Sound-localization deficits induced by lesions in the barn owl's auditory space map.

Barn owls possess a two-dimensional map of auditory space. The map appears in its final form in the external nucleus of the inferior colliculus (ICx) and is projected from there to the optic tectum (OT). To determine the role of the map in ICx or its projected version in OT in the localization of acoustic stimuli, head movements of three adult owls were recorded before and after lesioning parts of the map either in ICx or in OT. Small electrolytic lesions caused sound-localization deficits that were characterized by failures to turn, turns away from the sound source, increased response latenci

Primary malleus fixation: diagnosis and treatment.

Primary malleus fixation occurs in an otherwise normal middle ear without evidence of congenital deformity and without chronic inflammatory changes. It occurs in the latter decades of life and is frequently associated with sensorineural presbycusis. We believe it is a ligament ankylosis with osteoarthritis related to the aging process. The diagnosis of malleus fixation is facilitated through the use of a modified Siegle pneumatic otoscope in conjunction with the Zeiss binocular microscope. The literature pertaining to this subject as well as the more historical reports are reviewed. Goodhill h

Expression of GAD in the Cochlear Nucleus and Superior Olivary Complex and Auditory Brainstem Response Thresholds in Fmr1-Knockout Mice.

BACKGROUND: This study investigates audiogenic seizures in Fmr1 knockout (KO) mice compared with wild-type (WT) mice across different ages. The aim was to assess auditory thresholds and examine glutamic acid decarboxylase (GAD) expression in auditory pathways to clarify the mechanisms underlying seizure susceptibility in fragile X syndrome (FXS). METHODS: Fmr1 knockout (KO) and wild-type (WT) FVB mice of different ages were used. Audiogenic seizure susceptibility was evaluated using sound stimulation. Auditory brainstem response (ABR) testing was performed to assess auditory thresholds, and g

Effects of early childhood otitis media-related conductive hearing loss on speech perception, neural processing, and working memory.

Otitis media with effusion (OME) is a common childhood ear disease that can cause temporary conductive hearing loss during a critical period of auditory development. This condition is hypothesized to be related to deficits in neural, perceptual, and cognitive abilities. This study examined the relation between conductive hearing loss due to OME in early childhood and its effects on neural and perceptual speech processing, as well as working memory (WM) function in early school-aged children. A total of 21 children aged 6-8 years with normal hearing who had a history of OME in early childhood w

Comparing sound localization and speech-in-noise in unilateral, bilateral, single-sided deaf, and bimodal cochlear implant users.

Cochlear implant users do not experience spatial hearing with the same accuracy or acuity as typical hearing listeners. Two standard measures of spatial hearing are sound localization error and speech reception thresholds (SRTs) in spatially separated noise. While sound localization requires a complicated setup, SRTs may be measured with fewer loudspeakers and less testing time. If a predictive relationship between sound localization error and SRTs can be measured, then the spatial hearing abilities of cochlear implant users may be quickly assessed by measuring one task or the other. To test t

Increased vulnerability to noise exposure of low spontaneous rate type 1C spiral ganglion neuron synapses with inner hair cells.

The inner hair cells (IHCs) in the inner ear form synapses with auditory nerve fibers (ANFs) that send sound signals to the brain. ANFs have been grouped by their level of spontaneous firing rates (SRs) into high-, medium-, and low-SR ANFs. Based on their molecular profiles evaluated by RNAseq experiments, ANFs have been divided into three groups (1A, 1B, and 1C) that likely correspond to high-, medium-, and low-SR ANFs, respectively. In guinea pigs, the synapses between IHCs and low-SR ANFs have been shown to be more vulnerable to noise exposure compared to other ANF subtypes, but not in a st

Alpha9alpha10 knockout mice show altered physiological and behavioral responses to signals in masking noise.

Medial olivocochlear (MOC) efferents modulate outer hair cell motility through specialized nicotinic acetylcholine receptors to support encoding of signals in noise. Transgenic mice lacking the alpha9 subunits of these receptors (α9KOs) have normal hearing in quiet and noise, but lack classic cochlear suppression effects and show abnormal temporal, spectral, and spatial processing. Mice deficient for both the alpha9 and alpha10 receptor subunits (α9α10KOs) may exhibit more severe MOC-related phenotypes. Like α9KOs, α9α10KOs have normal auditory brainstem response (ABR) thresholds and weak MOC

Effect of shock wave power spectrum on the inner ear pathophysiology in blast-induced hearing loss.

Blast exposure can induce various types of hearing impairment, including permanent hearing loss, tinnitus, and hyperacusis. Herein, we conducted a detailed investigation of the cochlear pathophysiology in blast-induced hearing loss in mice using two blasts with different characteristics: a low-frequency dominant blast generated by a shock tube and a high-frequency dominant shock wave generated by laser irradiation (laser-induced shock wave). The pattern of sensorineural hearing loss (SNHL) was low-frequency- and high-frequency-dominant in response to the low- and high-frequency blasts, respect

Acute pharmacological inhibition of matrix metalloproteinase-9 activity during development restores perineuronal net formation and normalizes auditory processing in Fmr1 KO mice.

Individuals with Fragile X Syndrome (FXS) and autism spectrum disorder (ASD) exhibit cognitive impairments, social deficits, increased anxiety, and sensory hyperexcitability. Previously, we showed that elevated levels of matrix metalloproteinase-9 (MMP-9) may contribute to abnormal development of parvalbumin (PV) interneurons and perineuronal nets (PNNs) in the developing auditory cortex (AC) of Fmr1 knock-out (KO) mice, which likely underlie auditory hypersensitivity. Thus, MMP-9 may serve as a potential target for treatment of auditory hypersensitivity in FXS. Here, we used the MMP-2/9 inhib

Increased spontaneous firing rates in auditory midbrain following noise exposure are specifically abolished by a Kv3 channel modulator.

Noise exposure has been shown to produce long-lasting increases in spontaneous activity in central auditory structures in animal models, and similar pathologies are thought to contribute to clinical phenomena such as hyperacusis or tinnitus in humans. Here we demonstrate that multi-unit spontaneous neuronal activity in the inferior colliculus (IC) of mice is significantly elevated four weeks following noise exposure at recording sites with frequency tuning within or near the noise exposure band, and this selective central auditory pathology can be normalised through administration of a novel c

M-CSF and IL-34 expression as indicators for growth in sporadic vestibular schwannoma.

Macrophage colony stimulating factor and IL-34 are associated with clinical vestibular schwannoma progression. Investigating the biology behind vestibular schwannoma progression helps understanding tumor growth. Inflammation is important in the microenvironment of neoplasms. Macrophages are major players in the intratumoral infiltrate. These tumor-associated macrophages are known to stimulate angiogenesis and cell growth. M-CSF and IL-34 are cytokines that can regulate tumor-infiltrating macrophages. They are expressed by tumors and form potential targets for therapy. The goal of this study wa

Prefrontal mediation of emotion regulation in social anxiety disorder during laughter perception.

Social anxiety disorder (SAD) is characterized by negatively biased perception of social cues and deficits in emotion regulation. While negatively biased perception is thought to maintain social anxiety, emotion regulation represents an ability necessary to overcome both biased perception and social anxiety. Here, we used laughter as a social threat in a functional magnetic resonance imaging (fMRI) study to identify cerebral mediators linking SAD with attention and interpretation biases and their modification through cognitive emotion regulation in the form of reappraisal. We found that reappr

Evidence that hidden hearing loss underlies amplitude modulation encoding deficits in individuals with and without tinnitus.

Damage to auditory nerve fibers that expresses with suprathreshold sounds but is hidden from the audiogram has been proposed to underlie deficits in temporal coding ability observed among individuals with otherwise normal hearing, and to be present in individuals experiencing chronic tinnitus with clinically normal audiograms. We tested whether these individuals may have hidden synaptic losses on auditory nerve fibers with low spontaneous rates of firing (low-SR fibers) that are important for coding suprathreshold sounds in noise while high-SR fibers determining threshold responses in quiet re

Auditory Brainstem Response Latency in Noise as a Marker of Cochlear Synaptopathy.

UNLABELLED: Evidence from animal and human studies suggests that moderate acoustic exposure, causing only transient threshold elevation, can nonetheless cause "hidden hearing loss" that interferes with coding of suprathreshold sound. Such noise exposure destroys synaptic connections between cochlear hair cells and auditory nerve fibers; however, there is no clinical test of this synaptopathy in humans. In animals, synaptopathy reduces the amplitude of auditory brainstem response (ABR) wave-I. Unfortunately, ABR wave-I is difficult to measure in humans, limiting its clinical use. Here, using an

The impact of degree of hearing loss on auditory brainstem response predictions of behavioral thresholds.

OBJECTIVES: Diagnosis of hearing loss and prescription of amplification for infants and young children require accurate estimates of ear- and frequency-specific behavioral thresholds based on auditory brainstem response (ABR) measurements. Although the overall relationship between ABR and behavioral thresholds has been demonstrated, the agreement is imperfect, and the accuracy of predictions of behavioral threshold based on ABR may depend on degree of hearing loss. Behavioral thresholds are lower than ABR thresholds, at least in part due to differences in calibration interacting with the effec

The pupil response reveals increased listening effort when it is difficult to focus attention.

Recent studies have shown that prior knowledge about where, when, and who is going to talk improves speech intelligibility. How related attentional processes affect cognitive processing load has not been investigated yet. In the current study, three experiments investigated how the pupil dilation response is affected by prior knowledge of target speech location, target speech onset, and who is going to talk. A total of 56 young adults with normal hearing participated. They had to reproduce a target sentence presented to one ear while ignoring a distracting sentence simultaneously presented to

Comparison between a new implantable transcutaneous bone conductor and percutaneous bone-conduction hearing implant.

OBJECTIVES: Despite good results on osseointegration and limited skin reactions with percutaneous bone conductors, there remains room for improvement. Especially in children, adverse events with percutaneous bone conductors might occur more frequently. Transcutaneous bone conductors, if powerful enough, can provide a solution that minimizes adverse events and implant loss. This study compares a new transcutaneous bone conduction hearing aid, the Sophono Alpha 1 (Sophono), with the percutaneous BAHA system (BAHA). METHODS: In our tertiary referral center, 12 patients (age 5-12 yr) with congeni
