Experimental Charcot-Marie-Tooth type 1A: a cDNA microarrays analysis.

To reveal the spectrum of genes that are modulated in Charcot-Marie-Tooth neuropathy type 1A (CMT1A), which is due to overexpression of the gene coding for the peripheral myelin protein 22 (pmp22), we performed a cDNA microarray experiment with cDNA from sciatic nerves of a rat model of the disease. In homozygous pmp22 overexpressing animals, we found a significant down-regulation of 86 genes, while only 23 known genes were up-regulated, suggesting that the increased dosage of pmp22 induces a general down-regulation of gene expression in peripheral nerve tissue. Classification of the modulated

Integrating transcriptomics and machine learning to predict ferroptosis-related genes and analyzing the role of GOT1 in gastric cancer progression.

BACKGROUND: Gastric cancer (GC) is one of the leading causes of cancer-related mortality worldwide. Despite advances in treatment, the prognosis for advanced GC remains poor, highlighting the need for new therapeutic targets. Ferroptosis, a form of programmed cell death characterized by iron-dependent lipid peroxidation, has emerged as a potential pathway for cancer therapy. This study aims to identify ferroptosis-related genes in GC using transcriptomics and machine learning, and to validate the role of one key gene, Glutamic-Oxaloacetic Transaminase 1 (GOT1), in GC progression. METHODS: The

Environmental endocrine disruptors activate CBX2 to disrupt cholesterol homeostasis in PCOS.

Polycystic ovary syndrome (PCOS) is a common endocrine disorder influenced by genetic and environmental factors, yet the molecular mechanisms linking endocrine-disrupting chemicals (EDCs) to ovarian dysfunction remain unclear. Here, we establish a letrozole-induced PCOS-like rat model and integrate bulk RNA-seq, single-cell RNA-seq of human granulosa cells (GCs), and functional validation to identify CBX2 as a critical epigenetic regulator connecting EDC exposure to PCOS pathogenesis. Transcriptomic profiling revealed dysregulated cell cycle progression, disrupted cholesterol homeostasis, and

TSPO deficiency uncovers a novel role for porphyrins in adipocyte lipid metabolism.

The translocator protein (TSPO) is a mitochondrial outer membrane protein enriched in lipid-metabolic tissues, including white adipose tissue (WAT), but its physiological role in adipocytes remains poorly defined. Building on prior work that demonstrated a role for TSPO in mitochondrial fatty acid oxidation, we investigated whether TSPO contributes to adipocyte lipid homeostasis under metabolic stress. We employed a combination of in vivo and in vitro approaches using global Tspo knockout (Tspo-/-) mice, primary adipocyte cultures, and pharmacological TSPO-binding drugs to examine TSPO functio

Reduced KLF4 expression in endothelial cells triggers pulmonary vascular abnormalities associated with experimental congenital diaphragmatic hernia.

Congenital diaphragmatic hernia (CDH) is a rare anomaly with an incidence of 1 in 3000 live births and characterized by defective closure of the diaphragm, lung hypoplasia, and pulmonary vascular remodeling. Although several genes associated with the occurrence of CDH, no clear genetic component has been identified. Previously, we showed that CDH patients have vascular abnormalities already early in development, and that perivascular cells are affected in CDH. In the current study, we focused on early abnormalities in the nitrofen-induced CDH mouse model. Transcriptome analysis of FACS-sorted

An ancestral transmembrane transcription factor couples cell envelope regulation and the SOS response in Caulobacter crescentus.

The DNA damage (SOS) response in bacteria involves derepression of a set of genes in order to activate mechanisms to tolerate stress, repair DNA and slow down cell division. While some of these genes are well characterized, many genes exist which are clearly induced by DNA damage but for which the function is unclear. In Caulobacter crescentus, the toxin-antitoxin (TA) system higBA and a closely associated downstream transcription factor (higX) are strongly induced as part of the SOS response, but the role of higX is unknown. We show that, unexpectedly, HigX functions independently of HigBA as

An Lmx1a/b allelic series reveals the role of Lmx1 genes in cochlear nuclei development.

Lmx1a/b paralogous genes, which arose from the invertebrate Lmx1b-like gene, are critical for hearing in multiple vertebrate species, and mutations in these genes cause hearing deficits in humans. While the unique and redundant functions of Lmx1a/b in the inner ear are well established, their contribution to the development of the cochlear nuclei, which process and relay auditory information to the brain, is poorly understood. Since cochlear nuclei maturate postnatally, here we analyzed Lmx1a+/-;Lmx1b+/-, Lmx1a-/-, and Lmx1a-/-;Lmx1b+/- mice that survive past birth. Loss of Lmx1a reduced disti

The Charcot-Marie-Tooth Neuropathy (CMTX3) Complex Structural Variation Causes Differential SOX3 Spatiotemporal Expression.

Charcot-Marie-Tooth (CMT) neuropathy is a clinically and genetically heterogeneous group of diseases characterized by the length-dependent axonal degeneration of peripheral nerves. We previously mapped a rare form of X-linked CMT, CMTX3, to a 5.7-Mb interval on chromosome Xq26.3-q27.1 and excluded the coding region of all known genes in the linkage interval for mutations. Whole genome sequencing subsequently identified a 78-kb region of chromosome 8q24.3 that had been duplicated and inserted into the CMTX3 locus between the genes HAPSTR2 and SOX3. The 78-kb insertion, which contains a partial

Participation of the Transcriptional Regulator Fur in Modulating Biofilm Formation of the Salmonid Pathogen Yersinia ruckeri: Beyond Iron Homeostasis.

Biofilm formation by bacterial pathogens represents a major challenge in aquaculture, since biofilms promote persistence and increase tolerance to antimicrobial treatments. Yersinia ruckeri, the etiological agent of enteric redmouth disease in salmonids, contributes to recurrent infections in aquaculture. However, the molecular mechanisms governing biofilm development in this pathogen remain poorly understood. In this study, we investigated the role of the global transcriptional regulator Fur in the biofilm formation of Y. ruckeri. To this aim, a fur deletion mutant (Δfur) was phenotypically,

Automated high-throughput microscopy screening unveiled new Listeria monocytogenes genes involved in cell infection.

To uncover novel genetic factors required for Listeria monocytogenes cell infection, we developed an automated high-throughput microscopy screening pipeline that integrates GFP-expressing bacteria with machine learning-based image analysis. Using this approach, we screened a mariner transposon library comprising 4224 L. monocytogenes EGDe mutants and identified 58 with significantly reduced numbers of intracellular bacteria. Sequencing revealed 24 unique insertion sites corresponding to 14 genes, including previously known virulence factors and nine novel candidates not previously implicated i

PFBS disrupts lipid metabolism and mitochondrial function in human trophoblast cells.

Perfluorobutanesulfonic acid (PFBS) is an emerging short-chain per- and polyfluoroalkyl substance (PFAS), a group of persistent environmental contaminants associated with adverse reproductive outcomes. The placenta plays a critical role in the pathogenesis of pregnancy complications, and disrupted placentation is implicated in the mechanistic pathways linking PFBS exposure to these disorders. In particular, placental mitochondria function refines during pregnancy to optimize the dynamic growth of the fetus and placenta. Disruptions in mitochondrial function may therefore mediate the adverse ef

Single-cell RNA-sequencing co-expression analysis with CFTR and CF modifier genes in lung tissue.

While cystic fibrosis is caused by loss-of-function variants in the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR), other modifier genes have been shown to associate with disease severity. Co-expression of modifiers with CFTR in normal tissue indicates a cooperative relationship and suggests the potential for compensation in the presence of CFTR dysfunction. We examined the co-expression relationships with CFTR in the lung using single cell RNA sequencing to pinpoint cell types and their modifiers involved in the cystic fibrosis lung phenotype and support target prioritization for

Loss of peptidylarginine deiminase 4 mitigates maladaptive cardiac remodeling after myocardial infarction through inhibition of inflammatory and profibrotic pathways.

Inflammation and progressive fibrosis represent predictive risk factors for heart failure (HF) development following myocardial infarction (MI). Peptidylargininine deiminase 4 (PAD4) catalyzes the citrullination of arginine residues in polypeptides and has recently been identified as a contributor to HF pathogenesis. This study aimed to evaluate the role of PAD4 in monocytes / macrophages (Mo/Mφ) and cardiac fibroblasts (CFs) for cardiac repair following MI and HF progression. Cardiac Padi4 expression significantly increased in mice subjected to MI by permanent coronary artery ligation as well

Spatial transcriptomics reveals Inhba/Smad2/E2f4 axis in Lrp2high thecal cell proliferation in androgen-induced PCOS mice.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a common endocrine disorder characterized by elevated androgen levels and impaired follicular development. A hallmark of PCOS is the excessive proliferation of thecal cells (TCs), which are critical for androgen production. However, the molecular mechanisms underlying this aberrant cellular expansion remain incompletely understood. METHODS: A DHEA-induced mouse model was used to recapitulate the hormonal and ovarian features of human PCOS. Spatial transcriptomics was employed to profile gene expression in ovarian tissue at cellular resolution. D

The Inhibition of the Membrane-Bound Transcription Factor Site-1 Protease (MBTP1) Alleviates the p.Phe508del-Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) Defects in Cystic Fibrosis Cells.

Cystic Fibrosis (CF) is present due to mutations in the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) gene, the most frequent variant being p.phe508del. The CFTR protein is a chloride (Cl-) channel which is defective and almost absent of cell membranes when the p.Phe508del mutation is present. The p.Phe508del-CFTR protein is retained in the endoplasmic reticulum (ER) and together with inflammation and infection triggers the Unfolded Protein Response (UPR). During the UPR, the Activating Transcription Factor 6 (ATF6) is activated with cleavage and then decreases the expression of p

Loss of SLC30A10 manganese transporter alters expression of neurotransmission genes and activates hypoxia-inducible factor signaling in mice.

The essential metal manganese (Mn) induces neuromotor disease at elevated levels. The manganese efflux transporter SLC30A10 regulates brain Mn levels. Homozygous loss-of-function mutations in SLC30A10 induce hereditary Mn neurotoxicity in humans. Our prior characterization of Slc30a10 knockout mice recapitulated the high brain Mn levels and neuromotor deficits reported in humans. But, mechanisms of Mn-induced motor deficits due to SLC30A10 mutations or elevated Mn exposure are unclear. To gain insights into this issue, we characterized changes in gene expression in the basal ganglia, the main

Spermidine alleviates thymopoiesis defects and aging of the peripheral T-cell population in mice after radiation exposure.

The T cell aging process can be modified by genotoxic factors, including ionizing radiation, and metabolic controls, such as caloric restriction; the former accelerates and the latter retards the process. However, the mechanisms by which these systemic factors interact to cause T cell aging remain unclear. This study investigated the naïve T-cell pool, thymic cellularity, and transcriptome in mice irradiated with 3.8 Gy at 5 weeks of age and treated 13 months later with 30 mM spermidine (SPD), a metabolism regulator. The number of conventional naïve CD4 and CD8 T cells in the peripheral blood
