Harnessing epigenetic inhibitors: A promising approach for overcoming challenges in pancreatic ductal adenocarcinoma treatment.

Pancreatic ductal adenocarcinoma (PDAC) is an aggressive and often fatal form of malignancy frequently diagnosed at an advanced stage, posing significant challenges for treatment. The complex and intricate microenvironment of PDAC, characterized by heterogenous tumoral cells harbouring diverse mutations and epigenetic alterations, immune cells (primarily in an immunosuppressive state), cancer-associated fibroblasts (CAFs), low vascularization and extracellular matrix components supporting tumor growth, creates a physical barrier that impedes drug delivery and anti-tumoral immune cell responses

Preclinical Development of Magnetic Nanoparticles for Hyperthermia Treatment of Pancreatic Cancer.

Pancreatic ductal adenocarcinoma (PDAC) is a very challenging disease with a very poor prognosis. It is characterized by a dense desmoplastic stroma that hampers drug penetration and limits the effectiveness of conventional chemotherapy (CT). As an alternative, the combination of CT with hyperthermia (HT) has been proposed as an innovative treatment modality for PDAC. In previous works, we reported on the development of iron oxide magnetic nanoparticles (MNPs) that, when exposed to time-varying magnetic fields, exhibit strong HT responses that inhibited the growth of pancreatic cancers. We rep

Ultrasound imaging and regulated mechanotransduction for characteristics, regeneration, and therapeutics of bone.

Ultrasound imaging has been widely used in clinical diagnoses, such as B-mode and M-mode ultrasound imaging for cardiovascular, abdomen, OB-Gyn, and other soft tissue and organs in clinical diagnoses. Ultrasound imaging has traditionally been limited in its application to bone because of the high acoustic impedance and density of trabecular and cortical bone structure and density alterations, high wave reflection, absorption, scattering, and low penetration, which result in significant reflection and attenuation of ultrasonic energy in such mineral tissues. Recent advancements in quantitative

Cbfβ: A key regulator in skeletal stem cell differentiation, bone development, and disease.

The skeletal system comprises closely related yet functionally distinct bone and cartilage tissues, regulated by a complex network of transcriptional factors and signaling molecules. Among these, core-binding factor subunit beta (Cbfβ) emerges as a critical co-transcriptional factor that stabilizes Runx proteins, playing indispensable roles in skeletal development and homeostasis. Emerging evidence from genetic mouse models has highlighted the essential role of Cbfβ in directing the lineage commitment of mesenchymal stem cells (MSCs) and their differentiation into osteoblasts and chondrocytes.

Biomass accumulation in chondrocyte metabolic modelling: Incorporating extracellular matrix proxies to predict tissue engineering outcomes.

Metabolic modeling in chondrocytes plays a pivotal role in advancing our understanding of cellular function. These techniques have been used to study degenerative joint diseases (e.g. osteoarthritis), mechanotransduction, and more recently to optimize strategies for cartilage tissue engineering. Incorporating tissue formation into metabolic flux analysis is inherently challenging due to the complexity of linking metabolic activity to extracellular matrix (ECM) accumulation. Many ECM macromolecules are synthesized using metabolites derived from central carbon metabolism, but direct modeling of

Beyond the bleed: complications after aneurysmal subarachnoid hemorrhage. Pathophysiology, clinical implications, and management strategies: a review.

Aneurysmal subarachnoid hemorrhage is a critical condition with high case-fatality and lasting impacts on survivors. Acute events that are the direct result of aneurysm rupture, such as acute ischemia, elevated intracranial pressure, cerebral edema, seizures, and hydrocephalus, lead to early brain injury. A delayed cascade of processes, including a prominent systemic inflammatory response, may lead to secondary brain injury and delayed cerebral ischemia, which often further impairs recovery. Systemic complications, including cardiac and pulmonary dysfunction, fever, and electrolyte imbalances,

New Findings in Developmental Studies of Laryngeal Mound With a Reference to Its Attached Structures in Japanese Quail (Coturnix coturnix Japonica).

The laryngeal mound (LM) formed the caudal part of the pharyngeal floor, which varied in position, shape, and length at different ages. This work aimed to study the morphogenesis of the LM in the embryonic and post hatching periods grossly, histologically, and by scanning electron microscopy using forty-eight Japanese quails. The LM primordia appeared on the 8th day of incubation as a raised elevation carried on a deep median symmetrical sulcus (glottis primordium). As a result of rapid differential LM parts growth, LM took different shapes with advanced ages, finally ending in a heart shape.
