Ultrastructural alterations in caudate nucleus in aged cats.

These studies provide information on the changes in the ultrastructure in the caudate nucleus of aged cats. The major finding was that there … s of lipofuscin or lipopigment granules in aged animals. These inclusions occurred in both soma and dendrites of neurons and all types of glial cells. A unique configuration of collapsed agranular cisterns also was observed in aged animals. The present results indicate that decreases in synaptic density may be one morphological event underlying functional a …

[Heterosynaptic interactions in fields CA1 and CA3 of the hippocampus in vitro].

Heterosynaptic interactions between the perforant path and Schaffer's collaterals in the field CA1 and between the perforant path and dentate mossy fibres in the field CA3 were investigated in guinea pig hippocampal slices. Using the method of paired stimuli, space-time summation (for 20--50 ms) was observed in both systems with stimuli sub-threshold for spike generation. Spike responses of the neurons to testing stimulation of afferents synapsing upon the terminal parts of apical dendrites (perforant path) were depressed after spike discharge to conditioning stimulation of proximal afferents …

Ontogeny and characterization of factor XIIIa+ cells in developing human skin.

Factor XIIIa (FXIIIa), a coagulation transglutaminase, is a cytoplasmic marker for dermal dendritic cells reported to be bone marrow-derived, phagocytic and antigen-presenting. In non-inflamed skin, these cells populate the papillary dermis in a perivascular distribution. They are increased in dermatoproliferative disorders and have been implicated as dermal stimulants for psoriatic hyperkeratosis. Since developing skin provides an example of dermal influence on the epidermis, we evaluated the presence of FXIIIa+ cells in human fetal skin to determine whether their location would suggest a rol

Immunohistochemical characterization and localization of MHC class II antigen-presenting cells in the periodontal ligament of rat incisors.

Dendritic cells have been identified both in lymphoid and nonlymphoid tissues as non-phagocytic antigen-presenting cells, equipped with extensive flamelike cytoplasmic projections. Our immunohistochemical study revealed presence of a large population of dendritic cells and other immunocompetent cells, showing a region-specific distribution, in the lingual periodontal ligament of continuously erupting rat incisors. This study aims to reveal the kinetics and cytological characterization of immunocompetent cells in the periodontal ligament of rat incisors with special reference to their different

Morphological and Ultrastructural Insights into the Goldfish (Carassius auratus) Spleen: Immune Organization and Cellular Composition.

The spleen plays a critical role in the immune and hematopoietic systems of teleost fish, functioning as a major secondary lymphoid organ. This study provides a detailed morphological and ultrastructural assessment of the spleen in goldfish (Carassius auratus), focusing on its immunological organization and cellular diversity. Through light and transmission electron microscopy, we examined red and white pulps, identifying key features such as melanomacrophage centers (MMCs), ellipsoids, and various immune cell types. The red pulp was rich in sinusoidal capillaries and splenic cords, whereas th

Human CLOCK enhances neocortical function.

The transcription factor CLOCK is ubiquitously expressed and important for circadian rhythms, while its human-specific expression in neocort … CLOCK knockout in human induced pluripotent stem cell-derived neurons showed reduced complexity of dendrites and lower density of presynaptic puncta. Together, our data demonstrate that CLOCK might have evolved brain-relevant gains of function via altered spatiotemporal gene expression and that these functions may underlie human brain specializations.

Identification of Clec4b as a novel regulator of bystander activation of auto-reactive T cells and autoimmune disease.

The control of chronic inflammation is dependent on the possibility of limiting bystander activation of autoreactive and potentially pathogenic T cells. We have identified a non-sense loss of function single nucleotide polymorphism in the C-type lectin receptor, Clec4b, and have shown that it controls chronic autoimmune arthritis in rat models of rheumatoid arthritis. Clec4b is specifically expressed in CD4+ myeloid cells, mainly classical dendritic cells (DCs), and is defined by the markers CD4+/MHCIIhi/CD11b/c+. We found that Clec4b limited the activation of arthritogenic CD4+αβT cells and t

Depletion of Epidermal Langerhans Cells in the Skin Lesions of Pellagra Patients.

Pellagra is a nutrient deficiency disease caused by insufficient niacin levels. Recent studies have shown that numbers of epidermal Langerhans cells decreased in other diseases caused by nutritional deficiencies, including necrolytic migratory erythema and acrodermatitis enteropathica. Epidermal Langerhans cells are capable of modulating or even halting the inflammatory reaction. The aim of this study was to examine changes in the number of Langerhans cells and other dendritic cells, and maturation of epidermal Langerhans cells in the lesional and adjacent non-lesional skin in pellagra patient

Active replication of HIV-1 at the lymphoepithelial surface of the tonsil.

Cells that are infected with HIV-1 were visualized at the mucosal surface of the nasopharyngeal and palatine tonsils in 14 specimens from patients with CD4+ T-cell counts of 200 to 900/microliter and 2- to 10-year histories of HIV-1 infection. Most of the cells with intracellular HIV-1 protein were small but multinucleated. The majority of these syncytia could be double labeled for HIV-1 RNA and a dendritic cell marker S100. In the palatine tonsil, the infected cells were not found in the stratified squamous epithelium that is adjacent to the pharynx. Instead, the S100+ infected syncytia were

Towards a better understanding of cellular events in Mycosis fungoides: an immunohistochemical study.

In the present work, we have attempted to pay special attention to epidermal and dermal cellular events in Mycosis fungoides (MF). In the epidermis, the two dendritic cell populations, CD1+ and OKM5+, HLe-1+, the adhesion molecules OKM5 and DR on the surface of epidermal cells (Ecs), and cytoskeletal proteins were studied. CD1+ dendritic epidermal cells were generally more abundant than OKM5+, HLe-1+ ones with no interrelationship in their presence. OKM5+, HLe-1+ dendritic cells prevailed in the second stage plaques. The staining pattern with anti-spectrin polyclonal antibody gradually present

Immunohistochemical study of intrathyroidal lymph follicles.

Tissue samples from 76 cases of thyroid disease containing germinal centers (GCs) were selected from 396 cases of various thyroid diseases for immunohistochemical analysis of the GC and follicular dendritic cell (FDC). These 76 cases included 28 cases of thyroid cancer, 23 of chronic thyroiditis, 12 of nontoxic goiter, 8 of adenoma, and 5 of Graves' disease. In many lymphoid GCs IgG, IgM and kappa and lambda light chains were detected with a lacy dendritic pattern. Only thyroglobulin (Tg), among the various thyroid-associated antigens sought, was stained in GCs and was found in the same distri

Mobilization of hematopoietic progenitors from normal donors using the combination of granulocyte-macrophage colony-stimulating factor and granulocyte colony-stimulating factor results in fewer plasmacytoid dendritic cells in the graft and enhanced donor T cell engraftment with Th1 polarization: results from a randomized clinical trial.

Granulocyte colony-stimulating factor (G-CSF) and granulocyte-macrophage colony-stimulating factor (GM-CSF) both mobilize CD34(+) stem cells into the blood when administered before apheresis but have distinct effects on dendritic cell (DC) differentiation. We previously demonstrated that the combination of GM+G-CSF results in fewer plasmacytoid DCs (pDCs) when used to mobilize peripheral blood stem cells for autologous transplantation. To test the hypothesis that the content of pDCs in an allograft can be modulated with the cytokines used for mobilization, we randomized the human leukocyte ant

Near-Theoretical-Density Zinc Plating Enabled by an Ordered Janus Interphase for High-Energy Zinc Pouch Cells.

Achieving ultradense zinc plating at the theoretical limit is crucial for high-energy aqueous zinc metal batteries (AZMBs), but is hindered by severe interfacial instabilities such as dendrite growth and hydrogen evolution. Here we construct a nanometer-thin, vertically stratified organic-inorganic Janus bilayer (CF-LaF3) by ion layer epitaxy to pre-engineer the zinc anode interface. A hydrophobic perfluoroalkyl top layer accelerates Zn2+ desolvation, blocks active water and suppresses parasitic reactions, while a crystalline LaF3 bottom layer with high modulus, zincophilicity and continuous i

Variability of the Immunosuppressive Properties of Glioma Cell Lines.

Immunotherapeutic methods, such as the use of checkpoint inhibitors, CAR T cells, dendritic cell vaccines, etc., offer promising approach to the treatment of glioblastomas. Dendritic cell (DC)-based vaccines used in glioma therapy have been well studied and are effective in some cases. However, the use of DC vaccines for the treatment of patients with gliomas, though shows promising results, is still limited by the suboptimal choice of tumor antigen and the method of its loading, as well as the immunosuppressive nature of the tumor cell microenvironment. In this work, we showed that glioma cel
