essential for legal assessments, including refuting the “hipflask” defence. Markers with slower kinetics, such as ethyl glucuronide (EtG), ethyl sulphate (EtS), and uri…

Systematic citywide analysis reveals ecological connectivity of antimicrobial resistance genes across urban water systems.

Antimicrobial resistance (AMR) in drinking water raises public health concerns, while its anthropogenic sources, transmission dynamics, and health risks remain poorly understood, hindering the development of effective strategies to reduce human exposure. Here we conduct a systematic investigation of anthropogenic contributions to AMR across urban water compartments in a megacity, combining metagenomics and culturomics. We identify 1,309 antibiotic resistance genes (ARGs), and tracking their dynamics across microbial communities and fecal Enterobacteriaceae isolates indicates that ecological co…

Investigating the physical and chemical factors affecting the microbial status of water in the water distribution network of Babol City with an emphasis on the HPC index.

BACKGROUND AND PURPOSE: Safe and hygienic drinking water is crucial for public health. Secondary bacterial contamination, often caused by improper transport and distribution conditions, poses a significant threat. This study aimed to investigate the physical and chemical factors influencing the microbial status of drinking water in Babol's distribution network, focusing on the heterotrophic plate count (HPC) index. METHODS: Thirty-two water samples were collected randomly from Babol's rural areas and analyzed for HPC, residual free chlorine, turbidity, pH, and temperature. HPC was measured us…

Integrating multilevel, multidomain and multimodal neuroimaging factors to predict early alcohol exposure trajectories using explainable AI.

Various multilevel, multidomain factors at the individual-, family-, and environmental-level, and changes in neurobiology have been associated with the likelihood of developing alcohol use disorder (AUD) or binge drinking later in life. Prior studies have examined only limited subsets of these factors, typically focusing on cross-sectional associations with alcohol initiation, binge drinking, or AUD rather than exploring longitudinal alcohol use trajectories. Our study addresses these gaps by applying machine learning methods to a comprehensive set of multilevel, multidomain factors and multim…

Is facial emotion recognition deficit a predictor of riskyalcohol use in young adults?

OBJECTIVE: Social cognition skills, such as emotional processing, empathy, and mental state attribution, are critical for regulating behavior and decision-making, including alcohol consumption. However, there is no consensus on whether cognitive deficits are a risk factor for or a consequence of alcohol use disorders. Most studies on this issue have been conducted with people aged X 40 years with a long drinking history. This study investigated the associations between social cognition skills and Alcohol Use Disorders Identification Test (AUDIT) scores in young adults and considered the influe…

Chronic Alcoholism and Propofol Demand: The Impact of Alcohol Tolerance in Painless Gastrointestinal Endoscopy.

BACKGROUND AND AIM: Existing research suggests that chronic alcohol consumption may increase propofol requirements during anesthesia, though findings remain inconsistent. This study investigated the influence of chronic ethanol intake and alcoholic tolerance on propofol dosage in patients undergoing painless gastrointestinal endoscopy. METHODS: One hundred male patients with habitual alcohol consumption were enrolled. Drinking behavior was assessed using the Alcohol Use Disorders Identification Test (AUDIT), and alcohol tolerance was also assessed by self-reporting. Propofol was infused using…

Associations between Fecal Contamination of the Household Environment and Enteric Pathogen Detection in Children Living in Maputo, Mozambique.

Environmental exposure to enteric pathogens is generally assessed using fecal indicators but relationships between markers of fecal contamination and actual exposure to enteric pathogens remain poorly characterized. We investigated whether Escherichia coli and two human fecal markers (HF183 and Mnif) in urban Mozambican household soil and drinking water were associated with detection of eight bacteria, three viruses, and three protozoa measured by multiplex reverse-transcription PCR and soil transmitted helminths assessed by microscopy in stool samples from children. We used mixed-effects logi…

Relationship between age-related hearing loss and alcohol consumption in a Japanese population.

Age-related hearing loss (ARHL) has a multifactorial pathogenesis, and the influence of alcohol consumption on it is controversial. This cro … ssociated with reduced odds of ARHL at 4,000 Hz in women (OR 0.81; 95% CI 0.68-0.96). Assessment of drinking-related single nucleotide polymorphisms suggested that the effect of alcohol on ARHL may differ by genotype. Our findings suggest a sex-specific association between alcohol consumption and ARHL; heavy drinking is a potential risk factor in men, wher …

Research progress on the generation of NDMA by typical PPCPs in disinfection treatment of water environment in China: A review.

The drugs and personal care products in water sources are potential threats to the ecological environment and drinking water quality. In recent years, the presence of PPCPs has been detected in multiple drinking water sources in China. PPCPs are usually stable and resistant to degradation in aquatic environments. During chlorination, chloramination, and ozonation disinfection processes, PPCPs can act as precursor substances to generate N-nitrosodimethylamine (NDMA) which is the most widely detected nitrosamine byproduct in drinking water. This review provides a comprehensive overview of the im…

The Changing Relationship Between Hobby Engagement and Substance Use in Young People: Latent Growth Modelling of the Add Health Cohort.

Cross-sectional and some longitudinal evidence suggests doing hobbies can reduce substance use, but findings have been inconsistent, and whether associations differ across adolescence remains unclear. This study included 7454 Add Health participants (50% female, 77% White, age mean=14.95 and SD = 1.56). Participants were split into three groups, according to whether they were early (aged 11-14 at baseline), mid (aged 15-16), or late (aged 17-20) adolescents at baseline. The trajectories of binge drinking, marijuana, and tobacco use were analysed in latent growth models across Waves 1-5 (1994-2…

User-centred design, validation and clinical testing of an anti-choking mug for people with Parkinson's disease.

Oropharyngeal dysphagia, or difficulty initiating swallowing, is a frequent problem in people with Parkinson's disease (PD) and can lead to aspiration pneumonia. The efficacy of pharmacological options is limited. Postural strategies, such as a chin-down manoeuvre when drinking, have had some degree of success but may be difficult for people who have other limitations such as dementia or neck rigidity, to reproduce consistently. Using a user-centred design approach and a multidisciplinary team, we developed and tested an anti-choking mug for people with PD that helps angle the head in the opti…

How and when do work stressors and peer norms impact career entrants' alcohol-related behavior and its consequences?

Do the key drivers of alcohol misuse change as young adults transition from early to late stages of employee onboarding? To answer this question, a series of hypotheses were tested based on two waves of data collected from 1240 college graduates from four different universities in the United States who reported obtaining full-time employment following college graduation. Data on alcohol misuse and hypothesized mechanisms-peer drinking norms and work-related stressors-were collected during the early (i.e. first few months on the job: T1) and late (12 months following initial assessment: T2) sta…

Azithromycin induces read-through of the nonsense Apc allele and prevents intestinal tumorigenesis in C3B6F1 ApcMin/+ mice.

Therapeutic strategies that promote read-through of a mutant gene have proved effective for certain non-neoplastic diseases. However, the efficacy of this approach is unproven regarding neoplastic diseases with germline nonsense mutations, including familial adenomatous polyposis. Here we examined the cancer-preventive efficacy of the macrolide antibiotic azithromycin, with a reported read-through effect, on intestinal tumorigenesis in C3B6F1 ApcMin/+ mice harboring a nonsense Apc mutation resulting in a truncated Apc protein. Mice were given drinking water lacking azithromycin or containing 0…
