BACKGROUND: Human heart failure involves multicellular transcriptional remodeling, but public transcriptomic studies often remain disconnected from cell-type localization and perturbational interpretation.
METHODS: We developed a reproducible computational workflow integrating human left-ventricular bulk transcriptomes, donor-level cell-type pseudobulk results from a human heart-failure single-cell/single-nucleus atlas, external snRNA-seq support, curated module scoring, focused ligand-receptor prioritization and LINCS/L1000 perturbational matching.
RESULTS: Cross-cohort analysis identified

Osteosarcopenia combines low bone mass or skeletal fragility with sarcopenia, yet its metabolic biology is commonly assessed using single-time-point measurements. We conducted a structured narrative review of PubMed/MEDLINE, Web of Science Core Collection, Embase and Scopus from inception through 28 June 2026, supplemented by ScienceDirect, Google Scholar and citation chaining. Evidence was classified as direct human osteosarcopenia evidence, indirect human evidence, preclinical evidence or conceptual inference. Here, chronometabolomics denotes time-anchored or repeated metabolomic profiling u

The human immune response and factors that modulate it represent challenges and opportunities in many areas of medicine. Examples are the threat of transplant rejection and the reduction of this threat by the administration of a bacterially derived immune evasion enzyme. Streptococcus species secrete proteases of the IdeS/Mac family that efficiently degrade host immunoglobulin G (IgG) to help the bacteria evade the host's immune response. IdeS has recently received interest as a tool for transiently ablating circulating IgG in the context of diverse treatments, including transplantation and ge

Advances in generative artificial intelligence (GenAI) have prompted interest in its application in sensitive fields, including mental health. Yet the validity of its nonverbal social-cognitive abilities remains unclear. This study addresses two issues that probe GenAI's social intelligence: its capacity to interpret nonverbal channels (bodily gestures and vocal tone) with accuracy comparable to humans, and whether accuracy of emotion recognition is different by emotional valence. Emotion recognition accuracy of three Gemini GenAI models (Pro 1.5, Pro 2 (Experimental) and Flash 2) were evaluat

Over the past 30 years, the study of Mycobacterium tuberculosis has become a vibrant field that supports basic investigation into mycobacteria, wide-ranging studies of M. tuberculosis pathogenesis, and drug and therapeutic development. In this minireview, we highlight the multiple ways in which M. tuberculosis is a model. M. tuberculosis has emerged as a model for prokaryotic biology, revealing basic mechanisms of gene expression, genomic integrity, and mutagenesis that expand prokaryotic dogmas established in earlier model organisms. Next, we highlight the experimental systems used to model M

Plastic pollution has emerged as a critical global environmental challenge, yet the effects of the ingested plastic particles on ocular health remain largely unexplored. In this study, we investigated the impact of orally ingested polystyrene nanoplastics (PS-NPs) on the mouse retina. The in vivo experimental results showed that PS-NPs could penetrate the mouse retina within 2 h after gavage. Their levels increased at 4 h and remained detectable up to 24 h post-gavage. Prolonged exposure (28 days) to PS-NPs might disrupt the tight junctions of the inner blood-retinal barrier (iBRB). Moreover,

Motion assist robots have been studied to assist human motion for elderly, disabled, injured persons. The motion assist robots can be classified into two types: wearable robots which are directly attached to the user's body and external robots which assist the user's motion from the outside of the user. It is important for those robots to estimate motion intention of users in real-time to assist their daily motion effectively. This paper presents a method to estimate human lower-limb motion intention and its motion phase in real-time for the motion assist robots. By detecting motion phase in a

The global prevalence of ulcerative colitis (UC) and Crohn's disease (CD) is increasing, placing greater burdens on national health systems. The pathophysiology of diarrhea, the commonest debilitating symptom in patients with UC and CD, has been studied more extensively in UC, where it reflects defective colonic Na+ absorption combined with changes in colonic Cl- and K+ transport, which significantly reduce colonic water absorption. Dysfunctional ion transport in patients with UC is accompanied by abnormalities in tight junctional protein distribution and function, which cause the inflamed col

Rhesus macaques (Macaca mulatta) are the most common non-human primates living in captivity. The use of rhesus macaques as model objects is determined, first of all, by their phylogenetic and physiological closeness to humans, and, as a consequence, the possibility of extrapolating the obtained results to humans. Currently, it is known that a number of biochemical changes occur under various physiological conditions, including at the transcriptomic level. The real-time polymerase chain reaction is a widely used universal method for gene expression analysis. Carrying out such studies always req

PURPOSE: The measurement of passive muscle-tendon parameters (PMPs) is crucial for understanding and analyzing human movement. However, the common methods for measuring PMPs are inconsistent or involve invasive procedures.
METHODS: We propose a novel noninvasive method for estimating PMPs using a direct collocated optimal control algorithm. The optimal control algorithm was employed to determine PMPs of a fully in silico musculoskeletal simulation, of a mechanical analogue, and of human subjects in vivo. To eliminate the confounding effects of force-velocity and active muscle contraction, a q

The development and evaluation of effective therapeutic strategies for intervertebral disc degeneration (IVDD) and their successful incorporation into clinical practice remain challenging, largely due to the absence of an optimal preclinical animal model. While both induced and spontaneous IVDD animal models have provided valuable insights, they also present inherent limitations that restrict their translational applicability. Increasing evidence supports the use of sheep as a highly relevant large animal model, as spontaneous spine disorders in this species closely mirror many features of hum

The uterine tubes and uterus develop from the paramesonephric (Müllerian) ducts. Most experimental data are obtained in rodents. Since the (micro-)anatomy of the murine urogenital tract differs from that in humans, evaluation of the translatability of mouse data to human development is relevant. We studied the Müllerian ducts in serially sectioned female human embryos and fetuses between 5 and 15 weeks of development and prepared 3D-reconstructions to establish topographic relations. At 5 weeks of development, the dorsocranial peritoneal epithelium thickens locally to form a placode-like struc

INTRODUCTION: Chest Compression Synchronized Ventilation (CCSV) is a novel approach aimed at optimizing gas exchange and hemodynamics during cardiopulmonary resuscitation (CPR). However, its clinical value, safety profile and implementation barriers remain unclear. This study aimed to systematically synthesize existing evidence on the use of CCSV during cardiac arrest in animals and humans.
METHODS: We conducted a scoping review and systematically searched five databases (Medline, Embase, CENTRAL, Scopus, Web of Science) up to May 2025. Studies investigating CCSV or mechanistically related ve

Legionella pneumophila is an endosymbiotic bacterial species able to infect and reproduce in various protist and human hosts. Upon entry into human lungs, they may infect lung macrophages, causing Legionnaires' disease (LD), an atypical pneumonia, using similar mechanisms as in their protozoan hosts, despite the 2 hosts being separated by a billion years of evolution. In this study, we used experimental evolution to identify genes conferring host specificity to L. pneumophila. To this end, we passaged L. pneumophila in 2 different hosts-Acanthamoeba castellanii and the human macrophage-like ce

Tuberculosis (TB) remains a leading cause of death, but antibiotic treatments for tuberculous meningitis, the deadliest form of TB, are based on those developed for pulmonary TB and not optimized for brain penetration. Here, we perform first-in-human dynamic 18F-pretomanid positron emission tomography (PET) in eight human subjects to visualize 18F-pretomanid biodistribution as concentration-time exposures in multiple compartments (NCT05609552), demonstrating preferential brain versus lung tissue partitioning. Preferential, antibiotic-specific partitioning into brain or lung tissues of several

Lacking specimens is the biggest limitation of studying the mechanical behaviors of human corneal. Extracting stress-strain curves is the crucial step in investigating hyperelastic and anisotropic properties of human cornea. 15 human corneal specimens extracted from the small incision lenticule extraction (SMILE) surgery were applied in this study. To accurately measure the personalized true stress-strain curve using corneal lenticules, the digital image correlation (DIC) method and finite element method were used to calibrate the stress and the strain of the biaxial extension test. Chronic alcoholic liver disease has brought great harm to human health. Alcoholic fatty liver disease is the first stage in the progression of all chronic alcoholic liver diseases. At present, there is no cell model that fully matches the etiology (high-fat diet + alcohol) of human alcoholic fatty liver disease. We used 100 mM ethanol +6.25 μM PA to establish the ethanol combined with PA-induced mouse hepatocyte AFLD model (EP-AFLD hepatocyte model) and performed the RNA-seq transcriptome sequencing. Through bioinformatics analysis and comparison, we discovered that the EP-AFLD hepatocyte mode

Lateral gene transfer (LGT) is an important mechanism for genome diversification in microbial populations, including the human microbiome. While prior work has surveyed LGT events in human-associated microbial isolate genomes, the scope and dynamics of novel LGT events arising in personal microbiomes are not well understood, as there are no widely adopted computational methods to detect, quantify, and characterize LGT from complex microbial communities. We addressed this by developing, benchmarking, and experimentally validating a computational method (WAAFLE) to profile novel LGT events from

Toxoplasma gondii is the causative agent of toxoplasmosis, a disease that affects warm-blooded animals and one third of the human population worldwide. Pregnant women who have never been exposed to the parasite constitute an important risk group, as infection during pregnancy often leads to congenital toxoplasmosis, the most severe form of the disease. Current therapy for toxoplasmosis is the same as it was 50 years ago and has little or no effect when vertical transmission occurs. Therefore, it is urgent to develop new strategies to prevent mother-to-fetus transmission. The implementation of

Sulfate plays a pivotal role in numerous physiological processes in the human body, including bone and cartilage health. A role of the anion transporter SLC26A1 (Sat1) for sulfate reabsorption in the kidney is supported by the observation of hyposulfatemia and hypersulfaturia in Slc26a1-knockout mice. The impact of SLC26A1 on sulfate homeostasis in humans remains to be defined. By combining clinical genetics, functional expression assays, and population exome analysis, we identify SLC26A1 as a sulfate transporter in humans and experimentally validate several loss-of-function alleles.
