Development and optimization of human glucocerebrosidase-encoding mRNA for Gaucher disease therapy.

Gaucher disease (GD) is a rare autosomal-recessive lysosomal storage disorder caused by mutations in the GBA1 gene encoding the lysosomal hydrolase glucocerebrosidase (GCase). Mutations in GCase lead to glucosylceramide accumulation within macrophages. Current treatments, including enzyme replacement therapy (ERT) and substrate reduction therapy (SRT), alleviate symptoms but are limited by high cost, frequent dosing, and adverse effects, highlighting the need for novel strategies. To achieve higher expression levels and improved stability, we designed and optimized a series of hGBA1-mRNA by ch

Cryptococcal dumbbell inflammatory pseudotumor mimicking skull histiocytic sarcoma in immunocompetent patient.

BACKGROUND: Cryptococcus neoformans is an opportunistic fungus with marked neurotropism, classically affecting immunocompromised patients, even though occasionally reported in immunocompetent hosts. Skull involvement is exceptionally rare and may radiologically mimic malignant tumors, causing diagnostic delays. CASE DESCRIPTION: A 34-year-old immunocompetent male presented with a painless, progressive left parieto-occipital scalp swelling over 3 weeks. Imaging revealed a well-defined, dumbbell-shaped cystic mass beneath the scalp, causing lytic skull erosion and limited intracranial extension

Circulating exosomal miRNA in acute coronary syndrome: mechanisms, clinical relevance and future directions.

Despite significant advances in diagnosis and treatment, Acute Coronary Syndrome (ACS) remains one of the leading causes of morbidity and mortality globally. It comprises ST-elevation myocardial infarction, non-ST-elevation myocardial infarction, and unstable angina, caused by complex mechanisms such as plaque rupture, thrombosis, erosion, and microvascular dysfunction. The development of high-sensitivity cardiac troponins has greatly improved the early detection of ACS. However, concerns remain about specificity, timing, and prognostic accuracy. Exosomal miRNAs (ExomiRs) show promise as minim

Palatal Emetic Receptor Density in Evolutionary Perspective: A Synthesis from Neuroanatomy, Comparative Primatology, and Biological Anthropology.

The soft palate harbors a morphologically and functionally heterogeneous population of sensory receptors whose coordinated activation constitutes one of the most ancient and conserved protective reflexes in vertebrate biology: the emetic reflex. Despite decades of investigation into the neurochemistry and pharmacology of emesis, the quantitative topography of palatal receptor populations, comprising free nerve endings (FNEs), encapsulated mechanoreceptors, and epithelial taste receptor cells (TRCs), remains incompletely characterized, and their evolutionary significance has received comparativ

Dual-Responsive Biomimetic Nanoplatform for Cascade Synergistic Therapy of Endometrial Cancer.

Endometrial cancer is a common malignant tumor in the female reproductive system. Traditional chemotherapy has limitations such as poor targeting, dose-limiting toxicity, and treatment intensity restrictions. To address these issues, this study constructed an intelligent nanoplatform-GOD-Fe3+&DOX-Membrane@Polymer (GOD-Fe3+&DM@Poly.)-based on reversible addition-fragmentation chain transfer (RAFT) polymerization technology. The incorporation of pyridine disulfide bonds as polymer side chains through RAFT polymerization facilitated the precise covalent immobilization of glucose oxidase (GOD), ef

Enhanced FGF21 Delivery via Neutrophil-Membrane-Coated Nanoparticles Improves Therapeutic Efficacy for Myocardial Ischemia-Reperfusion Injury.

Acute myocardial infarction, a leading cause of death globally, is often associated with cardiometabolic disorders such as atherosclerosis and metabolic syndrome. Metabolic treatment of these disorders can improve cardiac outcomes, as exemplified by the GLP-1 agonist semaglutide. Fibroblast growth factor 21 (FGF21), a novel metabolic regulator, plays pivotal roles in lipid mobilization and energy conversion, reducing lipotoxicity, inflammation, mitochondrial health, and subsequent tissue damage in organs such as the liver, pancreas, and heart. Here, we test the therapeutic efficacy of FGF21 in

An electron microscopy study of Bacillus anthracis capsule reveals early expression and novel strand-like structures.

Capsule is a key virulence factor for Bacillus anthracis. In this study, we examined the onset of capsule expression and the ultrastructure of capsule expressed in vitro and in infected hosts. We determined by immunofluorescence and immunoelectron microscopy that capsule was produced as early as 15 min after onset of germination. Further analysis revealed bacilli were often fully encapsulated when they emerged from the exosporium. Using conventional fixative to prepare samples for electron microscopy resulted in loss of capsular material, but using a fixative containing ruthenium red and lysin

Stabilizing SnO2 Colloids via Phosphate Buffering for Efficient and Durable Perovskite Photovoltaics.

SnO2 nanoparticles (NPs) solutions are considered a highly efficient inks for fabricating electron transport layers in state-of-the-art solution-processed perovskite solar cells (PSCs). However, SnO2 colloids exhibit thermodynamic instability in aqueous solution due to strong van der Waals attractions between nanoparticles, often leading to aggregation and precipitation. Here, a phosphate-buffered synthesis strategy is reported that effectively stabilizes SnO2 colloids. The phosphate buffer maintains a stable pH during synthesis, dynamically regulating the electrostatic repulsion between nanop

Harnessing droplet microfluidics and morphology-based deep learning for the label-free study of polymicrobial-phage interactions.

Evaluating the impact of bacteriophages on bacterial communities is required to assess the future utility of phage therapy. Methods able to study bacterial polycultures in the presence of phages are useful to mimic evolutionary pressures found in natural environments and recapitulate complex ecological contexts. Bacteriophages can drive rapid genetic and phenotypic changes in host cells. However, the presence of other bacteria can also impact bacterial densities and community structure and classical methods remain lengthy and resource intensive. Here we introduce a microdroplet-based encapsula

Nanoparticle-Modified Nanofibers Induce Ferroptosis and Stimulate Antitumor Immunity for Melanoma Therapy.

Melanoma, a highly aggressive and therapy-resistant cutaneous malignancy, presents formidable clinical challenges owing to its propensity for early metastasis and frequent postoperative recurrence. Here, a nanofiber-based drug delivery system (5F/STFM/R-ES) is developed that synergistically induces ferroptosis and potentiates immunotherapy to inhibit melanoma growth and reduce the risk of metastasis and recurrence. To this end, mesoporous silica nanoparticles encapsulating resiquimod (R848) are surface-coated with Fe/Mn-based metal-polyphenol networks (TFM) to form STFM/R. Subsequently, STFM/R

Constructing Immunomodulator Biosynthesis Factory in Grafting-From DNA Hydrogel for Heart Valve Regeneration.

The development of heart valve prostheses with regenerative capabilities offers significant potential to overcome the limitations of existing commercial artificial valves in clinical practice. Immune modulation plays a crucial role in heart valve regeneration by reversing the coagulation and inflammatory microenvironment, thereby facilitating recellularization. In this study, a biosynthesis factory is constructed on decellularized heart valves (DHVs) to continuously convert the abundant heme in the blood into immunomodulators, supporting long-term immune modulation and tissue regeneration. Advancing cancer therapy: Nanomaterial-based encapsulation strategies for enhanced delivery and efficacy of curcumin.

Curcumin, a polyphenolic compound derived from Curcuma longa, has gained significant attention as a potential anticancer agent due to its anti-inflammatory, antioxidant, and antitumor properties. Despite its therapeutic potential, the clinical application of curcumin is limited by its poor aqueous solubility, rapid metabolism, and limited bioavailability. To address these limitations, various nanomaterial-based encapsulation strategies have been developed, including polymeric nanoparticles, liposomes, solid lipid nanoparticles, micelles, dendrimers, and hybrid nanomaterials. These formulations

TIGIT/PVR axis regulates anti-tumor immunity in hematologic malignancies.

Hematologic malignancy stands as a grave form of cancer characterized by its arduous treatment and heightened likelihood of recurrence. Over the recent years, immunotherapy has progressively evolved into a pivotal approach for addressing hematologic malignancies. As a novel inhibitory receptor of NK and T cells, TIGIT is similar to PD-1, and blocking TIGIT can play a huge anti-tumor effect. At present, target TIGIT is still in clinical trials. Within this context, the TIGIT/PVR axis, serving as a pivotal element within the immunomodulatory framework, assumes a critical role in tumor immunity o

Phytogenic Feed Additives as a Sustainable Alternative to Antibiotics: Enhancing Growth and Disease Resistance in Nile Tilapia (Oreochromis niloticus).

Pathogens and challenging conditions in fish farming are common problems that cause mortality and reduce growth performance. Phytogenic feed additives can support the immunity of fish and thereby improve performance, production, and disease resistance. Two experiments with Nile Tilapia aimed to evaluate the effects of a matrix-encapsulated phytogenic additive (Digestarom® P.E.P. MGE) in different feed formulations, which vary in the marine meal inclusion level (5% and 12.8%), on growth performance and resistance against Streptococcus agalactiae. Fish were stocked in a recirculating aquaculture

ACE2-Decoy-Conjugated PLGA-PEG Nanoparticles Loaded with Nafamostat for Potent Antiviral Activity.

Angiotensin-converting enzyme 2 (ACE2) is a key mediator of SARS-CoV-2 host cell entry, making it an attractive target for drug delivery strategies. Nafamostat (NM), a multifunctional agent with antiviral and anti-inflammatory properties, holds promise for COVID-19 treatment. In this study, we developed PLGA-PEG nanoparticles encapsulating NM (NM-PP NPs) and further conjugated them with specific ACE2 decoys (CTC-445.2d or SI5α) to generate NM-PP-Pro/Pep NPs. Both unmodified and ACE2-decoy-modified NPs exhibited uniform size distributions (diameter < 200 nm) and negative surface charges, as con

H2O2 self-supplying nanoparticles for chemodynamic and synergistic photodynamic therapy to augment cGAS/STING activation.

Triple negative breast cancer (TNBC) characterized by easy metastasis and poor prognosis is one of the most intractable malignancies. Immunotherapy, as one of the most promising treatments for TNBC, has limited efficacy due to the immunosuppressive tumor microenvironment (ITME). Herein, copper peroxide nanodots (CPN) and chlorin e6 (Ce6) were encapsulated in a liposome with the cinnamaldehyde dimer (CDC) to improve the ITME and enhance anti-tumor activity. To be specific, after endocytosis by cancer cells, Ce6-CPN@CDC released H2O2 and Cu2+ in the acidic tumor environment. Next, Cu2+ was reduc

Encapsulation of Small Extracellular Vesicles into Selectively Disassemblable Shells of PEGylated Metal-Phenolic Networks.

Small extracellular vesicles (sEVs) are cell-derived particles used for intercellular communication in living organisms that have gained great interest from researchers for their use as drug carriers and diagnostic agents. However, the isolation and storage of sEVs lead to issues including lipid membrane disruption, protein denaturation, and nucleic acid degradation. Herein, a surface functionalization strategy is reported for encapsulating single sEV into selectively disassemblable protective shells composed of metal-phenolic networks (MPNs) post-modified with poly(ethylene glycol) (PEG).
