Carrier-Free Nanoparticles Enhance Photothermal-Immune Therapy via Metabolic Reprogramming in Triple-Negative Breast Cancer.

A carrier-free co-assembled nanoplatform, designated as IR@PF-M NPs, was developed to enhance photothermal-immunotherapy against triple-negative breast cancer (TNBC) by reprogramming methionine metabolism. The nanoplatform was constructed through the self-assembly of the methionine adenosyltransferase 2A (MAT2A) inhibitor PF9366 with the near-infrared (NIR)-absorbing dye IR808, followed by surface modification with DSPE-PEG2000-Met. This formulation exhibited dual targeting capabilities, enabling tumor accumulation via the enhanced permeability and retention effect and selective uptake by tumo

Wrapping of adjacent nanoparticles with varying sizes and distances by membrane tethers possessing bilayer asymmetry.

Membrane tethers are long, slender membrane tubes that transport biological cargo (proteins and viruses) and engineered materials (particles and drugs) in organelles, vesicles, and cells. Nanoparticles with enhanced bioavailability and reduced toxicity are attracting great attention in nanomedicine for their ability to target membrane tethers, particularly tunneling nanotubes, which share similar structures and functions with membrane tethers. Nevertheless, the molecular mechanisms that control the cooperative wrapping of nanoparticles within and on membrane tethers are not well understood. Ou…

Multifunctional CuS-CDs@MnO2 nanoparticles and liposome-assisted amplification for triple-mode detection of enrofloxacin.

The residues of enrofloxacin (ENR) in animal-derived food and water environments have emerged as a growing public health concern in recent years. Herein, a tri-mode biosensor was designed by employing CuS-CDs@MnO2 as the signal probe and glutathione (GSH)-loaded liposomes for amplification. The manganese dioxide (MnO2) shell of the nanocomposite quenched the fluorescence of CuS-CDs, reduced its photothermal performance and also endowed it with oxidase-like activity. In the presence of ENR, a large amount of GSH was released after aptamer recognition and magnetic separation. Then, the shell of

Tetrandrine-based in situ vaccination via multifunctional Mn²⁺-hyaluronic acid hydrogel loading gelatinase-responsive nanoparticles.

In situ tumor vaccination (ISV) has emerged as a promising strategy for converting tumors into endogenous vaccine depots, but its application in breast cancer remains limited by poor intratumoral drug retention and insufficient microenvironment remodeling. Here, we developed an injectable Mn²⁺-coordinated hyaluronic acid hydrogel embedding gelatinase-responsive tetrandrine nanoparticles (TETNPs@Gel) as an MRI-trackable local ISV platform. TETNPs@Gel exhibited biphasic, pH-responsive drug release, accelerating tetrandrine liberation under mildly acidic conditions while limiting premature leakag…

Injectable Self-Healing Fucoidan-Chitosan Hydrogel Incorporating Polydopamine-Silver-Rutin Nanoparticles for Multifunctional Wound Dressing.

Excessive reactive oxygen species (ROS) and bacterial infection were the important obstacles to impaired wound healing. Herein, by the use of innate bioactivities of natural polysaccharides and flavonoid glycosides, a multifunctional hydrogel was engineered through the Schiff base reaction between oxidized fucoidan (oFu) and lysine-modified chitosan (CS-Ly) for wound dressing. Rutin and silver nanoparticle (AgNP)-modified polydopamine (PDA) nanoparticles were incorporated into the hydrogel matrix, thereby exhibiting favorable ROS scavenging ability and adhesive property. Moreover, the hydrogel…

Gold nanoparticles-modulated ECL/PEC aptasensor for the dual-mode detection of Alzheimer's disease biomarker.

A dual-mode electrochemiluminescent/photoelectrochemical (ECL/PEC) aptasensor modulated by optical and catalytic properties of gold nanoparticles (Au NPs) is constructed to determine soluble amyloid β oligomer (AβO), a critical Alzheimer’s disease (AD) biomarker. In this system, CdS quantum dots (QDs) are integrated onto an indium tin oxide electrode (ITO), acting as ECL emission source and PEC substrates. The developed sensing system enables signal transduction by exploiting the robust AβO binding ability of the developed aptamer and magnetic isolation as an enrichment strategy. The resultant…

Self-assembled supramolecular nanoparticles of α-mangostin for targeted STING activation and enhanced tumor immunotherapy.

Targeted activation of the stimulator of interferon genes (STING) signaling pathway represents a promising strategy to counteract immunosuppressive tumor microenvironments (TME) and elicit anti-tumor immunity. However, clinical translation of STING agonists has been strongly hindered by the challenges in achieving tumor-targeted STING activation. Here, we report a facile and efficient supramolecular self-assembly strategy for tumor-targeted STING activation of α-mangostin (MGT), a natural non-cyclic dinucleotide STING agonist. MGT can self-assemble into uniform supramolecular nanoparticles (MG…

Formulation and characterization of polyethylenimine-coated magnetopolymeric nanoparticles with potential application in thermal cancer therapies.

Use of superparamagnetic nanoparticles as heat generators contributed to the development of alternatives to conventional cancer therapies. These nanoparticle-based photothermal and/or magnetothermal therapies can kill malignant cells with negligible effects on surrounding tissues. Maghemite nuclei embedded into a poly(ε-caprolactone) nanomatrix were formulated by interfacial polymer disposition, and the resulting nanostructure was coated with polyethylenimine. DLS, TEM, EDX spectroscopy, electrophoretic and FTIR analysis were used to characterize the resulting ≈ 220 nm-sized (core/shell)/shell…

Pathology illustrates pathogenesis of indium lung diseases in rats induced by indium-tin oxide nanoparticles.

Indium lung disease remains an ultra-rare interstitial lung disease with no initiative effective in changing the trial. It is related to excessive protein deposition, the development of interstitial inflammation, and subsequent pulmonary fibrosis. This is a "destructive" pulmonary disease. The pathogenesis of indium lung disease remains unclear because the disease progresses continuously, making early diagnosis difficult. This study aimed to identify the histopathological features of pulmonary injury during and after Nano-ITO intratracheal instillation. Male Sprague-Dawley rats were intratrach

Green synthesis of CuO nanoparticles using mangrove leaves for targeted delivery of platinum (II) therapeutics.

Copper oxide nanoparticles (CuO NPs) synthesized via green chemistry using Avicennia marina mangrove leaf extract represent an environmentally sustainable approach to generating biocompatible nanomaterials for pharmaceutical applications. This study utilized the phytochemical-rich extract as both a reducing and capping agent to produce quasi-spherical monoclinic CuO NPs with a diameter range of 35-87 nm. The resulting nanoparticles possessed a specific surface area of 4.5 m² g⁻¹ with a mesoporous structure (pore volume of 0.018 cm³ g⁻¹), making them ideal candidates for drug loading applicatio…

Fabrication and statistical optimisation of Nifedipine-loaded alginate nanoparticles using honey as a natural stabilizer.

Nifedipine (NF) is widely used to manage hypertension and angina. However, its poor aqueous solubility limits oral delivery, warranting sustained, pH-responsive release systems. This study fabricated and optimised NF-loaded alginate nanoparticles (NF-loaded Alg-NPs) using honey as a green, natural stabiliser. NPs were prepared by ionotropic gelation and optimised via a 32 factorial design. The optimised formulation showed a mean particle size of 437 ± 4.58 nm, zeta potential of -10.98 ± 0.12 mV, and entrapment efficiency of 84.5 ± 0.8%. FTIR, XRD, DSC, and SEM confirmed successful, largely amo…

Cell Painting phenomics reveals size-dependent phenotypic responses to titanium dioxide nanoparticles in HepG2 cells.

Titanium dioxide nanoparticles (TiO₂NPs) are widely produced engineered nanomaterials with ongoing human exposure through consumer and occupational uses. Conventional in vitro assays often focus on cytotoxicity and may therefore overlook early or sublethal cellular perturbations. Here, we applied Cell Painting-based phenomics to resolve size-dependent sub-lethal phenotypic signatures of TiO2NP exposure in human HepG2 hepatocytes. Two TiO2NPs (<25 nm and <100 nm) were characterized by field emission scanning electron microscopy and evaluated following 24-hour exposure at five concentrations: 6.…

Size Effect of Nanoparticles on Targeting Injured Kidneys for Enhanced Intervention in Acute Kidney Injury.

Nanoparticles show great potential for the management of acute kidney injuries (AKIs). It is well known that nanoparticle size plays a critical role in targeting renal tubules due to the pore structure and kidney filtration threshold (KFT) of the glomerulus. However, AKI can compromise the physiological barriers, which might render the established KFT (∼6 nm) and size laws under healthy conditions not applicable in AKI conditions. Reinvestigating the nanoparticle size effect on targeting injured kidneys is thus essential for developing efficacious AKI nanomedicine. Herein, using different-size…
