[Whole genome sequencing and analysis of familial nonsyndromic congenital tooth agenesis].

Congenital tooth agenesis impairs masticatory function and aesthetics and adversely affects craniofacial development. Although largely considered genetic in origin, its exact etiology remains unclear. This study reports three familial cases of nonsyndromic congenital tooth agenesis (NSTA). Whole genome sequencing (WGS) revealed five pathogenic variants: filamins-B (FLNB) (c.5186C>A, p.Ser1729Ter), methylcrotonyl coenzyme a carboxylase 2 (MCCC2) (c.91C>T, p.Gln31Ter; c.484C>T, p.Gln162Ter; c.340C>T, p.Gln114Ter), laminin subunit alpha 2 (LAMA2) (c.1084A>T, p.Arg362Ter), cathepsin C (CTSC) (c.74

Genomic relationship between polyendocrine metabolic ovarian syndrome and bipolar disorder.

Women with bipolar disorder (BIP) have a higher risk of developing polyendocrine metabolic ovarian syndrome (PMOS). Shared genetic architecture may underlie this comorbidity. Valproate, a mood-stabilizer commonly used to treat BIP, increases the risk of PMOS. Still, the mechanism underlying PMOS in BIP remains unknown. Here, we aimed to identify genetic variants shared between BIP and PMOS, as well as their interaction with valproate. We used the results of large-scale genome-wide association studies of BIP (41,510 cases and 354,340 controls), and PMOS (3609 cases and 229,788 controls). Using

New clinical and genomic insights into paracoccidioidomycosis in Paraguay: A neglected endemic area.

Paracoccidioidomycosis (PCM) is a systemic fungal infection caused by Paracoccidioides spp., predominantly affecting individuals in Latin America. While extensive epidemiological and molecular studies have been conducted in Brazil, Colombia, and Venezuela, where PCM is a recognized public health concern, Paraguay remains largely overlooked. The scarcity of epidemiological and molecular data hinders accurate disease surveillance, early diagnosis, and effective treatment strategies. This study provides the first comprehensive epidemiological and molecular characterization of PCM in Paraguay. We

Genomic targets of hurricane-induced selection on clinging performance in an island lizard.

A central goal of evolutionary biology is to understand the molecular mechanisms that enable adaptive responses to novel environmental pressures. Extreme weather events like natural disasters can have catastrophic impacts on wildlife via habitat destruction, mortality,1,2,3,4,5,6 population displacement,6,7 and natural selection,1,2,5,8,9 even shaping broad biogeographic patterns of local adaptation on phylogenetic scales.10,11 However, few studies have investigated the functional changes in organismal performance or genomic targets associated with responses to such events.2,12,13,14 Here, we

[Genomic Characterization of SARS-CoV-2 by Next-Generation Sequencing Analysis].

The coronavirus disease-2019 pandemic has been shaped by successive waves of variants due to the high mutational capacity of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Monitoring the genomic diversity and mutational profiles of these variants and their clinical impact is essential to inform public health measures as well as vaccine and treatment strategies. This study aimed to analyze clinical samples detected as SARS-CoV-2 positive by real-time reverse transcription polymerase chain reaction (rRT-PCR) during two different periods of the pandemic in Türkiye using next-genera

Whole-Genome Characterization of Rodent Coronaviruses (RCoVs) in Thailand, 2024-2025.

Rodents are recognized as important reservoirs of coronaviruses, contributing to viral diversity and potential zoonotic emergence. However, data on rodent coronaviruses (RCoVs) in Thailand remained limited. This study aimed to determine the prevalence, genetic characteristics, and phylogenetic relationships of RCoVs circulating in rodent populations across Thailand. A total of 435 rodents were captured in 26 provinces across five geographic regions of Thailand between January 2024 and April 2025. Paired oral and rectal swabs (n = 870) from 435 rodents were collected and screened for RCoVs usin

Genomic Characterization of a Locally Transmitted Leishmania mexicana Isolate from Texas.

Cutaneous leishmaniasis, which is caused by Leishmania species transmitted by sandflies, is increasingly recognized in subtropical, higher-resource regions, including the southern United States. The first whole-genome sequence of Leishmania mexicana (L. mexicana) is reported from an autochthonous US case in a 3-year-old boy from Ellis County, Texas, with no travel history. Genomic DNA was sequenced using Illumina (San Diego, CA) technology. Phylogenomic analysis of nuclear and mitochondrial single-nucleotide polymorphisms (SNPs) confirmed the isolate as L. mexicana, clustering within the L. me

Insights into Genomic Dynamics and Plasticity in the Monkeypox Virus from the 2022 Outbreak.

The 2022 global mpox outbreak represented a turning point in the Monkeypox virus (MPXV) epidemiology, highlighting the incredible capability of this virus to adapt to different conditions, also in a non-endemic context. To investigate the genomic dynamics of MPXV 2022 strains, we performed whole-genome sequencing of 40 clinical samples from 16 Italian patients across multiple anatomical sites and timepoints between May and December 2022. Combining single-nucleotide analysis with detailed investigation of short tandem repeats (STRs), we explored inter- and intra-host viral dynamics. We identifi

Sero-genomic evidence for occult mpox exposure in healthy Nigerian adults.

The 2022 multi-country mpox (formerly monkeypox) outbreak, driven by mpox virus (MPXV) Clade IIb poses renewed threat to global public health. The cessation of smallpox vaccination has created large immunologically naïve cohorts, with uncertain implications for contemporary MPXV susceptibility. To assess whether residual vaccination-derived immunity influences exposure risk, we combine serological and phylodynamic analyses. Using a six-plex Luminex assay, we measure immunoglobulin G (IgG) binding to six MPXV antigens in 176 Nigerian adults comprising of 75 healthcare workers sampled in 2021 an

Pathogenicity and genomic characterization of Brazilian fowl adenovirus serotypes 1 and 11.

Fowl adenoviruses (FAdVs) are important pathogens affecting poultry worldwide. Different serotypes cause distinct disease syndromes in domestic chickens, including gizzard erosions (GE), inclusion body hepatitis (IBH), and hepatitis-hydropericardium syndrome (HHS). Despite the economic impact, the evaluation of pathogenicity and genomic features of Brazilian isolates has not yet been assessed. This study aimed to investigate the pathogenic characteristics and whole-genome features of two FAdV Brazilian isolates, BRMSA3762 (FAdV-1) and BRMSA3761 (FAdV-11). For this, pathogenicity was evaluated

Genomic Context and Insert Orientation in the Regulation of Transgene Expression in Adenoviral Vectors.

Adenoviral vectors are among the most efficient platforms for gene delivery; however, the level and pattern of transgene expression in these vectors are largely shaped by the viral genomic context. This review discusses the mechanisms of adenoviral transcription and alternative splicing and how they influence the expression of inserted expression cassettes. Particular attention is given to the role of insertion orientation and transgene placement within the E1 and E3 regions, as well as to the effects of viral regulatory elements, including the E1A enhancer. We analyze evidence on the use of i

Active Surveillance for Heartland virus in North Carolina: Clinical and Genomic Epidemiology.

BACKGROUND: Heartland virus (HRTV) is an emerging tick-borne virus capable of causing severe illness and death. The burden of disease is likely underestimated due to limited seroprevalence studies, lack of commercially available diagnostic tests, and an overlapping clinical syndrome with more commonly diagnosed bacterial diseases such as spotted fever group rickettsiosis or ehrlichiosis. METHODS: Active surveillance for Heartland virus disease was conducted at a large academic center from March to September 2024. Enrolled subjects included those who had testing sent for Ehrlichia polymerase c

Calibration-aware genomic prediction of paratuberculosis serostatus in indigenous Turkish goats.

Paratuberculosis, caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a chronic, incurable enteritis of ruminants in which late-onset clinical signs preclude timely intervention, making early genomic stratification a key control lever. We investigated the genetic architecture underlying the host antibody response to MAP - measured as anti-MAP ELISA serostatus - using genome-wide SNP data from 474 animals representing seven indigenous Turkish goat breeds sampled across 11 provinces (237 seropositive, 237 seronegative; estimated true seroprevalence 14.1%). Fourteen architecturally di

Cellulosimicrobium Sepsis: Identification of an Emerging Pathogen Using Whole-Genome Sequencing.

Cellulosimicrobium is an emerging but still rare pathogen that has exhibited a predilection for immunocompromised patients and those with indwelling medical devices. A case of an immunocompetent 89-year-old man with a history of hypertension who presented with 1 day of subjective fever and right-sided flank pain and was found to be bacteremic secondary to cholecystitis is presented in the present study. Piperacillin-tazobactam was initiated, and a percutaneous cholecystostomy tube was placed the following day. The blood-cultured pathogen was originally identified as Cellulomonas, but whole-gen

Contributions of whole-genome sequencing to the epidemiological monitoring of Campylobacter spp. in France.

Next-generation sequencing techniques have revolutionized the epidemiology and understanding of Campylobacter spp. infections. In this study, we present the results of a next-generation sequencing (NGS) analysis conducted in 2024 on 2,360 Campylobacter spp. strains isolated in France, including 1,959 Campylobacter jejuni, 358 Campylobacter coli, and 43 Campylobacter fetus strains. We describe the diversity of circulating strains and provide an overview of the main mechanisms of antibiotic resistance. We identified a variety of resistance mechanisms, particularly for resistance to group A penic

Genomic analysis of antibiotic resistance and virulence factors in the uropathogen Citrobacter koseri.

Citrobacter koseri is a significant bacterial cause of community-acquired urinary tract infections in female college students. Given limited previous attention to genomic analysis and molecular epidemiology of this species, whole-genome sequencing was carried out for eight uropathogenic C. koseri clinical isolates. Six complete and two draft assemblies are presented. Genome organization was largely conserved between isolates, and comparative analysis with archived genomes suggests that more than 90% of C. koseri genes are conserved across the species. These isolates contained relatively few mo
