P2X7 Signaling in Neuroinflammation: Glial Crosstalk, Redox Integration, and Therapeutic Targeting.

Extracellular ATP is a central danger signal linking cellular stress to neuroinflammation, yet the mechanisms governing the transition from physiological purinergic signaling to pathological neuroimmune activation remain incompletely understood. In this review, we integrate current evidence into a conceptual framework describing how ATP-dependent purinergic signaling may contribute to the shift from adaptive neuron-glia communication to maladaptive glial crosstalk and inflammatory amplification. Under physiological conditions, controlled ATP turnover primarily engages lower-threshold purinergi

A context-dependent METTL1-m7G-SLC7A11 axis links metabolic stress to epithelial fate in ulcerative colitis.

Ulcerative colitis (UC) is characterized by chronic intestinal inflammation accompanied by epithelial barrier dysfunction and profound metabolic stress; however, how metabolic cues are integrated to determine epithelial cell fate remains incompletely understood. Here, we identify a context-dependent METTL1-m7G-SLC7A11 regulatory axis that links metabolic stress to intestinal epithelial outcomes during UC progression. By integrating analyses of human UC tissues, DSS-induced acute and chronic colitis mouse models, and mechanistic in vitro experiments, we demonstrate that METTL1 enhances N7-methy

ZFP36L1 Enhances Microglial Ferroptosis in Ischemic Stroke by Reducing FTO-Mediated N6-Methyladenosine Demethylation of ACSL1 mRNA.

Microglia play an important role in ischemic stroke (IS). However, the molecular regulatory mechanisms underlying microglial ferroptosis in IS remain incompletely understood. In this study, blood samples were collected from 20 IS patients and 15 healthy volunteers. Microglia BV-2 subjected to oxygen-glucose deprivation/reoxygenation (OGD/R) and mice undergoing middle cerebral artery occlusion-reperfusion (MCAO/R) surgery were used to establish IS models. Our results revealed that in blood samples from IS patients and in OGD/R-treated BV-2 cells, ZFP36 ring finger protein like 1 (ZFP36L1) and A

Anomalous Sub-Band Gap Emission at Room Temperature in Mixed-Dimensional WS2/Cu2O Heterostructures.

Practical implementation of two-dimensional (2D) transition-metal dichalcogenides (TMDCs) in novel optoelectronics requires a fundamental understanding of carrier dynamics and interfacial states at 2D/3D heterostructures. Herein, a mixed-dimensional p-n junction based on monolayer tungsten disulfide ( WS 2 ) and cuprous oxide ( Cu 2 O ) is investigated. An anomalous emission (AE) at 1.66 eV is identified at room temperature, which is in the same spectral range as oxygen-related Cu 2 O defect luminescence, but is enhanced at the heterointerface under 457 nm excitation. While this sub-bandg

Redox-responsive and liver-targeted nanoplatform for paclitaxel delivery toward hepatocellular carcinoma therapy.

Conventional paclitaxel (PTX) chemotherapy for hepatocellular carcinoma (HCC) is severely restricted by insufficient tumor-targeting specificity, unsatisfactory therapeutic efficacy, and marked systemic toxicity. To address these drawbacks, a redox-responsive and liver-targeted nanoplatform was designed for efficient PTX delivery in HCC therapy. Herein, we synthesized a disulfide-bridged amphiphilic glycyrrhetinic acid-indomethacin prodrug conjugate (GA-ss-IND), which could spontaneously self-assemble into uniform nanomicelles driven by hydrophobic interaction and van der Waals forces, as veri

Synergistic reinforcement of bio-based non-isocyanate polyurethane by nanocellulose and lignin.

With the growing global demand for sustainable polymeric materials, bio-based non-isocyanate polyurethanes (NIPU) have attracted considerable attention as environmentally friendly alternatives to conventional polyurethanes. However, simultaneously achieving high performance and multifunctionality in NIPU remains challenging. Although nanocellulose (NCs) and lignin (Lig) have been widely employed as bio-based fillers, their potential synergistic effects in NIPU systems remain insufficiently understood. In this study, epoxidized soybean oil (ESO) was converted into cyclic carbonate soybean oil (

Sensitive RP-LC Method Enabling PTM-Specific Quality Control and MS-Compatible Characterization of Fc-Containing GLP-1 Therapeutics.

Fc-containing GLP-1 therapeutics exhibit complex post-translational modification (PTM) heterogeneity, necessitating advanced analytical methods for quality control (QC) and process analytical technology (PAT). We developed a reverse-phase liquid chromatography (RP-LC) method for the PTM-specific profiling of these biologics. Using dulaglutide (IgG4-Fc) as a model, critical parameters─including mobile-phase additives, acid concentration, and shallow gradients─were optimized to resolve PTM variants (e.g., hydroxylation, N-terminal truncation, disulfide reduction, glycosylation) within 40 min. Cyclic Peptides with RNase A Inhibitor Activity Based on Amino Acid Sequences Screened from the Fv-Antibody Library.

Ribonuclease A (RNase A) is a pancreatic enzyme that rapidly degrades single-stranded RNA and is widely used as a representative enzyme in studies of RNase-mediated RNA degradation. Therefore, effective RNase A inhibitors are needed to regulate RNase activity and protect RNA from enzymatic degradation. Fv-antibodies represent the antigen-binding region of immunoglobulin G (IgG) and are composed of complementarity-determining regions (CDRs) and framework regions (FRs). Previously, CDR3 sequences with RNase A inhibitory activity were screened using RNase A as a probe from an autodisplayed Fv-ant

Nanoarchitectonics of bridged palladium-polyelectrolyte colloidal complexes via reversible coordination-driven assembly and disassembly for closed-loop recovery.

Palladium (Pd) is a critical element in modern technologies, yet it faces supply risks and is often lost in industrial wastewater. In this study, a cystamine-modified polyacrylic acid (Cyst-PAA) was developed for rapid and targeted Pd recovery via S/N cooperative coordination. Binding of Pd(II) through amide N, amine N, and disulfide S atoms triggers intermolecular bridging, crosslinking polymer chains into a three-dimensional network within seconds that enables spontaneous precipitation and achieves over 93% removal by microfiltration in single-metal systems, delivering a water flux 77.5 time

Leveraging two-dimensional materials for high-sensitivity optical sensors: quasi-bound states in the continuum within hybrid metasurfaces.

Hybrid metal-dielectric metasurfaces supporting quasi-bound states in the continuum (qBICs) provide a powerful platform for high-Q optical sensing. In this work, we leverage two-dimensional materials as functional interlayers in a hybrid metal-dielectric metasurface to enable material-driven regulation of qBIC resonances for sensing applications. This approach introduces a material-assisted perturbation mechanism that enables tuning of the qBIC response within the same structural framework, without re-optimizing the overall geometry in the present design. Numerical simulations reveal that diff

Crosstalk of thioredoxin system and programmed cell death: from pathophysiology to novel therapy.

The thioredoxin (Trx) system, an integral component of cellular redox regulation, preserves protein dithiol-disulfide equilibrium through its conserved Cys-Gly-Pro-Cys active site and is involved in key cellular functions, including cell proliferation, apoptosis, and signal transduction. In cancer biology, the thioredoxin system plays dual roles: overexpression can suppress oxidative stress and promote tumor growth, whereas dysfunction can trigger programmed cell death (PCD). However, a critical area for future research is to delineate how Trx modulates the intricate networks of PCD, and to id

Mechanistic Interpretability of Fine-Tuned Protein Language Models for Nanobody Thermostability Prediction.

MOTIVATION: While Protein Language Models (PLMs) fine-tuned on biophysical data achieve high predictive accuracy, the physical principles underlying their predictions remain obscure. Deciphering these representations offers a unique opportunity to not only interpret model decisions but also to discover novel biophysical insights governing protein properties. Here, we present a framework using Sparse Autoencoders (SAEs) to extract mechanistic knowledge from PLMs fine-tuned for nanobody thermostability. RESULTS: We fine-tuned the ESM-2 model on the nanobody thermostability dataset, achieving su

Thermal processing under mild acidic conditions modulates metabolite profile and emulsifying functionality of Moringa oleifera leaf proteins.

This study investigated the effects of thermal treatment (30, 55, and 85 °C) under mild acidic conditions (pH 4.5) during protein precipitation on structural characteristics, emulsifying properties, metabolite profile and protein composition of Moringa leaf protein (MoLP) from two cultivars (local Thai and PKM-1). Moderate heating at 55 °C resulted in the highest protein recovery, reaching 4.59% and 2.47% for Thai and PKM-1 cultivars, respectively. In contrast, treatment at 85 °C reduced protein content to 54.8% and 50.1%. Structural analyses indicated heat-induced unfolding, thiol-disulfide r

Omicron RBD expressed in E. coli outperforms mammalian-expressed S1 spike protein in generating highly neutralizing anti-SARS-CoV-2 antibodies in mice.

SARS-CoV-2 variants are characterized by high transmissibility, rapid mutations, and the ability to evade immune responses, underscoring the urgent need for affordable and easily scalable vaccine platforms. We previously reported that the E. coli-expressed receptor-binding domain (RBD) from both the Omicron BA.5 and the Wuhan variants, administered at 30 μg x 4 doses, induces robust humoral and cellular immune responses in mice. Here, using Omicron BA.5 as a model, we demonstrate that a much lower 5 μg x 2 doses of E. coli-expressed RBD elicited a strong anti-SARS-CoV-2 response even without a

Comprehensive genomic analyses revealed the adaptation strategies of Exiguobacterium and its phage genomic diversity.

Exiguobacterium exhibits high species diversity and complex evolutionary patterns, with members widely distributed across diverse habitats. To elucidate the mechanisms enabling its high adaptability to various environments, 187 genomes of Exiguobacterium strains were analyzed using the phylogenomic and comparative genomics methods. Our analysis revealed that nearly all Exiguobacterium strains harbor genes encoding the utilization of diverse complex polysaccharides and proteinaceous, as well as intact glycolysis and tricarboxylic acid cycle pathways. These abilities suggest that the strains of

Phase and Defect Engineering of Transition Metal Dichalcogenides Via Carbon-Nanotube-Induced Photothermal Effect.

Molybdenum disulfide (MoS2), a representative transition metal dichalcogenide (TMD), exists in semiconducting 2H and metallic 1T phases; however, the chemically inert basal plane and intrinsically low conductivity of the stable 2H phase limit its sensing performance. To overcome these limitations, 1T-rich MoS2-decorated single-walled carbon nanotube heterostructures, i.e., SWCNT@1T-rich MoS2, were synthesized via flash-thermal shock (FTS), using SWCNTs as both conductive and photothermal supports. The intense pulsed light-driven FTS process enables ultrafast annealing, reaching temperatures up

Structural insights into the activation mechanism of the human metabolite receptor HCAR1.

Hydroxycarboxylic acid receptor 1 (HCAR1) is a class A G protein-coupled receptor (GPCR) that is activated by the endogenous metabolite l-lactate and that plays an important role in various metabolic and inflammatory disorders. HCAR1 uses distinct ligand recognition and self-activation mechanisms to mediate specific pathophysiological functions through Gαi/o and β-arrestin signaling pathways. To support effective drug development targeting HCAR1, we investigated ligand recognition and activation mechanisms through cryo-electron microscopy (cryo-EM) structures of the HCAR1-Gαi1 complex in the a

TPGS incorporated solanesol-paclitaxel prodrug nanoparticles for synergic therapy of cancer.

Paclitaxel (PTX) chemotherapy is severely limited by poor aqueous solubility, off-target toxicity, and P-glycoprotein-mediated multidrug resistance (MDR). Herein, we report a redox-responsive prodrug nanosystem, SOL-SS-PTX, constructed by conjugating PTX to the bioactive natural polyisoprenoid solanesol (SOL) via a disulfide bond. Unlike conventional polymer- or lipid-based carriers, SOL simultaneously serves as the hydrophobic self-assembly driver and a pharmacologically active co-agent, enabling synergistic antitumor activity. The prodrug spontaneously self-assembles into uniform nanoparticl
