Biomimetic siRNA therapeutics attenuate mitochondrial DNA damage and cytokine storm in sepsis.

During the progression of severe sepsis, the oxidized mitochondrial DNA (mtDNA) in macrophages is cleaved by flap-structure-specific endonuclease 1 (FEN1) into small fragments, which are subsequently released into the cytosol and extracellular space to activate multiple pro-inflammatory signaling pathways such as NLRP3 inflammasome, cGAS-STING, and TLR9-NF-κB. Herein, biomimetic nanocomplexes (NCs) partially cloaked with macrophage membrane (MM) are developed to efficiently deliver FEN1 siRNA (siFEN1) into macrophages for sepsis management. To construct the NCs, membrane-penetrating, helical p

Interplay of Ferroptosis, Cuproptosis, Autophagy and Pyroptosis in Male Infertility: Molecular Crossroads and Therapeutic Opportunities.

Male infertility is intricately linked to dysregulated cell death pathways, including ferroptosis, cuproptosis, pyroptosis, and autophagy. Ferroptosis, driven by iron-dependent lipid peroxidation through the Fenton reaction and inactivation of the GPX4/Nrf2/SLC7A11 axis, disrupts spermatogenesis under conditions of oxidative stress, environmental toxin exposure, or metabolic disorders. Similarly, cuproptosis-characterized by mitochondrial dysfunction and disulfide stress due to copper overload-exacerbates germ cell apoptosis via FDX1 activation and NADPH depletion. Pyroptosis, mediated by the

Cephalomannine as in vitro and in vivo anticancer agent in mesothelioma cells: mechanisms of action.

BACKGROUND: In our previous drug screening, cephalomannine, fludarabine, and crenolanib were among the most active compounds against permanent cell lines of Pleural Mesothelioma (PM). METHODS: Here, firstly we assessed the efficacy of these compounds on PM-xenograft NOD-SCID mice and evaluated the overall survival (OS) as major endpoint. Then, we evaluated in vitro the underlying mechanisms involving CPM (at 0.1 µM) as anticancer agent on REN (derived from PM) and MeT-5A, a mesothelial cell line often used as a non-malignant model of mesothelial cells. We measured the oxygen consumption rate (

NRF2 Antioxidant Response and Interferon-Stimulated Genes Are Differentially Expressed in SARS-CoV-2-Positive Young Subjects.

BACKGROUND: Several respiratory viruses, including Severe Acute Respiratory Syndrome-Coronavirus-2 (SARS-CoV-2), suppress nuclear factor-E2-related factor-2 (NRF2) antioxidant response, generating oxidative stress conditions to its advantage. NRF2 has also been reported to regulate the innate immune response through the inhibition of the interferon (IFN) pathway. However, its modulation in younger individuals and its correlation with the IFN response remain to be elucidated. METHODS: The NRF2 and redox-related genes expression was examined in nasopharyngeal swabs from children attending the p

Optimization on the extraction conditions of flavonoids from Suaeda glauca and the research of its hepatoprotection in mice.

Liver disease is a serious threat to health worldwide. Flavonoids from Suaeda glauca (SGF) is able to alleviate liver lipid peroxidation. However, it is unclear whether SGF could protect against liver glycogen accumulation, inflammation and fibrosis. In this study, the extraction conditions of SGF were optimized with response surface methodology. The qualitative analysis of components in SGF was carried out by a LC-MS/MS method. Moreover, SGF was administered orally to male mice given 10 % carbon tetrachloride (CCl4) for 4 weeks at doses of 25 and 50 mg/kg once daily for 4 weeks. The optimal e

Withaferin A Attenuates Muscle Cachexia Induced by Angiotensin II Through Regulating Pathways Activated by Angiotensin II.

Cachexia is a multifactorial syndrome characterized by severe muscle wasting and is a debilitating condition frequently associated with cancer. Previous studies from our group revealed that withaferin A (WFA), a steroidal lactone, mitigated muscle cachexia induced by ovarian tumors in NSG mice. However, it remains unclear whether WFA's protective effects are direct or secondary to its antitumor properties. We developed a cachectic model through continuous angiotensin II (Ang II) infusion in C57BL/6 mice to address this issue. Ang II infusion resulted in profound muscle atrophy, evidenced by si

The multifaceted role of SQSTM1/p62 in disc degeneration: A master regulator of cellular stress responses.

Intervertebral disc degeneration (IDD) is a key contributor to lumbar degenerative diseases and chronic low back pain. Accumulating evidence indicates that Sequestosome 1 (SQSTM1/p62), a multifunctional adaptor protein, plays a pivotal role in IDD pathogenesis through its regulation of autophagy, oxidative stress, inflammation, and programmed cell death. This review summarizes the multifaceted functions of SQSTM1 in the context of IDD, including its involvement in the autophagy-lysosome pathway, antioxidant defense via the Keap1-Nrf2 axis, activation of the NF-κB signaling and NLRP3 inflammaso

Tape strip expression profiling of juvenile dermatomyositis skin reveals mitochondrial dysfunction contributing to disease endotype.

Skin inflammation in juvenile dermatomyositis (JDM) can signal disease onset or flare, and the persistence of cutaneous disease can prevent complete disease remission. The noninvasive study of cutaneous expression signatures through tape stripping (TS) holds the potential to reveal mechanisms underlying disease heterogeneity and organ-specific inflammation. The objectives of this study were to (a) define TS expression signatures in lesional and nonlesional JDM skin, (b) analyze TS signatures to identify JDM disease endotypes, and (c) compare TS and blood signatures. Although JDM lesional skin

Yogurt alleviates alcohol-induced hepatocyte pyroptosis and inflammation via autophagy/P62/Nrf2/ROS/NLRP3 pathway.

Alcohol, one of the most widely popular and consumed beverages worldwide, often leads to acute alcoholic liver injury (AALI) when excessively consumed within a short timeframe. It can further induce alcoholic hepatitis, posing a serious risk to human health and public health safety. Pyroptosis, a typical mode of programmed cell death, is closely associated with the body's inflammatory response and plays a central role in AALI. However, the role of yogurt in acute alcohol-induced pyroptosis and inflammation is unclear. In this study, we found that yogurt and its metabolite d-lactate effectively

Therapeutic prospects of modulating TLR4/MAPK/ROS signalling in obesity-associated neuroinflammation.

Источник пометил публикацию как отозванную

It is becoming more widely acknowledged that obesity is a chronic low-grade inflammatory disease that has a significant influence on brain health in addition to metabolic problems. Adipose tissue growth, macrophage polarization, and cytokine release all contribute to systemic inflammation, which weakens the blood-brain barrier (BBB) and promotes immune-to-brain communication. Saturated fatty acids and gut-derived lipopolysaccharides activate Toll-like receptor 4 (TLR4) in the central nervous system, which triggers downstream nuclear factor-κB (NF-κB) and Mitogen-activated protein kinase (MAPK)

New Perspectives on Gastric Inflammaging: Integrating Multi-Omics Mechanisms and Gerotherapeutic Strategies in Chronic Gastritis.

Chronic gastritis (CG) is a highly prevalent, age-associated inflammatory disorder of gastric mucosa and a key precursor of gastric cancer in older adults. Beyond Helicobacter pylori infection and environmental insults, accumulating evidence indicates that chronic, low-grade inflammation coupled with aging biology, "gastric inflammaging", plays a central role in driving mucosal degeneration, atrophy, and malignant transformation. Here, we synthesize current mechanistic and multi-omics evidence to conceptualize CG as a tractable model of organ-specific inflammaging. We first summarize how hallm

Targeting Cathepsin B-mediated pyroptosis in fibroblasts alleviate ultraviolet-induced skin inflammation and injury.

Exposure to ultraviolet (UV) radiation is known to cause acute skin damage and inflammation, yet the underlying molecular mechanisms and cellular damage involved in this process remain poorly understood. In this study, we provide compelling evidence that UV-induced fibroblast pyroptosis not only triggers a cascade of macrophage inflammatory responses but also could represents a important mechanism in the pathogenesis of acute sunburn. We further investigate the intricate interplay between pyroptotic fibroblasts (Pyr-Fbs) and macrophages, elucidating the resultant alterations in macrophage phen

IL-6: A new target in crystal-induced arthritides - A narrative review.

Gout and calcium pyrophosphate deposition disease (CPPD) are two highly prevalent causes of inflammatory arthritis, characterized by the pathological deposition of monosodium urate (MSU) and CPP crystals, respectively, in joint tissues. These crystals can induce an intense inflammatory response, known as crystal-induced inflammation, which involves innate immunity and is highly dependent of the activation of interleukin 1β (IL-1β) following the recruitment of the NLRP3 inflammasome. In patients in whom first-line treatments (colchicine, prednisone, NSAIDs) are either ineffective or inappropria

Interplay Among Gut Microbiota-Derived TMAO, Autonomic Nervous System Dysfunction, and Heart Failure Progression.

The gut microbiota is crucial for metabolic homeostasis and cardiovascular health. Dysbiosis triggers a gut-brain-heart axis dysfunction: vagal signaling promotes neuroinflammation and cerebral damage, which in turn impairs cardiac function. This bidirectional cycle is further exacerbated by reduced cerebral perfusion. Trimethylamine-N-oxide (TMAO), a metabolite of dietary choline and L-carnitine, acts as a primary mediator in this network. Elevated TMAO levels-resulting from bacterial conversion and hepatic oxidation-are linked to atherosclerosis and heart failure. Mechanistically, TMAO activ

Molecular Insights into Cell-Mediated Immunity in Atypical Non-Ulcerated Cutaneous Leishmaniasis.

Leishmania (Leishmania) infantum chagasi infections range from asymptomatic (AS) to severe visceral leishmaniasis (VL). One of the manifestations is an atypical non-ulcerated cutaneous leishmaniasis (NUCL), which occurs in some locations of Central America with few cases of VL. We conducted a transcriptomic analysis of cell-mediated immunity (CMI) on blood samples from NUCL, AS, VL patients from Amapala, Honduras, and healthy controls. RNA-seq revealed a similar perturbation of gene expression in NUCL and AS. Eight gene signatures of CMI were found in NUCL involved in CD8+ T lymphocyte infiltr

Anti-IL-18 immunotherapy decreases inflammatory and vaso-occlusive responses in mice with sickle cell disease.

Sickle cell disease (SCD) is characterized by inflammatory and vaso-occlusive processes that drive acute crises and progressive organ damage. Interleukin-18 (IL-18), elevated in patients with SCD and mouse models, contributes to these pathological mechanisms. We evaluated the effects of acute and prolonged IL-18 blockade using the SK113AE-4 monoclonal antibody in Townes and Berkeley SCD mice. Acute IL-18 neutralization reduced tumor necrosis factor-alpha (TNF-α)-induced microvascular leukocyte recruitment and prevented hypoperfusion, indicating that modulation of inflammatory signaling improve
