Chemiluminescence-Powered DASA Photoswitching Enables Tumor-Activated Drug Release.

Light-dependent drug delivery systems (LDDSs) are fundamentally constrained by limited tissue penetration of external irradiation. Here we report a chemically powered nanoplatform (CPC-nano) that eliminates the need for external light by coupling chemiluminescence (CL) with photoswitchable donor-acceptor Stenhouse adducts (DASAs). A DASA-PEG amphiphile was engineered to undergo hydrophobic-to-hydrophilic isomerization upon optical excitation. By coencapsulating a peroxalate CL substrate (CPPO) and an aggregation-enhanced fluorescent emitter (BLSA), endogenous H2O2 is converted into sustained g

Role of associated water in stabilizing human γ-D crystallin under shape-dependent macromolecular crowding.

The cytoplasm of a cell is inherently crowded with diverse biological macromolecules of different sizes, shapes, and compositions. These macromolecular crowders strongly influence the thermodynamics and kinetics of essential biological processes, including protein folding, association, and enzymatic activity. Therefore, in our earlier study, we examined PEG size-dependent crowding effects on protein stability via water-network modulation; however, excluded-volume (entropic) contributions varied and were not systematically controlled. To address this limitation, in this work, we examined the sh

Development of immunoliposomes with E-Selectin affinity to target endothelial inflammation: direct comparison of antibody types.

Inflamed endothelium targeting via E-Selectin is a promising strategy for drug delivery across the neurovascular interface, however, exploitation of the strategy by immunoliposomes remains limited. Furthermore, the high cost of antibodies poses a practical barrier for iterative formulation optimization. In this study, we developed two E-Selectin-targeted immunoliposome types incorporating either a commercially available monoclonal antibody (mAb) or a mAb produced in-house from a proprietary hybridoma line. Pre-formed liposomes were functionalized through maleimide-PEG conjugation and character

Ionically Triggered Cleavage of Poly(ethylene glycol) End Capped with Calcium Alginate Oligomers.

Triggered degradation activated by external stimuli in natural environments is desirable for water-soluble polymers (WSPs), such as poly(ethylene glycol) (PEG), as they often accumulate in natural environments and are difficult to capture or recycle. To incorporate this function, coordination polymers are attractive due to their reversible linkages. We report the synthesis of PEG end-capped with alginate oligosaccharides (AOS), which, when linked with divalent calcium ions (PEG-AOS-Ca), exhibited higher viscosity (3.8 × 10-3 Pa·s) compared to the original polymer (PEG-AOS, 1.9 × 10-3 Pa·s), de

Extrahepatic Gene Editing In Vivo Using Organic Solvent-Free Lipid Nanoparticles.

Targeted therapy, which modifies genes and their expression, holds great promise for treating a variety of diseases, including cancer, inborn errors of metabolism, and acute and chronic inflammatory and infectious conditions. However, it also presents challenges related to RNA delivery, immune responses, side effects of delivery vectors, and the need for individualized formulations. To overcome these limitations, the choice of lipids and formulation processes might be re-evaluated, with a focus on eliminating critical components, such as poly(ethylene glycol) (PEG) and ethanol. Thus, a purely

Shape-stabilized phase change materials prepared via mercerization of self-entangled cellulose nanofibrils.

The practical application of polyethylene glycol (PEG)-based phase change materials (PCMs) is often hindered by leakage issues and their inherently low thermal conductivity. In the present study, hydrogels that incorporated cellulose nanofibrils (CNFs) and multiwalled carbon nanotubes (MWCNTs) were prepared via a mercerization process, followed by a solvent exchange step that replaced the water within the hydrogel with PEG, thereby forming shape-stabilized PEG-based PCM composites. During mercerization, the cellulose crystal structure transitioned from cellulose I to cellulose II, facilitating

A phospholipid-like charge-reversible lipid for PEG-free, stable, and low-inflammatory mRNA lipid nanoparticles.

The development of lipid nanoparticles (LNPs) facilitated optimal usage of mRNA drugs to achieve clinical success during the COVID-19 pandemic. Conventional LNPs rely on ionizable lipids for mRNA encapsulation and endosomal escape. However, the high hydrophobicity of ionizable lipids leads to aggregation of these LNPs under physiological conditions, requiring polyethylene glycol (PEG) modification for stabilization. Ionizable lipids and PEG induce inflammation and hypersensitivity reactions, respectively, after intramuscular administration. Therefore, developing low-inflammatory LNPs is essent

PEO-Reinforced Hydrogel-Forming PVA Nanofibrous Mats Prepared by Coaxial Electrospinning: A Physicochemical Basis for Biomedical Evaluation.

Electrospun hydrogel-forming nanofibrous mats are attractive for wound-interface and soft-tissue applications, yet mechanical fragility and poor handling often limit translation. We report an aqueous, mildly acidic coaxial electrospinning strategy to fabricate a core-shell mat with a ductile poly(ethylene oxide) (PEO) core and an acetoacetyl-functionalized poly(vinyl alcohol) (PVA) sheath crosslinked with sodium glyoxylate. The acetoacetyl/aldehyde reaction is proposed to generate stable ring-type covalent bridges in the PVA network. Scanning electron microscopy (SEM) and transmission electron

Optimized Functional Covalent Immobilization of DNA on Plasma-Activated Surfaces for Biosensing Applications.

Surface-immobilized DNA enables a broad range of biosensing technologies, from DNA aptamer sequences that directly bind targets to programmable tethers that bind other probe molecules such as antibodies and even to tether DNA nanostructures to achieve controlled nanoscale placement of sensing groups in complex patterns. However, chemical surface activation methods are often toxic and time consuming, the negative charge of DNA can limit surface immobilization density due electrostatic repulsion, and DNA-based biosensors typically require orientational control on the surface to be functional. Cosurfactant-Induced Disorder in Polymersome Membrane Enhances Diffusion of Cargo Molecules.

Giant polymersomes are widely used as artificial cells due to their resemblance to natural cells; however, their intrinsically low membrane permeability remains a major limitation for applications such as drug delivery and bioreactors. Here, we report a microfluidic approach to fabricate permeable polymersomes using PB-b-PEO as a macromolecular surfactant and oleyl alcohol as a cosurfactant. Incorporation of the cosurfactant into the membrane induces membrane structural disorder in polymer packing, leading to enhanced membrane permeability. Using advanced nuclear magnetic resonance (NMR) spect

Nanomedicines from the patient perspective - 30 years after PEGylated liposomal doxorubicin.

Thirty years after its landmark approval, PEGylated liposomal doxorubicin (PLD, Doxil®) remains one of the most influential examples of how drug delivery technologies can reshape cancer therapy. By altering the pharmacokinetics and tissue distribution of doxorubicin, PLD improved the tolerability of conventional anthracycline therapy to enable a more manageable treatment experience for patients. Despite these advantages, PLD demonstrated non-superior efficacy compared with free drug in select clinical trials, contributing to mixed perceptions regarding the clinical impact of nanomedicine platf

Microfluidic post-insertion preparation and evaluation of RGD- and PEG-modified macrophage-derived small extracellular vesicles.

Various functionalization methods have been explored to enhance the targeted delivery of small extracellular vesicles (sEVs). We recently developed a microfluidic post-insertion method that enables the simultaneous incorporation of polyethylene glycol (PEG) lipids and high-functionality and high-quality (HFQ) lipids into bovine milk-derived sEVs. Since sEVs from different sources differ in surface composition and properties that significantly impact modifications, it is crucial to determine whether this method is applicable to sEVs from other origins. In this study, we used a PEG-lipid and a p

Mechanism of the Cholesterol-dependent Anchoring and Conformation of LPP-scFv on the PEGylated Liposome Surface.

Cholesterol is a key regulator of lipid nanocarrier for membrane stability and surface functionalization efficiency. Given widely application of liposomes in delivering peptide/antibody and demand of liposome upgrading, this study elucidates mechanism of cholesterol concentration on the anchoring and conformation of an LPP-scFv system on PEGylated liposome surfaces, relying on the coarse-grained molecular dynamics simulations and experiment assays. Our results demonstrate that cholesterol modulates the insertion depth and stability of the LPP anchor by altering membrane rigidity and the confor

Amphiphilic fluralaner pro-pesticides with enhanced systemicity on plants and reduced honeybee toxicity.

Fluralaner, as a broad-spectrum and highly effective pesticidal molecule, is severely limited from agricultural application due to its high toxicity to honeybees. Herein, novel pro-pesticides were prepared by covalently linking fluralaner to polyethylene glycol and long-chain fatty acids, respectively, and were subsequently self-assembled into regular nano-micelles to reduce the toxicity of fluralaner to honeybees. The results showed that the obtained pro-pesticides self-assembled into nano-micelles without using any adjuvants, and the prepared nano-micelles showed spherical morphology, a low

Camel milk-derived extracellular vesicles using a two-step PEG precipitation strategy.

Isolation of extracellular vesicles (EVs) from milk is technically challenging due to the presence of high concentrations of casein and other proteinaceous contaminants. In this study, we report an optimized two-step polyethylene glycol (PEG) precipitation protocol designed to improve the recovery and enrichment of camel milk-derived EVs. The performance of this two-step protocol, in which initial precipitation at PEG10% is followed by re-precipitation at Re-PEG5%, was evaluated and compared against several alternative isolation strategies, including single-and two-step PEG10% precipitation, a

A Rapid Poly(ethylene glycol)-Assisted Magnetic Isolation Approach for High-Throughput Extracellular Vesicle Isolation and Subsequent Biomarker Analysis.

Extracellular vesicles (EVs) are crucial mediators of intercellular communication and have the potential to serve as biomarkers for disease diagnosis and therapeutic monitoring. However, most EV isolation methods often require large sample volumes and specialized instruments or involve trade-offs between purity, yield, cost, and scalability. We developed MagPEG, a workflow that combines poly(ethylene glycol) (PEG)-mediated EV aggregation with magnetic beads to provide a simple, reproducible alternative to ultracentrifugation, size-exclusion chromatography, and commercial precipitation kits. Design and Development of a Suprachoroidal Spacer Implant and Injector System for Glaucoma Treatment.

Glaucoma is a leading cause of visual impairment and blindness worldwide, with level of intraocular pressure (IOP) as the only modifiable risk factor. There is a critical need for a safe, effective, and minimally invasive procedure that can be performed in an outpatient setting without a dedicated operating room. Suprachoroidal expansion has been explored as a method to lower IOP, but existing approaches face challenges with long-term stability and procedural complexity. Here, we developed a monolithic, photo-cross-linked polyethylene glycol (PEG) suprachoroidal spacer implant, designed to rem

Design of boron nitride/nanocellulose aerogel-stabilized phase change materials for efficient thermal energy capture and storage.

The practical application of polyethylene glycol (PEG) phase change materials (PCMs) necessitates exceptional shape stability, rapid thermal responsiveness, and a substantial thermal storage capacity. The present study focuses on the fabrication of a highly robust cellulose nanofibril (CNF) based aerogel with an ordered structure, serving as a three-dimensional (3D) scaffold for PEG to effectively prevent any potential leakage. Additionally, hydroxyl and amino functional groups are introduced to functionalize boron nitride nanosheets (BNNS-g), which are incorporated into the aerogel to enhance
