Changes in the phosphorylation of neurofilament proteins in facial motoneurons following various types of nerve lesion.

This report defines the conditions for changes in the phosphorylation state of neurofilaments (NF) after facial nerve lesions. In adult cont … the proximal nerve stump. After botulinum toxin application into facial muscles (i.e. inhibition of synaptic transmission at motoric endplates) there was no increase in the amount of pNF after 4 and 10 days, but it appeared in facial nuclei 4 weeks after injection on both, the treated and the untreated side, i.e. during functional restitution. Selectively transecting t …

Depressant insect selective neurotoxins from scorpion venom: chemistry, action, and gene cloning.

The present study examines the similarity in the symptoms and binding properties between the depressant and excitatory insect-selective neurotoxins, derived from scorpion venom. A comparison of their primary structures and neuromuscular effects is presented. A new depressant toxin (LqhIT2) was purified from the venom of the scorpion Leiurus quinquestriatus hebraeus. The effects of this toxin on a prepupal housefly neuromuscular preparation mimic its effects on the intact insect, i.e, a brief period of repetitive bursts of regular junction potentials (JPs) is followed by reduced amplitude JPs e

omega-Conotoxin GVIA and nifedipine inhibit the depolarizing action of the fungal metabolite, destruxin B on muscle from the tobacco budworm (Heliothis virescens).

Recent studies on a group of fungal metabolites, collectively called the destruxins, have suggested that these compounds activate calcium influx in insect skeletal muscle. In this study, we have investigated the sensitivity of destruxin B to the voltage-dependent calcium channel antagonists; omega-conotoxin GVIA, nifedipine, diltiazem and methoxyverapamil on skeletal muscle from the lepidopteran insect pest, tobacco budworm (Heliothis virescens). At a concentration of 1.7 microM, destruxin B caused a rapid decrease in the transmembrane resting potential. The effect of destruxin B on insect mus

Inhibitors of calcium-dependent enzymes prevent the onset of afterdischarge in the peptidergic bag cell neurons of Aplysia.

Experiments with isolated bag cell neurons of Aplysia have produced evidence that changes in neuronal excitability may be brought about by t … sis of the effects of TFP showed that this inhibition occurs at concentrations which do not inhibit synaptic transmission or the endogenous bursting of another neurosecretory neuron, R15. Secretion of neuroactive peptides from the bag cell neurons was measured both electrophysiologically and biochemically. No attenuation of secretion could be observed at concentrations …

Preserved autism-associated behaviors and dopaminergic neuron hyperactivity in DAT-IRES-Cre mice after valproate exposure.

Dopamine (DA) signaling is implicated in neurodevelopmental disorders such as autism spectrum disorder (ASD). Cell-type-specific tools like DAT-IRES-Cre mice are essential for dissecting the underlying circuit mechanisms. However, as the transgenic Cre insertion may affect DA transmission, it is crucial to validate that these Cre mice recapitulate key physiological and behavioral phenotypes in established ASD models. Prenatal exposure to valproic acid (VPA) is a widely used ASD model that captures gene-environment interactions in etiology, and DA system dysfunction has been reported in this mo

Multiplex gray and white matter networks in autism spectrum disorder: differential topological alterations and transcriptomic associations.

Autism spectrum disorder (ASD) is classically conceptualized as a dysconnectivity syndrome. However, most studies have examined structural a … cortical circuit and the default mode network associated with genes implicated in cell adhesion and synaptic transmission. Notably, no significant group differences were observed in the multiplex participation coefficient, which indexes cross-layer integration, suggesting that the overall cross-layer connectivity distribution remains relatively stable. These findings d …

Activation of DMH GABAergic neurons, but not local GABAergic AgRP neurons, attenuates chronic stress-induced POMC neuron hyperactivity.

Identifying the neural circuits engaged and reshaped by chronic stress is critical for understanding how adaptive responses shift to maladaptive behaviors that contribute to stress-related disorders. Our previous work demonstrates that chronic unpredictable stress (CUS) induces a persistent increase in the firing activity of proopiomelanocortin (POMC) neurons in the arcuate nucleus (ARC). This hyperactivity is due, in part, to a reduction in GABAergic synaptic transmission onto POMC neurons, indicating a disruption in inhibitory control. However, the sources of GABAergic inputs responsible for…

Sensory maps in the telencephalic pallium of goldfish.

The functional organization of the teleost telencephalic pallium remains poorly understood, particularly regarding the presence of modality- … ptors markedly reduced sensory-evoked responses, indicating that these maps depend on glutamatergic synaptic transmission. Together, these findings reveal a more elaborate and functionally differentiated pallial organization than previously recognized and provide the first direct evidence for modality-specific topographic sensory maps in the teleost pallium. More broad …

Cell-specific expression of APP and GABABR1 isoforms in wild-type and the APP NL-G-F mouse model of Alzheimer's disease.

Amyloid precursor protein (APP) is a type I transmembrane protein that undergoes proteolytic processing to generate amyloid-β, the main component of amyloid plaques found in brains with Alzheimer's disease (AD). APP proteolytic processing also generates soluble APP alpha (sAPPα) which modulates synaptic transmission and neurite outgrowth through the γ-aminobutyric acid type B receptor (GABABR). sAPPα specifically binds the 1a isoform of GABABR subunit 1 (R1a) which contains two sushi domains absent in R1b. Here, we define the cell type-specific expression patterns of APP and GABABR1a in the br…

Compartment-specific analysis reveals disrupted astrocytic calcium transients in anesthetized Alzheimer's disease mice.

BackgroundAlzheimer's disease (AD) is characterized by presence of extracellular amyloid plaques, intracellular tau tangles, and extensive neuronal cell death. In addition to neurons, astrocytes modulate neuronal network activity through tripartite synapses. Also, astrocytes are increasingly recognized for their involvement in AD pathology. Aberrant astrocytic calcium signaling has been implicated in AD pathological processes, including disrupted synaptic transmission, dysregulated glutamate homeostasis, and impaired vascular function via astrocytic endfeet. Previous investigations have assess…

Heroin Regulates the Voltage-Gated Sodium Channel Auxiliary Subunit, SCN1b, to Modulate Nucleus Accumbens Medium Spiny Neuron Intrinsic Excitability and Cue-Induced Heroin Seeking.

Self-administration of addictive substances like heroin can couple the rewarding/euphoric effects of the drug with drug-associated cues, and … CN1b increased the intrinsic excitability of MSNs, but without altering glutamatergic and GABAergic synaptic transmission. While reducing NAc SCN1b levels had no effect on acquisition of heroin self-administration or extinction learning, we observed a significant increase in cue-reinstated heroin seeking, suggesting that NAc SCN1b normally limits cue-reinstated heroin …

Compensatory remodeling of a septo-hippocampal GABAergic network in the triple transgenic Alzheimer's mouse model.

BACKGROUND: Alzheimer's disease (AD) is characterized by a progressive loss of memory that cannot be efficiently managed by currently available AD therapeutics. So far, most treatments for AD that have the potential to improve memory target neural circuits to protect their integrity. However, the vulnerable neural circuits and their dynamic remodeling during AD progression remain largely undefined. METHODS: Circuit-based approaches, including anterograde and retrograde tracing, slice electrophysiology, and fiber photometry, were used to investigate the dynamic structural and functional remode

Exaggerated potassium current reduction by oxytocin in visceral sensory neurons following chronic intermittent hypoxia.

Oxytocin (OT) from the hypothalamus is increased in several cardiorespiratory nuclei and systemically in response to a variety of stimuli and stressors, including hypoxia. Within the nucleus tractus solitarii (nTS), the first integration site for cardiorespiratory reflexes, OT enhances synaptic transmission, action potential (AP) discharge, and cardiac baroreflex gain. The hypoxic stressor obstructive sleep apnea, and its CIH animal model, elevates blood pressure and alters heart rate variability. The nTS receives sensory input from baroafferent neurons that originate in the nodose ganglia. No…
