Asymmetry and Heterogeneity: Part and Parcel in Cardiac Autonomic Innervation and Function.

The cardiac autonomic nervous system (cANS) regulates cardiac adaptation to different demands. The heart is an asymmetrical organ, and in the selection of adequate treatment of cardiac diseases it may be relevant to take into account that the cANS also has sidedness as well as regional differences in anatomical, functional, and molecular characteristics. The left and right ventricles respond differently to adrenergic stimulation. Isoforms of nitric oxide synthase, which plays an important role in parasympathetic function, are also distributed asymmetrically across the heart. Treatment of cardi

Arrhythmogenic right ventricular cardiomyopathy in dogs.

Arrhythmogenic right ventricular cardiomyopathy (ARVC) is an inherited myocardial disease seen in dogs, cats, and humans. A common entity in Boxers and the related English bulldog, the disease is characterized by fatty or fibrofatty replacement of the myocardium, ventricular arrhythmias, and the potential for syncope or sudden death. In some individuals, concomitant left ventricular involvement results in systolic dysfunction and a progression to congestive heart failure. The clinical and pathological characteristics of ARVC share many similarities in dogs and humans, and Boxers serve as an im

Clostridium difficile shedding by healthy dogs in Nigeria and Malawi.

The objectives of this study were to determine the prevalence and characteristics of Clostridium difficile shedding in owned dogs in Nigeria and Malawi. Clostridium difficile was isolated from 31/120 (26%) dogs in Nigeria and 11/92 (12%) dogs in Malawi (p = 0.012). Overall, 22/42 (52%) isolates were toxigenic; 17/31 (55%) from Nigeria and 5/11 (45%) from Malawi. All toxigenic isolates possessing tcdA and tcdB, and only one also possessed cdtA/B. Sixteen different ribotypes were found, ten (63%) of which were non-toxigenic. Most isolates corresponded to ribotypes that have been previously ident

The piglet acute diarrhea model for evaluating efficacy of treatment and control of cryptosporidiosis.

Cryptosporidium spp. are ranked as the second leading pathogens causing life-threatening diarrhea in children under 2 years of age. Although Cryptosporidium hominis causes three quarters of the cases of cryptosporidiosis, studies on C. hominis are limited since natural disease due to C. hominis is host-restricted to humans only. In this mini-review, we demonstrate the successfully adoption, propagation, and utility of the C. hominis strain TU502, isolated originally from an infant with diarrhea in Uganda, in gnotobiotic piglets. The TU502 C. hominis strain and the gnotobiotic piglet model curr

Globozoospermia and lack of acrosome formation in GM130-deficient mice.

Globozoospermia is a common reproductive disorder that causes male infertility in humans, and the malformation or loss of acrosomes is the prominent feature of this disease. Although the acrosome is thought to be derived from the Golgi apparatus, the detailed molecular mechanisms remain unclear. GM130 is a cis-side localized Golgi matrix protein, whereas the physiological functions of this protein remain elusive. Here we showed that inactivation of GM130-caused male infertility in mouse model. The primary defects were the absence of acrosomes, round sperm heads, and aberrant assembly of the mit

Leishmania infection: painful or painless?

The complex life cycle and immunopathological features underpinning the interaction of Leishmania parasites and their mammalian hosts poses frequent poorly explored and inconclusively resolved questions. The altered nociceptive signals over the course of leishmaniasis remain an intriguing issue for nociceptive and parasitology researchers. Experimental investigations have utilized behavioral, morphological, and neuro-immune approaches in the study of experimental cutaneous leishmaniasis (CL). The data generated indicates new venues for the study of the pathological characteristics of nocicepti

A new humanized mouse model for alopecia areata.

Although alopecia areata (AA) is not life threatening, it may lead to severe psychological disturbances, reducing the quality of life in all ages. Thus, a new animal model is needed for shedding more light onto the pathogenesis of this cell-mediated, organ-specific autoimmune disease to identify more effective therapeutic strategies. Recently, we succeeded in developing a new humanized mouse model of AA, which includes transplantation of healthy human scalp skin obtained from normal volunteers on to severe-combined immunodeficient mice. This is followed by intradermal injection of either autol

Generation of mice with a conditional null Fraser syndrome 1 (Fras1) allele.

Fraser syndrome (FS) is an autosomal recessive disease characterized by skin lesions and kidney and upper airway malformations. Fraser syndrome 1 (FRAS1) is an extracellular matrix protein, and FRAS1 homozygous mutations occur in some FS individuals. FRAS1 is expressed at the epithelial-mesenchymal interface in embryonic skin and kidney. blebbed mice have a null Fras1 mutation and phenocopy human FS. Like humans with FS, they exhibit a high fetal and neonatal mortality, precluding studies of FRAS1 functions in later life. We generated conditional Fras1 null allele mice. Cre-mediated generalize

Pathological features of diseases associated with human T-cell leukemia virus type I.

In the early 1980s, the first human retrovirus, human T-cell leukemia virus type I (HTLV-I), was isolated and its characterization opened up the new field of human viral oncology. Adult T-cell leukemia/lymphoma (ATLL), which is associated with HTLV-I, is characterized clinically by the appearance of characteristic flower cells, a rapid clinical course, occasional skin lesions, lymphadenopathy and hepatosplenomegaly. Severe opportunistic infections are occasionally accompanied. In addition, HTLV-I infection is associated with autoimmune and reactive disorders, such as HTLV-I-associated myelopat

Mouse model of X-linked Alport syndrome.

X-linked Alport syndrome (XLAS) is a progressive disorder of basement membranes caused by mutations in the COL4A5 gene, encoding the alpha5 chain of type IV collagen. A mouse model of this disorder was generated by targeting a human nonsense mutation, G5X, to the mouse Col4a5 gene. As predicted for a nonsense mutation, hemizygous mutant male mice are null and heterozygous carrier female mice are mosaic for alpha5(IV) chain expression. Mutant male mice and carrier female mice are viable through reproductive age and fertile. Mutant male mice died spontaneously at 6 to 34 wk of age, and carrier f

Nosocomial Kikuchi's disease--a search for herpesvirus sequences in lymph node tissues using PCR.

BACKGROUND: Histiocytic necrotizing lymphadenitis, also known as Kikuchi's disease (KD), is a rare disease. Fever and lymphadenopathies with characteristic pathologic features are present. The etiology of this disease remains undetermined. Since the disorder is self-limiting, different viruses have been implicated as the causative agent. PATIENTS AND METHODS: Seven cases of KD were studied. Three patients acquired the disease nosocomially, three had community-acquired KD and one case was associated with systemic lupus erythematosus. PCR was performed on DNA extracted from lymph node tissues i

Therapeutic administration of fibronectin: current uses and potential applications.

Fibronectins are a class of multifunctional glycoproteins that exist in soluble and insoluble forms. These glycoproteins are found in nearly all body fluids and in tissue extracellular matrices. Alterations in the normal synthesis, release, and distribution of fibronectins are characteristic features of certain human pathophysiologic conditions. The therapeutic administration of plasma fibronectin to human patients with defects in corneal wound healing and critically ill adult patients with multisystem organ failure appears clinically beneficial, although confirmation is needed from additional

Human pancreatic secretion and intestinal motility: effects of ileal nutrient perfusion.

To study the effects of intraileal nutrients on human pancreatic secretion and gastrointestinal motility, nine healthy subjects were intubated with an oroileal multilumen tube for ileal perfusion, duodenal juice aspiration, and intestinal motility recording. The duodenum was perfused continuously with essential amino acids to induce submaximal stimulation of pancreatic enzyme secretion and fed motility pattern. Additional ileal perfusion with carbohydrate at quantities similar to those observed under physiological late postprandial conditions or fat at isocaloric loads significantly decreased

Sex-specific hormonal trajectories reveal an early neuroendocrine vulnerability window preceding menopausal depressive symptoms.

Depressive symptoms show marked sex differences in prevalence and symptom profiles, with sex hormones thought to play a key role. However, systematic evaluations of age-related hormone variation across the adult lifespan remain limited. We analyzed data from 8421 adults (4204 males and 4217 females) drawn from the National Health and Nutrition Examination Survey (NHANES) collected between 2013 and 2016. Estradiol and testosterone levels, along with depressive symptoms assessed using the Patient Health Questionnaire-9 (PHQ-9), were included in the analysis. Participants were stratified by decad

From neighborhoods to brain health: an outreach model linking health literacy, cognitive care, research, and the arts, in San Francisco.

Traditional community outreach, research recruitment, and engagement strategies in aging neuroscience often fail to target and capture the diversity of structural and environmental conditions that shape the risk of age-related neurocognitive disorders (NCDs), including Alzheimer's disease. In this manuscript, we describe the rationale, implementation, and outcomes of the Community Outreach Program (COP) of the UCSF Fein Memory and Aging Center (MAC), emphasizing its neighborhood-based initiative designed to engage vulnerable populations to improve brain health literacy, access to cognitive car

Sex shapes CD64 expression and vaccine-induced monocytic responses.

Sex-specific differences influence vaccine-induced immunity, with females generally mounting more robust immune responses than males. While enhanced antibody production in females is well established, the cellular mechanisms underlying these differences remain elusive. Monocytes and monocyte-derived dendritic cells contribute to vaccine-induced immunity by capturing antibody-antigen immune complexes via Fc gamma receptors (FcγRs), positioning them as potential mediators of sex-biased vaccine responses. Here, we investigated sex-specific differences in FcγRI (CD64)-expressing monocytic cells in

Uncovering the role of sociodemographic factors in sex/gender differences in emotional brain activation: an SDM-PSI meta-analysis of fMRI studies.

INTRODUCTION: The study of sex/gender (S/G) differences in neuroscience, particularly in emotional processing, has been hindered by methodological inconsistencies, often producing biased conclusions that overgeneralize brain differences between males and females. Moreover, many studies fail to consider how other sociodemographic factors interact with S/G to influence the brain. This study aims to address these gaps by investigating whether potential S/G effects in brain activation during emotion-evoking functional magnetic resonance imaging (fMRI) tasks are influenced by those factors. METHOD

Sex-dependent locus coeruleus vulnerability in Alzheimer's disease: gut dysbiosis as a driver and probiotic intervention as rescue.

Alzheimer's disease (AD) displays striking sex differences in incidence, progression, and resilience, yet the mechanisms that drive female-biased vulnerability remain incompletely understood. Emerging evidence indicates that gut dysbiosis, increasingly prevalent with ageing, acts as a systemic amplifier of neuroinflammation, vascular instability, and metabolic dysfunction. Here, we synthesize converging findings linking gut microbial alterations to noradrenergic pathology in the locus coeruleus (LC), one of the earliest brain regions affected in AD. We outline how dysbiosis-associated inflamma

XYomics: detecting sex-dependent molecular mechanisms in omics data.

Understanding sex-dependent differences in disease risk, manifestation, and treatment response is essential for precision medicine. While funding agencies now mandate consideration of Sex as a Biological Variable (SABV), existing bioinformatics tools lack systematic approaches to characterize sex-related molecular mechanisms. Current practices frequently treat sex as a confounding variable, which may obscure important biological differences such as sex-specific alterations, sex-dimorphic changes (opposite effects between sexes), and sex-modulated changes (different effect magnitudes). We prese
