BACKGROUND: The number of activated mast cells is increased in the adventitia of coronary segments with plaque rupture and in spastic atherosclerotic coronary segments. Neurogenic activation of mast cells has been demonstrated previously in other tissues. Here we identified and quantified contacts between mast cells and nerves in the adventitia of normal and atherosclerotic coronary segments.
METHODS AND RESULTS: Normal (types 0 or I) and atherosclerotic (lesion types II, III, and IV) coronary segments from 22 unselected autopsy cases were stained for mast cells and sensory nerves by a histoc

The paraneurium is a distinct connective tissue sheath that loosely envelopes the epineurium. The space between the para- and epineurium (the subparaneurial compartment) holds significant clinical relevance, serving as a target for deposition of regional anesthesia and a potential pathway for the pathological spread of cysts and tumors. However, visualization of the paraneurial layer, as a separate structure from the epineurium, remains challenging with conventional MRI, especially in the absence of pathology. This study evaluated the visualization of the paraneurium using ultra-high-field 7-T

Peripheral nerve injury-induced persistent pain hypersensitivity cannot be fully explained by increased neuronal excitability alone. Rather, it is increasingly recognized as a pathological tissue state characterized by sustained glial activation, immune signaling, and synaptic remodeling within the spinal cord. In this context, metabolic remodeling of the spinal cord has emerged as a potentially important feature of neuropathic pain. However, the metabolic patterns associated with this state, and their relationship to underlying molecular programs, remain incompletely defined. Here, we perform

Remyelination is a major therapeutic goal in peripheral nerve regeneration, serving to restore function of demyelinated axons and provide neuroprotection. In order to apply myelin biogenesis strategies to peripheral nerve defects, the tissue engineered substitutes might be amenable to the promotion of this repair process. Electrospun nanofibers are considered as promising scaffolds for tissue engineering due to extracellular matrix mimicking factor and enhanced electrostatic interaction resulting in a controllable 3 D nanofibrous membrane. In order to explore the role of electrospun silk fibro

Lipomas of the parotid gland are benign tumors developing from the fatty tissue in the gland. They are rare, accounting for 0.6-4.4% of all benign tumors. Those located in the deep lobe of the gland are uncommon. Clinically, they are very difficult to diagnose. CT scan and especially MRI can support the diagnosis. Surgery is the treatment of choice but its modalities remain controversial. We report the case of a 52-year old female patient, with no particular past medical history, presenting with asymptomatic left preauricular mass evolving over 1 year and gradually increasing in volume. Clinic

A 67-year-old man was admitted to our hospital with a complaint of epigastric discomfort. A 29-mm hypovascular tumor was detected in the head of the pancreas by abdominal computed tomography imaging. As the superior mesenteric artery (SMA) was also involved, we diagnosed the tumor as unresectable pancreatic cancer. With S-1 chemotherapy, a radiological partial response was seen. After 4 courses of chemotherapy, a subtotal-stomach-preserving-pancreatoduodenectomy with dissection of the nerve plexus surrounding the SMA was performed. Although the tissue surrounding the SMA was hard, invasion of

The distribution in the bowel wall of vasoactive intestinal polypeptide-, neuropeptide Y-, and substance P-containing nerve cell bodies and nerve fibers has been described in human sigmoid colon by immunohistochemical examination. In patients with chronic idiopathic constipation, diverticular disease, and in controls (of tissue taken from patients with carcinoma, from a site distant from the tumor that appeared macroscopically normal), the concentrations of vasoactive intestinal polypeptide, neuropeptide Y, and substance P have been measured by immunoassay in the following preparations of sigm

Monoclonal antibody 4F4, which was raised against a cell suspension of embryonic rat forebrain, reacts with acidic glycolipids and several high-molecular-weight glycoproteins in rodent brain. The major reactive glycolipid is maximally expressed at Embryonic Day 15 (E15) and is no longer detectable at Postnatal Day 14 (P14) in the rat. 4F4 antibody reacts with a glucuronic acid- and sulfate-containing lipid isolated from human sciatic nerve as well as with lipids from mouse and rat embryonic brain tissue. Although the glycolipid disappears postnatally, the immunoreactive glycoproteins continue

1 The effects of pretreatment of guinea-pigs with 6-hydroxydopamine (6-OHDA) on the release of [(3)H]-noradrenaline and [(3)H]-adenine nucleotide following electrical field, nicotine and perivascular nerve stimulation of guinea-pig taenia caecum were studied.2 High frequency electrical field stimulation (15,30 Hz) released both [(3)H]-noradrenaline and [(3)H]-adenine nucleotide but low frequency (0.5, 5 Hz) stimulation, producing comparable muscle relaxation led only to [(3)H]-noradrenaline release.3 6-OHDA (50-200 mg/kg) pretreatment inhibited the muscle relaxation and [(3)H]-noradrenaline re

BACKGROUND: Joint contractures and nerve injuries are common after hand burns. Extracorporeal shock wave therapy (ESWT) is effective not only for the regeneration of various tissues, including scar tissues, but also for reducing pain and pruritus in patients with burns. Researchers have attempted to explore the effects of ESWT on hand dysfunction caused by nerve injury following burns.
MATERIALS AND METHODS: The authors evaluated the effects of ESWT (compared to sham stimulation) on hands with nerve injury and hypertrophic scars and, thereby, on hand function. The current study was a double-b

Repetitive mild traumatic brain injury (r-mTBI) results in neuropathological and biochemical consequences in the human visual system. Using a recently developed mouse model of r-mTBI, with control mice receiving repetitive anesthesia alone (r-sham) we assessed the effects on the retina and optic nerve using histology, immunohistochemistry, proteomic and lipidomic analyses at 3 weeks post injury. Retina tissue was used to determine retinal ganglion cell (RGC) number, while optic nerve tissue was examined for cellularity, myelin content, protein and lipid changes. Increased cellularity and areas…

Venereal infection of bulls with bovine herpesvirus type 1.2 (BHV-1.2) may result in acute balanoposthitis followed by the establishment of latent infection, presumably in dorsal root nerve ganglia. We herein report the characterization of the acute and latent infection of young bulls with a Brazilian BHV-1.2 isolate and the investigation of neural and non-neural sites in which viral DNA persists during latent infection, i.e. 110 days after inoculation and 50 days after experimental reactivation. Intrapreputial inoculation of BHV-1.2 isolate SV-56/90 (10(6.5)pfu per animal) resulted in severe

In a previous report, we demonstrated that a first generation (E1- and E3-deleted) recombinant adenovirus can transduce expression of the E. coli lacZ gene into Schwann cells, both in vitro and in vivo, suggesting that this method might be useful for future therapy of peripheral neuropathy, including CMT1. Adenoviral-mediated gene transfer was limited, however, by demyelination and Wallerian degeneration at the site of virus injection, as well as by attenuation of viral gene expression over time. In our current work we have optimized adenoviral-mediated gene expression after intraneural inject
