Histaminase (diamine oxidase) is an enzyme produced at very high levels by the decidua of the placenta and is found to be associated with a number of human cancers. A procedure for the affinity chromatography purification of this enzyme is described. In this procedure, cadaverine-AH-sepharose was used to bind the enzyme in the placental extract. After extensive washing of the column with 2.5% Triton X-100 in 1 M NaCl, the enzyme was released from the column by 0.1 N chromotropic acid. To ascertain whether the content of endocrine markers is constant in small-cell carcinoma of the lung, levels of three markers of medullary thyroid carcinoma were studied in this tumor. Histaminase was increased in six of six primary tumors (three to 14,000 times), L-dopa decarboxylase in four of six (six to 30 times), and calcitonin in one of one (eight times) over levels in adjacent lung. Complexes of sodium pentacyanoamine ferrate (TPF) with amylnitrite and with butylnitrite showed hypotensive activity equal to that of sodium nitroprusside, as tested intravenously, in unanesthetized , instrumented Rhesus, monkeys. The complex with n-octylamine, on the other hand, had significantly lower activity. These complexes produced only marginal hypotensive effects when administered orally in a dose of 10 mg/kg to spontaneously hypertensive rats.

NADPH oxidases (NOXs) are attractive but challenging drug targets. Starting from M41 and its analog MC4876, we synthesized quinoline-based derivatives. SAR studies revealed that modifications at the quinoline C2 position and the amine-moiety influenced potency and isoform preference. Boc-protected analogs preferred NOX5, (compound 1a: IC50 = 1.6 μM), whereas deprotected derivatives favored NOX2. Aniline is an essential synthetic building block, with its manufacture reliant on fossil carbon feedstocks. While bioproduction could connect aniline manufacture to renewable carbon inputs, efficient metabolic pathway design remains a hurdle for aniline biosynthesis.
