Attachment and germination of Entomophaga maimaiga conidia on host and non-host larval cuticle.

The lepidopteran-specific fungal pathogen Entomophaga maimaiga is highly virulent against Lymantria dispar (gypsy moth) larvae, and other members of the family Lymantriidae. Numerous species in the subfamily Cuculliinae (Family Noctuidae) are not susceptible to E. maimaiga due to the inability of this fungus to penetrate the larval cuticle. Conidial attachment and germination were compared among five cuculliine species and L. dispar using bioassays and scanning electron microscopy. Although conidia were showered evenly across larvae during bioassays, on L. dispar conidia were most abundant on …

Evaluation of ELISA and Western Blot Analysis using three antigens to detect anti-Trichinella IgG in horses.

We assessed a serological method for detecting Trichinella infection in horses, specifically, an ELISA using three antigens to detect anti-Trichinella IgG (i.e. a synthetic tyvelose glycan-BSA (stg-BSA) antigen, an excretory/secretory (ES) antigen, and a crude worm extract (CWE) antigen). Serum samples were collected from 2502 horses (433 live horses from Romania and 2069 horses slaughtered in Italy and originating from Italy, Poland, Romania, and Serbia). Serum samples were also taken from horses experimentally infected with different doses of T. spiralis and T. murrelli larvae, as controls. …

Clinical, clinico-pathological and serological studies of Babesia ovis in experimentally infected sheep.

Clinical, clinico-pathological and serological studies were performed in sheep experimentally infected with Babesia ovis. Acute babesiosis occurred in all the lambs infested with adult Rhipicephalus bursa ticks and in one lamb infested with the larvae. The rate of parasitaemia and the degree of anaemia were not correlated. Decrease in the packed-cell volume ranged from 30 to 40%. Parasitized erythrocytes were not observed to block capillaries in the brain, which explained the absence of nervous symptoms in acute babesiosis. The kidneys were the most severely affected organs, exhibiting acute g…

Evaluating a CRISPR-based sterile male for Drosophila suzukii (Diptera: Drosophilidae) control.

Spotted-wing drosophila (SWD), Drosophila suzukii Matsumura, is a serious invasive pest of small fruits. The novel precision-guided Sterile … erformed predator feeding assays to address regulatory concerns of pgSIT technology. Green lacewing larvae, Chrysoperla rufilabris, that fed on wild or pgSIT SWD larvae developed similarly into pupae and adults. While pgSIT flies would not be present in the field at the larval stage, this test revealed no discernible impacts when predators fed on it. Ra …

Comparative lethal and sublethal effects of sulfoxaflor and acetamiprid on Cryptolaemus montrouzieri (Coleoptera: Coccinellidae): integrating IOBC classification and matrix population modeling.

Following regulatory restrictions on several neonicotinoids, sulfoxaflor, a sulfoximine insecticide proposed as a replacement, has been reported as highly effective against a broad range of sap-feeding pests. However, its impact on key biological control agents, such as the widely used mealybug destroyer Cryptolaemus montrouzieri Mulsant (Coleoptera: Coccinellidae), remains insufficiently investigated. In this study, we assessed the lethal and sublethal effects of recommended field rates of sulfoxaflor on larvae and adults of C. montrouzieri through direct spray treatments and exposure to fres…

Integrating environmental DNA, diving surveys and DNA barcoding to track endangered twaite shad (Alosa fallax) spawning sites.

The twaite shad (A. fallax), an anadromous fish species belonging to the Clupeidae family, has experienced population declines throughout Eu … on might be up to 400 km upstream from the sea. The demersal eggs hatch relatively quickly, and the larvae start a planktonic life in the river. Traditional monitoring methods have often proved inadequate due to the cryptic nature of A. fallax. With the goal of improving the efficacy of current biomonitoring designs, we tested an environmental DNA (eDNA …

Polylactic acid microplastics enhance copper bioavailability and host-microbiome toxicity during black soldier fly bioconversion of food waste: Mechanisms and multi-level impacts.

Biodegradable microplastics are increasingly entering food waste (FW) streams, yet their effects on heavy metal behavior during black soldier fly larvae (BSFL) bioconversion remain unclear. This study investigated the combined effects of polylactic acid microplastics (PLA-MPs, 1% w/w) and Cu (100-300 mg/kg ww) in a BSFL-based FW conversion system. The 100-200 mg/kg treatments represented upper-range Cu exposure scenarios relevant to Cu-rich food residues. PLA-MPs increased Cu accumulation in larvae by 14-32% and shifted Cu toward more bioactive fractions in larvae and frass, indicating enhance…

The internal larval anatomy of two rodent bot fly species: Cuterebra fontinella (Diptera: Oestridae) and Cuterebra americana.

Species within the genus Cuterebra Clark, 1815 (Diptera: Oestridae: Cuterebrinae) are obligate parasites of rodents and other mammals during the larval stage, causing cutaneous myiasis. The internal larval anatomy of Cuterebra species remains unstudied, despite its potential utility for understanding their parasitic adaptations and for comparative studies with other Oestridae. The present study describes the internal larval anatomy of the species Cuterebra fontinella Clark, 1827 and Cuterebra americana (Fabricius, 1775), using micro-computed tomography-based virtual histological sections and 3…

Analysis of Toxic Components in Secondary Metabolites of Entomopathogenic Fungi Clonostachys rosea (Hipocreales: Bionectriaceae) from Cephalcia chuxiongica (Hymenoptera: Pamphiliidae).

Clonostachys rosea, an entomopathogenic fungus that infects Cephalcia chuxiongica, is highly pathogenic and has significant potential for controlling the damage this pest causes to pine forests. To investigate the role of C. rosea secondary metabolites in fungal pathogenicity, we conducted toxicity assays using crude metabolite extracts. These assays evaluated the effects of different concentrations, larval developmental stages, and exposure methods on larval mortality. Gas chromatography-mass spectrometry (GC-MS) was subsequently employed to identify the chemical constituents of the crude ext…

Critical thermal maxima of Polistes life stages from different climates, with a critical evaluation of methods.

Ambient temperature is a crucial abiotic factor for ectotherms. It strongly influences development, life and abundance, as well as success in colonizing new habitats. In the eusocial paper wasps Polistes sp., colony-forming insects with open nests, the larvae and pupae have limited options to influence their own body temperature in response to high environmental temperatures. They are dependent on measures taken by the adults to keep it at tolerable levels. We determined the upper thermal limits (CTmax) in field populations of three paper wasp species (Polistes dominula, P. gallicus, P. biglum…

A novel real-time PCR for New World screwworm fly (Cochliomyia hominivorax) and its application in a non-destructive multiplex for efficient detection of screwworm flies.

Surveillance and diagnostics are critical for the early detection, containment and eradication of exotic pests. For the screwworm fly, this is usually via targeted surveillance and exclusion testing of trap-caught flies, as well as the identification of larvae associated with myiasis wounds. We present a specific and sensitive real-time polymerase chain reaction (PCR) assay for the detection of the New World screwworm fly, Cochliomyia hominivorax Coquerel (Diptera: Calliphoridae). The assay targets the cytochrome oxidase subunit 1 (cox1) gene from adult flies or larvae and retains high analyti…
