Molecular epidemiology of penicillinase-producing Neisseria gonorrhoeae isolates in France.

OBJECTIVES: Characterizing the molecular epidemiology of antibiotic resistance is crucial for a better understanding of the evolution and spread of resistance in Neisseria gonorrhoeae. Here, we examine the molecular epidemiology of penicillinase-producing N. gonorrhoeae (PPNG) isolates in France. METHODS: We investigated 176 PPNG isolates collected between 2010 and 2012 by the National Reference Centre in France. Genotyping was performed using the NG-MAST technique, blaTEM genes were Sanger-sequenced, and plasmids were characterized by PCR-typing. RESULTS: We revealed the existence of four m

Recent insights into the pathogenesis of severe sepsis.

OBJECTIVE: Severe sepsis remains the dominant challenge in the care of critically ill patients. Over the last 10 years a large body of research has modified our understanding of this condition. In this article, we review the evolution of our understanding of the molecular mechanisms responsible for the development of this clinical syndrome. DATA SOURCES: The authors undertook a critical review of the literature on the molecular basis of the pathogenesis of sepsis with particular emphasis on the role of cytokines, toll-like receptors, adhesion molecules, coagulation cascade molecules and the p

REACT-PXE: a consensus on diagnosis and future research concerning pseudoxanthoma elasticum (PXE).

Pseudoxanthoma elasticum (PXE) is an autosomal recessive disorder described more than 150 years ago in which calcification and fragmentation of the elastic fibers result in a variety of symptoms which differ greatly in presentation and severity between patients. The diagnosis of PXE can still be made in many patients based on clinical presentation. However, in the advent of genetic screening that followed upon the identification of the first causal gene, ABCC6, milder cases of PXE or patients with a less typical onset of disease surfaced. Moreover, it became clear in the last years that PXE ha

Genomic analysis of the main epidemiological lineages of Acinetobacter baumannii in Mexico.

Acinetobacter baumannii has emerged as a critical global health threat due to its exceptional survival skills in adverse environment and its ability to acquire antibiotic resistance, presenting significant challenges for infection treatment and control. The World Health Organization has classified carbapenem-resistant A. baumannii as a "Critical Priority" pathogen to guide research and the development of control and prevention strategies. Epidemiological surveillance methodologies provide the tools necessary for classifying A. baumannii into international clonal lineages, facilitating the anal

Snake- and Spider-Venom-Derived Toxins as Lead Compounds for Drug Development.

Snake and spider venoms have been developed by nature as a defense mechanism against predators or to immobilize their prey by blocking the cardiovascular, respiratory, and/or nervous systems. Consequently, predators are deterred from approaching their prey by painful sensations. At a molecular level, the targeted physiological systems are blocked or stimulated by peptide toxins which, once injected into the body, modulate, though not exclusively, important cell membrane ion channels and receptors. Millions of years of constant evolution have led to the evolvement of complex venom libraries of

Genetic characterisation of influenza A(H1N1)pdm09 viruses circulating in Assam, Northeast India during 2009-2015.

INTRODUCTION: Influenza A(H1N1)pdm09 virus, since its identification in April 2009, has continued to cause significant outbreaks of respiratory tract infections including pandemics in humans. In the course of its evolution, the virus has acquired many mutations with an ability to cause increased disease severity. A regular molecular surveillance of the virus is essential to mark the evolutionary changes that may cause a shift to the viral behavior. MATERIALS AND METHODS: Samples of Throat/Nasal swabs were collected from a total of 3715 influenza-like illness cases and screened by Real-time Re

Comparative transcriptome analysis reveals expression signatures of albino Russian sturgeon, Acipenseriformes gueldenstaedtii.

Albinism is a genetically inherited condition that is caused by a series of genetic abnormalities leading to a reduction in melanin production. Russian sturgeon is one of the most valuable freshwater fish species worldwide, and albino individuals have been found in fish farms. Due to its complicated genome and scarce genome-wide genetic resources, the underlying molecular basis of albinism in Russian sturgeon is unknown. In the present study, we first generated transcriptome profile of Acipenser gueldenstaedtii using pooled tissues, which provided reliable reference sequences for future molecu

The anuran Bauplan: a review of the adaptive, developmental, and genetic underpinnings of frog and tadpole morphology.

Anurans (frogs, toads, and their larvae) are among the most morphologically derived of vertebrates. While tightly conserved across the order, the anuran Bauplan (body plan) diverges widely from that of other vertebrates, particularly with respect to the skeleton. Here we address the adaptive, ontogenetic, and genetic bases of three such hallmark anuran features: (1) the absence of discrete caudal vertebrae, (2) a truncated axial skeleton, and (3) elongate hind limbs. We review the functional significance of each as it relates to the anuran lifestyle, which includes locomotor adaptations to bot

Colitis-Associated Carcinoma: The Quintessential Epithelial Neoplasia Driven by Chronic Inflammation.

Colitis-associated carcinoma (CAC) represents ~1% of colorectal carcinomas and has important differences from sporadic colorectal carcinoma (sCRC). The precursors and carcinomas that arise in the setting of IBD are uniquely challenging to visualize by endoscopy and diagnose via histology, and the rising prevalence of IBD amplifies the challenges of surveillance to informed management. Although in broad strokes, CAC and sCRC share molecular features (~85% chromosomal instability pathway 15% microsatellite instability high (MSI-H)), CAC has a distinct distribution of molecular abnormalities, inc

Time, tissue, and team: Reframing transbronchial biopsy in lung transplantation as an integrated diagnostic system.

Despite remarkable advances in transplant pathology, molecular diagnostics, imaging, and biomarker discovery, uncertainty remains an intrinsic feature of lung allograft evaluation. Traditionally, this uncertainty has been attributed to sampling limitations, overlapping histologic patterns, imperfect diagnostic criteria, insufficiently specific biomarkers, and interobserver variability. While these factors undoubtedly contribute, they overlook a fundamental dimension of clinical practice: diagnosis is not generated by pathological findings in a vacuum but through communication among the discipl

Mesenchymal transitions reduce lamin A expression and nuclear stiffness to enhance confined migration in glioblastoma.

The evolution of glioblastoma (GBM) is coupled with a proneural-to-mesenchymal transition (PMT) that exacerbates disease in part through heightened cell invasiveness and spread. PMT follows similar molecular and phenotypic features with epithelial to mesenchymal transitions (EMT) and promotes rapid GBM invasion despite tight physical barriers in the dense brain parenchyma. Recent studies suggest that efficient confined migration is key to navigating these physical constraints; however, the role of PMT in mediating rapid confined migration remain unclear. Using microchannel platforms, AFM, and

Resampling-Iteration-Evolution (RIE): A Rational Design System for Engineering High-Affinity Peptides against Small Molecules.

Peptides are attractive molecular recognition elements because of their compact size, ease of modification, and structural tunability. The design of high-affinity peptides for small molecules lags far behind that of antibodies and aptamers due to the lack of general and effective discovery and optimization strategies. Computational design offers a promising avenue to overcome this bottleneck. Herein, we report a computational design pipeline, termed resampling-iteration-evolution (RIE), for the rapid discovery of high-affinity molecular recognition peptides (MRPs) from large structural and seq

Prolonged Survival in Metastatic Pancreatic Acinar Cell Carcinoma Treated with Hepatic Arterial Infusion of Oxaliplatin: A Case Report with Longitudinal Molecular Profiling.

INTRODUCTION: Pancreatic acinar cell carcinoma (PACC) is a rare malignancy, representing 1-2% of pancreatic cancers. Although it generally has a better prognosis than ductal adenocarcinoma, optimal systemic management remains undefined. Hepatic arterial infusion (HAI) of chemotherapy has shown efficacy in colorectal liver metastases but is rarely reported in pancreatic cancer. CASE PRESENTATION: We present a 36-year-old man diagnosed with metastatic PACC after developing abdominal pain and hepatomegaly. Imaging revealed a pancreatic tail mass and multiple liver metastases. Differential epigenetic regulation underlies evolutionary transition of seasonal migration.

Seasonal migratory animals can evolve residency through adaptation. The threshold model of quantitative genetics predicts that residency evolves from migrants when genetic variation for lower liability, or a latent state underlying migratory propensity, is positively selected. However, it is unknown what cellular and molecular regulations underlie this process. The hypothalamus, which regulates physiology and behaviour of vertebrates by integrating internal and environmental conditions, is a strong candidate structure where migratory propensity may be regulated. To understand the hypothalamic

Genome organization and packaging, mutation impacts and quasi-periodic patterns in viral genomes.

A method described in this publication is able (i) to display features related to the general genome organization and packaging; (ii) to identify explicitly motifs responsible for these features and the distribution of motifs over the genome; the most pronounced motifs can be screened against available databases for further studies; (iii) to assess the impact of mutations; the salient mutations can be tried experimentally and used for genome editing; and (iv) to quantify the information that can be insightful for general studies of molecular evolution of living organisms. The method is based o

Very Late Extramedullary Relapse of Acute Myeloid Leukemia 13 Years After Allogeneic Transplant: A Case Report.

Relapse of acute myeloid leukemia (AML) typically occurs within the first 3 years following treatment, with very late relapses being rare and often presenting extramedullary. We report a unique case of extramedullary AML relapse occurring 13 years after allogeneic hematopoietic stem cell transplantation. The patient developed a myelosarcoma, prompting molecular and histological investigation to determine the origin and mutational landscape of the disease. Next-generation sequencing (NGS) was performed on archived bone marrow samples from the initial AML diagnosis in 2007 and on the relapsed tu

Drug-resistant Acinetobacter baumannii: mortality, emerging treatments, and future pharmacological targets for a WHO priority pathogen.

SUMMARYAcinetobacter baumannii has emerged as a formidable global health concern and is a major contributor to infection-related mortality in critically ill patients worldwide. This versatile Gram-negative bacterium is notorious for its highly plastic genome, which enables the rapid emergence and dissemination of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains, severely limiting the available treatment options. The genetic flexibility of A. baumannii underpins its arsenal of molecular mechanisms, enabling it to resist a range of antibiotics, from traditional agents to th

Navigating the landscape of EGFR TKI resistance in EGFR-mutant NSCLC - mechanisms and evolving treatment approaches.

Resistance to EGFR tyrosine kinase inhibitors (TKIs) remains a major obstacle in the clinical management of EGFR-mutant non-small-cell lung cancer (NSCLC). Despite the transformative therapeutic activity of the multiple iterations of EGFR TKIs, spanning from first-generation reversible inhibitors such as erlotinib and gefitinib to the current standard-of-care third-generation covalent inhibitor osimertinib, primary or acquired resistance to these agents inevitably emerges via diverse mechanisms. The advent of combination therapies that incorporate chemotherapy, anti-angiogenic agents, bispecif

The formation and propagation of human Robertsonian chromosomes.

Robertsonian chromosomes are a type of variant chromosome that is commonly found in nature. Present in 1 in 800 humans, these chromosomes can underlie infertility, trisomies and increased cancer incidence1-5. They have been recognized cytogenetically for more than a century6, yet their origins have remained unknown. Here we describe complete assemblies of three human Robertsonian chromosomes. We identified a common breakpoint in SST1, a macrosatellite DNA located on chromosomes 13, 14 and 21, which commonly undergo Robertsonian translocation. SST1 is contained within a larger shared homology d

Single-Cell Landscape of the Cochlea Revealed Cell-Type-Specific Diversification in Hipposideros armiger Based on PacBio Long-Read Sequencing.

Echolocation represents one of the most rapid adaptive sensorimotor modulation behaviors observed in mammals, establishing bats as one of the most evolutionarily successful mammals. Bats rely on high-frequency hearing for survival, but our understanding of its cellular molecular basis is scattered and segmented. Herein, we constructed the first single-cell transcriptomic landscape of the cochlea in Hipposideros armiger, a CF-FM bat, using a PacBio-optimized genome and compared it with the results obtained from unoptimized original genomes.
