Decreased auditory function in the C3H/lpr autoimmune disease mouse.

To better understand autoimmune-related inner ear disease, cochlear structure and function were evaluated in the C3H/lpr autoimmune strain mouse, a model for systemic lupus erythematosus. C3H/lpr mice were examined at ages from 2 to 12 months along with age-matched C3H/HeJ controls. Autoimmune disease onset occurred at 3-4 months of age as serum immune complexes, antinuclear antibodies, and spleen weights were significantly elevated. Auditory brainstem response (ABR) audiometry showed normal auditory thresholds in C3H/lpr mice at 4 months of age, but elevated thresholds by 6 months, particular

An assessment of cochlear hair-cell loss in insulin-dependent diabetes mellitus diabetic and noise-exposed rats.

OBJECTIVE: The purpose of this study was to investigate if insulin-dependent diabetes mellitus causes degenerative changes in the inner ear and whether these changes are exacerbated by noise exposure. METHODS: Insulin-dependent diabetes mellitus was induced in male rats using streptozotocin (65 mg/kg of body weight, intravenously). Half the animals were exposed to 95 dB of random noise for 12 hours per day over a period of 6 months. The cochleae were removed, fixed, decalcified, dissected, and the hair cells counted. RESULTS: A significant loss of outer hair cells was exhibited in both noise

Carbon dioxide laser stapedotomy. Thermal effects in the vestibule.

Small-fenestra stapedotomy has recently been popularized in efforts to reduce the incidence of sensorineural hearing loss following stapes surgery for otospongiosis. Lasers have been advocated as a tool to fenestrate the stapes footplate. Conversion of radiant energy from the laser into heat in the vestibule represents the greatest potential risk to the inner ear. Using a carbon dioxide laser with a focal point of 150 microns at 300 mm, fenestrations of the stapes footplate were performed in a series of 14 anesthetized cats. The laser power output ranged from 0.47 to 3.05 W, with pulse duratio

Immediate noise effects on cochlear vasculature in the guinea pig.

Nine guinea pigs were exposed to a 4 kHz narrow band noise at 120 dB for 30 minutes. The animals were killed immediately following exposure and the cochlear tissues prepared using a soft-surface preparation technique. Experimental and control animals were mixed and randomly assessed without prior knowledge of the groups of which the specimens belonged. The results were analysed by computer. Statistically significant differences in the vasculature between noise-exposed and control animals of a general nature included: more changes in spiral lamina than external wall vessels, more changes in ves

Integrative Analyses of Genes Associated With Otologic Disorders in Turner Syndrome.

Background: Loss or partial loss of one X chromosome induces Turner syndrome (TS) in females, causing major medical concerns, including otologic disorders. However, the underlying genetic pathophysiology of otologic disorders in TS is mostly unclear. Methods: Ear-related genes of TS (TSEs) were identified by analyzing differentially expressed genes (DEGs) in two Gene Expression Omnibus (GEO)-derived expression profiles and ear-genes in the Comparative Toxicogenomic Database (CTD). Subsequently, Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and Disease Ontology (DO) analys

Robotics, automation, active electrode arrays, and new devices for cochlear implantation: A contemporary review.

In the last two decades, cochlear implant surgery has evolved into a minimally invasive, hearing preservation surgical technique. The devices used during surgery have benefited from technological advances that have allowed modification and possible improvement of the surgical technique. Robotics has recently gained popularity in otology as an effective tool to overcome the surgeon's limitations such as tremor, drift and accurate force control feedback in laboratory testing. Cochlear implantation benefits from robotic assistance in several steps during the surgical procedure: (i) during the app

Distortion product otoacoustic emissions in very preterm infants: A longitudinal study.

BACKGROUND: Very preterm infants are at a greater risk of developing neurodevelopmental impairments such as neuro-motor delays, vision and hearing deficits (Roze and Breart, 2004; Saigal and Doyle, 2008) [1,2]. The hearing difficulties in preterm infants vary depending on the co-morbid conditions. However, prematurity itself is considered as a risk factor that influence the functioning of auditory system. AIM: The current study aims to compare the DPOAEs in very preterm infants and term infants at 1 month, 3 months and 6 months of age (corrected age in preterm infants). METHOD: DPOAEs were r

Adaptive bone modeling and remodeling in acute otitis media caused by non-typeable or type B Haemophilus influenzae or Moraxella catarrhalis.

Experimental studies have shown that acute otitis media caused by Streptococcus pneumoniae alters modeling dynamics in bone tissue structures surrounding the middle ear cavity. Initial resorption of bone is followed by formative activity, seen as massive osteoneogenesis. However, neither resorptive nor formative activity occurs in the otic capsule, supporting the existence of a perilymphatic zone of specialized bone. This study investigates adaptive bone modeling in acute otitis media caused by other bacteria frequently encountered in this disease. Seventy-five rats were inoculated with either

[Therapeutic approach in blast injuries of the ear].

UNLABELLED: Otic blast injury is caused by arrhythmic air blast wave. The perforation of the tympanic membrane is the commonet finding associated with lacerations of mucosa in the middle ear. Makki [6] published 34 cases of myringoplasty after war blast injuries. However, healing of such perforations is common; Kerr [7] noted a healing rate of 83% after the blast injury. The aim of the study was to evaluate different therapic procedures of otic blast injuries. There were 74 patients with bilateral otic blast injuries, who underwent otomycroscopic examination. The following parameters were note

Two-step voltage-sensor activation of the human KV7.4 channel and effect of a deafness-associated mutation.

KCNQ4-encoded KV7.4 voltage-gated potassium channels are expressed in hair-cells of the inner ear. Loss-of-function variants in KCNQ4 cause non-syndromic progressive hearing loss (DFNA2). KV7.4 pore opening requires voltage-dependent conformational changes (activation) of the voltage-sensor domains (VSDs); however, how fast charge displacement during VSD activation is coupled to slow channel opening is currently unclear. Here, we optically tracked KV7.4 VSD activation with voltage-clamp fluorometry, leveraging two fluorophores and pulsed excitation, and found that VSD activation comprises seve

Sudden onset bilateral hearing loss after forehead osteoma removal associated with superior semicircular canal dehiscence.

Sudden sensorineural hearing loss (SNHL) following non-otologic surgery is a rare but potentially devastating complication. We report a case of bilateral SNHL occurring after elective excision of a frontal bone osteoma under general anaesthesia. A 52-year-old woman with no prior hearing impairment developed profound bilateral hearing loss immediately on emergence from anaesthesia, with partial spontaneous improvement over hours and subsequent right-sided predominance with tinnitus. Imaging excluded ischaemic pathology. Audiometry demonstrated a transient bilateral noise-induced pattern, with p

CASZ1 regulates the maturation of outer hair cells and is required for hearing in mice.

The transcription factor ATOH1 is a master regulator of mechanosensory hair-cell (HC) development in the ear. Here, we report that its target gene Casz1 regulates the maturation of outer HCs (OHCs). Genetic deletion of Casz1 during (but not after) cochlear development in the mouse caused: hearing loss; disorganization of mechanosensory stereocilia bundles in OHCs; reduced F-actin density in OHC cuticular plates; progressive OHC loss; and mild morphological alterations in inner HCs. This deletion also altered gene expression, delaying downregulation of genes expressed in immature OHCs, includin

An engineered ferritin nanocage co-delivery system for targeted hair cells protection and hearing loss attenuation.

RATIONALE: Hearing loss is a common sensory disorder with few treatment options, mainly due to the blood-labyrinth barrier and lack of effective targeted delivery systems. Small molecules such as LLY283 and RG108 protect hair cells (HCs), but clinical use is limited by poor cellular uptake, off-target toxicity, and inefficient cochlear delivery. METHODS: We developed a ferritin (Fn)-based nanocage (LR@Fn) to co-deliver RG108 and LLY283 via local injection through the round window membrane. Fn offers natural HC targeting, a hollow cavity, and good biocompatibility. Drugs were loaded using a pH

Enhanced neurotropism of bovine H5N1 compared to the Vietnam H5N1 isolate in C57BL/6J mice.

In this study, we investigated differences in tissue tropism of two HPAI H5N1 strains, the isolate A/Vietnam/1203/2004 (VN1203) isolated from a fatal human case in 2004 and the bovine isolate A/Bovine/Ohio/B24osu-342/2024 (Bov342) isolated in 2024, in C57BL/6J mice. Infection via aerosols was uniformly lethal in mice. However, tissue tropism differed significantly: while VN1203 replication was largely restricted to the respiratory tract, Bov342 successfully replicated in the respiratory tract as well as various regions of the brain. Correspondingly, cytokine profiles in the brain differed sign

The Notch ligand Jagged1 plays a dual role in cochlear hair cell regeneration.

Hair cells (HCs) within the inner ear cochlea are specialized mechanoreceptors required for hearing. Cochlear HCs are not regenerated in mammals, and their loss is a leading cause of deafness in humans. Cochlear supporting cells (SCs) in newborn mice have the capacity to regenerate HCs, but persistent Notch signaling, presumably activated by SC-specific Notch ligand Jagged1 (JAG1), prevents SCs from converting into HCs. Here, employing an organoid platform, we show that while JAG1 participates in HC-fate repression, JAG1's primary function is to preserve the "progenitor-like characteristics" o

Establishment of a Cochlear Organoid Platform for Remodeling the Extracellular Matrix.

Irreparable injury and depletion of inner ear hair cells lead to permanent hearing impairment. Cochlear organoids serve as a robust in vitro platform for investigating the pathogenesis of deafness and exploring therapeutic approaches. However, the widely used commercial Matrigel in cochlear organoid culture suffers from issues related to consistency and structural stability. Here, we developed a cochlear organoid culture system using synthetic GelMA hydrogel, which promoted the spontaneous clustering and organization of cochlear stem cells, thereby accelerating organoid formation and facilitat

Identification of druggable binding sites and small molecules as modulators of TMC1.

Our ability to hear and maintain balance relies on the proper functioning of inner ear sensory hair cells, which translate mechanical stimuli into electrical signals via mechano-electrical transducer (MET) channels, composed of TMC1/2 proteins. However, the therapeutic use of ototoxic drugs, such as aminoglycosides and cisplatin, which can enter hair cells through MET channels, often leads to profound auditory and vestibular dysfunction. To date, our understanding of how small-molecule modulators interact with TMCs remains limited, hampering the discovery of novel drugs. Here, we propose a str

Gating of hair cell Ca2+ channels governs the activity of cochlear neurons.

Our sense of hearing processes sound intensities spanning six orders of magnitude. In the ear, the receptor potential of presynaptic inner hair cells (IHCs) covers the entire intensity range, while postsynaptic spiral ganglion neurons (SGNs) tile the range with their firing rate codes. IHCs vary the voltage dependence of Ca2+ channel activation among their active zones (AZs), potentially diversifying SGN firing. Here, we tested this hypothesis in mice modeling the human CaV1.3A749G mutation that causes low-voltage Ca2+ channel activation. We demonstrate activation of Ca2+ influx and glutamate
