Effects of molecular weight, end group of polylactide and coating thickness on corrosion rate and inhomogeneity of iron under biomimetic conditions.

Iron-based biodegradable stents hold significant promise for percutaneous coronary intervention. Our previous study has revealed that a polylactide (PLA) coating can not only accelerate iron corrosion but also mitigate corrosion inhomogeneity, presenting a metal-polymer composite stent (MPS). Nevertheless, current understanding of how PLA coating parameters regulate iron corrosion remains limited. This article represents the systematic and quantitative in vitro investigation into the effects of coating thickness, molecular weight, and end group of PLA on the corrosion rate and inhomogeneity of

Green Solvent Pretreatment of Biomass: Insights into Lignin Structural Evolution in Organosolv and Deep Eutectic Solvent Systems.

Lignocellulosic biomass is a key renewable resource for producing biofuels, biomaterials, and high-value chemicals, but its efficient utilization is hindered by the recalcitrant lignin-carbohydrate complex (LCC) network. Organosolv (OS) and deep eutectic solvent (DES) pretreatments have emerged as promising green strategies for biomass fractionation and lignin extraction. The present work summarizes recent advances in OS and DES pretreatment, with a particular focus on the mechanisms of lignin depolymerization and structural evolution, including changes in major bonds, functional groups, S/G r

[Advances in predisposing factors and pathogenesis of granulomatous lobular mastitis].

Granulomatous lobular mastitis (GLM) is a chronic inflammatory breast disease of unknown aetiology, characterised by granulomatous inflammation centred on the mammary lobules. Clinically, it presents with breast redness, swelling, and ulceration, with a protracted course prone to recurrence. Its development is closely associated with multiple factors including milk stasis, hyperprolactinemia, ductal injury, and microbial infection. The pathogenesis remains incompletely elucidated, with various hypotheses proposed such as autoimmune responses, delayed-type hypersensitivity, and autoinflammatory

Single-cell technologies in deciphering drug resistance of multiple myeloma: mechanistic insights and clinical translation prospects.

The greatest hindrance to long-lasting remission in multiple myeloma (MM) is drug resistance and is modulated by the complex interaction of tumor-intrinsic plasticity and microenvironmental co-evolution. Multi-omics of single cells has transformed the exploration of this complexity, and has solved the dynamics of clonal architecture and ecosystem at an unprecedented resolution. The present review summarizes critical relapse-driver revealed by single cell technologies, including existing resistant subclones and non-genetic programs, as well as inflammatory stromal remodeling and immune exhausti

Transfection of the free-living alga Chromera velia enables direct comparisons with its parasitic apicomplexan relative, Toxoplasma gondii.

Chromera velia is a photosynthetic, free-living alga closely related to the apicomplexan parasites, a phylum of intracellular pathogens responsible for many devastating diseases, including malaria, cryptosporidiosis and toxoplasmosis. With molecular and cellular landmarks clearly related to but distinguishable from those found in apicomplexan parasites, Chromera provides an opportunity to investigate the evolutionary origin of the structures and processes needed for intracellular parasitism. However, tools for defining localization and functions of gene products do not exist for Chromera, whic

Dynamical Simulation of Chiral-Induced Spin-Polarization and Magnetization.

Despite generally lacking ferromagnetic properties or strong spin-orbit coupling, electrons in chiral molecules exhibit unique spin-dependent transport behavior, known as chiral-induced spin selectivity (CISS). This phenomenon implies a profound connection between chirality and spin and draws attention to the link between chirality and magnetism. Experiments in recent years have shown that chirality can induce spin-polarizations and magnetizations, providing fresh insights into interpreting chirality-related biochemical processes and designing nanomagnetic devices. In this Letter, we present a

The expanding landscape of inositol phosphate signaling network in land plants.

The emergence of land plants from aquatic habitat over half a billion years ago marked a pivotal moment in Earth's history, profoundly altering both soil composition and atmospheric chemistry. This remarkable evolutionary transition was accompanied by the establishment of multiple signaling pathways that facilitated plant adaptation to terrestrial environments. Among these, signaling pathways based on myo-inositol-derived cellular messengers have recently gained significant attention. Combinatorial attachment of phosphate to the myo-inositol (hereafter inositol) ring produces a large array of

The molecular mechanism of fluoride export by the eukaryotic fluoride channel FEX.

Much of life on Earth, including plants, fungi, and bacteria, evolved to resist toxic environmental fluoride. In eukaryotes, the major resistance mechanism is fluoride export by membrane proteins known as FEX. Using electrophysiology and transport assays, we establish that FEX from plants and yeasts are highly selective fluoride channels. Fluoride transport activity depends on reversible sodium ion binding, but sodium itself is not transported. We determine a structure of a FEX protein, from pathogenic yeast Candida albicans, using cryo-EM. Bolstered by mutagenesis studies, this structure reve

Patient-derived organoid co-culture systems as next-generation models for bladder cancer stem cell research.

Three-dimensional patient-derived organoids (PDOs) have emerged as a powerful model for investigating the molecular and cellular mechanisms underlying bladder cancer, particularly in the context of cancer stem cells (CSCs) and drug screening. However, a significant limitation of conventional PDOs is the absence of tumor microenvironment (TME), which includes critical stromal, immune and microbial components that influence tumor behavior and treatment response. In this review, we provide a comprehensive overview of the recent advancements in PDO co-culture systems designed to integrate TME elem

An X-linked sex determination mechanism in cannabis and hop.

Sex chromosomes in cannabis and hop were identified a century ago because of their obvious visible differences in size (heteromorphy). However, we know little about the genes they contain that control the development of the inflorescences. Here we assembled genomes, with phased sex chromosomes, for hop and cannabis. The XY chromosomes share an origin prior to the divergence between the genera >36 MYA. Due to the inheritance patterns of the XYs, the male-specific region of the Y is highly-degenerated, with substantial gene loss, while the X shows faster rates of molecular evolution. Consistent

Cell membranes sustain phospholipid imbalance via cholesterol asymmetry.

Membranes are molecular interfaces that compartmentalize cells to control the flow of nutrients and information. These functions are facilitated by diverse collections of lipids, nearly all of which are distributed asymmetrically between the two bilayer leaflets. Most models of biomembrane structure and function often include the implicit assumption that these leaflets have similar abundances of phospholipids. Here, we show that this assumption is generally invalid and investigate the consequences of lipid abundance imbalances in mammalian plasma membranes (PM). Using quantitative lipidomics,

Systematic screens for fertility genes essential for malaria parasite transmission reveal conserved aspects of sex in a divergent eukaryote.

Sexual reproduction in malaria parasites is essential for their transmission to mosquitoes and offers a divergent eukaryote model to understand the evolution of sex. Through a panel of genetic screens in Plasmodium berghei, we identify 348 sex and transmission-related genes and define roles for unstudied genes as putative targets for transmission-blocking interventions. The functional data provide a deeper understanding of female metabolic reprogramming, meiosis, and the axoneme. We identify a complex of a SUN domain protein (SUN1) and a putative allantoicase (ALLC1) that is essential for male

Pyroptosis: Mechanisms and links with diabetic cardiomyopathy.

Diabetes mellitus (DM) is a chronic metabolic disease characterized by hyperglycaemia that seriously affects human health. Diabetic cardiomyopathy (DCM) is a major cardiovascular complication and one of the main causes of death in patients with DM. Although DCM attracts great attention, and new therapeutic methods are continuously developed, there is a lack of effective treatment strategies. Therefore, exploring and targeting new signalling pathways related to the evolution of DCM becomes a hotspot and difficulty in the prevention and treatment of DCM. Pyroptosis is a newly discovered regulate

A Challenging Case of Recurrent Fever.

Syndrome of undifferentiated recurrent fever (SURF) includes heterogeneous episodes of systemic inflammation without documented infection, without response to antibiotherapy, and characterized by a paucity of specific clinical or molecular criteria. Colchicine is an effective treatment with an impact on morbimortality. We describe a case of a previously healthy one-year-old male, with consanguineous ancestry, admitted four times due to recurrent fever, associated with nonspecific symptoms and an increase of inflammatory markers in a sepsis-like pattern. No consistent infection was documented,

Seahorse Male Pregnancy as a Model System to Study Pregnancy, Immune Adaptations, and Environmental Effects.

Seahorses, together with sea dragons and pipefishes, belong to the Syngnathidae family of teleost fishes. Seahorses and other Syngnathidae species have a very peculiar feature: male pregnancy. Among different species, there is a gradation of paternal involvement in carrying for the offspring, from a simple attachment of the eggs to the skin surface, through various degrees of egg coverage by skin flaps, to the internal pregnancy within a brood pouch, which resembles mammalian uterus with the placenta. Because of the gradation of parental involvement and similarities to mammalian pregnancy, sea

Synchronous presentation of ETV6::RUNX1 fusion positive concordant B-acute lymphoblastic leukaemia in identical twin toddlers.

Concordant leukaemia in identical twins is rare. The likelihood of concordance of leukaemia in twins is near 100% in infancy, around 10% from 1 to 6 years of age, and rare at a later age with variable latency. Reporting of new cases of concordant leukaemia in twins is encouraged to contribute to data pool of this infrequent but exceptional condition; especially when the theories with respect to evolution, natural history and molecular evidence explaining concordant leukaemia in identical twins are still evolving. We discuss identical pair of monochorionic twin toddlers who were detected to have

A Study on the Effect of Doping Metallic Nanoparticles on Fracture Properties of Polylactic Acid Nanofibres via Molecular Dynamics Simulation.

All-atom molecular dynamics simulations are conducted to elucidate the fracture mechanism of polylactic acid nanofibres doped with metallic nanoparticles. Extensional deformation is applied on polymer nanofibres decorated with spherical silver nanoparticles on the surface layer. In the obtained stress-strain curve, the elastic, yield, strain softening and fracture regions are recognized, where mechanical parameters are evaluated by tracking the stress, strain energy and geometrical evolutions. The energy release rate during crack propagation, which is a crucial factor in fracture mechanics, is
