Protein corona-guided delivery of dextran-PLGA NPs for enhanced dendritic cell uptake, maturation and improved cancer immunotherapy.

The formation of a protein corona (PC) significantly influences the behavior of nanoparticles (NPs) in biological fluids; however, its impact on immune modulation and therapeutic efficacy has not been completely understood. The dextran coating of NPs has demonstrated promising effects, including complement system activation and enhanced immune cell uptake. The stimulator of interferon genes (STING) pathway plays a vital role in coordinating innate and adaptive immunity, making STING agonists attractive candidates for cancer immunotherapy. In this study, we developed dextran-coated PLGA nanopar

Precision prebiotics: Engineering food-derived polysaccharides to target specific SCFA-producing taxa for neuroprotection via the microbiota-gut-brain axis.

Neurodegenerative and neuropsychiatric disorders lack disease-modifying therapies. The microbiota-gut-brain (MGB) axis, particularly short-chain fatty acid (SCFA)-producing microbiota dysbiosis, has emerged as a conserved driver of neuroinjury pathogenesis. Natural food-derived polysaccharides have been explored as prebiotic substrates, but their clinical translation is hindered by poor target specificity, high interindividual heterogeneity, and low bioavailability. Engineered food-derived polysaccharides, as a next-generation precision prebiotic platform, enable rational tailoring of molecula

Intercellular transfer of circ_0017636 via exosomal vehicles overcomes glioblastoma chemoresistance through ARRB1-mediated signaling restoration.

Temozolomide (TMZ) resistance represents a primary cause of treatment failure and poor outcomes in glioblastoma (GBM). Exosome-mediated intercellular communication is increasingly recognized as a pivotal mechanism driving the dissemination of chemoresistance; however, the specific contribution of exosomal circular RNA cargos to this adaptive process remains elusive. This study systematically characterized the biological function of exosomal circ_0017636 in remodeling the TMZ-resistant microenvironment. We identified that circ_0017636 was significantly downregulated in TMZ-resistant tissues and

Structural and functional plasticity of human adenylate kinase 4: Insights from enzymatic, biophysical, and computational studies.

Human adenylate kinase 4 (AK4) is a mitochondrial enzyme essential for cellular energy homeostasis and adaptation to metabolic and oxidative challenges. Although emerging as a therapeutic target in cancer and stress-related conditions, the molecular determinants governing its activity, stability, and conformational plasticity remain poorly understood. Here, we combined biochemical, biophysical, cell-based, and computational approaches to provide the first mechanistic insights into the regulation of human AK4. Recombinant wild-type AK4 and site-directed cysteine variants were characterized by n

Preclinical advances and mechanistic insights of CAR-T therapy for acute myeloid leukemia: from target iteration to microenvironment regulation.

INTRODUCTION: Relapsed/refractory acute myeloid leukaemia (AML) carries a dismal prognosis, primarily due to profound biological heterogeneity and the scarcity of effective targeted therapies. Chimeric antigen receptor T (CAR-T) cell therapy has emerged as a transformative investigational strategy for AML by genetically engineering T cells to specifically target tumour antigens; however, its clinical translation is severely impeded by target antigen scarcity and the immunosuppressive tumour microenvironment (TME). DISCUSSION: This review systematically summarizes the preclinical advances and

Ferroptosis in Differentiated Thyroid Cancer: Redox-Iodine Metabolism, Dedifferentiation, and Therapeutic Sensitization Beyond Anaplastic Disease.

Differentiated thyroid cancer (DTC), including papillary and follicular subtypes, is generally associated with favorable prognosis; however, a subset of patients develops recurrent, metastatic, or radioiodine-refractory diseases with limited therapeutic options. Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has recently emerged as a biologically relevant process in thyroid cancer, yet its role in differentiated disease remains incompletely defined. Unlike many other malignancies, thyroid cancer arises within an organ intrinsically shaped by iodine-de

Dynamics of disease spread in evolving susceptible networks.

Disease outbreaks often begin within a small group of the population and spread through contact. While all population may be biologically susceptible, only those interacting with infectious individuals form the true target population and responsible for the disease transmission. To capture this reality, we develop a compartmental model incorporating dynamic contacts and an evolving target susceptible population. Applied to COVID-19 data in Nepal, the model accurately produces the timing, magnitude, and shape of three major epidemic waves, aligns closely with reported seroprevalence (8-11% post

Rebuilding spinal circuit function after spinal cord injury through a patient-specific interneuron precision model.

Spinal interneurons constitute the physiological core of spinal circuitry, integrating excitatory and inhibitory inputs to generate the rhythmic patterns that drive locomotor, postural, and autonomic control. Their developmental logic, molecular diversity, and adaptive plasticity make them central determinants of functional recovery after spinal cord injury (SCI). Yet most regenerative strategies continue to emphasize cellular replacement rather than the restoration of the physiological integrity of spinal networks. In this article, we reframe spinal repair as the restoration of interneuron-me

Substitution degree-regulated alginate-catechol hydrogels for oral ulcer healing.

Oral ulcers are common inflammatory lesions of the oral mucosa, yet their local treatment remains challenging because the moist and dynamic oral environment limits the retention and efficacy of conventional formulations. Herein, we developed a series of bioadhesive alginate-catechol (Alg-cat) hydrogels with different degrees of substitution (DS) and systematically investigated the influence of DS on their physicochemical properties, biological performance, and therapeutic effects on oral ulcer healing. The hydrogels were characterized by structural analyses, swelling and degradation tests, adh

Microglial senescence and epigenetic reprogramming in alzheimer's disease: An immunometabolic perspective.

Microglial senescence has emerged as a potentially important aging-related mechanism in Alzheimer's disease (AD), shaped in part by epigenetic reprogramming and closely coupled to immunometabolic dysfunction. While microglia initially mount adaptive responses to amyloid-beta (Aβ), tau, and tissue stress, persistent exposure to chronic neurodegenerative cues may drive subsets of microglia toward senescence-like states characterized by altered chromatin regulation, transcriptional remodeling, stable cell-cycle arrest, and a sustained senescence-associated secretory phenotype (SASP). These change

Trained immunity in chronic rhinosinusitis: epigenetic reprogramming of innate immune memory as a driver of mucosal inflammation and recurrence.

Chronic rhinosinusitis (CRS) is a highly prevalent and debilitating inflammatory condition of the upper airway, affecting 5-28% of the global population and imposing a substantial socioeconomic burden. Despite major advances in endoscopic sinus surgery, pharmacological management, and targeted biologic therapies, long-term disease recurrence following treatment remains an unresolved clinical challenge. Current pathophysiological frameworks centered on adaptive type 2 immunity, eosinophilic inflammation, and pathogen persistence fail to fully account for the chronification and therapy resistanc

Cervical cancer survival analysis by integrating multi-omics data through an adaptive weighted deep neural network.

Accurately analyzing cervical cancer prognosis holds significant importance for enhancing clinical outcomes within the framework of precision medicine. Since single-omics data only capture a limited perspective of cervical cancer patients, integrating multiple types of omics data is essential to provide more comprehensive insights. However, capturing the key information in multi-omics data fusion remains challenging owing to the presence of a large number of redundant variables and a relatively small sample size. In this study, we propose a deep neural network called AWMP to better integrate m

Observed effects of improvisational theater on personal growth: A scoping review.

Drama has long been used as a modality for promoting personal growth; still, new approaches continue to emerge. This scoping review aimed to chart evidence on the administration of improvisational theater (a form of unscripted performance) for personal growth and to identify directions for future research and practice. Conducted in line with Joanna Briggs Institute (JBI) guidelines, this review included empirical studies examining the effects of theatrical improv on personal growth, with no limitations on participants, regions, or publication periods. Systematic searches of four databases (Psy

Benchmarking the impact of reference genome selection on taxonomic profiling accuracy.

BACKGROUND: Over the past decades, genome databases have expanded exponentially, often incorporating highly similar genomes at the same taxonomic level. This redundancy can hinder taxonomic classification, leading to difficulties distinguishing between closely related sequences and increasing computational demands. While some novel taxonomic classification tools address this redundancy by selecting a subset of genomes as references, insights regarding the impact of different reference genome selection methods across taxonomic classification tools are lacking. RESULTS: We systematically evalua

Bacterial Membrane Vesicles as Versatile Platforms for Systemic and Mucosal Vaccines.

Bacterial membrane vesicles (BMVs), encompassing outer membrane vesicles (OMVs) released from Gram-negative bacteria and extracellular vesicles (EVs) released from Gram-positive bacteria, have emerged as promising vaccine platforms owing to their intrinsic immunostimulatory properties and capacity to deliver a wide range of antigens. Although conventional vaccines effectively prevent infectious diseases, their long-term efficacy is often limited by antigenic variation and reliance on a restricted number of licensed adjuvants. BMVs, as self-adjuvanting systems, enable both antigen delivery and

DrugDL: dual-modal deep learning framework for multi-property drug prediction and targeted therapy discovery.

MOTIVATION: The accurate and robust representation of drug molecule features, the prediction of drug-target biomacromolecule interactions, and the determination of physicochemical properties are crucial in drug development. However, these tasks remain challenging due to issues such as the limited generalizability of single-modal representations, the absence of multitask prediction frameworks, and weak adaptability in cold-start scenarios. RESULTS: In this study, we present DrugDL, a framework for comprehensive drug molecule representation and the prediction of multiple downstream tasks, inclu

Dietary polyphenols and nutritional modulators in glioblastoma: mechanistic promise and translational barriers.

Glioblastoma is the deadliest primary brain tumor, and fewer than 7% of patients survive 5 years even after surgery, radiation, and chemotherapy. Several features make it difficult to treat: the tumor heterogeneity, it adapts quickly to resist drugs, and the blood-brain barrier keeps most agents from reaching it. These obstacles have renewed interest in adjuncts to standard care. Dietary polyphenols and certain nutritional compounds are among the candidates, since they can influence oxidative stress, inflammation, tumor metabolism, and the glioblastoma stem cells that drive recurrence. This re

Non-invasive neuromodulation in Alzheimer's disease: toward brain-heart coupling as a candidate framework for biomarker-guided adaptive intervention.

Alzheimer's disease (AD) has entered an era of biologically informed, disease-modifying therapy, yet durable clinical benefit remains limited, highlighting the need for complementary interventions. Noninvasive neuromodulation has emerged as a promising strategy, but the field is still largely organized by device categories, stimulation targets, and short-term cognitive outcomes. This framing alone does not readily account for the marked heterogeneity, stage dependence, and limited durability reported across studies. This narrative review synthesizes clinical, preclinical, and mechanistic evide

CD4+ T cells orchestrate the immune response to ALK-positive T-cell lymphoma.

Immunotherapy has revolutionized the treatment of solid cancers in recent years. However, T-cell lymphomas (T-NHLs) originate from immune cells themselves and are biologically heterogeneous, rendering investigations of immune checkpoint inhibitor (ICI) mechanisms of action complex. While case reports and individual Anaplastic Large Cell Lymphoma (ALCL) cases enrolled in T-NHL trials demonstrated favourable responses to ICI, hyperprogression was observed in other T-NHL subtypes. We therefore utilized a syngeneic mouse model of ALK+ ALCL to investigate immune surveillance and ICI-induced immune

TOPlus: An algorithm integrating prior genetic and environmental information for multi-trait synergic selection in maize hybrid breeding.

In maize hybrid breeding, synergic multi-trait selection of elite hybrids in specific target environments remains a major challenge. Enviromic data and functional gene knowledge are rapidly increasing; however, they have not been effectively integrated into crop breeding decisions. Here, we present TOPlus, a multi-trait hybrid prioritization framework comprising predictive and selective modules. The predictive module uses environmental information to improve phenotype prediction for untested genotypes and environments, whereas the selective module incorporates functional gene priors and multip
