Mouse Model of Thyroid Cancer Progression and Dedifferentiation Driven by STRN-ALK Expression and Loss of p53: Evidence for the Existence of Two Types of Poorly Differentiated Carcinoma.

Background: Thyroid tumor progression from well-differentiated cancer to poorly differentiated thyroid carcinoma (PDTC) and anaplastic thyroid carcinoma (ATC) involves step-wise dedifferentiation associated with loss of iodine avidity and poor outcomes. ALK fusions, typically STRN-ALK, are found with higher incidence in human PDTC compared with well-differentiated cancer and, as previously shown, can drive the development of murine PDTC. The aim of this study was to evaluate thyroid cancer initiation and progression in mice with concomitant expression of STRN-ALK and inactivation of the tumor

Does SGLT2 inhibition with dapagliflozin overcome individual therapy resistance to RAAS inhibition?

Individual patients show a large variation in their response to renin-angiotensin-aldosteron system (RAAS) inhibition (RAASi), both in surrogates such as albuminuria and in hard renal outcomes. Sodium-glucose co-transporter 2 inhibitors (SGLT2) have been shown to lower albuminuria and to confer cardiovascular and, possibly, renal protection. To establish whether individual therapy resistance to RAASi can be overcome by adding an SGLT2 inhibitor, we assessed individual albuminuria responses in patients exposed to both RAASi and the SGLT2 inhibitor dapagliflozin. We used data from a randomized c

Inhibition of the glucose transporter SGLT2 with dapagliflozin in pancreatic alpha cells triggers glucagon secretion.

Type 2 diabetes (T2D) is characterized by chronic hyperglycemia resulting from a deficiency in insulin signaling, because of insulin resistance and/or defects in insulin secretion; it is also associated with increases in glucagon and endogenous glucose production (EGP). Gliflozins, including dapagliflozin, are a new class of approved oral antidiabetic agents that specifically inhibit sodium-glucose co-transporter 2 (SGLT2) function in the kidney, thus preventing renal glucose reabsorption and increasing glycosuria in diabetic individuals while reducing hyperglycemia. However, gliflozin treatme

[Dapagliflozin, the first SGLT-2 inhibitor in the treatment of type 2 diabetes].

Dapagliflozin is the first novel sodium-glucose co-transporter-2 (SGLT2) inhibitor approved by the European Medicines Agency (EMA) for the treatment of type 2 diabetes. By inhibiting SGLT2, dapagliflozin blocks reabsorption of filtered glucose in the kidney, increasing urinary glucose excretion and reducing blood glucose levels. Its mechanism of action is independent of pancreatic β cell function and modulation of insulin sensitivity. The results of phase III clinical trials showed that dapagliflozin, at a dose of 5 or 10mg/day for 24 weeks as monotherapy in previously untreated patients, or a

Association of SGLT2 inhibitors use with a lower risk of biliary diseases in patients with type 2 diabetes mellitus: a retrospective cohort study.

BACKGROUND: Type 2 diabetes mellitus (T2DM) increases the risk of biliary diseases (BD). This study aimed to evaluate the association between sodium-glucose cotransporter-2 inhibitors (SGLT2i) use andBD risk in T2DM patients, compared with sulfonylureas. METHODS: We conducted a multi-center retrospective cohort study using electronic health records from Nanjing Medical University (January 2017-September 2022). Adults aged 18-75 years with T2DM newly prescribed SGLT2i or sulfonylureas were included. Propensity score with inverse probability of treatment weighting (IPTW) was applied to balance

Multifunctional Glycine-Carbon Dots Protect Against Arsenic Hepatotoxicity Through Redox Balance and PI3K/AKT Activation.

BACKGROUND: The theranostic potential of glycine-derived carbon dots (Gly/CDs) against arsenic-induced hepatotoxicity remains largely unexplored. This study aimed to synthesize Gly/CDs via a green, microwave-assisted method and to systematically evaluate their hepatoprotective efficacy and underlying mechanisms in a sodium arsenite (NaAsO2)-induced hepatic injury model. METHODS: Gly/CDs were synthesized using citric acid, urea, and glycine as precursors. Their physicochemical properties were characterized using transmission electron microscopy (TEM), fluorescence spectroscopy, Fourier-transfo

Effect of empagliflozin on copeptin levels in patients with recent acute coronary syndrome and newly detected dysglycaemia: a post-hoc analysis of the SOCOGAMI randomized controlled trial.

BACKGROUND: Copeptin, a surrogate marker for vasopressin secretion, is associated with cardiovascular disease, insulin resistance and dysglycaemia. The cardioprotective effects of sodium-glucose cotransporter 2 inhibitors (SGLT2i) may involve vasopressin modulation through fluid redistribution, but whether this effect persists long-term in high-cardiovascular-risk patients with newly detected dysglycaemia remains unknown. METHODS: In this post-hoc analysis of the SOCOGAMI double-blind, placebo-controlled trial, 42 patients (mean age 67.5 years, 19% females) with impaired glucose tolerance or

Effects of Dapagliflozin on the Bone of Patients with CKD on Dialysis.

The effects of gliflozins on bones of CKD patients on dialysis are unknown. Recently, SGLT2 expression in bone tissue of patients with CKD has been reported. We hypothesized that dapagliflozin may act in bone cells and modulate the Klotho-FGF23 axis. This study analyzed the effects of dapagliflozin on serum bone biomarkers and related outcomes. In this predefined post-hoc analysis of a previous RCT, patients on dialysis received (1:1) dapagliflozin 10 mg daily or standard care for 24 weeks. The primary endpoint was the change from baseline in serum Klotho and FGF23 levels, compared by ranked a

RIPK3/MLKL-mediated necroptosis promotes hepatic inflammation in chronic arsenic exposure through drinking water.

Chronic exposure to arsenic-contaminated drinking water is a major environmental risk factor for liver injury, yet its pathogenic mechanisms remain unclear. This study explores the role of RIPK3/MLKL-mediated necroptosis in arsenic-induced hepatotoxicity using a chronic in vivo mouse model. Mice were exposed to sodium arsenite (NaAsO2) at concentrations of 2.8, 8.4, and 25.2 mg/L in drinking water for 8 weeks. Arsenic exposure induced dose-dependent hepatocellular necrosis, elevated liver injury markers (ALT, AST, GST-α, GDH), and increased hepatic expression of p-RIPK3, p-MLKL and HMGB1, alon

SGLT2 inhibitors ameliorate myocardial hypertrophy in rats via AMPK/Nrf2-mediated ferroptosis suppression and antioxidant response.

This study investigated whether dapagliflozin, a sodium-glucose cotransporter 2 inhibitor, alleviates myocardial hypertrophy by modulating the AMP-activated protein kinase (AMPK)/nuclear factor E2-related factor 2 (Nrf2) signaling pathway, thereby reducing oxidative stress and ferroptosis. A rat model of myocardial hypertrophy was created using abdominal aortic stenosis. Rats were divided into blank, sham, surgery (Abdominal Aortic Coarctation, AAC), and dapagliflozin-treated groups (n = 4), with evaluations at 2, 4, 8, and 12 weeks post-surgery. In vitro, an isoproterenol-induced hypertrophic

Glucagon-like Peptide-1 Receptor Agonists and Risk of Tuberculosis in Type 2 Diabetes.

Tuberculosis remains a leading cause of illness and death worldwide, and individuals with type 2 diabetes have an increased risk of developing this infection. However, whether different glucose-lowering medications influence this risk is unclear. Here we show that use of glucagon-like peptide-1 receptor agonists is associated with a lower risk of tuberculosis compared with several commonly prescribed alternatives. We conducted a multicenter cohort study using TriNetX electronic health records from 143 healthcare organizations across multiple countries between 2017-2025. Patients receiving gluc

Metabolic Profiling of the EmDia Cohort by LC-MS Reveals Empagliflozin-Intake Associated Regulation of 1,5-anhydroglucitol and Urate.

The EmDia trial, designed to study the effects of the sodium glucose cotransporter-2 (SGLT2) inhibitor empagliflozin on cardiovascular comorbidities in type 2 diabetes mellitus (T2DM) patients, has been investigated for short-term metabolic alterations by a limited set of clinical assays. To expand on this data, we report on the development of a liquid chromatography-mass spectrometry (LC-MS)-based metabolomics approach employing an optimized metabolite separation by pentafluorophenyl chromatography. High-confidence metabolite annotation based on reference standards allows for fast and robust

Effect of dapagliflozin on sepsis-induced kidney injury in a rat model: modulation of the NLRP3 pathway.

INTRODUCTION: Sepsis-induced acute kidney injury (AKI) is a major cause of mortality in critically ill patients. Inflammation, oxidative stress, and apoptosis are key contributors to sepsis-related renal damage. Dapagliflozin (DPG), an SGLT2 inhibitor, has demonstrated anti-inflammatory and nephroprotective effects. This study aimed to investigate the renoprotective effects of DPG in a lipopolysaccharide (LPS)-induced sepsis model, focusing on inflammation, oxidative stress, and apoptosis in the kidneys. METHODS: Thirty-two male Wistar albino rats were divided into four equal groups: (1) Cont

The Beneficial Effect of the SGLT2 Inhibitor Dapagliflozin in Alleviating Acute Myocardial Infarction-Induced Cardiomyocyte Injury by Increasing the Sirtuin Family SIRT1/SIRT3 and Cascade Signaling.

Sodium-glucose cotransporter-2 inhibitors (SGLT2i) have a variety of cardiovascular and renoprotective effects and have been developed as novel agents for the treatment of heart failure. However, the beneficial mechanisms of SGLT2i on cardiac tissue need to be investigated further. In this study, we established a mouse model of acute myocardial infarction (AMI) using coronary artery constriction surgery and investigated the role of dapagliflozin (DAPA) in protecting cardiomyocytes from hypoxic injury induced by AMI. In vitro experiments were done using hypoxic cultured H9c2 ventricular cells t

Empagliflozin in patients with autosomal dominant polycystic kidney disease (EMPA-PKD): study protocol for a randomised controlled trial.

INTRODUCTION: Autosomal dominant polycystic kidney disease (ADPKD) is a hereditary condition that causes the formation of cysts primarily in the kidneys. The continuous growth of multiple cysts leads to the destruction of functional parenchyma, which may progress to end-stage kidney disease. Tolvaptan is the only drug specifically approved for slowing down the progression of ADPKD. Sodium-glucose transporter 2 inhibitors might provide additional benefits but there is currently no information on safety and outcome effects of SGLT2i in patients with ADPKD, as these patients were excluded in SGLT

SGLT2 inhibitor empagliflozin monotherapy alleviates renal oxidative stress in albino Wistar diabetic rats after myocardial infarction induction.

Acute kidney injury (AKI) is a sudden insult of the kidney that happens within a short period of time, which is associated with poor prognosis in diabetic patients with myocardial infarction (MI). Subclinical AKI is a condition in which tubular damage biomarkers [Neutrophil gelatinase-associated lipocalin (NGAL) and kidney injury molecule-1(KIM-1)] are positive even in the absence of elevated serum creatinine. Recent studies reported that SGLT-2 inhibitors could protect against subclinical AKI in diabetic patients by elevating the level of β-Hydroxybutyric acid (βOHB). This study aims to exami

Effect of dapagliflozin on urinary albumin excretion in patients with chronic kidney disease with and without type 2 diabetes: a prespecified analysis from the DAPA-CKD trial.

BACKGROUND: Reductions in albuminuria are associated with a subsequent lower risk of kidney failure in patients with chronic kidney disease. The SGLT2 inhibitor dapagliflozin significantly reduced albuminuria in patients with type 2 diabetes and normal or near-normal kidney function. Whether this effect persists in patients with chronic kidney disease with and without type 2 diabetes is unknown. We assessed the effects of dapagliflozin on albuminuria in patients with chronic kidney disease with and without type 2 diabetes in the dapagliflozin and prevention of adverse outcomes in chronic kidne

On-off-on fluorescent carbon dots from waste tea: Their properties, antioxidant and selective detection of CrO42-, Fe3+, ascorbic acid and L-cysteine in real samples.

In this work, we reported an economical plant-based hydrothermal method for one-pot green synthesis of water-soluble carbon dots (Tea-CDs) by using waste tea extract as a carbon source. The synthesized Tea-CDs were characterized by UV-visible, fluorescence, FT-IR, TEM, XPS and XRD. The Tea-CDs were found to remove hydroxyl and superoxide anion radical in vitro. In addition, the Tea-CDs exhibited bright blue fluorescence under UV-light (λex = 365 nm), and the fluorescence could be effectively quenched by CrO42- and Fe3+ ions. Meanwhile, the fluorescence of Tea-CDs-CrO42- and Tea-CDs-Fe3+ system
