Mechanisms of maladaptive eating behavior among individuals with obesity and pain: exploring rumination.

Upwards of 40% of adults in the United States (US) meet criteria for obesity, highlighting obesity as a critical, ongoing public health crisis. Pain affects 33-40% of individuals with obesity, contributing to increased distress and added burden which necessitates a higher degree of coping resources. Individuals with obesity often manage distress through maladaptive eating behaviors, yet it is unclear what mechanisms may contribute to these individual differences in eating behaviors. One mechanism that is relevant to both obesity and pain is rumination, an aspect of repetitive negative thinking

Individual and Sex Differences in Behavior and Cortical Excitability in the Rat Valproate Model of Autism.

The rodent prenatal valproate (VPA) treatment is a widely used animal model of idiopathic autism spectrum disorder (ASD). However, the presence of autistic-like symptoms is highly variable in treated offspring. The disruption of the excitation-inhibition balance of certain brain areas has been proposed as a main feature in both human ASD and the VPA model. The current study presents a detailed analysis of neural development, diverse behaviors, and neocortical excitability in a high number of individually identified VPA-treated rat offspring of both sexes. Neocortical excitability was assessed

ARDS management beyond the guidelines: a practical physiology-based approach to individualized care.

Practice guidelines for acute respiratory distress syndrome (ARDS) have established lung-protective ventilation as the standard of care, but optimized management requires further individualization to each patient's specific physiological characteristics and response to treatment. Beyond guidelines-level targets, clinicians must answer numerous practical bedside questions from how to start ventilating a newly intubated patient, to what to do when the driving pressure target is not met, and how to assess recruitability and set positive end expiratory pressure (PEEP). Integrating ventilator manag

Individual C. elegans neurons display differential sensitivity to smn- 1 silencing.

Spinal muscular atrophy is a neuromuscular disorder primarily caused by mutations in the SMN1 (Survival of Motor Neuron 1) gene. SMN1 is ubiquitously expressed and encodes a protein essential for the assembly of small nuclear ribonucleoproteins, key components of pre-mRNA splicing. The SMN protein also participates in several other fundamental cellular processes, including RNA transport, regulation of actin dynamics, transcription, and translation. While multiple hypotheses have been put forward to explain the selective motor neurons (MNs) vulnerability to SMN deficiency, the precise mechanism

Neurocognitive mechanisms of mental imagery-based fear conditioning in individuals with social anxiety.

Fear learning serves as a foundational framework for understanding anxiety disorders, with growing interest in the role of mental imagery in fear conditioning. The present study examined whether individuals with social anxiety can acquire fear responses toward neutral conditioned stimuli (CS) via imagined fear-related social scenarios acting as unconditioned stimuli (US). Employing an imagery-based fear conditioning paradigm integrated with event-related potentials (ERPs), we recruited 31 participants with high social anxiety and 30 with low social anxiety. Behaviorally, neutral faces, initial

Characterize Oral-to-Blood Microbial DNA Translocation in Individuals with Cocaine Use Disorder.

BACKGROUND: Cocaine disrupts gut barriers in animal models, potentially enabling microbial translocation and inflammation in the periphery and central nervous system (CNS), but its direct role in inducing inflammation remains controversial. This study aimed to determine if the oral cavity is a source of circulating microbial DNA translocation in individuals with current cocaine use disorder (CUD). RESULTS: A cross-sectional case-control study was conducted, comparing CUD and demographically matched non-drug controls. Ten CUD (via smoking or vaping) and 24 controls provided paired saliva and b

Attention-deficit/hyperactivity disorder in individuals with primary tic disorders and Tourette syndrome.

The co-occurrence of tic disorders and ADHD (attention-deficit/hyperactivity disorder) is common; however, the significant overlap in their symptoms and underlying mechanisms frequently poses questions for diagnosis and treatment. The interaction between tic disorders and ADHD involves intricate genetic and neurobiologic factors, particularly related to inhibitory processes and cortico-striato-thalamic pathways in the brain. While both conditions present shared and specific challenges, studies suggest that ADHD symptoms often pose a greater burden on patients than tics. Therefore addressing AD

Online gambling as a public health and regulatory challenge: moving beyond individual responsibility.

Online gambling has expanded faster than the public health systems and regulatory frameworks designed to contain its harms. Unlike venue-based gambling, digital platforms combine constant accessibility, rapid betting cycles, frictionless payments, algorithmic personalization, and intensive marketing, features that may heighten risk and hinder early detection of problematic use. In this perspective, we argue that online gambling should be understood not only as an individual behavioral disorder, but also as a public health and regulatory challenge with broader social consequences. Recent eviden

Beta Thalassemia Carrier Detection in Individuals with Borderline Normal Hemoglobin A2 Levels.

Transfusion-dependent β-thalassemia is a significant public health challenge in many countries. Prevention through carrier detection enables counseling for at-risk couples and informed decision-making. Beta-thalassemia carriers are identified by elevated hemoglobin A2 levels (≥3.6%). However, individuals with borderline HbA2 levels of 3.0-3.5% fall within a diagnostic gray zone. Identifying carriers in this group is crucial given the significant medical, financial, and psychosocial implications. Therefore, this study aimed to identify β-thalassemia carriers among individuals with borderline he
