A multidimensional coding architecture of the vagal interoceptive system.

Interoception, the ability to timely and precisely sense changes inside the body, is critical for survival1-4. Vagal sensory neurons (VSNs) form an important body-to-brain connection, navigating visceral organs along the rostral-caudal axis of the body and crossing the surface-lumen axis of organs into appropriate tissue layers5,6. The brain can discriminate numerous body signals through VSNs, but the underlying coding strategy remains poorly understood. Here we show that VSNs code visceral organ, tissue layer and stimulus modality-three key features of an interoceptive signal-in different dim

Function of astrocyte MyD88 in high-fat-diet-induced hypothalamic inflammation.

BACKGROUND: A growing body of evidence shows that hypothalamic inflammation is an important factor in the initiation of obesity. In particular, reactive gliosis accompanied by inflammatory responses in the hypothalamus are pivotal cellular events that elicit metabolic abnormalities. In this study, we examined whether MyD88 signaling in hypothalamic astrocytes controls reactive gliosis and inflammatory responses, thereby contributing to the pathogenesis of obesity. METHODS: To analyze the role of astrocyte MyD88 in obesity pathogenesis, we used astrocyte-specific Myd88 knockout (KO) mice fed a

Intravitreal S100B Injection Triggers a Time-Dependent Microglia Response in a Pro-Inflammatory Manner in Retina and Optic Nerve.

S100B is a glial protein, which belongs to calcium-binding protein family. Alterations of S100B level were noted in various neurodegenerative diseases. In a new glaucoma-like animal model S100B was injected intravitreally, which led to neuronal degeneration in retina and optic nerve. The pathological mechanisms are still unknown. Therefore, S100B protein was intravitreally injected in rats. At days 14 and 21, retina, optic nerve, serum, and aqueous humor were investigated. S100B injection led to an increase of retinal NF-κB at day 14. Furthermore, higher IL-1β levels in retina, serum, and aque

Identification of putative phosphoproteins in wheat spikes induced by Fusarium graminearum.

Phosphorylation and dephosphorylation events were initiated in wheat scab resistance. The putative FHB-responsive phosphoproteins are mainly involved in three functional groups and contain at least one tyrosine, serine, or threonine phosphorylation site. Fusarium head blight (FHB), caused by Fusarium graminearum, is a severe disease in wheat. Protein phosphorylation plays an important role in plant-pathogen interactions, however, a global analysis of protein phosphorylation in response to FHB infection remains to be explored. To study the effect of FHB on the phosphorylation state of wheat pro

Stochastic nanoroughness modulates neuron-astrocyte interactions and function via mechanosensing cation channels.

Extracellular soluble signals are known to play a critical role in maintaining neuronal function and homeostasis in the CNS. However, the CNS is also composed of extracellular matrix macromolecules and glia support cells, and the contribution of the physical attributes of these components in maintenance and regulation of neuronal function is not well understood. Because these components possess well-defined topography, we theorize a role for topography in neuronal development and we demonstrate that survival and function of hippocampal neurons and differentiation of telencephalic neural stem c

Pathogenic mechanisms for parathyroid hyperplasia.

Parathyroid hyperplasia is the cause of parathyroid gland enlargement in kidney disease (KD). Hypocalcemia, hyperphosphatemia, and vitamin D deficiency are critical contributors to the worsening of the hyperplastic parathyroid growth induced by KD. Reproduction of the features of human KD in the 5/6 nephrectomized rat model has shown that 80% of the mitogenic signals induced by KD in parathyroid cells that are aggravated by either high phosphate (P) or low calcium (Ca) diets occurred within 5 days after the onset of KD. Enhanced parathyroid expression of the potent growth promoter transforming

Protein kinase C cross-talk with gonadotrope progesterone receptor is involved in GnRH-induced LH secretion.

In the absence of progesterone (P), the anti-P at the receptor RU486 reduces basal and GnRH-stimulated LH secretion both in vivo and in vitro, demonstrating the existence of a ligand-independent activation of progesterone receptor (LIAPR). The aim of the present study was to determine which component of the intracellular LH secretory pathway activated by GnRH is responsible for LIAPR. To do this, anterior pituitary dispersed cells from female rats in proestrus, cultured in the presence of 17beta-estradiol, were incubated with activators or inhibitors of PKC, cAMP-PKA signalling pathways or int

Constitutively active protein kinase B enhances Lck and Erk activities and influences thymocyte selection and activation.

Protein kinase B (PKB), a serine threonine kinase is critically involved in cellular proliferation and survival. To characterize its role in T cell development in vivo, we have analyzed transgenic mice that express a membrane-targeted constitutively active version of PKB (myr PKB) in thymocytes and peripheral T cells. We report that myr PKB renders proliferative responses of thymocytes more sensitive to TCR signals by increased and sustained activation of Src kinase Lck and the extracellular signal-regulated kinase/mitogen-activated protein kinase pathway. In addition, the proliferative respon

Calcium-magnesium interactions in pancreatic acinar cells.

Although the role of calcium (Ca2+) in the signal transduction and pathobiology of the exocrine pancreas is firmly established, the role of magnesium (Mg2+) remains unclear. We have characterized the intracellular distribution of Mg2+ in response to hormone stimulation in isolated mouse pancreatic acinar cells and studied the role of Mg2+ in modulating Ca2+ signaling using microspectrofluorometry and digital imaging of Ca2+- or Mg2+-sensitive fluorescent dyes as well as Mg2+-sensitive intracellular microelectrodes. Our results indicate that an increase in intracellular Mg2+ concentrations redu

Membrane cofactor protein (MCP; CD46): isoform-specific tyrosine phosphorylation.

Membrane cofactor protein (MCP; CD46) is a widely expressed type 1 transmembrane glycoprotein that inhibits complement activation on host cells. It also is a receptor for several pathogens including measles virus, Streptococcus pyogenes, Neisseria gonorrhea, and Neisseria meningitidis. That MCP may have signaling capability was suggested by its microbial interactions. That is, binding of MCP on human monocytes by measles virus hemagglutinin or cross-linking by an anti-MCP Ab resulted in IL-12 down-regulation, while binding to MCP by Neisseria on epithelial cells produced a calcium flux. Ultraviolet light irradiation increases cellular diacylglycerol and induces translocation of diacylglycerol kinase in murine keratinocytes.

Cellular lipid metabolism can provide a variety of mediators of signal transduction, including diacylglycerols and inositol phosphates. These factors may be involved in the control of epidermal differentiation and proliferation because they are modulated by extracellular calcium, which also regulates the maturation phenotype of cultured keratinocytes. The effect of non-cytotoxic exposures to ultraviolet light on lipid metabolism was studied in cultured murine keratinocytes. Ultraviolet treatment of cultured murine keratinocytes growing in 0.05 mM Ca++ did not significantly change the total amo

Steric confinement-induced emission probe for monitoring protein conformations in live cells.

Protein conformational changes drive signal transduction to regulate cellular activities, yet monitoring of these changes in living cells remains challenging. Here, we introduce BIOSCE (BIOprobe based on Steric Confinement-induced Emission), a technique that enables tracking of individual protein conformations in living cells across millisecond-to-minute timescales. BIOSCE reports protein conformational changes via steric confinement-induced luminescence switching from non-luminescent to luminescent states. We demonstrate that BIOSCE rapidly senses calmodulin conformational changes triggered b

Intrinsic timing, not temporal prediction, underlies ramping dynamics in visual and parietal cortex during passive behavior.

Ramping neural activity is widely interpreted as a signature of predictive processing, but whether these signals truly reflect predictions or instead emerge from sensory mechanisms remains unclear. To address this question, we used two-photon calcium imaging across multiple cell types in visual and parietal cortex, while awake mice passively received repeated audiovisual stimuli presented under distinct temporal structures. Neurons segregated into two broad response classes: stimulus-activated (ramp-down) and stimulus-inhibited (ramp-up) populations with diverse temporal kinetics. Multiple fin

Decanoic acid extends lifespan and modulates metabolism in models of PLA2G6-associated neurodegeneration.

PLA2G6-associated neurodegeneration (PLAN) is a group of rare genetic disorders characterised by progressive neurodegeneration resulting from mutations in the PLA2G6 gene, encoding a calcium-independent phospholipase enzyme. Here, we have explored the effects of decanoic acid (DA), a medium-chain fatty acid, in the fruit fly Drosophila melanogaster models of PLAN and show that DA treatment significantly extends the lifespan, reduces bang sensitivity and improves resistance to heat shock stress. Transcriptional analysis showed that DA affects genes in key signalling pathways, including Insulin/

Failure to Observe Stomatal ABA Responses in a Fern and a Lycophyte.

Stomatal closure induced by abscisic acid (ABA) is an important mechanism by which plants reduce transpiration under drought stress and is well studied in seed plants. However, ABA-induced stomatal closure in ferns and lycophytes has been reported in some studies but not in others, even within the same species, such as the fern Nephrolepis exaltata and the lycophyte Selaginella uncinata. Here, ABA-induced stomatal behaviour and ABA signalling components, including guard cell potassium ion (K+) and calcium ion (Ca2+) fluxes and vacuoles, were analysed in the seed plant Vicia faba, the fern N. e

Mycobacterium tuberculosis sulfolipid-1 (Sl-1) increases the excitability of mouse and human TRPV1-positive sensory neurons in a YM254890-reversible fashion.

Cough is a hallmark sign of tuberculosis and key driver of transmission. While traditionally attributed to host-driven inflammation, we previously demonstrated that Mycobacterium tuberculosis lipid extract (Mtb extract) and its component sulfolipid-1 (SL-1) directly activate nociceptive neurons to induce cough in guinea pigs. However, the cellular mechanisms by which Mtb extract and SL-1 modulate nociceptive sensory neurons remain incompletely understood. Here, we show that Mtb extract enhances action potential (AP) generation in mouse nodose nociceptors via an SL-1-dependent mechanism. Using

Emerging Roles of Phospholipase C Beta Isozymes as Potential Biomarkers in Cardiac Disorders.

Phospholipase C (PLC) enzymes represent crucial participants in the plasma membrane of mammalian cells, including the cardiac sarcolemmal (SL) membrane of cardiomyocytes. They are responsible for the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) into 1,2-diacylglycerol (DAG) and inositol (1,4,5) trisphosphate (Ins(1,4,5)P3), both essential lipid mediators. These second messengers regulate the intracellular calcium (Ca2+) concentration, which activates signal transduction cascades involved in the regulation of cardiomyocyte activity. Of note, emerging evidence suggests tha
