When Pathways Converge: Iron, Lipid Peroxidation, and α-Synuclein in Ferroptosis-Driven Dopaminergic Neurodegeneration.

The selective degeneration of dopaminergic neurons is a hallmark of Parkinson's disease and related disorders. While multiple cell death pathways have been implicated, ferroptosis has recently emerged as a critical mechanism. This iron-dependent form of regulated cell death is driven by the accumulation of phospholipid hydroperoxides, leading to oxidative membrane damage. Dopaminergic neurons are intrinsically vulnerable to ferroptosis due to their high iron content, active dopamine metabolism (a source of reactive oxygen species), and relatively low antioxidant defenses. Here we synthesize ev

Organoids for Metabolic Disease Modeling.

Inherited metabolic diseases (IMDs) are a diverse group of rare genetic disorders that disrupt metabolic pathways, leading to severe clinical manifestations. Disease models ranging from complex animal models to simple in vitro systems have provided insights into IMDs, but each has limitations. Organoids, three-dimensional in vitro models, bridge this gap by replicating key metabolic functions that are absent in most simple 2D cell models. While organoids do not fully mimic organ complexity, they effectively model disease-specific metabolic defects, as seen in methylmalonic acidemia, Wilson's d

Structural basis for transport and inhibition of nucleotide sugar transport in pathogenic fungi.

GDP-Mannose transporters are Golgi-localised solute carriers that are essential for the virulence of pathogenic fungi, serving as critical components of fungal glycosylation pathways. However, the mechanism by which nucleotide sugars are recognised and transported across the Golgi membrane remains unclear, hindering efforts to develop effective inhibitors that could serve as distinct antifungal agents. Here, we present cryo-EM structures of the GDP-Mannose transporter, Vrg4, from Candida albicans in complex with nanobodies in both the cytoplasmic and Golgi-facing states. Structural comparisons

Liprin-α family proteins and their cellular functions.

Scaffold proteins are critical for the spatial organization and assembly of subcellular compartments through interactions with specific binding partners. The liprin-α family of scaffold proteins are implicated in diverse cellular functions across both neuronal and non-neuronal cell types. All four liprin-α isoforms contribute to presynaptic active zone formation, promoting synaptic vesicle accumulation and neurotransmitter release. Additionally, there is evidence that liprin-α plays a role in postsynaptic cellular processes such as dendritic growth, spinogenesis, and AMPA receptor trafficking.

Perioperative Pain Management in Urogynecology: A Review.

Postoperative pain after pelvic reconstructive surgery is inseparably tied to care quality, patient experience, and demands on the health care system. Pelvic reconstructive surgeons have considerably advanced our knowledge of postoperative pain after procedures and of perioperative interventions to address postoperative pain. This narrative aims to provide helpful information to surgeons as we continue to prioritize patient-centric outcomes, lower costs, and the risks of long-term opioid use. In this review, we present an overview of pain processing pathways. Then we discuss the natural histor

Mechanical regulation of cell memory.

Cell memory describes the capacity of a cell to retain induced biological changes following withdrawal of initiating signals. Accumulating evidence reveals that mechanical stimuli in the microenvironment not only drive immediate structural reorganization in cells and extracellular matrix (ECM), but also can induce long-term changes in cell phenotype and identity. These effects underlie critical biological phenomena, including cell migration, differentiation and reprogramming. In this Review, we present an integrative framework for understanding how mechanical signals modulate cellular structur

Prenylation: A key regulatory point in tumor metabolism and a potential therapeutic target.

Prenylation is an important post-translational lipid modification that enhances protein hydrophobicity and regulates subcellular localization. It plays a critical role in multiple signaling pathways and metabolic processes and is closely associated with the development and progression of various diseases, particularly in tumor initiation, progression, and metastasis. However, its systemic regulatory roles and therapeutic vulnerabilities in cancer remain incompletely understood. In this review, we summarize the molecular mechanisms of protein prenylation and its biological functions, with a par

Clonal evolutionary analysis reveals patterns of malignant transformation of Intraductal Papillary Mucinous Neoplasms of the pancreas.

Intraductal papillary mucinous neoplasms (IPMNs) are critical precursors to pancreatic ductal adenocarcinoma, a highly lethal cancer characterized by late detection and rapid progression. Here we integrate multi-region whole-genome and transcriptome sequencing to trace the evolution of IPMN, constructing detailed phylogenetic trees to provide insights into subclonal architectures and progression pathways. Our analysis identifies two distinct evolutionary trajectories: one driven by a single ancestral clone, and another involving multiple independent ancestral clones. We further explore the rol

Epitranscriptomic regulation in the progression of cancer and platinum resistance.

Epitranscriptomic regulation has emerged as a critical mechanism in cancer biology, particularly in the development of chemoresistance. RNA modifications including N6-methyladenosine (m6A), 5-methylcytosine (m5C), N1-methyladenosine (m1A), 7-methylguanosine (m7G), pseudouridine (Ψ), and A-to-I editing dynamically control mRNA stability, splicing, translation, and degradation. RNA-modifying proteins called 'writers,' 'erasers,' and 'readers' regulate post-transcriptional networks to enable tumor adaptation and chemoresistance. In platinum-resistant tumors, epitranscriptomic changes modulate DNA

Preserving perianastomotic venous collaterals in arteriovenous grafts: A paradigm shift in outflow management.

Surgical management of perianastomotic venous collaterals during the creation of vascular access remains a critical yet often overlooked step. Traditionally, these branches are ligated to optimize hemodynamics and direct the entire flow into the primary venous outflow tract. However, pathophysiological analysis suggests that this practice may limit the overall resilience of the access. Perianastomotic collaterals serve a functional role as essential compensatory outflow pathways; in the presence of stenosis or obstruction of the primary venous segment, their patency maintains blood flow, reduc

Mechanism and therapeutic potential of mitophagy in spinal cord injury.

Mitochondrial dysfunction is a critical factor in secondary injury following spinal cord injury (SCI). Mitophagy is an essential mechanism for mitochondrial quality control. Proper and timely activation of mitophagy clears damaged mitochondria, reduces oxidative stress and cell death, and provides neuroprotection. However, excessive activation can cause energy depletion and worsen injury. The effects of mitophagy depend on the specificity of its spatial and temporal activation as well as the cellular microenvironment. This review summarizes novel therapeutic strategies targeting mitophagy, inc

Case Report: An incidentally discovered HPV-associated endocervical adenocarcinoma presenting as pseudomyxoma peritonei.

This case report describes a 48-year-old woman with occult HPV-associated endocervical adenocarcinoma (Silva pattern B) presenting as bilateral ovarian metastases and pseudomyxoma peritonei-like gelatinous ascites, despite normal cervical morphology and resolved HPV type 45 infection. Initial misdiagnosis as primary ovarian mucinous carcinoma was revised based on histopathology and immunohistochemistry (diffuse p16 positivity, ER-negative and PR-negative, Ki-67 index of 95%), confirming metastatic endocervical adenocarcinoma. The absence of appendiceal lesions excluded true PMP, attributing as

Nickel-iron-cobalt (hydr)oxide-decorated carbon nanotube forest for the oxygen-evolution reaction.

In this study, an oxygen-evolution reaction (OER) electrocatalyst was developed by first growing a vertically aligned carbon nanotube forest (CNT F), and a NiFeCo (hydr)oxide coating was co-electrodeposited onto the CNT forest. X-ray diffraction, scanning electron microscopy, and X-ray photoelectron spectroscopy confirm that the metal (hydr)oxide is distributed across the high-surface-area CNT scaffold. The performance is attributed to the synergy between the multi-metal (hydr)oxide supported on CNT F, which together provide abundant active sites, efficient charge transport pathways, and stron

Beyond antimicrobials: a host-directed precision pharmacotherapy framework for septic shock.

Despite progress in antibiotic therapy and supportive care, septic shock continues to pose a substantial mortality burden in intensive care units, highlighting persistent limitations in current pathogen-focused treatment approaches. A central issue is the heterogeneity of host immune responses to infection, manifesting as diverse clinical phenotypes-such as immunosuppression and hyperinflammation-that drive variability in outcomes. Thus, evolving the therapeutic paradigm from infection control alone to precise modulation of the host response represents a critical next step for improving progno

Cyclic sealing and drainage on an oceanic transform fault.

Oceanic transform faults have been considered conservative, shear-dominated boundaries, yet their proximity to magmatic systems implies fluid involvement. In this work, we discovered tidally modulated tremor at the Gofar transform fault along the East Pacific Rise. Tremor amplitude correlates with semidiurnal tides during periods of sparse seismicity and low in situ compressional to shear wave velocity ratio (Vp/Vs), but this correlation weakens following earthquake swarms accompanied by high Vp/Vs. We propose a valve-like sealing-drainage dynamic process where sealing traps volatiles and boos

Thioredoxin-interacting protein in radiation-induced pyroptosis and autophagy: Unveiling mechanisms and implications for preventing radiation-induced lung injury.

Radiation-induced lung injury is a serious complication of thoracic radiotherapy, wherein the inflammatory cell death process of pyroptosis and the catabolic process of autophagy are concurrently activated, yet the master regulator coordinating their crosstalk remains unknown. This study identified thioredoxin-interacting protein (TXNIP) as this key regulator. We discovered that TXNIP, upregulated by radiation, does not independently promote pyroptosis and autophagy but directly couples them by competitively binding to NLRP3. This competition displaces Beclin-1 from NLRP3, simultaneously facil

Novel Inflammatory Biomarkers for Poor Ovarian Response Detected by Follicular Fluid Olink Proteomics.

Although inflammatory and immune mechanisms are known to play a crucial role in the pathogenesis of poor ovarian response (POR), their specific functions remain unclear. This study aimed to compare inflammatory proteins in the follicular fluid of POR patients and those with normal ovarian reserve using Olink proteomics to identify differentially expressed proteins (DEPs) and their related pathways. Using the Olink Reveal platform, bioinformatics, and machine-learning models, we identified 50 DEPs. Correlation and protein-protein interaction analyses revealed relationships between key proteins,

Advances in osteosarcoma research: Pathogenesis and emerging therapeutic strategies.

Osteosarcoma is the most common primary malignant bone tumor in children and adolescents, and continues to pose formidable clinical challenges due to its high invasiveness and propensity for early metastasis. Although recent advancements in multimodal treatment, such as neoadjuvant chemotherapy combined with limb-salvage surgery, have significantly improved local control, the 5-year survival rate for patients with metastatic or recurrent disease remains dismally low at 20-30%. In this review, we present a comprehensive synthesis of current knowledge regarding the molecular and genetic underpin

The therapeutic potential of compounds from natural products for alleviating depression by targeting P2X7 receptor-mediated proinflammatory signaling pathways.

Depression is a prevalent neuropsychiatric disorder with high incidence and causing severe disability, representing a clinically unmet challenge and thus demanding more effective therapeutics. Neuroinflammation in the central nervous system (CNS) is a pathological feature of depression and, with increasing recognition, it is also a critical depression-driving mechanism. In the CNS, the P2X7 receptor for extracellular ATP is expressed in microglia and astrocytes, and acts as a key mediator of neuroinflammation. Besides medicinal chemistry efforts in developing novel CNS-penetrable P2X7 antagoni
