A quantitative study of the relative contribution of different retinal sectors to the innervation of various thalamic and pretectal nuclei in goldfish.

The contribution of retinal ganglion cells situated in different retinal quadrants to the innervation of eight nontectal, retinorecipient targets was examined in goldfish. In some fish, cobaltous-lysine was used to selectively fill severed intraretinal ganglion cell axons and the number of filled axons within each nucleus was determined. In other fish, either the dorsal or ventral or nasal or temporal retina was ablated and the remaining axons from the intact retina were filled with cobalt. The density of the cobalt-filled axons within the retinorecipient targets was quantified with a microden…

Decoding the roles of bipolar, amacrine, and horizontal cells.

The retina consists of several specialized cell types, crucial for visual processing. Bipolar cells, amacrine cells, and horizontal cells are retinal interneurons, playing pivotal roles in transmitting and modulating visual information within the retina. In this chapter, we first highlight the significance of bipolar cells in relaying signals from photoreceptors to ganglion cells, initiating visual signaling. Recent consensus indicates the existence of different types of bipolar cells in various species, leading us to focus on their functional diversity and morphologic characteristics. Second,…

Diversity and Feature Selectivity of Primate Retinal Ganglion Cells.

The primate retina transmits visual information to the brain through parallel pathways carried by diverse retinal ganglion cell (RGC) types, each of which encodes specific features in the visual environment. These parallel channels provide input to a variety of brain regions to support both conscious and reflexive visual functions. Although the midget, parasol, and small bistratified pathways have dominated classical models of primate vision, accumulating evidence points to a broader diversity of wide-field RGC types whose functions are less well understood. Recent breakthroughs in single-cell…

Rod/Cone Gap Junctions: Plastic Electrical Synapses at the First Visual Synapse.

Electrical coupling mediated by gap junctions is a defining feature of almost every neuron in the retina, as it is throughout the central nervous system. By allowing direct current flow between neurons, electrical coupling helps establish functional circuits, allows signals to be shared, and improves the signal-to-noise ratio, thereby regulating neuronal activity at the dendritic, single-neuron, and network levels. Remarkably, electrical coupling is not static or constitutive. It shows a high degree of plasticity, changing in strength on time scales ranging from milliseconds to days, thereby p…

Retinal Transcriptome-Wide Association Study Identifies Novel Alzheimer's Disease Risk Genes.

INTRODUCTION: Alzheimer's disease (AD) is the leading cause of dementia worldwide. The retina shares molecular pathways with the brain, yet no study has systematically linked retinal gene expression to AD risk. METHODS: We performed transcriptome-wide association studies (TWAS) using two independent retinal eQTL panels (Strunz et al., n = 311; EyeGEx, n = 406) and a large meta-analyzed AD genome-wide association study (GWAS) (Bellenguez et al., 111,326 cases, 677,663 controls). Genes were further validated with GWAS in the independent Alzheimer's Disease Sequencing Project (ADSP) using a matc…

The human fovea.

While the fovea occupies only about 0.1% of the total retinal surface area, it is arguably the most important region of the human retina. The human fovea is characterized by unique anatomic specializations relative to the rest of the retina, which include an absence of inner retinal layers and the accompanying vasculature, an increased density of cone photoreceptors, a near-absence of rod photoreceptors, and a unique "private-line" midget ganglion cell circuitry. These anatomic specializations are responsible for our high-acuity vision. Unfortunately, as a result, even minor disruptions to fov…

Maf-family bZIP transcription factor NRL interacts with RNA-binding proteins and R-loops in retinal photoreceptors.

RNA-binding proteins (RBPs) perform diverse functions including the regulation of chromatin dynamics and the coupling of transcription with RNA processing. However, our understanding of their actions in mammalian neurons remains limited. Using affinity purification, yeast-two-hybrid and proximity ligation assays, we identified interactions of multiple RBPs with neural retina leucine (NRL) zipper, a Maf-family transcription factor critical for retinal rod photoreceptor development and function. In addition to splicing, many NRL-interacting RBPs are associated with R-loops, which form during tra…

Circulating Cell-Free DNA as an Epigenetic Biomarker for Early Diabetic Retinopathy: A Narrative Review.

Diabetic retinopathy (DR), a complication of type 2 diabetes mellitus (T2DM), is typically asymptomatic in its early stages. Diagnosis typically relies on routine fundoscopy for the clinical detection of microvascular abnormalities. However, permanent retinal damage may occur well before clinical signs are appreciable. In the early stages of DR, the retina undergoes distinct epigenetic changes, including DNA methylation and histone modifications. Recent evidence supports unique epigenetic 'signatures' in patients with DR compared to non-diabetic controls. These DNA 'signature' sequences may be…

Emerging role of circular RNAs in diabetic retinopathy.

Diabetic retinopathy (DR), a significant complication that affects the retina of individuals with diabetes, poses a severe threat to their visual health. DR is classified into stages ranging from non-proliferative to proliferative forms. As the disease progresses, pathological neovascularization and hemorrhage in the retina or vitreous can occur, potentially leading to vision impairment or blindness. Current treatments for DR include intravitreal injections of anti-vascular endothelial growth factor drugs and surgical interventions such as laser photocoagulation. However, these treatments are …

Zebrafish cdh23 Affects Rod Cell Phototransduction Through Regulating Ca2+ Transport and MAPK Signaling Pathway.

Mutations in the pathogenic gene CDH23 are known to cause Usher syndrome, affecting both auditory and visual functions. Our previous results … and eosin (H&E) staining to assess their histological changes. The results showed that the cdh23-/- retina was morphologically indistinguishable from the control. Apoptosis was assessed using TUNEL staining, which revealed an increase in total cell death in the cdh23-/- retina. Our results revealed that the cell death was induced by Ca2+ and MAPK signali …

Retinal neurovascular coupling evaluation with circular-scan OCTA.

Neurovascular coupling (NVC) is the mechanism by which neuronal activity regulates blood flow. Detection of abnormalities in NVC may be clinically useful as they could signal early retinal dysfunction before structural damage becomes apparent. While optical coherence tomography angiography (OCTA) enables high-contrast, non-invasive functional imaging of the retina, its conventional form cannot detect light-evoked hemodynamic responses, which are the key indicators of NVC. Visualizing and quantifying NVC-driven hemodynamics in the human retina remains challenging due to motion artifacts and ins…

Alternative Splicing Dysregulation in Retinitis Pigmentosa: Pathogenic Mechanisms and Therapeutic Opportunities.

Retinitis pigmentosa (RP) represents a genetically heterogeneous group of inherited retinal dystrophies characterized by progressive photoreceptor degeneration and irreversible vision loss. Among the diverse pathogenic mechanisms, dysregulation of alternative splicing has emerged as a pivotal driver, particularly in RP cases caused by mutations in splicing factors or cis-regulatory elements. Alternative splicing governs transcript diversity and fine-tunes gene expression, with more than 95% of human multi-exon genes undergoing this process. Disruption of precise splicing patterns in the retina…

Intravitreal AAV-IKV mediated delivery of decorin inhibits choroidal neovascularization, fibrosis, inflammation and elevates autophagy.

Age-related macular degeneration (AMD) is the most common cause of blindness in the elderly. The exudative or wet form of AMD is caused by choroidal neovascularization (CNV) and subsequently a macular edema. Wet AMD can be effectively treated with anti-vascular endothelial growth factor (VEGF) therapies. However, despite treatment, more than half of patients continue to lose vision due to a lack of compliance with frequent intravitreal injections, failure to adequately respond to anti-VEGF therapy and emergence of fibrotic scars underneath the retina. In this study we investigated the use of o…

Peptide-based hydrogel co-assembled with antibody-drug for enhanced retinal cell uptake and attenuated experimental autoimmune uveitis.

Effective treatment of chronic posterior ocular diseases such as uveitis, diabetic retinopathy, and age-related macular degeneration requires improvements in targeted drug delivery strategies. This study introduces a novel injectable drug delivery system co-assembled with a peptide-based hydrogel and secukinumab (SEK), an IL-17A neutralising monoclonal antibody, targeting retinal pigmented epithelium (RPE) cells. Compared to a SEK solution, the SEK loaded hydrogel significantly enhanced the protein uptake (3.7 times higher) by RPE cells in an inflammatory state after 24 hours of treatment and …

Lytic photoreceptor cell death caused by Rab escort protein deficiency in Drosophila.

Choroideremia (CHM) is a rare X-linked recessive form of inherited retinal degeneration caused by the deficiency of the Rab escort protein 1 (REP1)-encoding CHM gene. REP1 is essential for the post-translational prenylation of the key players in intracellular membrane trafficking, the Rab GTPases. In this study, we aimed to analyze the mechanisms of retinal degeneration caused by Rep deficiency using the Drosophila retina as a model system. Rab GTPases lost their membrane association ability and diffused into the cytoplasm, and the accumulation of unprenylated Rab6 and Rab7 was observed in Rep…

Retinal Ganglion Cell Transplantation: Differentiation and Quantification Strategies.

Glaucoma is a leading cause of irreversible blindness worldwide. Current treatments focus on reducing intraocular pressure but cannot restore lost visual function once it is lost due to retinal ganglion cell (RGC) degeneration and death. Recent advances suggest that transplantation of stem cell-derived RGCs could offer new therapeutic approaches for glaucoma and vision restoration. Here, we present a detailed protocol for differentiating human RGCs from embryonic stem cells using both three-dimensional retinal organoid and two-dimensional culture approaches. Following differentiation, we descr…
