Modeling and dynamics of Brucella infection-induced macrophage apoptosis inhibition.

Brucella is an intracellular bacterium that causes the widespread zoonotic disease brucellosis. Within their hosts, Brucella has evolved various immune evasion strategies, including the ability to survive and replicate within host cells, especially within macrophages. This leads brucellosis from an acute phase to a chronic phase that is difficult to cure. In order to explore the key factors for the survival of Brucella and the mechanisms of persistent chronic infection, a mathematical model is developed to characterize the interactions between Brucella and macrophages, which incorporates a sat

Comparative immunological roles of TEP1 in Anopheles gambiae and Biomphalaria glabrata: implications for malaria and schistosomiasis control.

Malaria and schistosomiasis represent two of the most significant global parasitic diseases in terms of public health burden. These diseases are transmitted through Anopheles mosquitoes and freshwater snails, respectively. Although their transmission mechanisms differ, both pathogens critically interact with thioester-containing proteins (TEPs) during immune evasion and clearance within their invertebrate hosts. This review compares the activation mechanisms and functional divergences of TEPs in Anopheles gambiae and Biomphalaria glabrata in the context of host anti-infective immunity. We focu

Influence of hosts on the oviposition and hatching of engorged females Amblyomma cajennense sensu stricto (Acari: Ixodidae) with notes on new host records.

The parasite-host relationships of Amblyomma cajennense sensu stricto, one of the species of the Amblyomma cajennense complex that occurs in the state of Maranhão, are poorly known and are restricted to its association with horses and humans. This study evaluated, for the first time, the effect of hosts on the oviposition of A. cajennense engorged females, naturally collected from wild animals and humans, as well as those experimentally fed on cattle and highlights the importance of the relationship between A. cajennense and different hosts in maintaining tick populations in the Amazon biome.

Exposing the hidden: establishing immunity to Plasmodium liver stage infection.

Plasmodium, the parasite which causes malaria, has evolved with - and plagued - humans since our species emerged. Decades of intervention efforts have concluded that the blueprint for success is to hamper transmission between Anopheles mosquitoes and humans. Before causing symptomatic blood-stage infection, Plasmodium must first develop in the liver, which serves as a hidden gateway into the human host. Blocking parasite entrance or exit through the liver stage prevents both illness and onward transmission. Robust immunity against the pre-erythrocytic stages of Plasmodium falciparum fails to d

25 Years of Ebola Virus VP35 Research.

Ebola virus (EBOV) is a highly virulent non-segmented, negative-sense RNA virus and a causative agent of severe, often fatal disease in humans. EBOV encodes 7 genes and viral protein 35 (VP35) plays a key role in EBOV pathogenesis. VP35 is an EBOV polymerase cofactor that facilitates host immune evasion, viral RNA synthesis, and nucleocapsid assembly. Over the past 25 years since the first recognition that VP35 can antagonize innate immune responses, considerable progress has been made to define the molecular mechanisms by which VP35 mediates its multiple functions and interactions with host f

Emerging Arthropod-Borne Viruses Hijack the Host Cell Cytoskeleton During Neuroinvasion.

Arthropod-borne viral infections, ranging from asymptomatic to fatal diseases, are expanding from endemic to nonendemic areas. Climate change, deforestation, and globalization favor their spread. Although arboviral manifestations mainly determine the onset of generalized symptoms, distinct clinical signs have been assessed, depending on the particular arthropod-borne virus (arbovirus) involved in the infectious process. A number of arboviruses cause neuroinvasive diseases in vertebrate hosts, with acute to chronic outcomes. Long-term neurological sequelae can include cognitive dysfunction and

Pathogenicity and Virulence of Coxiella burnetii: Focus on Q fever.

Coxiella burnetii is responsible for Q fever, a zoonosis that mainly manifests as a mild acute infection and may persist in specific individuals causing severe complications, including endocarditis and vascular infections. Transmission mainly occurs through the inhalation of aerosols, underlining bacterial resistance to environmental stress. C. burnetii is an intracellular gram-negative pathogen that can infect and survive in monocytes and macrophages and replicate within acidic compartments. C. burnetii expresses several virulence factors, including lipopolysaccharide and a type IV secretion

Mechanisms of Virulence of Mycobacterium abscessus and Interaction with the Host Immune System.

Mycobacterium abscessus is a non-tuberculosis fast-growing mycobacterium that has recently become a serious concern due to its rapidly increasing prevalence worldwide, mainly in individuals with a high susceptibility to pulmonary infections, for example, patients with cystic fibrosis, bronchiectasis, chronic obstructive pulmonary disease, and previous tuberculosis infection. According to present estimations, at least 20% of patients with cystic fibrosis are infected with M. abscessus. This bacterium is extremely resistant to most drugs, leading to a severe and difficult-to-treat infection. Tha

Recent advances in research on Mycobacterium tuberculosis virulence factors and their role in pathogenesis.

Mycobacterium tuberculosis (Mtb) is the causative agent of tuberculosis (TB) in humans and animals. Mtb invades the host's lungs via airborne transmission, infecting macrophages and causing TB. In some cases, the infection can spread to other tissues and organs. Despite the availability of several drugs for TB treatment, the emergence of multidrug-resistant TB has led to high morbidity and mortality rates worldwide. Therefore, it is urgent to discover new anti-tuberculosis drugs for more effective treatment. Recent studies have shown that Mtb virulence factors play a crucial role in its pathog

Caseinolytic Protease P: A Therapeutic Nexus in Infection, Inflammation, and Immunity.

Caseinolytic protease P (ClpP) is a highly conserved serine protease that plays a pivotal role in protein homeostasis and quality control in bacteria, mitochondria of mammalian cells, and plant chloroplasts. As the proteolytic core of the ATP-dependent Clp protease complex, ClpP partners with regulatory ATPases (e.g., ClpX, ClpA) to degrade misfolded, damaged, or regulatory proteins. In bacteria, ClpP is crucial for survival under host-imposed stresses and modulates virulence through degradation of transcriptional regulators and signaling proteins, contributing to immune evasion, dormancy, and

Triggering infection: When microbial pathogens are in cahoots.

Due to the presence of gut microbiota, enteric pathogens such as Salmonella are rarely alone in their battleground with the host. In a recent study published in Nature, Jaswal et al. revealed that Candida albicans, a fungus frequently found in the gut, facilitates the intestinal colonization and infection of Salmonella.

Enigmatic roles of Vibrio cholerae hemolysin/cytolysin in the bacterial pathogenesis and host-pathogen interactions.

Vibrio cholerae cytolysin (VCC; also known as V. cholerae hemolysin) is a β-barrel pore-forming toxin (β-PFT) secreted by the cholera pathogen V. cholerae. VCC acts to disrupt the selective permeability barrier function of the target cell membranes. Monomeric VCC molecules bind to and form heptameric transmembrane water-filled pores or channels in the lipid bilayer of the membranes, thus resulting in colloid-osmotic lysis of the target cells. Apart from its pore-forming function, VCC can activate an array of signaling cascades leading to the diverse responses that include programmed cell death

Differentially expressed extracellular matrix genes functionally separate ameloblastoma from odontogenic keratocyst.

BACKGROUND: Ameloblastoma and odontogenic keratocyst (OKC) are odontogenic tumors that develop from remnants of odontogenic epithelium. Both display locally invasive growth characteristics and high predilection for recurrence after surgical removal. Most ameloblastomas harbor BRAFV600E mutation while OKCs are associated with PATCH1 gene mutation but distinctive indicators of ameloblastoma growth characteristics relative to OKC are still unclear. The aim of this study was to assess hub genes that underlie ameloblastoma growth characteristics using bioinformatic analysis, ameloblastoma samples a

FcRγIIA response duality in leishmaniasis.

Leishmania is responsible for a neglected tropical disease affecting millions of people around the world and could potentially spread more due to climate change. This disease not only leads to significant morbidity but also imposes substantial social and economic burdens on affected populations, often exacerbating poverty and health disparities. Despite the complexity and effectiveness of the immune response, the parasite has developed various strategies to evade detection and manipulates host cells in favor of its replication. These evasion strategies start at early stages of the infection by

A biological invasion modifies the dynamics of a host-parasite arms race.

By imposing novel selection pressures on both participants, biological invasions can modify evolutionary 'arms races' between hosts and parasites. A spatially replicated cross-infection experiment reveals strong spatial divergence in the ability of lungworms (Rhabdias pseudosphaerocephala) to infect invasive cane toads (Rhinella marina) in Australia. In areas colonized for longer than 20 years, toads are more resistant to infection by local strains of parasites than by allopatric strains. The situation reverses at the invasion front, where super-infective parasites have evolved. Invasion-induc
