We describe the cloning, expression pattern and functional overexpression analysis of Xotx5b, a new member of the Otx gene family in Xenopus laevis. Early expression of Xotx5b resembles that of Xotx2, being detected in the organizer region at early gastrula stage, and, shortly after, also in anterior neuroectoderm. During neurula stages Xotx5b exhibits a changing and dynamic pattern of expression. After neural tube closure, Xotx5b is expressed in the eye and pineal gland, both involved in photoreception. The primary structure of the cone-rod homeobox protein (CRX), a member of the OTX-like homeobox protein family, in the rat was deduced from the cDNA nucleotide sequence. The deduced protein consisted of 299 amino acid residues with motifs conserved in mammalian CRXs, and was 98% identical to mouse CRX. Northern blot analysis showed that Crx mRNA levels in the rat retina were constant under a daily light-dark cycle. These findings suggest that the expression mechanism of rat Crx in the retina was different from that in the pineal, where Crx mRNA exhibits a daily expression rhythm.

Hedgehog (Hh) proteins control many developmental events by inducing specific cell fates or regulating cell proliferation. The Patched1 (Ptc1) protein, a binding protein for Hh molecules, appears to oppose Hh signals by repressing transcription of genes that can be activated by Hh. Sonic hedgehog (Shh), one of the vertebrate homologs of Hh, controls patterning and growth of the limb but the early embryonic lethality of ptc1(-)(/)(-) mice obscures the roles of ptc1 in later stages of development.
