Comparison of the effects of vitamin A and its analogs upon rabbit ear cartilage in organ culture and upon growth of the vitamin A-deficient rat.

A study was conducted to explore the relationship between the effects of vitamin A upon cartilage and the biological role of vitamin A in maintaining growth and life. Retinol, retinoic acid, alpha-retinoic acid, and ROB-7699 (a cyclopentyl analog of retinoic acid) were highly effective in promoting the lysis of the extracellular matrix of cartilage grown in organ culture in vitro. Retinoic acid and its two analogs were quantitatively more active than was retinol in bringing about lysis of matrix and release of proteoglycan into the culture medium. A bioassay was then conducted to determine the…

PRP-Derived Exosomes Promote Cartilage Repair by Suppressing Oxidative Stress and Inflammatory Signaling in Chondrocytes: An In Vitro and Ex Vivo Study.

Osteoarthritis (OA) is a degenerative musculoskeletal condition characterized by progressive cartilage degradation accompanied by joint inflammation, for which current treatments offer only symptomatic relief without halting progression. This study compared the regenerative and anti-inflammatory properties of platelet-rich plasma (PRP) and its exosome-enriched derivative (PRP-Exos) on human chondrocytes and cartilage explants under IL-1β stimulated inflammatory conditions. PRP-derived exosomes (PRP-Exos) were extracted using an aqueous two-phase separation method and subsequently evaluated for…

Association of Wearable Sensor-Derived Gait Measures With Cartilage Damage Over Two Years Among Adults With or at Risk of Knee Osteoarthritis.

OBJECTIVE: Gait affects knee loading. Modifying gait could reduce load and protect against cartilage loss. Our objective is to look for modifiable gait parameters and determine their relation to worsening cartilage damage. METHODS: We studied participants from the Multicenter Osteoarthritis Study (MOST) ages 45 to 90 years with, or at risk for, knee osteoarthritis (OA). Gait assessment used inertial measurement units (APDM, Inc) on the pelvis and ankles during a 20-m walk. Knee magnetic resonance imaging (MRI) was acquired at baseline and two years later. Cartilage damage worsening was assess…

Development of a gene-activated scaffold using niosomes and minicircle DNA as a novel non-viral delivery platform to enhance cartilage repair.

Gene therapy combined with advanced biomaterial-based delivery systems represents a powerful strategy to enhance chondrogenic differentiation of mesenchymal stem cells (MSCs), enabling the development of next-generation regenerative therapies for cartilage repair. In this context, gene-activated biomaterials provide a versatile tool for spatially and temporally regulating cell fate within three-dimensional (3D) microenvironments. Here, we combine collagen type I/type II-hyaluronic acid (CI/CII-HyA) scaffold with a novel non-viral gene delivery platform based on niosomes (DP20CQ) to deliver the…

The zone of calcified cartilage: A human explant culture model and a proof-of-concept transcriptomic study of 3D laser-cut interface tissue.

OBJECTIVE: The cartilage-bone interface, including the zone of calcified cartilage (ZCC), is a thin layer of calcified tissue with a low cell number, thus making it technically challenging to study. The aim of this study was to develop methods to selectively study the ZCC for gene expression analysis and transcriptomic studies. DESIGN: We set-up a method to isolate ZCC explants comprising the ZCC and a thin layer of subchondral bone from human osteochondral tissue (n=4 patients) for tissue culture. The cellular response to a pro-inflammatory cytokine cocktail or TGF-ß1 (72 h) was compared to …

The Impact of varus and valgus alignment on knee cartilage quality assessed by magnetic resonance imaging: insights from the IMI-APPROACH cohort.

BACKGROUND: Lower limb malalignment increases the risk of unicompartmental knee osteoarthritis (KOA). This study investigates the association between knee cartilage quality, assessed via MRI-based T2 mapping, and lower limb malalignment. It also examines whether cartilage quality is more influenced by bony or intra-articular malalignment. METHODS: In this cross-sectional analysis of 156 knees from the IMI-APPROACH cohort, tibiofemoral cartilage T2 values were measured using high-resolution MRI, distinguishing superficial and deep layers. Malalignment was categorized into entire leg, bony, and…
