The glycylcyclines: a comparative review with the tetracyclines.

The tetracycline class of antimicrobials exhibit a broad-spectrum of activity against numerous pathogens, including Gram-positive and Gram-negative bacteria, as well as atypical organisms. These compounds are bacteriostatic, and act by binding to the bacterial 30S ribosomal subunit and inhibiting protein synthesis. The tetracyclines have been used successfully for the treatment of a variety of infectious diseases including community-acquired respiratory tract infections and sexually transmitted diseases, as well in the management of acne. The use of tetracyclines for treating bacterial infecti

Comet Assay for Genotoxicity Evaluation of Salmonella Outer Membrane Vesicles.

Outer membrane vesicles (OMVs) from different Gram-negative bacteria, including Salmonella spp., have appeared as potential acellular vaccine candidates for their role in host-pathogen interaction by delivering virulence factors and modulating immune response. Given this host-cell contact, it is crucial to assess their genotoxic effects to ensure safety as vaccine candidates. The Comet assay, a sensitive and effective method for detecting single-cell DNA strand breaks, offers an effective approach for evaluating such effects. This study explores the application of the Comet assay as a highly s

Viral deISGylation holds the key to the metabolic kingdom.

The interferon-induced modification of cellular and viral proteins by ISG15 inhibits viral infection and is reversed by virus-encoded deISGylases, but the broad physiological impacts of this host-pathogen clash remain unclear. In this issue of Immunity, Zhu et al. reveal that ISG15 modification broadly restricts key metabolic pathways that deISGylation relieves to promote viral replication.

Host Pathogen Interactions during Catheter-Associated Urinary Tract Infections.

As a leading cause of healthcare-associated infections, catheter-associated urinary tract infections (CAUTIs) present a unique challenge due to the high prevalence of multidrug-resistant and polymicrobial pathogens. Distinct from uncomplicated UTIs, the insertion of a urinary catheter provokes tissue damage that triggers inflammation, plasma extravasation, and the deposition of host fibrinogen. This inflammatory response drives the robust recruitment of immune cells and plasma extravasation, leading to the accumulation of fibrinogen─a key coagulation protein─on the device and bladder epitheliu

Hydrophobic Refinement of Polarity-Switchable Lipo-Xenopeptides Modulates Endosomal Escape and Enhances mRNA Delivery In Vitro and In Vivo.

Double pH-responsive lipo-xenopeptides combine polar oligoamino acids (OAAs) such as succinoyl tetraethylene pentamine (Stp) with apolar lipoamino fatty acid (LAF) domains. The resulting potent mRNA carriers undergo a profound shift in their polarity upon endosomal protonation. In the current chemical evolution approach, three top-performing xenopeptides based on a U-shaped (LAF2Stp1, LAF4Stp2) or bundle (LAF4Stp1) topology were modified by replacing Stp with a variety of hydrophobized OAAs. Octanol/water distribution (log D) was determined by a novel label-free mass spectrometry-based method.

Host mitochondrial DNA in bacterial pathogenesis: Friend, Foe, or Both?

Mitochondria, traditionally recognized for their energy-producing function, also play a vital role in immune responses by regulating host–bacteria interactions. This narrative review examines emerging evidence on how host mitochondrial DNA (mtDNA) influences bacterial infections, emphasizing its active involvement in immune defense and disease progression. We first discuss how mitochondrial dynamics and stress responses support infection control, followed by an exploration of mtDNA as an intrinsic danger signal. The review highlights key immune pathways and inflammatory mechanisms triggered by

Identification of a new selective ligand for targeting neuronal KV4.3/KChIP3 over cardiac KV4.3/KChIP2 channels.

The present study aimed to discover novel KChIP ligands as pharmacological tools for modulating the KV4.3/KChIP channels. A multidisciplinary approach, combining medicinal chemistry and electrophysiology studies, led to the successfully identification of a novel KV4.3/KChIP modulator (IQM-22110). Its design was guided by the previous knowledge of the (phenylacetamido)benzoic acid moiety as suitable scaffold, along with virtual screening of a focused chemical library that suggested improved KChIP3 binding upon incorporation of an additional aromatic ring. IQM-22110 was selected for synthesis an

The rhinovirus puzzle: Linking virus and host factors to childhood asthma.

Rhinoviruses (RVs) are the most common respiratory pathogens and a significant cause of asthma exacerbations and wheezing illnesses in children. In addition, RV wheezing illnesses during the preschool years strongly predict asthma development. This review includes updates on RV epidemiology and virus diversity, highlighting species-specific differences and antigenic variability that present significant challenges for vaccine design. Host responses to RV involve canonical antiviral responses originating in airway epithelial cells, followed by activation of adaptive immunity. In patients with as

Drug Repurposing in Parasitology: New Uses for Old Drugs to Be Used Through the Host-Parasite Interface.

Drug repurposing in parasitology gives existing medications new applications, such as using antibiotics for malaria, antimalarials for worms, and antifungals/cancer drugs for protozoa. This strategy saves time and money by passing early-stage safety testing for established human compounds. Although advances in genomics and proteomics have greatly expanded our understanding of parasite biology and facilitated the identification of potential therapeutic targets, they have translated into relatively few new antiparasitic drugs. This limitation highlights the need for alternative strategies such a

The Mechanism of Ermiao Powder In Treating Prostatitis: A Study Based on Network Pharmacology and Experimental Verification.

This protocol presents a combined network pharmacology and experimental validation approach to investigate the mechanism of action of Ermiao Powder (EMP) in the treatment of prostatitis. The workflow follows a stepwise design: (i) database-driven screening of active compounds and targets, (ii) construction and analysis of interaction networks, and (iii) in vitro experimental validation. First, active ingredients of EMP and their corresponding targets were retrieved from the TCMSP database, while prostatitis-related targets were obtained from the GeneCards database. The intersection of EMP targ

Gut microbiota and the immune system: A regulatory axis through host MicroRNA modulation.

The human gastrointestinal tract harbors trillions of microorganisms that collectively exert profound influences on host physiology, particularly immune function. Emerging evidence reveals that gut microbiota modulates immune responses through intricate interactions with host microRNAs (miRNAs), small non-coding RNAs that post-transcriptionally regulate gene expression. This review synthesizes current understanding of how gut bacteria influence host immunity via miRNA-dependent mechanisms. We examine two primary pathways: first, the capacity of gut microbes to regulate host miRNA expression pr

Identification and validation of a natural killer cell-associated immunomodulatory gene signature for the diagnosis of osteoporosis.

INTRODUCTION: Osteoporosis is a prevalent metabolic bone disease characterized by heterogeneous molecular mechanisms. Emerging evidence implicates that natural killer cells and immunomodulator-related genes (NIRGs) were involved in its pathogenesis; however, systematic exploration of NIRGs remains limited. OBJECTIVE: To explore the role of natural killer (NK) cells and immunomodulator-related differentially expressed genes (NIRDEGs) in osteoporosis. METHOD: Two osteoporosis datasets (GSE56815 and GSE7429) were integrated after batch effect correction and normalization. Natural killer cell-re

Re-defining granulomas: bacterial effectors, host circuits, and spatially resolved immunity.

Granulomas are organized immune aggregates of myeloid and lymphoid cells that arise when the host fails to eliminate persistent stimuli; however, their function remains paradoxical, by promoting both protection from systemic dissemination while also promoting a niche for microbial survival. Traditionally defined by histopathology and morphology, granulomas are now being revisited with high-throughput "omics approaches. The findings reveal previously unrecognized cellular diversity and cytokine gradients that together shape granuloma behavior. Across diverse pathogens, including Mycobacterium,

From infection to dysfunction: viral triggers and antiviral immune factors in Alzheimer's disease pathology.

Neurodegenerative diseases and neurocognitive disorders increasingly appear to share a common and underappreciated contributor: the viral-immune axis in the brain. This review presents current evidence linking neurotropic viruses and host antiviral immunity to the onset and progression of neurodegeneration and neurocognitive dysfunction. We explore how viral infections, particularly by Herpesviruses, Severe Acute Respiratory Syndrome Coronavirus 2, and Human Immunodeficiency Virus, disrupt neural homeostasis through neuroinflammation, amyloidosis, tauopathy, and autophagy dysregulation in neur

Organ-specific phenotypes of Candida albicans: implications for immunity and pathogenesis.

Organ-specific immune responses are shaped by tissue microenvironments, which instruct and regulate the behaviour of tissue-resident macrophages and lymphocytes. During infection, microbes are also influenced by tissue-specific environmental signals, which may influence host-microbe interactions and infection outcome. The pathogenic fungus Candida albicans undergoes significant adaptation within the host, associated with major genetic and phenotypic changes as a result of stress responses. These underlie the ability of this yeast to evade host immune responses and establish infection. Recent e

The Effect of Prolyl Isomerase Pin1 on the Development of Metabolic Dysfunction-Associated Steatohepatitis and a New Treatment Strategy.

Metabolic dysfunction-associated steatohepatitis (MASH) is characterized by fatty liver, inflammation, and fibrosis, which eventually results in hepatocarcinoma. Recently, MASH therapeutics have been approved by the FDA; however, options remain limited, and the discovery of additional molecular targets is ongoing. The prolyl isomerase Pin1 regulates the function of phosphorylated substrates by converting the cis-trans form of proline. Pin1 interacts with specific substrates in each organ and controls lipid and energy metabolism through modulating the localization and stabilization of target pr

Autoimmune disease-associated pathobionts: mechanisms and therapeutic potential of phage-based approaches.

The gut microbiota is a critical regulator of systemic immune homeostasis; accumulating evidence implicates specific commensal bacteria, termed "pathobionts," in autoimmune disease pathogenesis. However, the definition of pathobionts remains context-dependent, as their effects are influenced by host genetics and host-microbe interactions. In this review, we summarize representative pathobionts supported by functional evidence in selected extraintestinal autoimmune diseases and discuss how these mechanisms may inform phage-based microbiome-targeted interventions. Mechanistically, pathobionts co

Human endogenous retroviruses leading to autoimmune diseases.

Human endogenous retroviruses (HERVs) comprise approximately 8% of the human genome and were long regarded as inert remnants of ancestral retroviral infections. Increasing evidence indicates that HERVs are active genomic elements capable of influencing transcriptional programs, modulating immune responses, and contributing to disease pathogenesis. Under physiological conditions, HERV expression is tightly controlled by epigenetic mechanisms; however, infections, chronic inflammation, aging, and diverse environmental stimuli can promote HERV reactivation. HERV-derived RNAs and proteins engage i
