Competitive exclusion strategies utilizing healthy caecal microbiota impair Salmonella enterica serovars colonization in broilers.

Avian non-typhoidal Salmonella (NTS) is the second most prevalent foodborne zoonotic pathogen worldwide, which is transmitted through poultry products. Human-infecting serovars exhibit genotypic and phenotypic diversity, which affect bacterial virulence, such as susceptibility to antimicrobial drugs. The emergence of antimicrobial-resistant strains further reduces treatment efficacy, underscoring the need for alternatives in food-producing animals. One promising strategy is the use of competitive exclusion (CE) products, formulated with naturally occurring microorganisms derived from the gut m

Biosynthetic gene clusters in Pseudomonas viridiflava have a fitness cost during Arabidopsis thaliana infection.

UNLABELLED: Specialized or secondary metabolites mediate biotic interactions, including virulence and defense. In plant-pathogenic Pseudomonas, certain specialized metabolites can enhance colonization of plant hosts, yet their broader contribution to plant-microbe interactions and the relative importance of different metabolites remain unclear. Specialized metabolites are products of enzymes encoded in biosynthetic gene clusters (BGCs), whose prediction from genome sequences has become routine but whose functional roles are rarely tested experimentally. Here, we characterize the BGC repertoire

Microbiome Continuum: Connecting Environmental, Host-Associated and Engineered Microbiomes Through One Health and Artificial Intelligence.

Microbiomes are increasingly understood as interconnected ecological systems spanning soil, water, plants, animals, humans and engineered environments rather than as isolated host-associated communities. This microbiome continuum provides a mechanistic perspective for One Health by linking microbial cells with the movement of metabolites, genes, signalling molecules and ecological functions across environmental boundaries. Here, we critically examined the biological processes that maintain this continuity, including dispersal, horizontal gene transfer (HGT), metabolic exchange, environmental e

Transplant of cryopreserved mosquito gut microbiota reveals strain-specific patterns of microbial acquisition and assembly.

Mosquitoes are important vectors of human pathogens, and their gut microbiome influences traits such as survival, fecundity, pathogen resistance, insecticide resistance, and vector competence. Manipulating this microbiome offers a promising strategy for reducing mosquito-borne disease. While 16S rRNA gene amplicon sequencing has revealed global patterns of gut microbial diversity, the factors driving these patterns remain poorly understood. Here, we used a community-transplantation approach that allows the collection, cryopreservation, and transfer of whole adult microbial communities into axe

Phage therapy for multidrug-resistant pulmonary infections: bridging precision matching, lung delivery, host immunity, and clinical translation.

Chronic pulmonary infections caused by multidrug-resistant (MDR) bacteria are a growing global health problem, and the declining effectiveness of conventional antibiotics increases the need for alternative antimicrobial strategies. Unlike broad-spectrum antibiotics, bacteriophages are highly specific, and their use therefore offers not only antibacterial activity but also the possibility of reshaping the airway microbial ecosystem in a targeted manner. In this review, we reframe phage therapy as a precision microbiome-modulating strategy for chronic pulmonary infections. We first review precis

2'-FL's Depiction of the Human Physiological Function Landscape.

2'-fucosyllactose (2'-FL) is a major fucosylated human milk oligosaccharide characterized by a terminal α1,2-linked fucose residue, which supports its interactions with gut microbes, host glycan-recognition systems, and pathogen-binding sites. This review proposes that 2'-FL acts mainly through a hierarchical host-microbiota mechanism: it first shapes gut microbial composition, then influences microbial metabolites such as short-chain fatty acids, and subsequently contributes to intestinal barrier integrity, immune homeostasis, and broader physiological outcomes. Current evidence is strongest

Characterization of a Novel OTU-like Deubiquitinase from Babesia microti: Implications for Babesiosis Treatment.

Babesia microti is the primary agent of human babesiosis, an emerging tick-borne disease with limited treatment options and growing evidence of drug resistance. Deubiquitinases (DUBs) play critical roles in protein homeostasis and host-pathogen interactions, yet none have been characterized in B. microti. Here, we report the first molecular cloning, expression, and functional characterization of an OTU-like cysteine protease from B. microti (bm-OTU). The recombinant bm-OTU protein (~22 kDa) was expressed in E. coli, purified to high homogeneity, and exhibited ~93% solubility under native condi…

Koala MHCII association with chlamydia infertility remains equivocal: a need for new research approaches.

Chlamydiosis is a common infectious disease impacting koalas and is a major cause of population decline due to resulting mortality and infertility. Polymorphisms of major histocompatibility complex (MHC) genes influence chlamydial disease outcomes in several species but koala studies have produced variable results. We aimed to identify the MHC II DAB and DBB repertoire of koalas from Liverpool Plains, NSW, a population heavily impacted by chlamydiosis. We compared variants between two studies, age cohorts and chlamydial infertility groups. Four DBB and eight DAB alleles were identified. The me

Interactions between Gender and Sepsis-Implications for the Future.

Sex and gender dimorphisms are found in a large variety of diseases, including sepsis and septic shock which are more prevalent in men than in women. Animal models show that the host response to pathogens differs in females and males. This difference is partially explained by sex polarization of the intracellular pathways responding to pathogen-cell receptor interactions. Sex hormones seem to be responsible for this polarization, although other factors, such as chromosomal effects, have yet to be investigated. In brief, females are less susceptible to sepsis and seem to recover more effectivel

Differential microRNAs expression during ex vivo infection of canine and ovine placental explants with Trypanosoma cruzi and Toxoplasma gondii.

Trypanosoma cruzi and Toxoplasma gondii are two zoonotic parasites that constitute significant human and animal health threats, causing a significant economic burden worldwide. Both parasites can be transmitted congenitally, but transmission rates for T. gondii are high, contrary to what has been observed for T. cruzi. The probability of congenital transmission depends on complex interactions between the pathogen and the host, including the modulation of host cell gene expression by miRNAs. During ex vivo infection of canine and ovine placental explants, we evaluated the expression of 3 miRNAs

A model-specific role of microRNA-223 as a mediator of kidney injury during experimental sepsis.

Sepsis outcomes are heavily dependent on the development of septic organ injury, but no interventions exist to interrupt or reverse this process. microRNA-223 (miR-223) is known to be involved in both inflammatory gene regulation and host-pathogen interactions key to the pathogenesis of sepsis. The goal of this study was to determine the role of miR-223 as a mediator of septic kidney injury. Using miR-223 knockout mice and multiple models of experimental sepsis, we found that miR-223 differentially influences acute kidney injury (AKI) based on the model used. In the absence of miR-223, mice de…

Methods for studying mucin-microbe interactions.

Mucin glycoproteins are the major structural components of mucus, imparting its gel consistency. Not only do mucins form a protective barrier that traps debris and keeps pathogenic bacteria at a safe distance from host cells, but they display structurally complex glycans that feed beneficial microbes and directly interact with microbial pathogens to regulate gene expression and virulence. This chapter provides a conceptual framework for mucin-microbe interaction studies and integrated protocols spanning the purification of biologically relevant mucins, isolation of complex glycan pools for mec

Novel internalin P homologs in Listeria.

Listeria monocytogenes (Lm) is a bacterial pathogen that causes listeriosis in immunocompromised individuals, particularly pregnant women. Several virulence factors support the intracellular lifecycle of Lm and facilitate cell-to-cell spread, allowing it to occupy multiple niches within the host and cross-protective barriers, including the placenta. One family of virulence factors, internalins, contributes to Lm pathogenicity by inducing specific uptake and conferring tissue tropism. Over 25 internalins have been identified thus far, but only a few have been extensively studied. Internalins co

Genome dynamics of Bartonella grahamii in micro-populations of woodland rodents.

BACKGROUND: Rodents represent a high-risk reservoir for the emergence of new human pathogens. The recent completion of the 2.3 Mb genome of Bartonella grahamii, one of the most prevalent blood-borne bacteria in wild rodents, revealed a higher abundance of genes for host-cell interaction systems than in the genomes of closely related human pathogens. The sequence variability within the global B. grahamii population was recently investigated by multi locus sequence typing, but no study on the variability of putative host-cell interaction systems has been performed. RESULTS: To study the populat
