Neuromuscular changes in a rat model of colitis.

Intracolonic administration of Trichinella spiralis larvae in rats causes colitis with features similar to ulcerative colitis, notably with inflammation predominantly limited to the colonic mucosa. Our aim was to characterize the functional and neurochemical changes occurring within the myenteric (MP) and submucosal plexuses (SMP) during T. spiralis-induced colitis. Infected rats had decreased body weight, altered stool consistency and elevated myeloperoxidase activity, 6 and 14 days post-infection (PI). Responses to acetylcholine and KCl in circular muscle strips were reduced in infected tiss

The usefulness of the resting electrocardiogram for characterizing acute Chagas' heart disease in the rat.

The present experiment was undertaken to characterize the resting electrocardiogram of rats in the acute stage of experimental T. cruzi infection. One-hundred-and-eighty-seven newly-weaned male albino rats were infected intraperitoneally with 1000 parasites/g body weight. Thirty-two similar but non-infected rats served as controls. Groups of eight randomly chosen rats were killed on day 8, and on days 15, 18, 22, 25 and 29 after infection. Groups of 8 control rats were killed on days 8, 15 and 22. Electrocardiographic changes were observed in 25 of 48 (52%) infected rats. P wave abnormalities

Postweaning diet affects offspring metabolic outcomes more than maternal diet in male and female mice.

The pervasiveness of obesity necessitates expanded pursuits to understand mechanistic drivers of the condition. Maternal diet during conception and gestation can program offspring towards excessive weight gain. Furthermore, the association between obesity and hypertension suggests that maternal programming may impact offspring vascular reactivity. We investigated whether a maternal high fat+sucrose diet (HFD) prior and during pregnancy and lactation affected offspring body composition, metabolic parameters, and vascular reactivity. Using C57BLJ/6 mice, male and female offspring from control (l

The relationship between age related changes in strength and fitness with body size, shape and composition.

Handgrip strength (HGS), cardiorespiratory fitness (CRF) and body size, shape, and composition are all related to cardiometabolic health and are associated in cross-sectional settings. Their longitudinal relationship is less clear. We used observational data from the Study of Health in Pomerania at baseline (SHIP-TREND-0; 2008-2012) and follow-up (SHIP-TREND-1; 2016-2019) with 1,214 men and 1,293 women. HGS was measured with a hand dynamometer. CRF was assessed using cardiopulmonary exercise testing. Linear regression models were adjusted appropriately. Several sensitivity analyses were perfor

Role of cystathionine-γ-lyase in hypoxia-induced changes in TASK activity, intracellular [Ca2+] and ventilation in mice.

Cystathionine-γ-lyase (CSE) is a multifunctional enzyme, and hydrogen sulfide (H2S) is one of its products. CSE and H2S have recently been proposed to be critical signaling molecules in hypoxia-induced excitation of carotid body (CB) glomus cells and the chemosensory response. Because the role of H2S in arterial chemoreception is still debated, we further examined the role of CSE by studying the effects of hypoxia on TASK K+ channel activity, cell depolarization, [Ca2+]i and ventilation using CSE+/+ and CSE-/- mice. As predicted, hypoxia reduced TASK activity and depolarized glomus cells isola

Peripubertal Caffeine Exposure Impairs Longitudinal Bone Growth in Immature Male Rats in a Dose- and Time-Dependent Manner.

This study investigated the dose- and time-dependent effects of caffeine consumption throughout puberty in peripubertal rats. A total of 85 male SD rats were randomly divided into four groups: control and caffeine-fed groups with 20, 60, or 120 mg/kg/day through oral gavage for 10, 20, 30, or 40 days. Caffeine decreased body weight gain and food consumption in a dose- and time-dependent manner, accompanied by a reduction in muscle and body fat. In addition, it caused a shortening and lightening of leg bones and spinal column. The total height of the growth plate decreased sharply at 40 days in

Plant-Based Functional Formulae Mitigate Western Diet-Induced MASLD by Modulating Hepatic Oxidative Stress, Inflammation as Well as Nrf-2/HO-1 and NF-κB Gene Expression.

Psyllium has long been used to improve satiety and control weight. Leek and dill leaves are abundant in bioactive substances that have been confirmed to have anti-inflammatory and antioxidant properties. Our goal was to assess the hepatoprotective effects of two formulae (dill and psyllium or leek and psyllium) against oxidative stress and inflammatory markers in a metabolic dysfunction-associated steatotic liver disease (MASLD) rat model. Twenty four rats were used in this study. Rats were given a high-fat, high-sucrose diet (HF/HS), a Western diet, to create a MASLD model. The two formulae s

Assessment of change in drug-induced parkinsonism in daily-life gait using vision-based machine learning.

Drug-induced parkinsonism is a potentially reversible movement disorder that may be subtle and difficult to monitor in residential dementia care. Vision-based machine learning analysis of gait may provide unobtrusive longitudinal monitoring around medication changes. We conducted a two-center cohort study of 61 residents with dementia receiving psychotropic medication. Hallway gait sequences were recorded using ceiling-mounted cameras and converted to three-dimensional body pose. A TabPFN classifier estimated UPDRS-III item 29 gait severity for each walk. We evaluated associations between acut

Direct comparison of the effects of functionally equivalent dosing regimens of methamphetamine, methcathinone, and α-pyrrolidinopropiophenone on behavioral and neurochemical markers of stimulant-induced neurotoxicity.

Given that previous studies have provided mixed results on cathinone neurotoxicity, we compared the effects of 'functionally equivalent' dosing regimens of methamphetamine (METH), methcathinone, and α-pyrrolidinopropiophenone (α-PPP) on behavioral and neurochemical markers associated with dopaminergic alterations and cognitive function. Male Swiss Webster mice received METH (5 mg/kg), methcathinone (80 mg/kg), or α-PPP (80 mg/kg), using a 'binge'-like dosing regimen (q2h × 4, intraperitoneal). Body weight and temperature were recorded during dosing. Behavior was assessed 2-4 days later, follow

Parthenolide enhances imatinib activity in gastrointestinal stromal tumor models through stress-associated apoptosis.

Imatinib remains a mainstay of treatment for gastrointestinal stromal tumors (GISTs), yet responses may be incomplete or short-lived. We examined whether parthenolide (PTL) enhances imatinib activity in GIST models and characterized the cellular changes associated with combined treatment. Public GIST transcriptomic datasets were analyzed as hypothesis-generating resources. GIST-T1 and GIST-882 cells were treated with PTL, imatinib, or their combination. Cell growth, migration, colony formation, apoptosis, reactive oxygen species, mitochondrial function and ultrastructure, ER-stress-associated

Sub-acute intraperitoneal quercetin administration causes dose-dependent organ histopathology and oxidative stress in healthy rats: dissociation between serum biomarkers and tissue-level structural injury.

Quercetin is widely used in dietary supplements, yet its sub-acute organ safety profile in healthy organisms remains incompletely characterized. This study evaluated dose-dependent effects of intraperitoneal quercetin administration in healthy female Wistar rats (n = 6/group) receiving normal saline (control) or quercetin (5, 10, 20, or 30 mg/kg) every 72 h for 21 days. Endpoints included body and organ weights, serum biochemistry, hematology, systemic and tissue oxidative stress markers, and semi-quantitative multi-organ histopathology. Quercetin at ≥ 20 mg/kg significantly reduced percent we

Reduced Oligodendrocyte Density and Axonal Caliber Associated With Mitochondrial Alterations in the White Matter of Chronically-Starved Mice.

OBJECTIVE: Anorexia nervosa (AN) is a severe eating disorder associated with extreme weight loss, hyperactivity, and amenorrhea. Neuroimaging studies revealed brain atrophy and disruption of white matter integrity in the corpus callosum (CC) of patients with AN. However, the underlying pathophysiological mechanisms remain unclear. Emerging evidence indicates that starvation induces changes in mitochondrial metabolism and dynamics. We hypothesize that disturbances in white matter integrity arise from modifications in oligodendrocytes, associated with changes in the morphology of myelinated fibe

Associations of baseline and changes in the triglyceride glucose-weight adjusted waist index and cardiovascular disease risk: evidence from middle-aged and older individuals.

BACKGROUND: Existing researches have predominantly focused on the implications of dynamic alterations in the triglyceride-glucose (TyG) index and traditional obesity measures for cardiovascular disease (CVD) risk. However, the application of the weight-adjusted waist index (WWI), which incorporates the dynamically changing body composition factors of weight and waist circumference, alongside the TyG index for predicting CVD risk, remains unexplored. This study explores the relationships between baseline TyG-WWI index and its dynamic changes with CVD risk. METHODS: Subjects were drawn from the

Use of Photoacoustic Imaging to Study the Effects of Anemia on Placental Oxygen Saturation in Normoxic and Hypoxic Conditions.

We aimed to evaluate fetal and placental oxygen saturation (sO2) in anemic and non-anemic pregnant rats throughout gestation using photoacoustic imaging (PAI). Female Sprague-Dawley rats were fed an iron-restricted or iron-replete diet before and during pregnancy. On gestational days 13, 18, and 21, PAI was coupled with high resolution ultrasound to measure oxygenation of the fetus, whole placenta, mesometrial triangle, as well as the maternal and fetal faces of the placenta. PAI was performed in 3D, which allowed sO2 to be measured within an entire region, as well as in 2D, which enabled sO2

Carbonic anhydrase 3 increases during liver adipogenesis even in pre-obesity, and its inhibitors reduce liver adipose accumulation.

The abnormal lipid metabolism in the liver that occurs after high caloric intake is the main cause of nonalcoholic fatty liver disease (NAFLD). Differences between samples from healthy livers and livers from individuals with NAFLD indicate that changes in liver function occur during disease progression. Here, we examined changes in protein expression in a fatty liver model in the early stages of obesity to identify potential alterations in function. The proteins expressed in the liver tissue of pre-obese rats were separated via SDS/PAGE and stained with Coomassie brilliant blue-G250. Peptide m

Impact of Laparoscopic Sleeve Gastrectomy on Cardiovascular Pharmacotherapy in Left Ventricular Assist Device Patients.

Left ventricular assist device (LVAD) implantation is increasingly utilized in patients with advanced heart failure and morbid obesity. Laparoscopic sleeve gastrectomy (LSG) can facilitate weight loss in this population and can ultimately change the pharmacokinetics of heart failure therapeutics. In this study, we aimed to explore the changes in cardiovascular pharmacotherapy post LSG intervention. We conducted a retrospective observational cohort study of morbidly obese LVAD patients between 2013 and 2019 at the University of Florida with available pharmacotherapeutic data at 1 and 6 months.
