Mycobacteriophage Yasnaya_Polyana and its engineered lytic derivative: specificity of regulatory motifs and lytic potential.

INTRODUCTION: The growing prevalence of multidrug-resistant Mycobacterium tuberculosis and nontuberculous mycobacteria (NTM) highlights the urgent need for alternative therapeutic approaches. Mycobacteriophages, viruses that selectively infect mycobacteria, have emerged as promising tools. Here, we report the isolation and characterization of a new subcluster K4 phage, Yasnaya_Polyana, with a focus on its regulatory motifs and engineered lytic variant. METHODS: The phage was isolated by enrichment on Mycobacterium smegmatis mc(2)155, followed by genome sequencing and functional annotation. St

Non-invasive longitudinal monitoring of vascularization in tissue-engineered grafts by ultrasound localization microscopy.

Tissue engineering is a rapidly advancing field aimed at regenerating damaged tissues by combining cells, biomaterials, and bioactive molecules. Adequate vascularization is a critical determinant of successful tissue regeneration, as it supports both the regenerative process and the maintenance of physiological functions in the newly formed tissue. Traditional methods for evaluating vascularization, such as histological and immunohistochemical techniques, are inherently invasive and limited to endpoint analysis. In this study, we innovatively explored the potential of ultrasound localization m…

Dual engine-driven bionic microneedles for early intervention and prolonged treatment of Alzheimer's disease.

The microneedle (MN) delivery system presents an attractive administration route for patients with Alzheimer's disease (AD). However, the passive drug delivery mode and low drug loading of MNs often result in unsatisfactory therapeutic efficiency. To address these dilemmas, we developed dual engine-drive bionic MNs for robust AD treatment. Specifically, free rivastigmine (RVT) and RVT particles were co-loaded within the MNs to construct the valve and chambers of the guava, respectively, which can serve as an active engine to promote drug permeation by generating capillary force. Regeneration Process of an Autologous Tissue-Engineered Trachea (aTET) in a Rat Patch Tracheoplasty Model.

The treatment of long-tracheal lesion is difficult because there are currently no viable grafts for tracheal replacement. To solve this problem, we have developed an autologous Tissue-Engineered Trachea (aTET), which is made up of collagenous tissues and cartilage-like structures derived from rat chondrocytes. This graft induced successful long-term survival in a small-animal experiment in our previous study. In this study, we investigated the regeneration process of an aTET to attain reproducible success. We prepared an aTET by using a specially designed mold and performed patch tracheoplasty

Brainwide silencing of prion protein by AAV-mediated delivery of an engineered compact epigenetic editor.

Prion disease is caused by misfolding of the prion protein (PrP) into pathogenic self-propagating conformations, leading to rapid-onset dementia and death. However, elimination of endogenous PrP halts prion disease progression. In this study, we describe Coupled Histone tail for Autoinhibition Release of Methyltransferase (CHARM), a compact, enzyme-free epigenetic editor capable of silencing transcription through programmable DNA methylation. Using a histone H3 tail-Dnmt3l fusion, CHARM recruits and activates endogenous DNA methyltransferases, thereby reducing transgene size and cytotoxicity.

Efficient Rescue of Retinal Degeneration in Pde6a Mice by Engineered Base Editing and Prime Editing.

Retinitis pigmentosa (RP) is a complex spectrum of inherited retinal diseases marked by the gradual loss of photoreceptor cells, ultimately leading to blindness. Among these, mutations in PDE6A, responsible for encoding a cGMP-specific phosphodiesterase, stand out as pivotal in autosomal recessive RP (RP43). Unfortunately, no effective therapy currently exists for this specific form of RP. However, recent advancements in genome editing, such as base editing (BE) and prime editing (PE), offer a promising avenue for precise and efficient gene therapy. Here, it is illustrated that the engineered

Sustainable treatment scheme for in-situ remediation of contaminated drains using engineered natural systems.

Ensuring water security in resource-constrained, densely populated regions is a significant challenge globally. Due to insufficient treatment infrastructure, untreated sewage discharge into drainage channels is prevalent, especially in developing countries. This leads to the pollution of already dwindling water bodies and threatens future water availability. In this context, in-situ treatment within drains using nature-based systems is an attractive option. This study evaluates microbial bioremediation and phytoremediation as engineered natural solutions for in-stream treatment of municipal wa

Controlling Metal-Support Interactions to Engineer Highly Active and Stable Catalysts for COx Hydrogenation.

This perspective focuses on the modulation of metal-support interaction (MSI) in catalysts for COx hydrogenation, highlighting their profound impact on catalytic performance. Firstly, it outlines different strategies, including the use of highly reducible oxides and moderate reduction treatments, which induce the classical strong metal-support interaction (SMSI) effect and the electronic metal-support interaction (EMSI) effect. Morphology engineering and crystalline phase manipulation of oxides presented as effective methods to control EMSI are also discussed. The discrimination of SMSI and EM…

Phase Engineered CuxS-Ag2S with Photothermoelectric Activity for Enhanced Multienzyme Activity and Dynamic Therapy.

The insufficient exposure sites and active site competition of multienzyme are the two main factors to hinder its therapeutic effect. Here, a phase-junction nanomaterial (amorphous-crystalline CuxS-Ag2S) is designed and prepared through a simple room temperature ion-exchange process. A small amount of Ag+ is added into Cu7S4 nanocrystals, which transforms Cu7S4 into amorphous phased CuxS and produces crystalline Ag2S simultaneously. In this structure, the overhanging bonds on the amorphous CuxS surface provide abundant active sites for optimizing the therapeutic activity. Meanwhile, the amorph

Engineered Carrier-Free Nanosystem-Induced In Situ Therapeutic Vaccines for Potent Cancer Immunotherapy.

In situ vaccines that can stimulate tumor immune response have emerged as a breakthrough in antitumor therapy. However, the immunosuppressed tumor microenvironment and insufficient infiltration of immune cells lead to ineffective antitumor immunity. Hence, a biomimetic carrier-free nanosystem (BCC) to induce synergistic phototherapy/chemotherapy-driven in situ vaccines was designed. A carrier-free nanosystem was developed using phototherapeutic reagents CyI and celastrol as raw materials. In vitro and in vivo studies have shown that under NIR light irradiation, BCC-mediated photo/chemotherapy

Isopropanol production using engineered Corynebacterium glutamicum from waste rice straw biomass.

Isopropanol, a well-known biofuel, is a widely used precursor for chemical products that can replace nonrenewable petroleum energy. Here, engineered Corynebacterium glutamicum that can effectively utilize all xylose and glucose in agricultural waste rice straw to produce isopropanol was described. First, codon mutations were introduced into transporters and glycolytic-related genes to decrease the glucose preference of C. glutamicum. A more energetically favorable xylose oxidative pathway was constructed that replaced traditional xylose isomerization pathways, saving twice the number of enzyma
