[Role of β2-Adrenergic Receptor Agonist in the Cognitive Impairment Associated with Neurodegenerative Diseases].

Studies have revealed the neuropathological mechanism of the cognitive impairment associated with neurodegenerative diseases. However, the therapies for these cognitive disorders are limited, and the prevalence of cognitive impairment is expected to increase significantly in the future, which proves the necessity of new therapeutic agents. In recent years, the pharmacological activity of β2-adrenergic receptor(β2-AR)has been extensively studied, which has demonstrated that β2-AR agonist has therapeutic effects on the cognitive impairment associated with several common neurodegenerative diseases, inclu

Modeling cognitive dysfunction in neurofibromatosis-1.

Cognitive dysfunction, including significant impairments in learning, behavior, and attention, is found in over 10% of children in the general population. However, in the common inherited cancer predisposition syndrome, neurofibromatosis type 1 (NF1), the prevalence of these cognitive deficits approaches 70%. As a monogenic disorder, NF1 provides a unique genetic tool to identify and dissect mechanistically the molecular and cellular bases underlying cognitive dysfunction. In this review, we discuss Nf1 fly and mouse systems that mimic many of the cognitive abnormalities seen in children with

Increased Serum Neuropeptide Galanin Level Is a Predictor of Cognitive Dysfunction in Patients with Hip Fracture.

BACKGROUND: Hip fracture is a common occurrence in elderly populations and is frequently followed by various levels of cognitive dysfunction, leading to adverse functional outcomes. Risk stratification of hip fracture patients to identify high-risk subsets can enable improved strategies to mitigate cognitive complications. The neuropeptide galanin has multiple neurological functions, and altered levels are documented in dementia-type and depression disorders. The present study investigated the association of serum neuropeptide galanin levels in hip fracture patients with the occurrence of cogn

Divergent Long-Term Recovery After Long COVID in Tobacco Use Disorder and Healthy Controls: A Longitudinal Study.

INTRODUCTION: Although tobacco use disorder (TUD) and long COVID (LC) have been associated to neuropsychiatric disturbances, it remains unclear how tobacco dependence affects the recovery of clinical, cognitive, and biological processes over time. METHODS: This study compared individuals with tobacco use disorder and healthy controls using longitudinal assessments from pre-infection baseline through two post-COVID follow-ups over 12 months. Comprehensive assessments covered clinical, neuropsychiatric, and cognitive functioning, tobacco dependence and exposure, and peripheral biomarkers reflec

Kynurenic acid signaling expands in human and nonhuman primates and impairs dorsolateral prefrontal cortical cognition that is key to mental illness.

Cognitive deficits from dorsolateral prefrontal cortex (dlPFC) dysfunction are common in neuroinflammatory disorders, including long-COVID, schizophrenia and Alzheimer's disease, where impairments are correlated with kynurenine inflammatory signaling. Kynurenine synthesis from tryptophan is increased under conditions of inflammation, then further metabolized to kynurenic acid (KYNA) in brain, where it blocks NMDA and α7-nicotinic receptors (nic-α7Rs). These receptors are essential for neurotransmission in dlPFC, suggesting that KYNA may contribute to higher cognitive deficits in these disorder

Combined transcranial electrical stimulation (tES) and cognitive training (CT) for cognitive impairment: Evidence from clinical applications and basic research.

Cognitive impairment is a serious pathological feature of neuropsychiatric disorders, making the exploration of effective treatments urgent. Recent research shows that combined transcranial electrical stimulation (tES) and cognitive training (CT) can reduce cognitive deficits. This review summarizes studies on neurological disorders that use both clinical patients and rodent models to highlight the underlying neural mechanisms. In patients, this combined approach improves cognitive domains such as attention, working memory, and executive function. Improvements have been observed in patients wi

Bipolar disorder and obsessive compulsive disorder: The comorbidity does not further impair the neurocognitive profile.

BACKGROUND: The comorbidity of bipolar disorder (BD) and obsessive-compulsive disorder (OCD) has been widely described. Several studies have investigated the cognitive profiles of BD and OCD patients, but studies that compare BD, BD-OCD, and OCD patients in neuropsychological domains do not exist. The purpose of this study was to compare set-shifting, decision making, and central coherence among BD, BD-OCD, and OCD patients. METHODS: A battery of neuropsychological tests was administered to 68 patients (22 BD, 26 BD-OCD, 20 OCD). The Young Mania Rating Scale and Hamilton Depression Rating Sca

Electroconvulsive therapy-related cognitive impairment and choice of anesthesia: the tipping point.

INTRODUCTION: Electroconvulsive therapy (ECT) is among the most effective treatments of several life-threatening psychiatric disorder. Despite effective therapy, ECT-induced seizure could cause several adverse effects including cognitive disorders and memory impairment. Drugs such as thiopental, which have been prescribed for anesthesia required for ECT, are known as drugs with cognitive effects. This pilot randomized clinical trial tried to assess the feasibility of using a lower dose of thiopental in combination with remifentanil instead of a higher challenging dose of a single drug with cog

Neurocognitive and clinical predictors of functional outcome in patients with schizophrenia and bipolar I disorder at one-year follow-up.

OBJECTIVE: Many studies have reported that cognitive ability may be predictive of the functional outcome for patients with schizophrenia. However, no study has prospectively examined these aspects in schizophrenia and bipolar disorders simultaneously. The present study attempted to analyze if neurocognition and clinical status predicts the real-life functioning for patients with schizophrenia or bipolar I disorder, using a longitudinal design. METHOD: Forty-seven schizophrenic and 43 bipolar I outpatients were assessed twice with a neurocognitive battery (Executive Functions, Working Memory,

Comparison of cognitive style in bulimia nervosa and depression.

This study investigated the global and specific cognitive style associated with bulimia nervosa. Three groups of women (women with bulimia nervosa, women with major depression, and controls) completed measures of eating disorder severity, depression, dysfunctional cognitions and irrational beliefs. The control group was found to report significantly lower levels of cognitive distortions and irrational beliefs overall than both women with bulimia nervosa and women with depression. However, no difference was found between the latter two groups. Furthermore, the pattern of individual cognitions a

Neurobiological substrates of major psychiatry disorders: transdiagnostic associations between white matter abnormalities, neuregulin 1 and clinical manifestation.

BACKGROUND: Schizophrenia, bipolar disorder and major depressive disorder are increasingly being conceptualized as a transdiagnostic continuum. Disruption of white matter is a common alteration in these psychiatric disorders, but the molecular mechanisms underlying the disruption remain unclear. Neuregulin 1 (NRG1) is genetically linked with susceptibility to schizophrenia, bipolar disorder and major depressive disorder, and it is also related to white matter. METHODS: Using a transdiagnostic approach, we aimed to identify white matter differences associated with NRG1 and their relationship t

A Critical Review of Behavioral and Emotional Disinhibition.

The theory of "disinhibition" has been very influential in psychiatry and neurology for over a century. Disinhibition has been used to explain clinical findings in many neurological and psychiatric disorders including dementia, traumatic brain injury, attention deficit hyperactive disorder, substance abuse, impulsivity in personality disorders, and neurodevelopmental disorders. In addition, disinhibition has been used as a unifying theory to link clinical observations with cognitive findings, and even cellular findings. This review discusses the origins and history of the theory of disinhibiti

Loss of emotional insight in behavioral variant frontotemporal dementia or "frontal anosodiaphoria".

Loss of insight is a prominent clinical manifestation of behavioral variant frontotemporal dementia (bvFTD), but its characteristics are poorly understood. Twelve bvFTD patients were compared with 12 Alzheimer's disease (AD) patients on a structured insight interview of cognitive insight (awareness of having a disorder) and emotional insight (concern over having a disorder). Compared to the AD patients, the bvFTD patients were less aware and less concerned about their disorder, and they had less appreciation of its effects on themselves and on others. After corrective feedback ("updating"), th

[Trichloroethylene intoxication presenting with temporal seizures].

INTRODUCTION: Trichloroethylene (TCE) is extensively used as a degreasing agent in the metal industry and as a solvent for organic compounds. Its neurotoxicity (with respect to both the peripheral and central nervous systems) is now widely acknowledged. OBSERVATION: Here, we report the case of a 32-year-old male drug addict presenting temporal seizures after "huffing" TCE (i.e. voluntary inhalation). The patient also deve loped a "psycho-organic syndrome" which included cognitive dysfunction (with memory disorders in particular) and personality changes. These disorders had not been noticed by

Child physical abuse: risk for psychopathology and efficacy of interventions.

The emerging research on child physical abuse (CPA) is reviewed herewith. Incidence of CPA may be as high as 11% for adolescents in the US. Child physical abuse survivors vary in their symptom picture of severity and type. Children who have been physically abused may experience symptoms of post-traumatic stress disorder, anxiety disorders, depressive disorders, or disruptive behavior disorders. Additionally, developmental disturbances include problems with attachment, social skills, and affect regulation. Chronic abuse is associated with more severe psychopathology.
