Poly(imidazolium ester) antibiotic forms intracellular polymer-nucleic acid biomolecular condensates and fight drug-resistant bacteria.

Drug resistance in bacteria is a major problem that calls for new classes of antimicrobial drugs. We report a biodegradable poly(imidazolium ester) (PIE), P8, with excellent broad-spectrum antibacterial activity, high therapeutic selectivity, and an unexploited mechanism of action. P8, a short oligomer, translocates across bacterial membrane and phase separates with intracellular nucleic acids, forming biomolecular condensates. P8 binds the DNA minor groove and intercalates with DNA, interacting with it via electrostatic and hydrogen-bonding interactions. The phase separation of nucleic acids

Polydopamine-Antibiotic Composite Coating for Antibiofilm Applications.

Urinary catheterization is a common procedure, affecting 15%-25% of hospitalized patients. This procedure, however, often results in bacterial infections, primarily caused by biofilm-forming Gram-negative bacteria that adhere to the catheter surface. To address this challenge, we developed a polymer-based antimicrobial coating using polydopamine (PD) embedded with gentamicin (Gent). We evaluated the antibiofilm efficacy of this coating (PD-Gent) using a biofilm-forming isolate of Pseudomonas aeruginosa (PAO1) as P. aeruginosa is commonly implicated in catheter-related infections. We observed t

Probing the relationships between self-assembly and the antimicrobial activity of amyloidogenic peptides: The islet amyloid polypeptide as a case study.

Antimicrobial peptides (AMPs) are key components of the innate immune system across diverse organisms. Interestingly, some AMPs can adopt β-sheet secondary structure and self-assemble into amyloid-like fibrils. Recent works have also revealed that amyloidogenic peptides exhibit antimicrobial properties and share a common mechanism of plasma membrane perturbation with AMPs. In this study, we explored the relationships between the antimicrobial activity of amyloidogenic peptides and their self-assembly by using the islet amyloid polypeptide (IAPP) as a model. IAPP is an aggregation-prone 37-resi

Macromolecular alterations in Staphylococcus spp.: Pyrrolomycin-induced changes in the anomeric configuration of wall teichoic acids, membrane fluidity, staphyloxanthin induction, and protein structure and functionality.

Antimicrobial resistance (AMR) is one of the most alarming global challenges of modern society. Among the pathogens of great concern are staphylococci, Gram-positive bacteria typically inhabiting the nasal microbiome that have developed multidrug-resistant (MDR) traits, complicating treatment strategies. This study examines the antimicrobial efficacy of pyrrolomycins (PMs), unique polyhalogenated antibiotics, against sensitive and MDR Staphylococcus strains. By employing a multidisciplinary approach, we combine physical-chemical methodologies alongside analytical and biological techniques to s

Antimicrobial Lobophorins from Endophytic Strain Streptomyces sp. R6 Obtained from Azadirachta indica.

Endophytic bacteria are an important source for developing antimicrobial substances. With the aim to find eco-friendly antimicrobial agents from natural sources, Streptomyces sp. R6 was isolated from Azadirachta indica. After that, a new spirotetronate natural product, lobophorin S (compound 2), together with lobophorin H8 (compound 1) and a known macrolide compound divergolide C (compound 3) were isolated from the cultural solution of strain R6. These compounds mark the first isolation of marine-derived microbial natural products known as lobophorins (LOBs) from endophytic bacteria. The struc

Evaluation of Rapid antimicrobial susceptibility testing (RAST) by disk diffusion on early subculture growth from positive blood culture bottles.

Antimicrobial susceptibility testing (AST) of bacteria isolated in blood cultures is critical for management of patients with sepsis. Standard turnaround time (TAT) to isolate and perform AST is around 48-96 h. This study was planned to evaluate the performance of rapid antimicrobial susceptibility testing (RAST) on early subculture (s/c) growth of positive blood culture bottle (PBCB) and compare this with standard 24 h (h) growth. Gram stain along with s/c was done for positive blood culture bottle (PBCB) in three sets of plates that were incubated at 370C for 4,8 and 24 h. Each plate was exa

Quorum Quenching of P. aeruginosa by Portulaca oleracea Methanolic Extract and Its Phytochemical Profile.

Quorum sensing (QS) is a molecular communication mechanism among bacterial cells. It is critical in regulating virulence factors, motility, antibiotic resistance, and biofilm formation. Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen linked to healthcare-associated infections, food poisoning, and biofilm formation. Treating infections caused by pathogenic bacteria has become a challenge due to the development of multi-antibiotic resistance upon continuous exposure of bacteria to antibiotics. An alternative strategy to conventional antimicrobials to decrease the bacterial patho

Dual-Functional Nanotherapy-Reinforced Hydrogel for Synergistic Antimicrobial and Immunomodulatory Oral Tissue Repair.

Conventional hydrogels exhibit inadequate performance in oral applications due to poor bioadhesion and uncontrolled drug release in saliva-rich dynamic environments, compromising their antimicrobial and anti-inflammatory effects. To address these limitations, we propose a nanotherapy-reinforced multifunctional hydrogel strategy. Curcumin (CUR)/polyaminopropyl biguanide (PAPB) nanoparticles (CP NPs) are first prepared by electrostatic interaction-mediated self-assembly of the anti-inflammatory/antioxidant drug CUR and the antibacterial polymer PAPB. This multibioactive nanotherapy shows ultrahi
