Targeting neutrophil extracellular traps in bacterial infections: mechanistic insights and translational perspectives.

Neutrophil Extracellular Traps (NETs) are web-like structures composed of DNA fibers, histones, and granular proteins released by the neutrophils activated upon various stimuli. Recognized as a host immune defense mechanism, NETs also play a significant role in several diseases, especially in bacterial infections. Research indicates that excessive NET formation contributes to various pathological outcomes, including multiple organ dysfunction. Once activated, NETs generally remain in a hyperinflammatory phenotype. In this review, we delineate how bacterial pathogens both induce excessive NET f

Computational methods, clinical evidence, and translational readiness for artificial intelligence for HER2 assessment using silver in situ hybridization (SISH).

Accurate determination of human epidermal growth factor receptor 2 (HER2) gene amplification is a cornerstone of precision oncology in breast cancer and an expanding range of solid tumors. While immunohistochemistry (IHC) remains the first-line screening modality, in situ hybridization (ISH) techniques are essential for resolving equivocal cases and providing quantitative assessment of HER2 gene status. Among ISH methods, silver-enhanced in situ hybridization (SISH) has emerged as a clinically relevant bright-field alternative to fluorescence in situ hybridization (FISH), offering permanent st

FBXW7 Gene Mutation and Expression in Colorectal Cancer (CRC): A Systematic Review from Molecular Mechanisms to Clinical Translation.

In the context of precision oncology, understanding the molecular drivers of colorectal cancer (CRC) is critical for improving prognosis and guiding targeted therapy. FBXW7 is a tumor suppressor that plays a pivotal role in CRC by regulating the degradation of key oncogenic proteins, influencing tumor initiation, growth, therapeutic response, and metastatic behavior. Mutations in FBXW7 occur in 6-10% of CRC. Despite its biological relevance, the prognostic and predictive role of FBXW7 in CRC remains unclear, with inconsistent findings across studies. This systematic review collects and analyze

Amniotic Fluid-Derived Stem Cells: An Overlooked Source of Stem Cells for Translational Research.

Amniotic fluid-derived stem cells (AFSCs) represent a promising yet underutilized resource for research and clinical applications. While AFSCs share phenotypic and functional characteristics with stem cells derived from somatic tissues such as bone marrow, adipose tissue, placenta, and umbilical cord, their unique developmental origin grants them several superior qualities. These include enhanced multipotency, tissue-specific genotypic profiles, and the ability to form single-cell colonies. Such features position AFSCs as highly valuable for translational research and tissue engineering. This

Reversal of a synthetic opioid overdose: Insights from a validated translational model.

Synthetic opioids are linked to >90 % of opioid overdose deaths in the United States. A FDA's validated translational model of synthetic opioid overdose, expanded to include intranasal reversal agents, was used to compare the effectiveness of FDA-approved intranasal naloxone (4 mg) and intranasal nalmefene (3 mg nalmefene hydrochloride, 2.7 mg base). In the absence of intervention, simulations using various intravenous doses of fentanyl and carfentanil predicted an incidence of cardiac arrest ranging from 18 % to 90 % in chronic opioid users and from 40 % to 96 % in opioid naïve subjects. Comparative Evaluation of Bleomycin- and Collagen-V-Induced Models of Systemic Sclerosis: Insights into Fibrosis and Autoimmunity for Translational Research.

Systemic sclerosis (SSc) is a complex autoimmune disease characterized by fibrosis, immune dysregulation, and vascular dysfunction, yet its pathogenesis remains incompletely understood. This study compares two widely used animal models of SSc-the bleomycin-induced fibrosis model and the collagen-V-induced autoimmune model-to evaluate their ability to replicate key disease features. In the bleomycin model, consistent cardiac fibrosis was observed across treatment groups despite variability in fibrosis in the skin and lungs, suggesting organ-specific differences in susceptibility. The collagen-V

Beneficial effects of recombinant CER-001 high-density lipoprotein infusion in sepsis: results from a bench to bedside translational research project.

BACKGROUND: Sepsis is characterized by a dysregulated immune response and metabolic alterations, including decreased high-density lipoprotein cholesterol (HDL-C) levels. HDL exhibits beneficial properties, such as lipopolysaccharides (LPS) scavenging, exerting anti-inflammatory effects and providing endothelial protection. We investigated the effects of CER-001, an engineered HDL-mimetic, in a swine model of LPS-induced acute kidney injury (AKI) and a Phase 2a clinical trial, aiming to better understand its molecular basis in systemic inflammation and renal function. METHODS: We carried out a

A patient advocating for transparent science in rare disease research.

300 million people live with at least one of 6,000 rare diseases worldwide. However, rare disease research is not always reviewed with scrutiny, making it susceptible to what the author refers to as nontransparent science. Nontransparent science can obscure animal model flaws, misguide medicine regulators and drug developers, delay or frustrate orphan drug development, or waste limited resources for rare disease research. Flawed animal models not only lack pharmacologic relevance, but also give rise to issue of clinical translatability. Sadly, these consequences and risks are grossly overlooke

Translational Research in Vitiligo.

Vitiligo is a disease of the skin characterized by the appearance of white spots. Significant progress has been made in understanding vitiligo pathogenesis over the past 30 years, but only through perseverance, collaboration, and open-minded discussion. Early hypotheses considered roles for innervation, microvascular anomalies, oxidative stress, defects in melanocyte adhesion, autoimmunity, somatic mosaicism, and genetics. Because theories about pathogenesis drive experimental design, focus, and even therapeutic approach, it is important to consider their impact on our current understanding ab
