The pyrazolo[3,4-d]pyrimidine derivative CLM24 has an antineoplastic effect in aggressive dedifferentiated thyroid cancer in vitro.

PURPOSE: Compounds with a pyrazolo[3,4-d]pyrimidine nucleus (CLM24, CLM3, and CLM29) with inhibitory activities towards RET protein kinases, EGFR, VEGFR and as antiangiogenic agents had been evaluated in preceding researches in experimental models of thyroid cancer (TC) in vitro and in vivo. Here, we designed a study to evaluate the antineoplastic activity of CLM24 in vitro in primary dedifferentiated thyroid carcinoma (DeDTC) cells and in the AF cell line (a continuous ATC cell line previously obtained from a patient-derived cell culture and immortalized in vitro). METHODS: We investigated in

Induction of apoptosis by oridonin in nonfunctioning pituitary adenoma cells.

Nonfunctioning pituitary adenoma (NFPA) is one of the major subtypes of pituitary adenomas (PA) and its primary treatment is surgical resection. However, normal surgery fails to remove lesions completely and there remains in lack of frontline treatment, so the development of new drugs for NFPA is no doubt urgent. Oridonin (ORI) has been reported to have antitumor effects on a variety of tumors, but whether it could exhibit the same effect on NFPA requires to be further investigated. The effects of ORI on pituitary-derived folliculostellate cell line (PDFS) cell viability, colony formation, pro

[High tumorigenicity of human acute monocytic leukemic cell Line SHI-1 in nude mice and its mechanism].

This study was purposed to explore the tumorigenicity of a novel human monocytic leukemic cell line SHI-1 in nude mice and its mechinism. The tumorigenicity in mice was evaluated in sixteen nude mice subcutaneously injected with the SHI-1 cell line. The tumor specimen was studied by the conventional pathologic examination. The mononuclear cells (MNC) of the tumor was assayed by RHG banding, the transcription of MLL-AF6 fusion gene and the VEGF gene was detected by RT-PCR. Gelatin zymography method was used to study the expression of MMP-9 and MMP-2 in the supernatant of the SHI-1 cell line. Ma

Breaking the Mechanical Compass: Disrupting Durotaxis to Halt Fibrosis and Metastasis.

Durotaxis is the directed migration of cells along tissue stiffness gradients and is emerging as a fundamental mechanobiological process orchestrating progression in diseases such as fibrosis and cancer. Despite compelling in vitro evidence demonstrating durotactic behavior across multiple cell types, translating these findings to in vivo contexts has remained challenging because of the inherent complexity of isolating biomechanical cues from the myriad biochemical and architectural features that collectively define tissue properties. In their recent Nature Cell Biology study, Al-Hilal and col

CACUL1 promotes hepatocellular carcinoma progression through enhanced tumor cell proliferation and macrophage-mediated immune suppression.

BACKGROUND: Although cullin-family E3 ubiquitin ligases regulate diverse oncogenic pathways, the role of CACUL1 in hepatocellular carcinoma (HCC) remains poorly characterized. METHODS: We performed pan-cancer expression and survival analysis using TCGA and GEO databases, validated findings through immunohistochemistry, and conducted functional studies in HCC cell lines using CRISPR-mediated CACUL1 knockout. Transcriptome sequencing, functional assays (CCK-8, wound-healing, transwell invasion), and single-cell RNA-sequencing were performed to elucidate mechanisms. RESULTS: Pan-cancer analysis

PRMT5-mediated arginine methylation of HSP90AA1 drives esophageal squamous cell carcinoma progression.

Dysregulated HSP90AA1 chaperone activity is a hallmark of multiple cancers; however, its post-translational regulation in esophageal squamous cell carcinoma (ESCC) remains poorly defined. Here, we defined PRMT5-dependent symmetric dimethylation of HSP90AA1 at arginine 182 (R182) as a critical molecular switch driving ESCC progression. HSP90AA1 physically interacted with PRMT5, and genetic or pharmacological inhibition of PRMT5 markedly reduced HSP90AA1 R182 methylation, accompanied by suppression of epithelial-mesenchymal transition (EMT) programs. HSP90AA1 depletion significantly impaired ESC

KLF5-mediated transcriptional activation of miR-203a inhibits EMT and increases cisplatin sensitivity by targeting SNAI2 in tongue cancer.

Distant metastasis, cancer recurrence, and resistance to chemotherapy together are responsible for poor prognosis and create challenges for effective treatment of tongue squamous cell carcinoma (TSCC) or tongue cancer. Consequently, identifying key molecular regulators that simultaneously modulate tumor progression and drug resistance is crucial to improving therapeutic outcomes. In this study, we investigated hub transcription factors (TFs) that regulate multiple tumorigenic and drug-resistance-associated processes and elucidated their downstream regulatory molecular mechanisms by integrating

Unraveling the complexity of glioblastoma microenvironment: A comparative study of 3D and 2D cultures in response to combination drug therapy.

Glioblastoma multiforme (GBM) represents one of the most prevalent and treatment-resistant forms of brain cancer. This investigation explored the therapeutic potential of a combined Erlotinib and Imatinib regimen through experimental studies conducted in both two-dimensional and three-dimensional cell culture systems. Cellular viability and apoptotic responses were evaluated 24- and 48-hours post-treatment utilizing MTT assays and flow cytometry, while gene expression analysis of Bcl-2 and VEGF was performed using qRT-PCR. Furthermore, the scratch assay was employed to assess the impact of dru

Targeting PADI2 as a potential therapeutic strategy against metastasis in oral cancer via suppressing EMT-mediated migration and invasion and CCL3/5-induced angiogenesis.

Oral squamous cell carcinoma (OSCC) is a prevalent and aggressive malignancy, with metastasis being the leading cause of death in patients. Unfortunately, therapeutic options for metastatic OSCC remain limited. Peptidylarginine deiminases (PADI) are implicated in various tumorigenesis and metastasis processes across multiple cancers. However, the role of PADI2, a type of PADI, in OSCC is not well understood. This study aimed to explore the impact of PADI2 on epithelial-mesenchymal transition (EMT), angiogenesis, and OSCC metastasis. The effect of PADI2 on EMT was evaluated using cell lines by

Ocoxin Increases the Antitumor Effect of BRAF Inhibition and Reduces Cancer Associated Fibroblast-Mediated Chemoresistance and Protumoral Activity in Metastatic Melanoma.

Whereas the prevalence of several cancer types is decreasing, skin malignancies are growing more common every year. Malignant melanoma is the most aggressive form of skin cancer with high metastatic capacity. In most cases, malignant melanoma shows acquired therapy resistance. We evaluated the ability of Ocoxin, a natural compound-based antioxidant and anti-inflammatory nutritional complement, to exert an antitumor effect in melanoma. To do so, the cytotoxicity of Ocoxin in a panel of BRAF-mutated murine and human melanoma cell lines was tested alone and in combination with BRAF inhibitor Vemu

Role of dendritic cell-mediated immune response in oral homeostasis: A new mechanism of osteonecrosis of the jaw.

Dendritic cells are an important link between innate and adaptive immune response. The role of dendritic cells in bone homeostasis, however, is not understood. Osteoporosis medications that inhibit osteoclasts have been associated with osteonecrosis, a condition limited to the jawbone, thus called medication-related osteonecrosis of the jaw. We propose that disruption of the local immune response renders the oral microenvironment conducive to osteonecrosis. We tested whether zoledronate (Zol) treatment impaired dendritic cell (DC) functions and increased bacterial load in alveolar bone in vivo

Hydrogen sulfide modulates epithelial-mesenchymal transition and angiogenesis in non-small cell lung cancer via HIF-1α activation.

Hydrogen sulfide (H2S) has been frequently implicated in tumor progression. However, the exact regulation mechanism of H2S in human non-small cell lung cancer (NSCLC) has not been fully elucidated. Here, analysis of NSCLC biopsies and adjacent non-tumor tissues revealed selectively high levels of endogenous H2S-producing enzymes, cystathionine-beta-synthase (CBS), cystathionine-gamma-lyase (CSE) and 3-mercaptopyruvate sulfurtransferase (MPST). Similarly, quantitative real-time PCR (qRT-PCR) and western blot results showed that NSCLC cell lines (A549, 95D) expressed higher levels of CBS, CSE an

SIRT1 inhibition by melatonin exerts antitumor activity in human osteosarcoma cells.

Melatonin, the main secretory product of the pineal gland, has potent antitumor activity against various types of cancer. However, the molecular mechanisms underlying the effects of melatonin remain largely unknown. SIRT1, a conserved nicotinamide adenine dinucleotide (NAD(+))-dependent deacetylase, has been implicated in modulating transcriptional silencing and cell survival and plays a key role in carcinogenesis through the deacetylation of important regulatory proteins. In this study, we assessed the antitumor activity of melatonin against human osteosarcoma cells (9607 cell line) and explo

Abelson interactor 1 (ABI1) and its interaction with Wiskott-Aldrich syndrome protein (wasp) are critical for proper eye formation in Xenopus embryos.

Abl interactor 1 (Abi1) is a scaffold protein that plays a central role in the regulation of actin cytoskeleton dynamics as a constituent of several key protein complexes, and homozygous loss of this protein leads to embryonic lethality in mice. Because this scaffold protein has been shown in cultured cells to be a critical component of pathways controlling cell migration and actin regulation at cell-cell contacts, we were interested to investigate the in vivo role of Abi1 in morphogenesis during the development of Xenopus embryos. Using morpholino-mediated translation inhibition, we demonstra

Decreasing mitochondrial fission ameliorates HIF-1α-dependent pathological retinal angiogenesis.

Angiogenesis plays a critical role in many pathological processes, including irreversible blindness in eye diseases such as retinopathy of prematurity. Endothelial mitochondria are dynamic organelles that undergo constant fusion and fission and are critical signalling hubs that modulate angiogenesis by coordinating reactive oxygen species (ROS) production and calcium signalling and metabolism. In this study, we investigated the role of mitochondrial dynamics in pathological retinal angiogenesis. We showed that treatment with vascular endothelial growth factor (VEGF; 20 ng/ml) induced mitochond

Myeloperoxidase induces monocyte migration and activation after acute myocardial infarction.

INTRODUCTION: Myocardial infarction (MI) is a significant contributor to morbidity and mortality worldwide. Many individuals who survive the acute event continue to experience heart failure (HF), with inflammatory and healing processes post-MI playing a pivotal role. Polymorphonuclear neutrophils (PMN) and monocytes infiltrate the infarcted area, where PMN release high amounts of the heme enzyme myeloperoxidase (MPO). MPO has numerous inflammatory properties and MPO plasma levels are correlated with prognosis and severity of MI. While studies have focused on MPO inhibition and controlling PMN

Atg5 deficiency in macrophages protects against kidney fibrosis via the CCR6-CCL20 axis.

BACKGROUND: Autophagy is a lysosome-dependent degradation pathway that regulates macrophage activation, differentiation, and polarization. Autophagy related 5 (Atg5) is a key protein involved in phagocytic membrane elongation in autophagic vesicles that forms a complex with Atg12 and Atg16L1. Alterations in Atg5 are related to both acute and chronic kidney diseases in experimental models. However, the role of macrophage-expressed Atg5 in acute kidney injury remains unclear. METHODS: Using a myeloid cell-specific Atg5 knockout (MΦ atg5-/-) mouse, we established renal ischemia/reperfusion and u

Cytomegalovirus Infection Triggers the Secretion of the PPARγ Agonists 15-Hydroxyeicosatetraenoic Acid (15-HETE) and 13-Hydroxyoctadecadienoic Acid (13-HODE) in Human Cytotrophoblasts and Placental Cultures.

INTRODUCTION: Congenital infection by human cytomegalovirus (HCMV) is a leading cause of congenital abnormalities of the central nervous system. Placenta infection by HCMV allows for viral spread to fetus and may result in intrauterine growth restriction, preeclampsia-like symptoms, or miscarriages. We previously reported that HCMV activates peroxisome proliferator-activated receptor gamma (PPARγ) for its own replication in cytotrophoblasts. Here, we investigated the molecular bases of PPARγ activation in infected cytotrophoblasts. RESULTS: We show that onboarded cPLA2 carried by HCMV particl

Dyad of CD40/CD40 ligand fosters neuroinflammation at the blood-brain barrier and is regulated via JNK signaling: implications for HIV-1 encephalitis.

Human immunodeficiency virus 1 (HIV-1) infection may result in activation of peripheral monocytes followed by their infiltration into the CNS, where the release of proinflammatory mediators causes neurologic disease. Previously, we detected high levels of soluble CD40 ligand (CD40L) in CSF and plasma of HIV-infected patients with cognitive impairment. We now show that CD40, a receptor for CD40L, is highly expressed in brain endothelial cells of patients affected by HIV-1 encephalitis (HIVE), suggesting an important role for the CD40/CD40L dyad in regulating blood-brain barrier (BBB) functions.
