Primary palmar hyperhidrosis locus maps to 14q11.2-q13.

Primary palmar hyperhidrosis (PPH) is a unique disorder of unknown cause. It is characterized by excessive perspiration of the eccrine sweat gland in the palm, sole, and the axilla. It is presumed that PPH results from overactivation of the cholinergic sympathetic nerve or dysfunction of the autonomic nervous system. There have been no genetic studies on the disease. We performed a linkage analysis of 11 families including 42 affected and 40 unaffected members using genome-wide DNA polymorphic markers to identify the disease locus. Diagnosis of their PPH was made by direct inspection, intervie

Beta2-agonist and anticholinergic drugs in the treatment of lung disease.

The lungs are innervated by both the sympathetic and parasympathetic nervous systems, which entails the activation of adrenergic and muscarinic receptors, respectively. Both the adrenergic and muscarinic receptors are G-protein-coupled receptors, and they share many similar signal transduction molecules. These receptors are widely expressed in the lung and the specific receptor expression can vary among the species. The location and the subtype of receptor expressed are important in the regulation of normal airway function. Acetylcholine released from the parasympathetic fibers activates the M…

Evidence-based care for the elderly with isolated systolic hypertension.

This paper reviews the epidemiology, pathophysiology and clinical significance of isolated systolic hypertension (ISH) in the elderly. Aging is associated with structural and functional changes in the arterial tree. Intimal thickening, migration of small muscle cells to the intima, medial fibrosis, and elastic fiber degeneration result in increased arterial stiffness and ISH. The augmented systemic vascular resistance in the elderly is mediated by increased arterial stiffness. Aging is correlated with overactivity of the sympathetic nervous system, reduced neuronal plasma norepinephrine uptake…

Review article: hepatorenal syndrome--how to assess response to treatment and nonpharmacological therapy.

Hepatorenal syndrome (HRS) is a complex syndrome. In addition to severe reduction of renal function due to renal vasoconstriction, there is impairment in systemic haemodynamics, activation of the renin-angiotensin and sympathetic nervous systems and antidiuretic hormone, vasoconstriction of the brain, muscle and skin, and dilutional hyponatraemia. Treatment in patients with type 2 HRS, the most frequent form of HRS, is directed towards managing refractory ascites. Paracentesis is the treatment of choice. TIPS is also effective but is more expensive, is associated with higher incidence of hepat…

Nodular osteochondrogenic activity in soft tissue surrounding osteoma in neurogenic para osteo-arthropathy: morphological and immunohistochemical study.

BACKGROUND: Neurogenic Para-Osteo-Arthropathy (NPOA) occurs as a consequence of central nervous system injuries or some systemic conditions. They are characterized by bone formation around the main joints. METHODS: In order to define some biological features of NPOAs, histological and immunohistological studies of the soft tissue surrounding osteoma and Ultrasound examination (US) of NPOA before the appearance of abnormal ossification on plain radiographs were performed. RESULTS: We have observed a great number of ossifying areas scattered in soft tissues. US examination have also shown scat

Physiologic and hemodynamic basis of ventricular assist devices.

Heart failure is a particularly complex disorder with etiology that is primary in nature or secondary to other systemic diseases, including hypertension, diabetes, and atherosclerosis. The pathogenesis appears to result, in part, from extensive abnormal interactions among tissues, such as the heart, vasculature, kidney, lungs, and sympathetic nervous system. Improvements in understanding this complex disorder, particularly factors that contribute to cardiac cell cycle alterations, gene activation and re-expression resulting in cardiac remodeling and, eventually, maladaption are paramount. Clin…

Effect of ganglionic blockade on catecholamine secretion in exercised dogfish.

A brief bout of vigorous exercise results in significant increases in plasma epinephrine (E) and norepinephrine (NE) in the dogfish, Squalus acanthias. Since the presence of a functioning sympathetic nervous system in dogfish is in doubt, experiments were undertaken to show whether or not exercise-induced catecholamine (CA) secretion is under autonomic neurogenic control. Changes in plasma E and NE in a control group of exercised fish were compared with changes in fish exercised while under the influence of ganglionic blockade. Ganglionic blockade was induced in dogfish by hexamethonium infusi…

Chronic psychosocial stress in tree shrews: effect of the substance P (NK1 receptor) antagonist L-760735 and clomipramine on endocrine and behavioral parameters.

RATIONALE: Substance P and its preferred receptor, the neurokinin 1 receptor (NK(1)R), have been proposed as possible targets for new antidepressant therapies, although results of a recently completed phase III trial failed to demonstrate that the NK(1)R antagonist MK-869 is more effective than placebo in the treatment of depression. METHODS: In the present study, we compared the effects of the NK(1)R antagonist L-760735 with the tricyclic antidepressant clomipramine on endocrine and behavioral parameters in chronically stressed tree shrews. Animals were subjected to a 7-day period of psychos

Endothelial semaphorin 6D controls immune responses under cold stress through regulation of sympathetic innervation.

Environmental temperature significantly influences immune responses. Cold exposure suppresses host defense against infections and exacerbates autoimmune and allergic conditions. However, the molecular mechanisms underlying temperature-dependent immune regulation remain unclear. In this study, we evaluated the cold-activated sympathetic modulation of immune responses. We presented that semaphorin 6D (Sema6D), an axon guidance molecule, is required for proper sympathetic nerve distribution. While both wild-type and Sema6d-/- mice developed experimental autoimmune encephalomyelitis (EAE) similarl

Interplay between Skin Sympathetic Nerve Activity and Gut Microbiota, the Neurocardiac Axis in Acute Coronary Syndrome.

Our previous study demonstrated increased skin sympathetic nerve activity (SKNA) in acute coronary syndrome (ACS) patients and its correlation with ventricular arrhythmias. In the current study, we explored specific gut microbial profiles and their relationships with SKNA in ACS patients. Fifteen ACS patients and age- and sex-matched controls were included in this cross-sectional study. Demographic and clinical data were recorded, and SKNA was measured by neuECG. The gut microbial profiles were analyzed by 16S rRNA gene sequencing of the V3-V4 region. Statistically significant differences in a

The NE-adrenergic receptor axis drives osteoarthritis progression via neurovascular cross-talk: a dual-responsive hydrogel strategy.

Osteoarthritis (OA) progression is strongly driven by the dysregulated interplay between the sympathetic nerve and abnormal vascularization, severely affecting patients' physical and mental well-being. In this study, temporomandibular joint osteoarthritis (TMJ-OA) was induced in mice using a unilateral anterior crossbite (UAC) model. We found that sympathetic nerve activation promoted pathological angiogenesis and vasoconstriction, whereas blocking sympathetic nerves inhibited both processes. Furthermore, during disease progression, hyperinnervation by sympathetic nerves in the subchondral bon

Peripheral cholecystokinin promotes thermogenesis via vagal afferent signalling, hypothalamic oxytocin and sympathetic outflow to brown adipose tissue.

Thermoregulation is essential for survival in homeothermic animals. Vagal sensory nerves are well known to detect visceral signals and regulate feeding and metabolic functions, but their role in thermoregulation remains poorly understood. Cholecystokinin (CCK), a gut hormone released postprandially, activates vagal sensory nerves via CCK-A receptors (CCK-AR). Although exogenous CCK has been reported to induce thermogenesis in intrascapular brown adipose tissue (iBAT), whether this effect depends on vagal pathways and specific central circuits remains incompletely understood. Here, we assessed

Systematic understanding of acute effects of dexmedetomidine on rat carotid sinus baroreflex-mediated sympathetic arterial pressure regulation.

Dexmedetomidine (DEX) is a centrally acting α2-adrenergic agonist widely used for sedation in emergency and intensive care settings. We examined the effects of DEX on baroreflex-mediated sympathetic regulation of arterial pressure (AP) to distinguish its central and peripheral effects. In anesthetized male Wistar-Kyoto rats (n = 8), baroreflex-mediated changes in sympathetic nerve activity (SNA), AP, and heart rate (HR) were evaluated before and after intravenous DEX administration. DEX (2.5 μg/kg) significantly reduced the response ranges of AP (64.4 ± 9.2 to 5.8 ± 3.0 mmHg; P < 0.001), HR (4

Sympathetic hyperactivity mediates the association between obstructive sleep apnea and hypertension: age effect.

BACKGROUND: Sympathetic hyperactivation is one of underlying mechanisms of obstructive sleep apnea (OSA) associated hypertension. The association between OSA and hypertension is stronger in young and middle-aged than older adults. We hypothesized that sympathetic hyperactivity mediates the association between OSA and hypertension and this effect is moderated by age. METHODS: OSA was defined as an apnea-hypopnea index (AHI) ≥5 events/hour based on an overnight polysomnography. Time under 90% oxygen (T90) was used to assess hypoxic severity. Twenty-four-hour urinary norepinephrine was used to a

Single acupressure on ST36 does not modulate skin blood flow and sympathetic nerve activity during and after cold stimulation of the hand.

Acupressure on ST36 (Zusanli), located on the lower leg, commonly used to optimize autonomic nerve activity and systemic blood flow in traditional Chinese medicine. However, its efficacy remains controversial. This study hypothesized that a single acupressure on ST36 attenuates sympathetic nerve activity and mitigate skin vasoconstriction in response to cold stimulation. Twelve participants received either 5-min acupressure or sham pressure in the right ST36; subsequently, their left hand was immersed in cold water (10°C) for 10 min. Heart rate (HR), arterial pressure, skin temperature, and bl

Eight weeks of running, but not stationary cycling, training attenuates the vestibulosympathetic reflex in healthy adults.

A possible mechanism for exercise training-induced orthostatic intolerance is attenuation of the vestibulosympathetic reflex (VSR) due to enhanced stimulation of the otolith organs during running. We tested the hypothesis that the VSR is attenuated after 8 wk of running, but not stationary cycling. Arterial blood pressure, heart rate, calf blood flow, and muscle sympathetic nerve activity (MSNA) were recorded in 37 healthy, sedentary subjects (14 males and 23 females, 25 ± 6 years old) during baseline and head-down rotation (HDR) before and after 8 wk of endurance training. Subjects were assig

[18F]MFBG PET/CT imaging of myocardial sympathetic innervation in healthy controls and patients with parkinson's disease: dosimetry and pharmacokinetics.

PURPOSE: Cardiac noradrenergic denervation visualized by meta-[123I]iodobenzylguanidine ([123I]MIBG) imaging supports the diagnosis of Parkinson's disease (PD). Recently, meta-[18F] fluorobenzylguanidine ([18F]MFBG) PET demonstrated favorable imaging characteristics compared with [123I]MIBG scintigraphy for neuroendocrine tumors. We assessed [18F]MFBG dosimetry and myocardial pharmacokinetics in healthy controls and PD patients. METHODS: For dosimetry, three controls underwent sequential whole-body [18F]MFBG PET/CT up to 6 hours postinjection. Five controls and three PD patients underwent 120

Hypertension-induced heart failure disrupts cardiac sympathetic innervation.

About 26 million people worldwide live with heart failure (HF), and hypertension is the primary cause in 25% of these cases. Autonomic dysfunction and sympathetic hyperactivity accompany cardiovascular diseases, including HF. However, changes in cardiac sympathetic innervation in HF are not well understood. We hypothesized that cardiac sympathetic innervation is disrupted in hypertension-induced HF. Male and female C57BL6/J mice were infused with angiotensin II (ANG II) for 4 wk to generate hypertension leading to HF; controls were infused with saline. ANG II-treated mice displayed HF phenotyp
