Grasping Thin Polypropylene Sutures Even With Less Traumatic Instruments May Decrease Their Tensile Strength.

OBJECTIVE: Anastomotic false aneurysms can form due to damaged polypropylene sutures (PS). This study aimed to examine whether pinching PS with modern less traumatic vascular instruments affects their tensile strength (TS). METHODS: Fifteen pairs each of 3-0 Prolene and 4-0 and 5-0 Optilene, as well as 18 pairs each of 6-0 and 7-0 Surgipro II PS were tested. Half of the sutures were compressed with a Jacobson Micro Needle Holder (NH) and half with DeBakey atraumatic tissue forceps (TF) for two seconds using a standardised pressure force of 40.8 ± 5.0 g. The pressure was based on the mean pinc

Prenatal exposure to Antiseizure Medications for all indications and the risk of neurodevelopmental disorders in offspring: a nationwide population-based cohort study in South Korea.

PURPOSE: Antiseizure medications (ASMs) are used during pregnancy for both epilepsy and non-epilepsy indications, yet the neurodevelopmental safety of prenatal ASM exposure remains uncertain, particularly in Asian populations. We evaluated the risk of neurodevelopmental disorders (NDDs) following in utero ASM exposure using a nationwide Korean database. METHODS: This population-based cohort study (2009-2021) used the Korea National Health Insurance Database to identify 3663,052 mother-infant pairs. ASM exposure was defined as ≥1 prescription between the last menstrual period and delivery. Usi

Predicting Antibody-Antigen Mutation ΔΔG via Side-Specific Protein Language Models and Paired Geometric Graph Learning.

Mutation-induced changes in binding free energy (ΔΔG) at antibody-antigen interfaces are important for antibody optimization, mutational scanning, and viral immune escape assessment. However, computational prediction remains challenging because antibodies and antigens have distinct sequence backgrounds, mutation effects are often localized at interfaces, and related complexes may remain across training and evaluation partitions. We present AbAgMut-GNN as a task-oriented paired graph framework that coordinates established sequence and geometric learning components around explicit comparison of

Pesticide and metabolite residue mixtures in subtropical agroecosystem soils: The inconvenient truth.

The presence and accumulation of multiple pesticide residues in soil threaten biodiversity and soil health, yet depth-resolved evidence on pesticide-metabolite mixtures in subtropical agroecosystems remains underexplored. Here, we present the most comprehensive paired-depth assessment of parent pesticides and metabolites in Indian soils to date. We analyzed 181 compounds (63 insecticides, 41 fungicides, 32 herbicides, 45 metabolites) in 140 paired surface (0-5 cm) and subsurface (15-20 cm) soil samples collected during the 2024 growing season from 70 sites representing major land uses in the H

Potential molecular communication of blood and synovium through ligand-receptor interactions in osteoarthritis.

INTRODUCTION: Osteoarthritis (OA) is a common degenerative joint disease resulting from the breakdown of multiple joint tissues, remains a leading cause of disability with limited therapeutic options. Synovitis is one of the reasons of OA progression, while communication between blood and synovium during disease process is still unclear. METHODS: We used transcriptomic datasets from blood and synovium of healthy controls and OA patients to investigate potential molecular crosstalk between blood and synovium in OA pathogenesis through ligand-receptor pairs. RESULTS: Ligand-receptor pair analy

A minimum of 16° of rotation of transcervical femoral neck fractures leads to arterial collapse: an anatomical cadaveric study.

PURPOSE: Avascular necrosis (AVN) of the femoral head is a significant complication following femoral neck fractures. Rotational deformity has been proposed as a mechanism of vascular compromise, yet the degree of rotation required to interrupt intracapsular blood flow remains undefined. This cadaveric study quantified the degree of external rotation required to produce angiographic occlusion of the femoral circumflex arteries. METHODS: Twelve cadaveric hips underwent catheterization of the deep femoral artery with radiocontrast injection under fluoroscopy to visualize the medial and lateral f

GiGCN: a network-based framework for uncovering synthetic lethal and viable genetic interactions.

Genetic interactions (GIs) underpin the functional connectivity of genes and pathways, and are important for dissecting genotype-phenotype relationships and identifying therapeutic targets for diseases. However, the scale of the human genome restricts systematic experimental interrogation of GIs. Existing computational tools focus on predicting synthetic lethality (SL) and synthetic viability (SV), the two primary forms of GIs, yet their accuracy and biological interpretability are compromised by inadequate modeling of the molecular mechanisms behind positive and negative interactions, as well

Flow Orchestrated Regulatory Genomics Engine (FORGE): A Configurable Nextflow Pipeline for End-to-End snMultiome Analysis.

UNLABELLED: Single-nucleus multiome assays jointly profile gene expression and chromatin accessibility, yet their analysis typically requires bespoke chaining of modality-specific tools, creating barriers to reproducibility, scalability, and regulatory interpretation. We present FORGE (Flow Orchestrated Regulatory Genomics Engine), a configurable workflow that automates standalone snRNA-seq and snATAC-seq analysis, integrates the pair through complementary linear and nonlinear latent-variable models, and carries them through regulatory-network inference and differential testing. Clonal overlap and convergent clustering of T cell receptor signatures in Crohn's disease in monozygotic twins.

INTRODUCTION: The dysregulated immune response in Crohn's disease might result from disturbances in the T cell receptor (TCR) repertoire. To investigate this hypothesis, we compared the peripheral TCR repertoire within T cell subsets in twin pairs concordant and discordant for Crohn's disease. METHODS: We performed TCRα and TCRβ sequencing on peripheral flow-sorted CD4+ memory gut-homing (integrin α4β7), non-gut-homing (integrin α4β7-), and regulatory T cells (Tregs) from Dutch monozygotic Crohn's disease concordant twins (n = 8), monozygotic Crohn's disease discordant twins (n = 8), and heal

Simplified definition and imaging-based stratification of at-risk MASH using magnetic resonance elastography.

BACKGROUND: Accurate identification of patients with metabolic dysfunction-associated steatohepatitis (MASH) at risk of disease progression (at-risk MASH) is critical for guiding resmetirom and semaglutide therapy. Inconsistent definitions of at-risk MASH complicate clinical risk classification and comparisons across studies. This study aimed to standardize the definition of at-risk MASH and propose optimized MRE-based liver stiffness (LS-MRE) cutoffs to improve noninvasive risk stratification and better guide pharmacotherapy. METHODS: In this prospective study, 413 patients with suspected or

A blood-based metabolomics study of cardiovascular diseases in Swedish twins reveals apolipoprotein B biology as important for myocardial infarction.

Cardiovascular diseases are the leading cause of death globally. Consequently, metabolomics studies have in recent years aimed at identifying relevant biomarkers, yet no studies have been performed in twin populations, which reduce confounding due to genetic and environmental factors. We included 11,217 twins (age at intake, 47-94 years (mean = 65)) from the Swedish Twin Registry, holding data on 173 nuclear magnetic resonance metabolomic biomarkers and nationwide register-based data on diagnoses of ischemic stroke, ischemic heart disease, myocardial infarction, angina pectoris, and coronary a

ProptoView: AI-based digital exophthalmometry using multi-view facial images in a multinational validation study.

BACKGROUND: Accurate proptosis measurement is vital for managing thyroid eye disease (TED) and other orbital conditions. However, current approaches have certain limitations: the Hertel exophthalmometer is convenient but imprecise, while computed tomography (CT) is accurate but costly and exposes patients to radiation. METHODS: We developed ProptoView, an AI-based digital exophthalmometer, using 5676 images from 2516 eyes across 1258 visits of 763 TED patients with CT and Hertel measurements. For external validation, we used an additional 644 images from 648 eyes of 324 patients with TED and

Platelet Depletion Alters MCP-1 Measurements in Plasma From Children With Dengue.

BACKGROUND: Platelets are increasingly recognized as contributors to the cytokine storm in dengue, yet their residual presence in plasma may bias cytokine measurements. We compared MCP-1 levels between paired regular plasma (RP) and platelet-poor plasma (PPP) in children with dengue and evaluated how depletion influences cytokine quantification. METHODS: We first conducted a systematic review of plasma isolation methods in pediatric dengue studies. We then collected paired RP and PPP from 16 hospitalized children with dengue (11 with warning signs and 5 with severe dengue). The PPP was obtain

Comparative transcriptome analysis to reveal key drought stress-responsive genes in sorghum (Sorghum bicolor (L.) Moench).

Flowering pathways are accelerated for the rapid production of flowers and seeds in response to drought in certain varieties of sorghum (Sorghum bicolor (L.) Moench). The objective of the present study was to identify potential drought-responsive genes that affect flowering time in sorghum under drought stress. Sorghum germplasm accessions representing early-, intermediate-, and late-flowering groups were selected, and drought stress was applied to 25-day-old seedlings in the drought-stressed (DS) group by withdrawing water, while those in the control (WW) group were well watered. At anthesis

Expression of long non‑coding RNAs DIAPH3‑AS1, HMMR‑AS1, and SPATA3‑AS1 in patients with colorectal cancer: association with chemotherapy response and clinicopathological features.

BACKGROUND: Antisense long non-coding RNAs (lncRNAs) may fine-tune expression of their overlapping sense genes in cancer. DIAPH3-AS1, HMMR-AS1, and SPATA3-AS1 remain uncharacterized in colorectal cancer (CRC), and SPATA3-AS1 has not been studied in any malignancy. METHODS AND RESULTS: Ninety CRC patients were enrolled, with paired tumor biopsies collected before and after capecitabine-based chemotherapy; 71 healthy individuals served as controls. Expression of the three lncRNAs was measured by qRT-PCR and normalized to β-actin (2 - ΔCt method). Group and paired comparisons used the Mann-Whitn

Sex-specific DNA methylation changes in placental thyroid hormone regulatory genes following prenatal exposure to flame retardants.

Prenatal exposure to organophosphate esters (OPEs) and polybrominated diphenyl ethers (PBDEs) has been linked to disrupted fetal thyroid hormone (TH), though the underlying mechanisms remain unclear. Based on the S-MBCS cohort, we analyzed OPE metabolite concentrations in maternal urine during early pregnancy and PBDE levels in cord plasma. Methylation of five TH regulatory genes, namely deiodinase type 3 (DIO3), solute carrier family 16 member 2 (SLC16A2), solute carrier organic anion transporter family member 1C1 (SLCO1C1), thyrotropin-releasing hormone (TRH), and transthyretin (TTR), was qu
