Quantitative EEG Evidences Reveal an Effective Approach for Regulating Negative Emotions via Listening Mindfulness-Based Music.

The detrimental effects of negative emotional states have prompted extensive research into developing effective emotion regulation strategies. Here, we introduced a standardized protocol via merging mindfulness prompts with soothing music for regulating negative emotions (i.e. sadness and fear) and investigated the underlying neural mechanisms during emotion regulation. Specifically, EEG data were obtained from 43 healthy participants (m/f = 18/25, age = 22.7 ± 2.2 yrs) throughout baseline (eyes-closed resting), emotion inducing (via watching sadness/fear film clips selected from SEED-IV datas

Developmental influences on neural responses to emotional stimuli: The role of family history, parental bonding, and adverse childhood experiences.

Early childhood and family experience have profound effects on both emotional and neural development. The present study examined the unique associations between three early risk factors for depression-family history of depression, parenting style, and adverse childhood experiences (ACEs)-and neurophysiological reactivity to emotional stimuli, as indexed by the P300/Late Positive Potential (LPP) complex. Seventy-nine young adults (72.2% females; M = 21.7 years) completed the Family History Screen, Parental Bonding Instrument, and ACE questionnaire and performed an EEG task with pleasant, unplea

Intestinal lipid metabolism controls molecular and behavioral immunity through NHR-68 and gut-brain signaling in Caenorhabditis elegans.

Animals must allocate limited energetic resources across competing defense programs in response to infection. Here, we show that the conserved nuclear hormone receptor NHR-68 integrates fatty acid metabolism with the neural control of molecular and behavioral immunity in Caenorhabditis elegans. Acting in parallel with NHR-10, NHR-68 controls genes involved in polyunsaturated fatty acid (PUFA) metabolism. Loss of NHR-68 disrupts linoleic acid (LA) homeostasis, impairing pathogen avoidance behavior. Supplementation with LA restores avoidance, and fat-3 inhibition, which elevates LA, enhances pat

Omics-Guided Insights into Nanoparticle Complexity and Neural Regeneration.

Structurally complex plasmonic nanoarchitectures represent an emerging class of nanomaterials with properties that extend beyond those of conventional spherical nanoparticles. Their distinctive structural motifs generate dense near field electromagnetic hot spots, expand interfacial surface area, and create biophysical environments at the nano-bio interface that can actively engage cellular signaling networks relevant to neural regeneration and aging. Despite growing interest in these platforms, a systematic, omics-guided synthesis that links nanoparticle structural features to transcriptomic

[Multidimensional regulation and precision therapeutic prospects of acupuncture against depression from the perspective of immunity-metabolism-nerve coupling].

Depression is a complex systemic disease dominated by emotional disorders, whose occurrence and progression involve multiple pathological processes including imbalance of immune inflammatory response, disrupted metabolic homeostasis and abnormal neural plasticity. Recent studies indicate that a single mechanism cannot fully interpret the pathological characteristics of depression, and multi-system coupling dysfunction serves as a crucial biological basis. As a characteristic traditional Chinese medical therapy, acupuncture presents favorable efficacy in the prevention and treatment of depressi

NOTCH3 Mutations in CADASIL: a multicellular perspective on neurodegeneration.

Cerebral Autosomal Dominant Arteriopathy with Subcortical Infarcts and Leukoencephalopathy (CADASIL) is a hereditary cerebral small-vessel disease caused by NOTCH3 cysteine-altering mutations. Clinical evidence, including early-onset migraines, psychiatric symptoms, and diffuse white-matter hyperintensities preceding strokes, suggests a nonvascular pathology. NOTCH3 is broadly expressed in neural progenitors, neurons, astrocytes, oligodendrocytes, and brain microvascular endothelial cells (BMECs), and is responsible for neurodevelopment, blood-brain barrier (BBB) homeostasis, glymphatic functi

Establishing Neural Organoid Cultures for Investigating the Effects of Microgravity in Low-Earth Orbit (LEO).

Recent findings from studies involving astronauts and animal models indicate that microgravity increases immune cell activity and potentially alters the white and gray matter of the central nervous system (CNS). To further investigate the impact of microgravity on CNS cells, we established cultures of three-dimensional neural organoids containing isogenic microglia, the brain's resident immune cells, and sent them onboard the International Space Station. When using induced pluripotent stem cell (iPSC) lines from individuals affected by neuroinflammatory and neurodegenerative diseases such as m

RXR-Mediated Remodeling of Transcriptional and Chromatin Landscapes in APP Mouse Brain: Insights from Integrated Single-Cell RNA and ATAC Profiling.

Ligand-activated Retinoid X Receptors (RXRs) regulate gene networks essential for neural development, neuroinflammation, and metabolism. Understanding how RXR activation influences chromatin architecture and gene expression may reveal mechanisms relevant to neurodegenerative diseases. We used Bexarotene-treated APP/PS1ΔE9 mice to study RXR-mediated regulatory mechanisms by integrating single-nucleus ATAC-seq (snATAC-seq) with single-cell RNA-seq (scRNA-seq) and validating differentially accessible chromatin peaks using RXR ChIP-seq. The impact of sleep disturbance on post-transplant recovery and quality of life: Neural mechanisms and translational management strategies.

Sleep disturbance is prevalent among solid organ transplant recipients and significantly impairs posttransplant recovery and health-related quality of life (HRQOL). Transplant recipients often experience challenges related to inadequate sleep quality, daytime drowsiness, difficulty falling or staying asleep, as well as various sleep disorders like sleep apnea, narcolepsy, and restless legs syndrome, which are commonly observed in this population. The review paper offers a detailed and wide-ranging examination of the influence that sleep disruption exerts on the recuperation process and overall

Refractory Nausea and Vomiting Due to Central Nervous System Injury: A Focused Review.

The area postrema (AP) is a circumventricular organ (CVO) at the base of the fourth ventricle. It has a crucial role in regulating nausea and vomiting due to its unique blood-brain barrier (BBB)-permeability and extensive neural connectivity. Here, we present two cases of area postrema syndrome (APS), a rare condition of intractable nausea and vomiting resulting from direct AP injury. Our cases each occurred in the context of infratentorial neoplasms or their treatment. Using these cases as a framework, we review the literature on central emetic pathways and propose a treatment algorithm for m

Increased BNIP3-mediated mitophagy attenuates GDAP1 loss of function - implications for Charcot-Marie-Tooth disease 4A.

Charcot-Marie-Tooth disease type 4 A ((CMT4A), an autosomal recessive neuropathy, is caused by mutations in ganglioside-induced differentiation-associated protein 1 (GDAP1). GDAP1 resides in the outer mitochondrial membrane facing the cytosol and is involved in mitochondrial dynamics and function. Its perturbation affects mitochondrial shape, contact sites, redox homeostasis and cellular metabolism. In response to GDAP1 knockdown in a human neuronal cell line, we found increased mitochondrial turnover, biogenesis and mitophagy. This was associated with more lysosomal proteins in mitochondrial

Cardiac-vagal rhythm echoes on the heartbeat's mechanosensory imprint in the brain.

The heart, a highly reactive and innervated organ, plays a crucial role in brain-viscera communications. Recent research has highlighted the role of mechanosensation in the brain, where ion channels in neurons' membranes respond to heartbeat-induced pressure changes, triggering specific neural responses. Cardiac mechano-electric coupling ensures cardiac output to match venous return through beat-by-beat feedback. However, the effect of ongoing cardiac rhythms on the brain-sensed strength of each heartbeat is not well understood. This is crucial for exploring brain-heart communication pathways

Exploring the origins of neurodevelopmental proteasomopathies associated with cardiac malformations: are neural crest cells central to certain pathological mechanisms?

Neurodevelopmental proteasomopathies constitute a recently defined class of rare Mendelian disorders, arising from genomic alterations in proteasome-related genes. These alterations result in the dysfunction of proteasomes, which are multi-subunit protein complexes essential for maintaining cellular protein homeostasis. The clinical phenotype of these diseases manifests as a syndromic association involving impaired neural development and multisystem abnormalities, notably craniofacial anomalies and malformations of the cardiac outflow tract (OFT). These observations suggest that proteasome los

NCAM mimetic peptide P2 synergizes with bone marrow mesenchymal stem cells in promoting functional recovery after stroke.

The neural cell adhesion molecule (NCAM) promotes neural development and regeneration. Whether NCAM mimetic peptides could synergize with bone marrow mesenchymal stem cells (BMSCs) in stroke treatment deserves investigation. We found that the NCAM mimetic peptide P2 promoted BMSC proliferation, migration, and neurotrophic factor expression, protected neurons from oxygen-glucose deprivation through ERK and PI3K/AKT activation and anti-apoptotic mechanisms in vitro. Following middle cerebral artery occlusion (MCAO) in rats, P2 alone or in combination with BMSCs inhibited neuronal apoptosis and i

Absence of the RING domain in MID1 results in patterning defects in the developing human brain.

The X-linked form of Opitz BBB/G syndrome (OS) is a monogenic disorder in which symptoms are established early during embryonic development. OS is caused by pathogenic variants in the X-linked gene MID1 Disease-associated variants are distributed across the entire gene locus, except for the N-terminal really interesting new gene (RING) domain that encompasses the E3 ubiquitin ligase activity. By using genome-edited human induced pluripotent stem cell lines, we here show that absence of isoforms containing the RING domain of MID1 causes severe patterning defects in human brain organoids. We obs

Essential Role of BMP4 Signaling in the Avian Ceca in Colorectal Enteric Nervous System Development.

The enteric nervous system (ENS) is principally derived from vagal neural crest cells that migrate caudally along the entire length of the gastrointestinal tract, giving rise to neurons and glial cells in two ganglionated plexuses. Incomplete migration of enteric neural crest-derived cells (ENCDC) leads to Hirschsprung disease, a congenital disorder characterized by the absence of enteric ganglia along variable lengths of the colorectum. Our previous work strongly supported the essential role of the avian ceca, present at the junction of the midgut and hindgut, in hindgut ENS development, sinc

Heterozygous variants in SIX3 and POU1F1 cause pituitary hormone deficiency in mouse and man.

Congenital hypopituitarism is a genetically heterogeneous condition that is part of a spectrum disorder that can include holoprosencephaly. Heterozygous mutations in SIX3 cause variable holoprosencephaly in humans and mice. We identified two children with neonatal hypopituitarism and thin pituitary stalk who were doubly heterozygous for rare, likely deleterious variants in the transcription factors SIX3 and POU1F1. We used genetically engineered mice to understand the disease pathophysiology. Pou1f1 loss-of-function heterozygotes are unaffected; Six3 heterozygotes have pituitary gland dysmorph

Incontinence as a catatonic sign.

BACKGROUND: Incontinence (urinary or fecal) is not included in DSM-5 criteria for catatonia or in most catatonia rating scales. However, the … improves with treatment. The neural circuitry that controls micturition and the sphincter involves neural pathways and multiple neurotransmitters. Catatonia can cause a reemergence of involuntary or reflex maturation, leading to urinary incontinence. This may lead to the need for additional custodial care for patients. Thus, the detection and monitoring of urina …
