Drive for thinness in black and white preadolescent girls.

This study examined racial differences in drive for thinness, a motivational variable implicated in the etiology of eating disorders. Subjects included 613 black and white preadolescent girls from one of three National Heart, Lung, and Blood Institute (NHLBI) Growth and Health Study centers. Instruments included the Drive for Thinness Scale, a Criticism about Weight scale, the Self-Perception Profile for Children, a Sexual Maturation index, and 3-day food diaries. Black girls reported significantly greater drive for thinness than white girls. Drive for thinness was significantly associated wit

The molecular genetics of albinism and piebaldism.

BACKGROUND: Oculocutaneous albinism (OCA) is an autosomal-recessive genetic disorder defined by hypomelanosis in the eyes, hair, and skin. Piebaldism is an autosomal-dominant congenital leukoderma associated with a white forelock. The molecular pathogeneses of these congenital pigmentary disorders have been clarified in recent years and are briefly reviewed here. OBSERVATIONS: The pathologic gene mutations causing OCA and piebaldism are as follows. When a mutated tyrosinase gene produces inactive, less active, or temperature-sensitive tyrosinase, its phenotype is tyrosinase-negative (type I-A

Changes in placental ultrasonic appearance. II. Pathologic significance of Grade III placental changes.

Changes in placental texture and structure on ultrasonic examination have been suggested as correlating to advancing fetal pulmonary maturity. The association of preterm appearance of Grade III changes in the placenta in the complicated pregnancy suggests that these changes are associated with a decrease in placental function and premature senescence of the placenta. Placentas were routinely graded on all obstetric examinations performed during a 1-year period of time. Those pregnancies in which Grade III changes in the placenta prior to term were reviewed. A high incidence (78%) of perinatal

Congenital abnormalities: "is it wise to have another child?".

Congenital abnormalities affect some five per cent of all live births, and are a major factor in childhood morbidity and mortality. Most physicians, therefore, encounter a variety of developmental disorders, and must frequently deal with problems faced by a handicapped patient and his family. This task is becoming increasingly difficult with the growth and sophistication of genetic knowledge and changes in society's attitude toward medical practice. Although congenital abnormalities are classified according to a variety of presumed genetic or environmental 'causes', it is important to remember

CHOPS Syndrome: A Rare Malformation Syndrome with de novo AFF4 Gene Variant.

INTRODUCTION: CHOPS syndrome is a rare, multisystem disorder caused by heterozygous pathogenic variants in the AFF4 gene. The acronym CHOPS is characterized by Cognitive impairment and coarse facial features (C), Heart defects (H), Obesity (O), Pulmonary involvement (P), and Short stature/skeletal dysplasia (S). The syndrome is caused by variants in AFF4, which encodes a protein critical for RNA polymerase II elongation and transcriptional regulation. CASE PRESENTATION: A 6-year-old male patient with CHOPS syndrome was diagnosed with a heterozygous de novo c.764C>T (p.Ala255Val) variant in AF

Impaired nutrient absorption, reduced bone mass and alterations in the gut microbiome contribute to postnatal growth retardation in a mouse model of MWS.

Mowat‒Wilson syndrome (MWS), a rare genetic disorder caused by heterozygous loss-of-function mutations in ZEB2, is characterised by significant growth retardation with unclear mechanisms. In this study, we developed a Zeb2 haploinsufficient (Zeb2+/-) mouse model that recapitulates key features of MWS, including reduced body weight, impaired intestinal development and skeletal hypoplasia. RNA sequencing revealed significant downregulation of nutrient digestion and absorption pathways in the duodenum of Zeb2+/- mice, which was associated with reduced body fat and bone mass loss. Additionally, Ze

Natural Products in the Treatment of Retinopathy of Prematurity: Exploring Therapeutic Potentials.

Retinopathy of prematurity (ROP) is a vascular disorder affecting the retinas of preterm infants. This condition arises when preterm infants in incubators are exposed to high oxygen levels, leading to oxidative stress, inflammatory responses, and a downregulation of vascular endothelial growth factors, which causes the loss of retinal microvascular capillaries. Upon returning to room air, the upregulation of vascular growth factors results in abnormal vascular growth of retinal endothelial cells. Without appropriate intervention, ROP can progress to blindness. The prevalence of ROP has risen,

Efficacy and Safety of Pegvisomant in the Treatment of Acromegaly.

Pegvisomant, the first and currently only clinically available growth hormone receptor antagonist, is an effective therapeutic option for the medical treatment of acromegaly, a rare disorder characterized by excessive growth hormone secretion. With now over 20 years of real world experience, its safety and efficacy is well-established. However, several aspects of its clinical use are still controversially discussed. The high cost of pegvisomant has limited its use in several countries, and recent studies have reported a lower efficacy than the initial clinical trials. A reported increase in tu

Disruption of Endochondral Ossification and Extracellular Matrix Maturation in an Ex Vivo Rat Femur Organotypic Slice Model Due to Growth Plate Injury.

Postnatal bone fractures of the growth plate (GP) are often associated with regenerative complications such as growth impairment. In order to understand the underlying processes of trauma-associated growth impairment within postnatal bone, an ex vivo rat femur slice model was developed. To achieve this, a 2 mm horizontal cut was made through the GP of rat femur prior to the organotypic culture being cultivated for 15 days in vitro. Histological analysis showed disrupted endochondral ossification, including disordered architecture, increased chondrocyte metabolic activity, and a loss of hypertr

Nutritional rickets presenting with developmental regression: a rare presentation of rickets.

Rickets is a disorder of defective mineralisation of the growth plate. Vitamin D deficiency remains the leading cause of nutritional rickets worldwide. We present the case of a 3.5-year-old breastfed boy who presented with dental abscess when a history of developmental regression was noted. Clinical assessment revealed hypotonia, poor growth and stunting. Biochemistry identified hypocalcaemia (1.63mmol/L, [normal range (NR) 2.2-2.7mmol/L]), severe vitamin D deficiency (25hydroxyvitamin D 5.3nmol/L, [NR > 50nmol/L]) with secondary hyperparathyroidism (Parathormone 159pmol/L, [NR 1.6-7.5pmol/L])

Endometriosis Cell Proliferation Induced by Bone Marrow Mesenchymal Stem Cells.

Endometriosis is an estrogen-dependent gynecological disorder that affects 10% of reproductive-aged women and causes pelvic pain and infertility. Bone marrow-derived stem cells (BMDCs) are known to engraft endometriosis in association with lesion growth; however, they do not undergo significant clonal expansion. The indirect effects of BMDCs on endometriosis growth and cell proliferation are not well characterized. Here, we demonstrate that BMDCs' co-culture increased endometrial stromal cell proliferation. In vitro studies using endometrial cells showed that BMDCs increased cell proliferation

An update on the pathogenesis, inflammation, and metabolism in hirsutism and polycystic ovary syndrome.

Hirsutism is a common endocrine disorder, defined as increased growth of terminal hairs in a male pattern. Hirsutism is most often caused by polycystic ovary syndrome (PCOS), whereas only 5% patients are diagnosed with rare endocrine diseases. PCOS may be considered a multiorgan disease causing not only increased adrenal and ovarian sex hormone secretion but also changed secretion of gonadotrophins, growth hormone, and adrenocorticotrophic hormone (ACTH) from the pituitary. The majority of patients with PCOS are insulin resistant and PCOS is characterized by an increased inflammatory state wit

[Paradoxal growth hormone and cortisol response to hypoglycemia caused by endogenous hyperinsulinemia: a case report].

Growth hormone (GH) together with cortisol are two important counter-regulatory hormones maintaining normal glycemia. Hyperinsulinemic hypoglycemia is a disorder of this counter-regulation described in neonates. We report here a rare case of reversible dissociated hypopituitarism secondary to an insulinoma in a 38-year-old man referred for investigation of hypoglycemic episodes. Hormonal investigations were in favour of dissociated anterior pituitary failure, with growth hormone and corticotroph deficiency. The hypothalamic-pituitary MRI was normal. The fasting test argued in favour of a hyper

A neuroprotective peptide antagonizes fetal alcohol exposure-compromised brain growth.

We evaluated a 9-amino-acid peptide, SALLRSIPA (SAL), an agonist of activity-dependent neurotrophic factor (ADNF), for its protective properties against fetal alcohol-related brain growth retardation, using an established liquid diet model of alcohol-related neurodevelopmental disorder (ARND) in C57BL/6 mice. Alcohol exposure during neurulation reduced body weight, head size, and specifically brain weight and volume. Major gross brain deficits include underdevelopment of brain areas, cortical thinning, ventricle enlargement, and restricted midline neural tissue growth leading to openings at th

Does it always have to be Perthes' disease? What is epiphyseal dysplasia?

The epiphyseal dysplasias are classified as osteochondral dysplasias in a heterogeneous group of skeletal dysplasias. The common feature of these hereditary conditions is short stature attributable to skeletal dysplasia at various sites. Multiple epiphyseal dysplasia and spondyloepiphyseal dysplasia are of interest to the authors, because the two syndromes are similar in their morphologic features and at first glance suggest the diagnosis of Perthes' disease. Spondyloepiphyseal dysplasia (Wiedemann-Spranger syndrome) is a disorder of growth in which there is an abnormality of enchondral ossifi

Connected to Others and Kind to Self: Social Connection and Self-Affiliation Change in Open-Ended Psychotherapy.

OBJECTIVE: Social connection predicts health, but remains largely underexplored as an outcome of psychotherapy. A friendly relationship to oneself is linked to positive relationships to others, but it is not clear how they influence each other across time during and after treatment. This study evaluates change in a measure of social connection and support, the Quantity and Quality of Social Relations (QQSR), and investigates its longitudinal associations with the Structural Analysis of Social Behavior Introject self-affiliation. METHODS: 370 adult outpatients in Norway with various presenting

Oxidative Stress Drives Cell Cycle Stalling, Apoptosis and Metabolic Suppression in Cystatin B Deficient EPM1 Patient iPSCs.

Cystatin B (CSTB) is an inhibitor of cysteine proteases, particularly cathepsins. Biallelic loss-of-function mutations in the CSTB gene are causative for progressive myoclonic epilepsy type 1 (EPM1), a neurodegenerative disorder characterised by stimulus-sensitive myoclonus and generalised tonic-clonic seizures. Pathomechanisms underlying the disease progression include disturbed proteostasis, increased oxidative stress, neuroinflammation and increased neuronal apoptosis. CSTB downregulation can also lead to cell cycle defects and reduced cell proliferation. Conversely, overexpression of CSTB
