The Needle-Free Frontier: Redefining the Limits of Transdermal Insulin Delivery.

Wei and colleagues reported a pH-responsive, charge-switchable polyzwitterionic polymer carrier that achieves non-invasive transdermal insulin delivery without external energy input. The approach leverages adaptive compatibility with the cutaneous microenvironment to shuttle functional macromolecules across intact skin and achieve definitive systemic therapeutic outcomes.

RBFOX1 Dysfunction Unlocks APOE4-Associated Microglial Genesis and Exacerbates Alzheimer's Pathology in Human Cerebral Organoids.

Alzheimer's disease (AD) pathogenesis is strongly influenced by APOE4, though how cooperative genetic factors modulate this relationship remains unclear. While genomic studies have tentatively linked RBFOX1 to AD susceptibility, its functional synergy with APOE4 has never been experimentally defined. We engineered APOE3 or APOE4 isogenic human cerebral organoids with CRISPR/Cas9-mediated RBFOX1 knockout. Remarkably, RBFOX1 depletion selectively triggered robust microglial generation exclusively in APOE4 organoids. Time-course gene expression revealed that this APOE4-specific effect correlated

Elimination of Fusobacterium nucleatum With Biomimetic Nanoparticles to Reverse Tumor Immunosuppression and Enhance Colorectal Cancer Therapy.

Tumor-associated microbes like Fusobacterium nucleatum (F. nucleatum) critically contribute to immunosuppression and hinder cancer therapy. To overcome this challenge, we developed a microbe-targeted immune reprogramming strategy utilizing tumor cell membrane-camouflaged Ag@MSN-PpIX nanoparticles (M-MAP). Under ultrasound irradiation, M-MAP specifically eliminates intratumoral F. nucleatum, thereby directly reversing F. nucleatum-mediated immunosuppression while preserving gut microbiota homeostasis. Crucially, the killed F. nucleatum acts as an immunostimulant, promoting robust activation of

Spherical Nucleic Acid Stabilized Cage Type Three-Dimensional Electrochemiluminescence Probe for Sensitive and Amplification-Free Diagnosis of Infectious Diseases.

Accurate nucleic acid-based pathogen diagnosis is critical for clinical treatment and epidemic control. However, present gold-standard testing methods based on nucleic acid amplification are limited by their time and labor-intensive nature. Hence, we reported an amplification-free assay that facilitates rapid, sensitive, and point-of-care detection of infectious diseases. Herein, spherical nucleic acid (SNA)-stabilized cage-type three-dimensional electrochemiluminescence (ECL) reporter probe, constructed utilizing Ru(dcbpy)3Cl2-doped NH2-MIL-101 (Fe) metal-organic framework, was combined with

Synthesis of Tris(Dimethylamino)Phosphine-Based InP Quantum Dots and Their Application in Light-Emitting Diodes: Progress and Perspectives.

Indium phosphide (InP)-based quantum dots (QDs) have emerged as promising cadmium-free alternatives for next-generation optoelectronic applications, particularly in quantum dot light-emitting diodes (QLEDs). Tris(dimethylamino)phosphine ((DMA)3P) has gained attention as a low-toxicity alternative to conventional precursors like tris(trimethylsilyl)phosphine ((TMS)3P) or toxic phosphine gas (PH3) in the synthesis of InP QDs. However, InP core/shell QDs synthesized using (DMA)3P and their corresponding QLEDs currently exhibit inferior optical and electronic performance compared to their (TMS)3P-

Customizing 3D Hierarchically Macroporous Covalent Organic Framework Superstructures for Improved Biosensing Interface in Highly-Sensitive Electrochemical Biosensors.

The construction of an optimized biosensing interface is critically important for electrochemical biosensing. Herein, we achieve the controllable construction of macroporous covalent organic framework (COF) nanostructures through a template-assisted assembly growth strategy, resulting in well-organized morphologies that are well-suited as advanced electrode materials. These macroporous structures with unique pore microenvironments support efficient immobilization of biorecognition elements via structure-confined supramolecular interactions. Notably, the 3D hierarchically macroporous COF supers

Sunlight-Driven Fixation of CO2 to Cyclic Carbonates Using Carbon Dots as a Photothermal Catalyst.

Fixation of CO2 through photocatalytic cycloaddition with epoxides to synthesize cyclic carbonates is an important but challenging process. In this work, carbon dots (CDs) synthesized from gallic acid and polyethylenimine are used for the efficient catalytic cycloaddition of CO2 with epoxides in the absence of any solvent, additives, and halides, and importantly upon irradiation by natural sunlight. Specifically, carbon dots generated thermal energy and electrons upon solar irradiation, which together with their surface N-sites activated the inert CO2. Meanwhile, epoxides were activated by the

Enhancing Optogenetics-Based Cancer Therapy Via Nanotechnology.

Optogenetics represents a promising frontier in precision cancer therapy by enabling spatiotemporal control over cellular behavior. However, its clinical application is limited by inefficient delivery of optogenetic components and poor tissue penetration of visible light. Recent advances in nanotechnology offer solutions to these challenges. Nanoscale drug delivery systems enhance the targeted delivery of optogenetic tools, while light-conversion nanomaterials enable deep-tissue activation. Besides, the integration of nanotechnology with optogenetics further facilitates the development of engi

Arterial Perivascular Space-Mediated Solute Transport in the Mouse Brain.

The perivascular space (PVS) around cerebral arteries supports nutrient delivery and waste clearance, yet its transport mechanisms remain poorly defined. Here, we establish arterial PVS as a major solute transport route in the brain, exhibiting marked region-specific functionality. Using in vivo imaging, we show that solute movement within arterial PVS is not driven by cerebrospinal fluid (CSF) bulk flow, but follows bidirectional, arterial-pulsation-dependent dynamics. Parenchymal solutes diffuse into the vascular PVS network, where arterial pulsations propel them along perivascular pathways

Hydrogel Adhesives for Gastrointestinal Perforation: Design Principles and Applications.

Gastrointestinal (GI) perforation, as an acute digestive condition, is difficult to heal spontaneously and requires prompt surgical intervention or bioactive adhesives to promote wound closure. Among various types of tissue adhesives, hydrogel adhesives have attracted tremendous attention and have been used in the clinic due to their atraumatic nature, good biocompatibility, and tunable physicochemical properties. Despite their promise, the bioadhesive applications with engineered hydrogels still face challenges in the wet and acidic gastric environment. This review outlines the mainstream des

M2 Macrophage-Mimetic and ROS-Sensitive Hybrid Nanoplatform for Targeted Alleviation of Rheumatoid Arthritis.

Rheumatoid arthritis (RA) constitutes the most prevalent inflammatory arthropathy globally, which is typically marked by synovitis and deteriorating joint damage. Regrettably, RA remains incurable owing to insufficient therapeutic response along with severe side reactions of available treatments. To overcome these formidable obstacles, an M2 macrophage membrane-cloaked, ROS-sensitive lipid-gated, and herbal anti-arthritic agent celastrol (Cel)-loaded hybrid silica nanosystem (Cel@M2-L-MSN) was established to reshape the intricate RA microenvironment. Benefiting from cell membrane camouflaging,

Electrochemical Valence-Regulated Biomimetic Nanozymes for Breast Cancer Metabolism Inhibition and Potentiated Catalytic Immunotherapy.

Manganese oxide (MnOx) nanozymes have garnered significant attention for their ability to respond to the tumor microenvironment (TME) and stimulator of interferon genes (STING) pathway activation. However, they often suffer from a weak direct tumor-killing capacity due to low enzymatic activity and from attenuated anti-tumor immunity resulting from STING activation-induced excessive programmed death-ligand 1 (PD-L1) expression. In this study, we fabricated MnOx nanozymes with precisely controllable valence states via electrochemical valence regulation to modulate their oxidase-like activity. N

Engineered Nature-Based Sonosensitizer for Broad-Spectrum Infectious Tissue Repair via Sono-Ionic Therapy.

Repairing infectious tissues, particularly bone and wound infections, poses significant challenges to clinicians due to the lack of effective treatment modalities. Sonodynamic therapy, which utilizes sonosensitizers to generate reactive oxygen species (ROS) under ultrasound (US) energy for antibacterial purposes, still lacks suitable sonosensitizers with good biocompatibility that can simultaneously promote the repair of various infected tissues. In this study, we constructed a novel sonosensitizer composed of hemoglobin derived from nature and zeolitic imidazolate framework (ZIF)-8, which exh

MiR-29a/b Suppresses CD8+ T Cell Effector Function and Intestinal Inflammation.

The role of CD8+ T cells in the pathogenesis of ulcerative colitis (UC) remains unclear. Similarly, the posttranscriptional regulation of the highly heterogenic CD8+ T cell populations and their effector function in IBD also remains poorly understood. Here, we find that miR-29a and -29b (miR-29a/b) regulate T cell fate, and their expression is higher near damaged colon tissue in patients with IBD compared to controls. In mice, we find that miR-29a/b suppresses the differentiation of CD8+ T cells and the secretion of pro-inflammatory and chemotactic factors during severe colitis by inhibiting t

Modulating Lysine Crotonylation in Ulcerative Colitis Maintains Mitochondrial Homeostasis: Modulating Crotonylation in Ulcerative Colitis.

Ulcerative colitis (UC) is a chronic and persistent clinical condition that is challenging to cure. Lysine crotonylation (KCr), a recently discovered post-translational modification (PTM), alters protein structure, stability, localization and activity in a variety of processes including cell differentiation and organism development. This study was designed to elucidate the pathophysiological relevance of KCr in UC and uncover potential underlying mechanisms involved. PTM proteomics was employed to track dynamic alterations in KCr sites and protein level in the colon tissue of dextran sulfate s

Spatial Isolation of Single Copper(I) Sites for Cascade Enzyme-Like Catalysis and Simultaneous Ferroptosis/Cuproptosis Boosted Immunotherapy.

Nanozyme-based immunogenic cell death (ICD) inducers that effectively induce a strong immune response via enzyme-like process have attracted great attention, but how to ensure controllable active sites and maximize site utilization remains a problem. Here, we report a structurally well-defined and highly functional single-site copper(I) nanomodulators termed CuNTD, constructed by precisely anchoring atomically dispersed self-assembly S-Cu(I)-S sites onto a two-dimensional Ti3C2 surface. Leveraging Cu+ with a higher catalytic efficiency than Cu2+, CuNTD generates reactive oxygen species (ROS) s

Global Stress Responses Identify the Functionally Divergent Regulators Required for Candida auris Commensalism and Pathogenicity.

Given its global distribution and high transmissibility in the environment, Candida auris poses a serious threat to global public health. However, the underlying mechanisms of its adaptive strategies remain poorly understood. Here we delineate the pan-genome structures of 1,306 representative C. auris isolates collected from 28 countries. In addition to the clade-related genetic diversity and highly variable pan-genomes, we identify the key regulatory modules and genes specific to C. auris in response to 32 different host microenvironment-mimicking stresses. Through comparative analysis with e
