Origin and diversity of human retroviruses.

Simian immunodeficiency viruses, simian T‑cell lymphotropic viruses, and simian foamy viruses from nonhuman primates have crossed the species barrier to humans at several time points, leading to the HIV and human T lymphotropic virus epidemic and to sporadic cases of human infections with simian foamy viruses, respectively. Efficient infection and spread in humans differs between simian foamy virus, simian lymphotropic virus, and simian immunodeficiency virus, but seems also to differ among the different viruses from the same simian lineage, as illustrated by the different spread of HIV‑1 M, N…

Ancient origin and molecular features of the novel human T-lymphotropic virus type 3 revealed by complete genome analysis.

Human T-lymphotropic virus type 3 (HTLV-3) is a new virus recently identified in two primate hunters in Central Africa. Limited sequence analysis shows that HTLV-3 is distinct from HTLV-1 and HTLV-2 but is genetically similar to simian T-lymphotropic virus type 3 (STLV-3). We report here the first complete HTLV-3 sequence obtained by PCR-based genome walking using uncultured peripheral blood lymphocytes from an HTLV-3-infected person. The HTLV-3(2026ND) genome is 8,917 bp long and is genetically equidistant from HTLV-1 and HTLV-2, sharing about 62% identity. Phylogenetic analysis of all gene r

Inhibition and augmentation of progesterone production during pregnancy: effects on parturition in rhesus monkeys.

OBJECTIVES: Uterine quiescence during mammalian pregnancy is attributed to progesterone. However. systemic progesterone levels remain elevated in primates before parturition. Epostane, a selective 3beta-hydroxysteroid dehydrogenase inhibitor, and progesterone (with or without epostane) were administered to late pregnant rhesus monkeys to clarify the role of progesterone in primate parturition. STUDY DESIGN: On days 122 to 132 of gestation (term 167 days), 11 rhesus monkeys (Macaca mulatta) with timed pregnancies were divided into three treatment groups: (1) epostane alone (10 mg/kg subcutaneo…

What is the situation of human T cell lymphotropic virus type II (HTLV-II) in Africa? Origin and dissemination of genomic subtypes.

Human T cell lymphotropic virus type II (HTLV-II) and its two genomic subtypes, A and B, which differ by 3 to 6% at the nucleotide level (depending on the gene studied), were until recently considered to be endemic only in certain Indian tribes in the Americas and were therefore considered mainly as a "New World virus." First, the evidence of HTLV-II antibodies and later characterization of isolates from sex workers or individuals living in large West and Central African cities suggested that HTLV-II subtype A could have been imported recently in Africa. However, the findings of HTLV-II infect

Another murine preparation, OKT4A, was initially found to prolong allograft survival in non-human primates. Limited clinical trials revealed that this mAb was well tolerated, that most recipients produced an antimurine response, and that only 26% of patients suffered rejection episodes during the first 3 post-operative months. Another murine preparation, …

Thalamotomy for the alleviation of levodopa-induced dyskinesia: experimental studies in the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-treated parkinsonian monkey.

This work set out to test the hypothesis that thalamotomy in the area of the thalamus which receives the input from the medial segment of the globus pallidus would decrease or prevent levodopa-induced dyskinesia. Peak dose dyskinesia is a major problem in the treatment of parkinsonian patients with levodopa therapy but this remains the best pharmacological agent for treating the condition. The hypothesis was derived from previous work which has suggested that reduced pallidal inhibition of the thalamus results in dyskinesia [Crossman (1990) Movement Dis. 5, 100-108]. Brain weight across the adult lifespan in great apes.

Reduction in brain weight is a hallmark feature of human aging, which is exacerbated in pathologies, such as Alzheimer's disease and other dementias. However, it remains unclear whether a decrease in brain weight with age reflects a vulnerability unique to the human lineage or represents a consequence of an extension of aging processes shared with other primates. To address this question, we examined age-related changes in brain weight across our closest living relatives, the great apes (chimpanzees, bonobos, gorillas, and orangutans). The sample spanned the full adult age range in each specie…

Allospecific splenic Tr1 cells drive effector T cell exhaustion through up-regulated Areg-EGFR signaling to promote transplant tolerance.

Inducing stable tolerance to transplants remains a challenge in immunology. Previously, we induced tolerance to allogeneic islets in nonhuman primates by preemptive alloantigen delivery to antigen-presenting cells in situ. Here, mass cytometry phenotyping with incorporated donor-derived MHC-I peptide-loaded MHC-II tetramers revealed accumulation of allospecific CD4+ T cell clusters in the spleen of tolerant recipients. Areg+Tr1 regulatory and terminally exhausted EGFRhi T (Tex) cells represented the predominant allospecific subsets. Trajectory analysis showed that antigen-experienced effector …

Transcranial alternating current stimulation can disrupt or reestablish neural entrainment in parkinsonian motor cortex.

BACKGROUND: In Parkinson's disease, motor network electrophysiology frequently exhibits excessive beta oscillations. The cornerstone of ther … model. METHODS: We recorded neuronal spike activity in the motor cortex in parkinsonian non-human primates during tACS to determine how stimulation-induced electric fields affect spike timing. RESULTS: Strong electric fields entrained neural activity at the stimulation frequency but altered the preferred spiking phase. Conversely, weak fields disrupted …

Noninvasive Suppression of Responses to Threat Using Focused Ultrasonic Waves.

The processing of threat- and fear-related responses is known to involve the amygdala and the bed nucleus of the stria terminalis. Malignant activity in these nuclei is associated with several important disorders, including generalized anxiety disorder and post-traumatic stress disorder. A noninvasive approach to modulate these deep brain circuits could provide novel treatment options for these disorders, but current noninvasive approaches cannot access these targets directly. This study modulates these nuclei using transcranial low-intensity focused ultrasound in nonhuman primates. The ultras…

Phase 1/2a Clinical Trial Protocol for Lentiviral Vector-Based Retinal Gene Therapy to Slow the Progression of Retinitis Pigmentosa.

Retinitis pigmentosa (RP) is a group of inherited retinal dystrophies (IRDs) that cause progressive rod and cone photoreceptor cell death. C … models of RP and achieved stable, safe, and long-term transgene expression over 5 years in nonhuman primates. Based on these findings, we conducted a Phase 1/2a investigator-initiated clinical trial using SIV-hPEDF to slow the disease progression and prevent blindness in RP patients, and we here outline the study protocol.

Evaluating southern pigtail macaques as an immunological model for human antibody responses.

Antibody specificity is primarily derived from immunoglobulin (Ig) genes, which exhibit extensive genetic polymorphism both within and across species. Non-human primates (NHP) such as pigtail macaques are often used as preclinical models for immunological studies because of their genetic proximity to humans, despite a comparatively poor characterisation of their immunoglobulin loci and its impact on antibody specificity. To determine how pigtail macaque antibody responses differ from humans, we annotated the immunoglobulin loci of two pigtail macaques and compared BCR sequences and antigen bin…

Triazole 187 is a biased KOR agonist that suppresses itch without sedation and induces anxiolytic-like behaviors in mice.

Kappa opioid receptor agonists are clinically used to treat pruritus and have therapeutic potential for the treatment of pain and neuropsychiatric disorders. We have previously shown that triazole 1.1 is a G protein signaling-biased KOR agonist, that can suppress itch without producing signs of sedation in mice. This profile was recapitulated in rats and non-human primates; however, triazole 1.1 had limited potency as an antipruritic. Here we describe a more potent, G protein signaling-biased agonist, triazole 187. Triazole 187 is a potent antipruritic agent and does not decrease spontaneous l…

Plasticity of synapses innervating spinal motor neurons after spinal cord injury in rhesus monkeys.

Patients with incomplete spinal cord injuries can experience partial recovery of motor function, a phenomenon believed to be linked to neural plasticity, as evidenced by numerous studies in rodent models. However, postinjury synaptic plasticity in non-human primates, which better mirror human neuroanatomy, is less well understood. This study investigated the effects of T10 thoracic spinal cord hemisection or over-hemisection (transection beyond the posterior median sulcus) injuries in 7 rhesus monkeys. Analysis of lumbar motor neurons (MNs) innervating gluteal and crural muscles showed increas…

Transgenic mouse models for investigating human DUX4 expression during development and its roles in FSHD pathophysiology.

Facioscapulohumeral muscular dystrophy (FSHD) is an autosomal dominant myopathy caused by aberrant expression of the double homeobox 4 (DUX4) retrogene, affecting skeletal muscles primarily in the face, shoulder and limbs. In healthy individuals, DUX4 is expressed in early development and subsequently silenced in most somatic tissues. The spatiotemporal pattern of DUX4 misexpression beyond the cleavage stage in FSHD is poorly understood because DUX4 is not well conserved beyond primates. Here, we generated Cre reporter mouse lines with human DUX4 regulatory elements to investigate the cell lin…
